    Gel-Free 3D Tumoroids with Stem Cell Properties Modeling Drug Resistance to Cisplatin and Imatinib in Metastatic Colorectal Cancer

    Researchers have developed several three-dimensional (3D) culture systems, including spheroids, organoids, and tumoroids with increased properties of cancer stem cells (CSCs), also called cancer-initiating cells (CICs). Drug resistance is a crucial issue involving recurrence in cancer patients. Many studies on anti-cancer drugs have been reported using 2D culture systems, whereas 3D cultured tumoroids have many advantages for assessing drug sensitivity and resistance. Here, we aimed to investigate whether Cisplatin (a DNA crosslinker), Imatinib (a multiple tyrosine kinase inhibitor), and 5-Fluorouracil (5-FU: an antimetabolite) alter the tumoroid growth of metastatic colorectal cancer (mCRC). Gene expression signatures of highly metastatic aggregative CRC (LuM1 cells) vs. low-metastatic, non-aggregative CRC (Colon26 and NM11 cells) were analyzed using microarray. To establish a 3D culture-based multiplexing reporter assay system, LuM1 was stably transfected with the Mmp9 promoter-driven ZsGreen fluorescence reporter gene, which was designated as LuM1/m9 cells and cultured in NanoCulture Plate®, a gel-free 3D culture device. LuM1 cells highly expressed mRNA encoding ABCG2 (a drug resistance pump, i.e., CSC/CIC marker), other CSC/CIC markers (DLL1, EpCAM, podoplanin, STAT3/5), pluripotent stem cell markers (Sox4/7, N-myc, GATA3, Nanog), and metastatic markers (MMPs, Integrins, EGFR), compared to the other two cell types. Hoechst efflux stem cell-like side population was increased in LuM1 (7.8%) compared with Colon26 (2.9%), both of which were markedly reduced by verapamil treatment, an ABCG2 inhibitor. Smaller cell aggregates of LuM1 were more sensitive to Cisplatin (at 10 μM), whereas larger tumoroids with increased ABCG2 expression were insensitive. Notably, Cisplatin (2 μM) and Imatinib (10 μM) at low concentrations significantly promoted tumoroid formation (cell aggregation) and increased Mmp9 promoter activity in mCRC LuM1/m9, while not cytotoxic to them. On the other hand, 5-FU significantly inhibited tumoroid growth, although not completely. Thus, drug resistance in cancer with increased stem cell properties was modeled using the gel-free 3D cultured tumoroid system. The tumoroid culture is useful and easily accessible for the assessment of drug sensitivity and resistance

    新型コロナウイルス感染拡大により生活様式が変化したが,新型コロナウイルス感染拡大が大学職員の健康管理に及ぼした影響は明らかでない。本研究では40歳以上の大学職員1608名を対象に,新型コロナウイルス感染拡大前後の職員健康診断データを比較検討した。年齢を調整すると,感染拡大前と比較して,感染拡大後ではALT(β= 0.04, P = 0.02),HDL コレステロール(β= -0.07, P = 0.0002),LDL コレステロール(β= 0.04, P = 0.02)が悪化した一方で,HbA1c(β= -0.04, P = 0.02)は改善した。BMI,腹囲,AST,γ-GT,血糖値,中性脂肪,収縮期血圧,拡張期血圧,尿酸については感染拡大前後で変化を認めなかった。大学職員の健康管理において,感染拡大による職員個人のライフスタイル変化を考慮した生活指導が重要と考えられる。Although the spread of COVID-19 brought radical change to our lifestyle, its impact on health management of university staff is unknown. We enrolled 1608 Hiroshima University staff aged 40 years or more and compared their health checkup data before and after the spread of COVID-19. After adjustment for age, values of alanine aminotransferase (β= 0.04, P = 0.02), high-density lipoprotein cholesterol (β= -0.07, P = 0.0002), and low-density lipoprotein cholesterol (β= 0.04, P = 0.02) were worsened due to the spread of COVID-19. However, hemoglobin A1c (β= -0.04, P = 0.02) was improved, and values of body mass index, waist circumference, aspartate transaminase, γ-glutamyl transpeptidase, blood glucose, triglyceride, systolic and diastolic blood pressure, and uric acid were unchanged by the spread of COVID-19. For better physical health management of the university staff, individual lifestyle changes due to the spread of COVID-19 need to be considered

