The RAB2B-GARIL5 Complex Promotes Cytosolic DNA-Induced Innate Immune Responses
Cyclic GMP-AMP synthase (cGAS) is a cytosolic DNA sensor that induces the IFN antiviral response. However, the regulatory mechanisms that mediate cGAS-triggered signaling have not been fully explored. Here, we show the involvement of a small GTPase, RAB2B, and its effector protein, Golgi-associated RAB2B interactor-like 5 (GARIL5), in the cGAS-mediated IFN response. RAB2B-deficiency affects the IFN response induced by cytosolic DNA. Consistent with this, RAB2B deficiency enhances replication of vaccinia virus, a DNA virus. After DNA stimulation, RAB2B colocalizes with stimulator of interferon genes (STING), the downstream signal mediator of cGAS, on the Golgi apparatus. The GTP-binding activity of RAB2B is required for its localization on the Golgi apparatus and for recruitment of GARIL5. GARIL5 deficiency also affects the IFN response induced by cytosolic DNA and enhances replication of vaccinia virus. These findings indicate that the RAB2B-GARIL5 complex promotes IFN responses against DNA viruses by regulating the cGAS-STING signaling axis

Mobile DHHC palmitoylating enzyme mediates activity-sensitive synaptic targeting of PSD-95
Protein palmitoylation is the most common posttranslational lipid modification; its reversibility mediates protein shuttling between intracellular compartments. A large family of DHHC (Asp-His-His-Cys) proteins has emerged as protein palmitoyl acyltransferases (PATs). However, mechanisms that regulate these PATs in a physiological context remain unknown. In this study, we efficiently monitored the dynamic palmitate cycling on synaptic scaffold PSD-95. We found that blocking synaptic activity rapidly induces PSD-95 palmitoylation and mediates synaptic clustering of PSD-95 and associated AMPA (α-amino-3-hydroxy-5-methyl-4-isoxazole propionic acid)-type glutamate receptors. A dendritically localized DHHC2 but not the Golgi-resident DHHC3 mediates this activity-sensitive palmitoylation. Upon activity blockade, DHHC2 translocates to the postsynaptic density to transduce this effect. These data demonstrate that individual DHHC members are differentially regulated and that dynamic recruitment of protein palmitoylation machinery enables compartmentalized regulation of protein trafficking in response to extracellular signals
Preprint typeset using L ATEX style emulateapj v. 02/07/07 THE ORIGIN OF LARGE PECULIAR MOTIONS OF STAR-FORMING REGIONS AND SPIRAL STRUCTURES OF OUR GALAXY
Recent VLBI (Very Long Baseline Interferometer) observations determined the distances and proper motions of star-forming regions in spiral arms directly. They showed that star-forming regions and young stars have large peculiar motions, as large as 30 km s −1 with complex structures. Such a large peculiar motion is incompatible with the prediction of the standard theory of quasi-stationary spiral arms. We use a high-resolution, self-consistent N-body+hydrodynamical simulation to explore how the spiral arms are formed and maintained, and how star-forming regions move. We found that arms are not quasi-stationary but transient and recurrent, as suggested in alternative theories of spiral structures. Because of this transient nature of the spiral arms, star-forming regions exhibit a trend of large and complex non-circular motions, which is qualitatively consistent with the VLBI observations. Owing to this large non-circular motion, a kinematically estimated gas map of our Galaxy has a large systematic errors of ∼ 2 − 3 kpc in the distance from the Sun
Novel germline variant of TMEM127 gene in a patient with familial pheochromocytoma
Pheochromocytomas (PCCs) and paragangliomas (PGLs) are rare tumours with a heterogeneous genetic background. Up to 40% of apparently sporadic PCC/PGL cases carry 1 of the 12 gene germline mutations conferring genetic susceptibility to PCC/PGL. Although the precise mechanisms are unclear, TMEM127 is one of the rare responsible genes for PCC/PGL. Here we report the case of a patient with familial PCC having a novel TMEM127 variant (c.119C > T, p.S40F). In silico prediction analysis to evaluate the functional significance of this variant suggested that it is a disease-causing variant. A PCC on the left side was considered to be the dominant lesion, and unilateral adrenalectomy was performed. The histopathologic findings were consistent with benign PCC.
