Evaluation of the anti-inflammatory effect of chalcone and chalcone analogues in a zebrafish model
[[abstract]]The aim of this study was to investigate novel chalcones with potent anti-inflammatory activities in vivo. Chalcone and two chalcone analogues (compound 5 and 9) were evaluated using a caudal fin-wounded transgenic zebrafish line “Tg(mpx:gfp)” to visualize the effect of neutrophil recruitment dynamically. Results showed that treatment with compound 9 not only affected wound-induced neutrophil recruitment, but also affected Mpx enzymatic activity. Moreover, protein expression levels of pro-inflammatory factors (Mpx, NFκB, and TNFα) were also regulated by compound 9. Taken together, our results provide in vivo evidence of the anti-inflammatory effects of synthesized chalcone analogues on wound-induced inflammation.[[notice]]補正完畢[[incitationindex]]SCI[[booktype]]紙本[[booktype]]電子

FAM129B, an antioxidative protein, reduces chemosensitivity by competing with Nrf2 for Keap1 binding.
BackgroundThe transcription factor Nrf2 is a master regulator of antioxidant response. While Nrf2 activation may counter increasing oxidative stress in aging, its activation in cancer can promote cancer progression and metastasis, and confer resistance to chemotherapy and radiotherapy. Thus, Nrf2 has been considered as a key pharmacological target. Unfortunately, there are no specific Nrf2 inhibitors for therapeutic application. Moreover, high Nrf2 activity in many tumors without Keap1 or Nrf2 mutations suggests that alternative mechanisms of Nrf2 regulation exist.MethodsInteraction of FAM129B with Keap1 is demonstrated by immunofluorescence, colocalization, co-immunoprecipitation and mammalian two-hybrid assay. Antioxidative function of FAM129B is analyzed by measuring ROS levels with DCF/flow cytometry, Nrf2 activation using luciferase reporter assay and determination of downstream gene expression by qPCR and wester blotting. Impact of FAM129B on in vivo chemosensitivity is examined in mice bearing breast and colon cancer xenografts. The clinical relevance of FAM129B is assessed by qPCR in breast cancer samples and data mining of publicly available databases.FindingsWe have demonstrated that FAM129B in cancer promotes Nrf2 activity by reducing its ubiquitination through competition with Nrf2 for Keap1 binding via its DLG and ETGE motifs. In addition, FAM129B reduces chemosensitivity by augmenting Nrf2 antioxidative signaling and confers poor prognosis in breast and lung cancer.InterpretationThese findings demonstrate the important role of FAM129B in Nrf2 activation and antioxidative response, and identify FMA129B as a potential therapeutic target. Reduced expression of alpha-1,2-mannosidase I extends lifespan in Drosophila melanogaster and Caenorhabditis elegans
Exposure to sub-lethal levels of stress, or hormesis, was a means to induce longevity. By screening for mutations that enhance resistance to multiple stresses, we identified multiple alleles of alpha-1,2-mannosidase I (mas1) which, in addition to promoting stress resistance, also extended longevity. Longevity enhancement is also observed when mas1 expression is reduced via RNA interference in both Drosophila melanogaster and Caenorhabditis elegans. The screen also identified Edem1 (Edm1), a gene downstream of mas1, as a modulator of lifespan. As double mutants for both mas1 and Edm1 showed no additional longevity enhancement, it appeared that both mutations function within a common pathway to extend lifespan. Molecular analysis of these mutants revealed that the expression of BiP, a putative biomarker of dietary restriction (DR), is down-regulated in response to reductions in mas1 expression. AMiBA: Broadband Heterodyne CMB Interferometry
The Y. T. Lee Array for Microwave Background (AMiBA) has reported the first
science results on the detection of galaxy clusters via the Sunyaev Zel'dovich
effect. The science objectives required small reflectors in order to sample
large scale structures (20') while interferometry provided modest resolutions
(2'). With these constraints, we designed for the best sensitivity by utilizing
the maximum possible continuum bandwidth matched to the atmospheric window at
86-102GHz, with dual polarizations. A novel wide-band analog correlator was
designed that is easily expandable for more interferometer elements. MMIC
technology was used throughout as much as possible in order to miniaturize the
components and to enhance mass production. These designs will find application
in other upcoming astronomy projects. AMiBA is now in operations since 2006,

