    Probing dynamics of dark energy with latest observations

    We examine the validity of the Λ\LambdaCDM model, and probe for the dynamics of dark energy using latest astronomical observations. Using the Om(z)Om(z) diagnosis, we find that different kinds of observational data are in tension within the Λ\LambdaCDM framework. We then allow for dynamics of dark energy and investigate the constraint on dark energy parameters. We find that for two different kinds of parametrisations of the equation of state parameter ww, a combination of current data mildly favours an evolving ww, although the significance is not sufficient for it to be supported by the Bayesian evidence. A forecast of the DESI survey shows that the dynamics of dark energy could be detected at 7σ7\sigma confidence level, and will be decisively supported by the Bayesian evidence, if the best fit model of ww derived from current data is the true model.Comment: 4.5 pages, 3 figures, 1 table; references adde

    A polymer ruthenium complex pbbp-RuCl3 has been easily synthesized from the direct coordination of RuCl3 with polymer constituted by tridentate 2,6-bis(benzimidazol-2-yl)pyridine unit (pbbp). As a heterogeneous catalyst, pbbp-RuCl3 showed high efficiency in aerobic oxidative self-condensation of primary amines to imines.     RITA: Boost Autonomous Driving Simulators with Realistic Interactive Traffic Flow

    High-quality traffic flow generation is the core module in building simulators for autonomous driving. However, the majority of available simulators are incapable of replicating traffic patterns that accurately reflect the various features of real-world data while also simulating human-like reactive responses to the tested autopilot driving strategies. Taking one step forward to addressing such a problem, we propose Realistic Interactive TrAffic flow (RITA) as an integrated component of existing driving simulators to provide high-quality traffic flow for the evaluation and optimization of the tested driving strategies. RITA is developed with consideration of three key features, i.e., fidelity, diversity, and controllability, and consists of two core modules called RITABackend and RITAKit. RITABackend is built to support vehicle-wise control and provide traffic generation models from real-world datasets, while RITAKit is developed with easy-to-use interfaces for controllable traffic generation via RITABackend. We demonstrate RITA's capacity to create diversified and high-fidelity traffic simulations in several highly interactive highway scenarios. The experimental findings demonstrate that our produced RITA traffic flows exhibit all three key features, hence enhancing the completeness of driving strategy evaluation. Moreover, we showcase the possibility for further improvement of baseline strategies through online fine-tuning with RITA traffic flows.Comment: 8 pages, 5 figures, 3 table

    The change of non-alcoholic fatty liver disease is associated with risk of incident diabetes

    Background & aimsThe effect of change in non-alcoholic fatty liver disease (NAFLD) status on incident diabetes has not been well studied. We aimed to investigate the association of NAFLD development and remission with the risk of incident diabetes during a median of 3.5-year follow-up.MethodsA total of 2690 participants without diabetes were recruited in 2011-2012 and assessed for incident diabetes in 2014. Abdominal ultrasonography was used to determine the change of NAFLD. 75 g oral glucose tolerance test (OGTT) was performed to determine diabetes. NAFLD severity was assessed using Gholam’s model. The odds ratios (ORs) for incident diabetes were estimated by logistic regression models.ResultsNAFLD was developed in 580 (33.2%) participants and NAFLD remission occurred in 150 (15.9%) participants during a median of 3.5-year follow-up. A total of 484 participants developed diabetes during follow-up, including 170 (14.6%) in consistent non-NAFLD group, 111 (19.1%) in NAFLD developed group, 19 (12.7%) in NAFLD remission group, and 184 (23.2%) in sustained NAFLD group. The development of NAFLD increased the risk of incident diabetes by 43% (OR, 1.43; 95%CI, 1.10-1.86) after adjustment for multiple confounders. Compared with sustained NAFLD group, remission of NAFLD reduced the risk of incident diabetes by 52% (OR, 0.48; 95%CI, 0.29-0.80). The effect of NAFLD alteration on incident diabetes was not changed after adjustment for body mass index or waist circumference, change of body mass index or waist circumference. In NAFLD remission group, participants with non-alcoholic steatohepatitis (NASH) at baseline were more likely to develop diabetes (OR, 3.03; 95%CI, 1.01-9.12).ConclusionsNAFLD development increases the risk of incident diabetes, whereas NAFLD remission reduces the risk of incident diabetes. Moreover, presence of NASH at baseline could attenuate the protective effect of NAFLD remission on incident diabetes. Our study suggests that early intervention of NAFLD and maintenance of non-NAFLD are important for prevention of diabetes

