    A plant transformation vector containing the gene dehd for the development of cultivars resistant to monochloroacetic acid

    A dehalogenase D gene (dehD) capable of degrading monochloroacetic acid (MCA) has been previously isolated from Rhizobium sp. RC1 and characterized. The 804-bp dehD gene was inserted into pCAMBIA under the control of the cauliflower mosaic virus 35S promoter and designated pCAMdehD, with a total size of 10,592 bp. The pCAMdehD was introduced into tobacco (Nicotiana benthamiana) via Agrobacterium-mediated transformation. The integration and expression of dehD in N. benthamiana was confirmed by PCR and reverse transcription PCR, respectively. MCA-resistant transformants were selected in tissue cultures containing 60 µg/L MCA. Analysis of plants using a leaf-painting assay revealed that transgenic N. benthamiana was resistant to 4.0 g/L MCA compared with 2.0 g/L for non-transformed controls. The use of dehD could thus be advantageous for herbicide-resistant plant breeding systems, and it is also a suitable marker gene for plant-transformation studies.     Oral cancer secretome: Identification of cancer-associated proteins

    This study aims to identify cancer-associated proteins in the secretome of oral cancer cell lines. We have successfully established four primary cell cultures of normal cells with a limited lifespan without human telomerase reverse transcriptase (hTERT) immortalization. The secretome of these primary cell cultures were compared with that of oral cancer cell lines using 2DE. Thirty five protein spots were found to have changed in abundance. Unambiguous identification of these proteins was achieved by MALDI TOF/TOF. In silico analysis predicted that 24 of these proteins were secreted via classical or nonclassical mechanisms. The mRNA expression of six genes was found to correlate with the corresponding protein abundance. Ingenuity Pathway Analysis (IPA) core analysis revealed that the identified proteins were relevant in, and related to, cancer development with likely involvements in tumor growth, metastasis, hyperproliferation, tumorigenesis, neoplasia, hyperplasia, and cell transformation. In conclusion, we have demonstrated that a comparative study of the secretome of cancer versus normal cell lines can be used to identify cancer-associated proteins.Article Link: http://onlinelibrary.wiley.com/doi/10.1002/elps.201300126/abstrac

    Ultrasensitive Detection of Cancer Biomarkers in the Clinic by Use of a Nanostructured Microfluidic Array

    Multiplexed biomarker protein detection holds unrealized promise for clinical cancer diagnostics due to lack of suitable measurement devices and lack of rigorously validated protein panels. Here we report an ultrasensitive electrochemical microfluidic array optimized to measure a four-protein panel of biomarker proteins, and we validate the protein panel for accurate oral cancer diagnostics. Unprecedented ultralow detection into the 5-50 fg.mL(-1) range was achieved for simultaneous measurement of proteins interleukin 6 (IL-6), IL-8, vascular endothelial growth factor (VEGF), and VEGF-C in diluted serum. The immunoarray achieves high sensitivity in 50 min assays by using off-line protein capture by magnetic beads carrying 400 000 enzyme labels and similar to 100 000 antibodies. After capture of the proteins and washing to inhibit nonspecific binding, the beads are magnetically separated and injected into the array for selective capture by antibodies on eight nanostructured sensors. Good correlations with enzyme-linked immunosorbent assays (ELISA) for protein determinations in conditioned cancer cell media confirmed the accuracy of this approach. Normalized means of the four protein levels in 78 oral cancer patient serum samples and 49 controls gave clinical sensitivity of 89 and specificity of 98 for oral cancer detection, demonstrating high diagnostic utility. The low-cost, easily fabricated immunoarray provides a rapid Serum test for diagnosis and personalized therapy of oral cancer.     Measurement of the quasi-elastic axial vector mass in neutrino-oxygen interactions

    The weak nucleon axial-vector form factor for quasi-elastic interactions is determined using neutrino interaction data from the K2K Scintillating Fiber detector in the neutrino beam at KEK. More than 12,000 events are analyzed, of which half are charged-current quasi-elastic interactions nu-mu n to mu- p occurring primarily in oxygen nuclei. We use a relativistic Fermi gas model for oxygen and assume the form factor is approximately a dipole with one parameter, the axial vector mass M_A, and fit to the shape of the distribution of the square of the momentum transfer from the nucleon to the nucleus. Our best fit result for M_A = 1.20 \pm 0.12 GeV. Furthermore, this analysis includes updated vector form factors from recent electron scattering experiments and a discussion of the effects of the nucleon momentum on the shape of the fitted distributions.Comment: 14 pages, 10 figures, 6 table