THE EFFECT OF TENNIS RACKET STRING VIBRATION ABSORBER PLACEMENT ON TENNIS RACKET VIBRATIONS
The purpose of this research was to analyze the vibration effect of tennis racket caused by impact on different locative vibration absorber and locations. In this experiment, The tennis racket was fixed and kept the face horizontalized to the ground with 50 Ib on strings. A tennis ball was dropped from one meter height to impact three locations of the racket. The different locative vibration absorber was fixed on the racket. It was installed an accelerometer to capture the vibration signal of the handle of the racket. After the experiment, the data was used logarithmic decrement to calculate damping ratio. It was illustrated when the vibration absorber in Back and Tip to have obvious effect. But in Side, it could not effectively decrease the vibration compared with the racket without absorber
AMiBA: scaling relations between the integrated Compton-y and X-ray derived temperature, mass, and luminosity
We investigate the scaling relations between the X-ray and the thermal
Sunyaev-Zel'dovich Effect (SZE) properties of clusters of galaxies, using data
taken during 2007 by the Y.T. Lee Array for Microwave Background Anisotropy
(AMiBA) at 94 GHz for the six clusters A1689, A1995, A2142, A2163, A2261, and
A2390. The scaling relations relate the integrated Compton-y parameter Y_{2500}
to the X-ray derived gas temperature T_{e}, total mass M_{2500}, and bolometric
luminosity L_X within r_{2500}. Our results for the power-law index and
normalization are both consistent with the self-similar model and other studies
in the literature except for the Y_{2500}-L_X relation, for which a physical
explanation is given though further investigation may be still needed. Our
results not only provide confidence for the AMiBA project but also support our

DNA topoisomerases participate in fragility of the oncogene RET
Fragile site breakage was previously shown to result in rearrangement of the RET oncogene, resembling the rearrangements found in thyroid cancer. Common fragile sites are specific regions of the genome with a high susceptibility to DNA breakage under conditions that partially inhibit DNA replication, and often coincide with genes deleted, amplified, or rearranged in cancer. While a substantial amount of work has been performed investigating DNA repair and cell cycle checkpoint proteins vital for maintaining stability at fragile sites, little is known about the initial events leading to DNA breakage at these sites. The purpose of this study was to investigate these initial events through the detection of aphidicolin (APH)-induced DNA breakage within the RET oncogene, in which 144 APHinduced DNA breakpoints were mapped on the nucleotide level in human thyroid cells within intron 11 of RET, the breakpoint cluster region found in patients. These breakpoints were located at or near DNA topoisomerase I and/or II predicted cleavage sites, as well as at DNA secondary structural features recognized and preferentially cleaved by DNA topoisomerases I and II. Co-treatment of thyroid cells with APH and the topoisomerase catalytic inhibitors, betulinic acid and merbarone, significantly decreased APH-induced fragile site breakage within RET intron 11 and within the common fragile site FRA3B. These data demonstrate that DNA topoisomerases I and II are involved in initiating APH-induced common fragile site breakage at RET, and may engage the recognition of DNA secondary structures formed during perturbed DNA replication
AMiBA Wideband Analog Correlator
A wideband analog correlator has been constructed for the Yuan-Tseh Lee Array
for Microwave Background Anisotropy. Lag correlators using analog multipliers
provide large bandwidth and moderate frequency resolution. Broadband IF
distribution, backend signal processing and control are described. Operating
conditions for optimum sensitivity and linearity are discussed. From
observations, a large effective bandwidth of around 10 GHz has been shown to
provide sufficient sensitivity for detecting cosmic microwave background