    Kuhuang alleviates liver fibrosis by modulating gut microbiota-mediated hepatic IFN signaling and bile acid synthesis

    Background: Liver fibrosis is a common outcome of the pathological progression of chronic liver disease; however, no specific and effective therapeutic agent has been approved for its treatment. We investigated the effects of Kuhuang on liver fibrosis and the underlying mechanisms of action.Materials and methods: To induce hepatic fibrosis, either 3,5-diethoxycarbonyl-1,4-dihydro-collidine (DDC) diet was administered, or bile duct ligation (BDL) surgery was performed on C57BL/6 mice. Kuhuang was orally administered to mice for 7 days before and after bile duct ligation or 4 weeks with a DDC diet. Hematoxylin and eosin, Sirius red staining, and immunohistochemical analyses were performed to evaluate hepatic pathology. Hepatic interferon-β (IFN-β) levels were measured using an enzyme-linked immunosorbent assay. RNA sequencing was performed to examine the gene expression profiles of liver tissues. The mRNA expression of inflammatory, profibrotic, and bile acid (BA)-related genes was further validated by qRT-PCR. A targeted metabolomics assay revealed the alteration of the hepatic bile acid (BA) composition. The composition of the gut microbiota was determined via 16S rRNA sequencing.Results: Treatment with Kuhuang attenuated liver fibrosis and reduced the inflammatory response in bile duct ligation and DDC mouse models. In addition, the hepatic IFN signaling pathway was activated following Kuhuang treatment. Kuhuang treatment also significantly decreased hepatic levels of both primary and secondary BAs. In addition, Kuhuang treatment altered gut microbiota composition, with an increased abundance of interferon-inducing Akkermansia and decreased abundance of bile salt hydrolase-producing Lactobacillus, Clostridium, and Bifidobacterium. Furthermore, the abundance of Akkermansia was positively correlated with the hepatic mRNA expression levels of Ifna4, Ifnb, and Isg15, whereas that of Lactobacillus, Clostridium-sensu-stricto-1, and Bifidobacterium was positively correlated with levels of bile acid synthesis-related genes.Conclusion: Our results suggest that Kuhuang plays a protective role during the progression of liver fibrosis, potentially by altering the composition of the gut microbiota, which consequently activates interferon signaling and inhibits bile acid synthesis in the liver

    Biological and genomic analysis of a symbiotic nitrogen fixation defective mutant in Medicago truncatula

    Medicago truncatula has been selected as one of the model legume species for gene functional studies. To elucidate the functions of the very large number of genes present in plant genomes, genetic mutant resources are very useful and necessary tools. Fast Neutron (FN) mutagenesis is effective in inducing deletion mutations in genomes of diverse species. Through this method, we have generated a large mutant resource in M. truncatula. This mutant resources have been used to screen for different mutant using a forward genetics methods. We have isolated and identified a large amount of symbiotic nitrogen fixation (SNF) deficiency mutants. Here, we describe the detail procedures that are being used to characterize symbiotic mutants in M. truncatula. In recent years, whole genome sequencing has been used to speed up and scale up the deletion identification in the mutant. Using this method, we have successfully isolated a SNF defective mutant FN007 and identified that it has a large segment deletion on chromosome 3. The causal deletion in the mutant was confirmed by tail PCR amplication and sequencing. Our results illustrate the utility of whole genome sequencing analysis in the characterization of FN induced deletion mutants for gene discovery and functional studies in the M. truncatula. It is expected to improve our understanding of molecular mechanisms underlying symbiotic nitrogen fixation in legume plants to a great extent

    Estimation of HIV-1 incidence among five focal populations in Dehong, Yunnan: a hard hit area along a major drug trafficking route