    Search for the W-exchange decays B0 --> Ds(*)- Ds(*)+

    We report a search for the decays B0DsDs+B^{0} \to D_{s}^{-} D_{s}^{+}, B0DsDs+B^{0} \to D_{s}^{*-} D_{s}^{+}, B0DsDs+B^{0} \to D_{s}^{*-} D_{s}^{*+} in a sample of 232 million Υ(4S)\Upsilon(4S) decays to \BBb ~pairs collected with the \babar detector at the PEP-II asymmetric-energy e+ee^+ e^- storage ring. We find no significant signal and set upper bounds for the branching fractions: B(B0DsDs+)<1.0×104,B(B0DsDs+)<1.3×104{\cal B}(B^{0} \to D_{s}^{-} D_{s}^{+}) < 1.0 \times 10^{-4}, {\cal B}(B^{0} \to D_{s}^{*-} D_{s}^{+}) < 1.3 \times 10^{-4} and B(B0DsDs+)<2.4×104{\cal B}(B^{0} \to D_{s}^{*-} D_{s}^{*+}) < 2.4 \times 10^{-4} at 90% confidence level.Comment: 8 pages, 2 figures, submitted to PRD-R

    Measurement of the B+ --> p pbar K+ Branching Fraction and Study of the Decay Dynamics

    With a sample of 232x10^6 Upsilon(4S) --> BBbar events collected with the BaBar detector, we study the decay B+ --> p pbar K+ excluding charmonium decays to ppbar. We measure a branching fraction Br(B+ --> p pbar K+)=(6.7+/-0.5+/-0.4)x10^{-6}. An enhancement at low ppbar mass is observed and the Dalitz plot asymmetry suggests dominance of the penguin amplitude in this B decay. We search for a pentaquark candidate Theta*++ decaying into pK+ in the mass range 1.43 to 2.00 GeV/c2 and set limits on Br(B+ --> Theta*++pbar)xBr(Theta*++ --> pK+) at the 10^{-7} level.Comment: 8 pages, 7 postscript figures, submitted to Phys.     Evidence for the η_b(1S) Meson in Radiative Υ(2S) Decay

    We have performed a search for the η_b(1S) meson in the radiative decay of the Υ(2S) resonance using a sample of 91.6 × 10^6 Υ(2S) events recorded with the BABAR detector at the PEP-II B factory at the SLAC National Accelerator Laboratory. We observe a peak in the photon energy spectrum at E_γ = 609.3^(+4.6)_(-4.5)(stat)±1.9(syst) MeV, corresponding to an η_b(1S) mass of 9394.2^(+4.8)_(-4.9)(stat) ± 2.0(syst) MeV/c^2. The branching fraction for the decay Υ(2S) → γη_b(1S) is determined to be [3.9 ± 1.1(stat)^(+1.1)_(-0.9)(syst)] × 10^(-4). We find the ratio of branching fractions B[Υ(2S) → γη_b(1S)]/B[Υ(3S) → γη_b(1S)]= 0.82 ± 0.24(stat)^(+0.20)_(-0.19)(syst)

    Isoprene hotspots at the Western Coast of Antarctic Peninsula during MASEC′16

    Isoprene (C5H8) plays an important role in the formation of surface ozone (O3) and the secondary organic aerosol (SOA) which contributed to the climate change. This study aims to determine hourly distribution of tropospheric isoprene over the Western Coast of Antarctic Peninsula (WCAP) during the Malaysian Antarctic Scientific Expedition Cruise 2016 (MASEC′16). In-situ measurements of isoprene were taken using a custom-built gas chromatography with photoionization detector, known as iDirac. Biological parameters such as chlorophyll a (chl-a) and particulate organic carbon (POC) were compared to the in-situ isoprene measurements. Significant positive correlation was observed between isoprene and POC concentrations (r2 = 0.67, p < 0.001), but not between isoprene and chl-a. The hotspots of isoprene over maritime Antarctic were then were investigated using NAME dispersion model reanalysis. Measurements showed that isoprene mixing ratio were the highest over region of King George Island, Deception Island and Booth Island with values of ∼5.0, ∼0.9 and ∼5.2 ppb, respectively. Backward trajectory analysis showed that air masses may have lifted the isoprene emitted by marine algae. We believe our findings provide valuable data set of isoprene estimation over the under sampled WCAP