    Deaths at work: analysis of branches of activity

    Introdução: Sabe-se que as estatísticas referentes aos indicadores gerais e específicos relacionados ao trabalho e à sua mortalidade são escassos, o que pode ser em decorrência da carência de computação destes dados, pois uma parcela grande de trabalhadores não se enquadra como segurados da previdência social e, portanto, não é incluída nas estatísticas. Este estudo visa a analisar as taxas de morte relacionada ao trabalho por ramo de atividade, verificando em que ramos a morte é mais prevalente de acordo com o CNAE (Classificação Nacional de Atividades Econômicas) e discutir acerca das possíveis causas correlacionadas.  Objetivos: Analisar e discutir acerca das principais taxas de morte relacionadas ao trabalho por ramo de atividade, no âmbito previdenciário. Método: revisão literária na base de dados Scielo nos meses de outubro, novembro e dezembro de 2019 e análise do Anuário Estatístico de Acidentes do Trabalho de 2017. Resultados: obtém-se, para o ano de 2015, 3,9% de mortalidade. Em 2016, houve leve redução, com 3,7%. Dentre os CNAE com maior mortalidade, no ano de 2015, temos 1.071 (34 mortes), 4.120 (107 mortes), 4.711 (45 mortes), 4.744 (46 mortes), 4.930 (301 mortes), 8.011 (62 mortes). Conclusão: as mortes em decorrência do trabalho mostram maiores índices entre os ramos de Transporte Rodoviário, Construção Civil e Atividades de Vigilância, Segurança e Investigação. Fatores como violência no trânsito e urbana podem ser causas que contribuem diretamente para estes números.Introduction: Statistics related to general and specific indicators of work and its mortality are known to be scarce. This may be due to the lack of data input since a large portion of workers do not qualify for Social Security coverage and therefore do not enter the statistics. This study aims to analyze the most prevalent work-related death rates by sector according to the CNAE (National Classification of Economic Activities) and to discuss the possible correlated causes. Objectives: To analyze and discuss the main work-related death rates by type of activity in the scope of social security. Method: Literary review in the Scielo database in the months of October, November, and December 2019 and analysis of the Statistical Yearbook of Accidents at Work in 2017. Results: The year 2015 had a 3.9% mortality rate. In 2016, there is a slight reduction to 3.7%. Among the CNAE with the highest mortality rates in 2015, we have 1071 (34 deaths), 4120 (107 deaths), 4711 (45 deaths), 4744 (46 deaths), 4930 (301 deaths), and 8011 (62 deaths). Conclusion: Deaths due to work show higher rates among the sectors of Road Transport, Civil Construction, and Surveillance, Security, and Investigation Activities. Factors such as traffic and urban violence can be a direct contributing factor to these numbers

    Surgical resection combined with perioperative chemotherapy for a patient with locally recurrent, previously stage IV thymic small-cell carcinoma : A case report

    An 83-year-old Japanese man visited our hospital with dyspnea and general fatigue. Computed tomography (CT) revealed a tumor in the anterior mediastinum, bilateral pleural effusion, pericardial fluid, and multiple liver nodules. We performed a CT-guided tumor biopsy, and the patient was diagnosed with thymic small-cell carcinoma, Masaoka–Koga stage classification IVb. The patient received four cycles of carboplatin and etoposide, and all lesions disappeared on CT. However, after 6 months, CT revealed a recurrent tumor in the anterior mediastinum. After one cycle of rechallenge chemotherapy, we performed extended total thymectomy followed by another three cycles of chemotherapy. More than 2.5 years after the last chemotherapy session, the patient’s carcinoma did not recur. Thus, this case suggests that salvage surgery may be a treatment option for local recurrence of thymic carcinoma after complete remission with chemotherapy, even in patients with stage IV cancer

    Gene Expression Profile of Mesenchymal Stem Cells from Paired Umbilical Cord Units: Cord is Different from Blood

    Mesenchymal stem cells (MSC) are multipotent cells which can be obtained from several adult and fetal tissues including human umbilical cord units. We have recently shown that umbilical cord tissue (UC) is richer in MSC than umbilical cord blood (UCB) but their origin and characteristics in blood as compared to the cord remains unknown. Here we compared, for the first time, the exonic protein-coding and intronic noncoding RNA (ncRNA) expression profiles of MSC from match-paired UC and UCB samples, harvested from the same donors, processed simultaneously and under the same culture conditions. The patterns of intronic ncRNA expression in MSC from UC and UCB paired units were highly similar, indicative of their common donor origin. The respective exonic protein-coding transcript expression profiles, however, were significantly different. Hierarchical clustering based on protein-coding expression similarities grouped MSC according to their tissue location rather than original donor. Genes related to systems development, osteogenesis and immune system were expressed at higher levels in UCB, whereas genes related to cell adhesion, morphogenesis, secretion, angiogenesis and neurogenesis were more expressed in UC cells. These molecular differences verified in tissue-specific MSC gene expression may reflect functional activities influenced by distinct niches and should be considered when developing clinical protocols involving MSC from different sources. In addition, these findings reinforce our previous suggestion on the importance of banking the whole umbilical cord unit for research or future therapeutic use