    <p>Abstract</p> <p>Background</p> <p>Since 1989 when the first 146 HIV positives in China were identified, Dehong Prefecture had been one of the areas hardest-hit by HIV in China. The local and national governments have put substantial financial resources into tackling the HIV epidemic in Dehong from 2004. The objective of this study was to track dynamic changes in HIV-1 prevalence and incidence among five focal populations in Dehong and to assess the impact of HIV prevention and control efforts.</p> <p>Methods</p> <p>Consecutive cross-sectional surveys conducted in five focal populations between 2004 and 2008. Specimens seropositive for HIV were tested with the BED IgG capture enzyme immunoassay to identify recent seroconversions (median, 155 days) using normalized optical density of 0.8 and adjustments.</p> <p>Results</p> <p>From 2004 to 2008, estimated annual HIV incidence among injecting drug users (IDUs) decreased significantly [from 15.0% (95% CI = 11.4%-18.5%) in 2004 to 4.3% (95% CI = 2.4%-6.2%) in 2008; trend test P < 0.0001]. The incidence among other focal populations, such as HIV discordant couples (varying from 5.5% to 4.7%), female sex workers (varying from 1.4% to 1.3%), pregnant women (0.1%), and pre-marital couples (0.2 to 0.1%) remained stable. Overall, the proportion of recent HIV-1 infections was higher among females than males (P < 0.0001).</p> <p>Conclusions</p> <p>The HIV epidemic in Dehong continued to expand during a five-year period but at a slowing rate among IDUs, and HIV incidence remains high among IDUs and discordant couples. Intensive prevention measures should target sub-groups at highest risk to further slow the epidemic and control the migration of HIV to other areas of China, and multivariate analysis is needed to explore which measures are more effective for different populations.</p

    Glycosylation in cancer and infection : the role of sialic acid

    Sialic acids (SA), a group of nine-carbon backbone monosaccharides are abundantly expressed in vertebrates. They are usually linked to the terminal of glycan chains and play crucial roles in many biological processes, including cell adhesion, cell-cell interactions, immune modulation, cancer cell migration and invasion, as well as viral infections. To analyze and monitor SA expression, antibodies and glycan-binding lectins are typically used. However, high costs and poor stability limit the application in SA analysis. To overcome these drawbacks, an imprinting technique was used to synthesize an alternative SA receptor – SA molecularly imprinted polymers (SA-MIPs). Fluorescent molecules are embedded into the MIPs, facilitating the detection of MIPs binding to cells by flow cytometry and fluorescence microscopy. Firstly, core-shell SA imprinted MIPs were used to analyze SA expression in a panel of breast cancer cell lines. The SA expression of these cell lines was also tested by using the two glycan-binding lectins, MAL andSNA, which recognize α2,3 and α2,6 SA, respectively. Our results show that breast cancer cell lines express α2,3 and α2,6 SA dissimilarly, and hence present different SA-MIP binding patterns. The specificity of SA-MIPs was further verified by an inhibition assay using two pentavalent SA conjugates that interfere with the SAMIPs.Furthermore, the SA-MIP synthesis protocol has been improved by using silica-coated polystyrene particles. The polystyrene core particles are lighter and smaller, increasing MIP suspensibility and augmenting MIP-cancer cell interactions. The cancer cell binding properties and the specificity have been verified by using thirteen different cancer cell lines, showing that the SA-MIPs can be used as effective tools for SA expression analysis. The SA-MIPs were used to analyze the SA expression of in vitro cultured cells treated with soluble cytokines to mimic the tumor microenvironment. The SA expression of two cancer cell lines stimulated with soluble cytokines was analyzed by using lectins and SA-MIPs. The MIPbinding data correlated well with lectin staining results, demonstrating the potential of SA-MIPs to be used in the analysis of overexpressed SA in the tumor microenvironment. Furthermore, the involvement of SA in the infection of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) was assessed. The viral surface receptor-binding domain (RBD) recognizes and conjugates with receptors on host cells, triggering the infection. Although the interaction between the RBD and host cells has been extensively studied, the mechanism behind this reaction is not fully determined. In this study, the interaction between the viral RBD and a panel of human cell lines from tissues susceptible to viral infection was tested. Moreover, the role of SA in this interaction has also been tested and evaluated
