    From Multiview Image Curves to 3D Drawings

    Reconstructing 3D scenes from multiple views has made impressive strides in recent years, chiefly by correlating isolated feature points, intensity patterns, or curvilinear structures. In the general setting - without controlled acquisition, abundant texture, curves and surfaces following specific models or limiting scene complexity - most methods produce unorganized point clouds, meshes, or voxel representations, with some exceptions producing unorganized clouds of 3D curve fragments. Ideally, many applications require structured representations of curves, surfaces and their spatial relationships. This paper presents a step in this direction by formulating an approach that combines 2D image curves into a collection of 3D curves, with topological connectivity between them represented as a 3D graph. This results in a 3D drawing, which is complementary to surface representations in the same sense as a 3D scaffold complements a tent taut over it.     Gene-modified T cells for adoptive immunotherapy of renal cell cancer maintain transgene-specific immune functions in vivo

    Abstract BACKGROUND: We have treated three patients with carboxy-anhydrase-IX (CAIX) positive metastatic renal cell cancer (RCC) by adoptive transfer of autologous T-cells that had been gene-transduced to express a single-chain antibody-G250 chimeric receptor [scFv(G250)], and encountered liver toxicity necessitating adaptation of the treatment protocol. Here, we investigate whether or not the in vivo activity of the infused scFv(G250)(+) T cells is reflected by changes of selected immune parameters measured in peripheral blood. METHODS: ScFv(G250)-chimeric receptor-mediated functions of peripheral blood mononuclear cells (PBMC) obtained from three patients during and after treatment were compared to the same functions of scFv(G250)(+) T lymphocytes prior to infusion, and were correlated with plasma cytokine levels. RESULTS: Prior to infusion, scFv(G250)(+) T lymphocytes showed in vitro high levels of scFv(G250)-chimeric receptor-mediated functions such as killing of CAIX(+) RCC cell lines and cytokine production upon exposure to these cells. High levels of IFN-gamma were produced, whilst production of TNF-alpha, interleukin-4 (IL-4), IL-5 and IL-10 was variable and to lower levels, and that of IL-2 virtually absent. PBMC taken from patients during therapy showed lower levels of in vitro scFv(G250)-receptor-mediated functions as compared to pre-infusion, whilst IFN-gamma was the only detectable cytokine upon in vitro PBMC exposure to CAIX. During treatment, plasma levels of IFN-gamma increased only in the patient with the most prominent liver toxicity. IL-5 plasma levels increased transiently during treatment in all patients, which may have been triggered by the co-administration of IL-2. CONCLUSION: ScFv(G250)-receptor-mediated functions of the scFv(G250)(+) T lymphocytes are, by and large, preserved in vivo upon administration, and may be reflected by fluctuations in plasma IFN-gamma levels

    Identification of Giardia lamblia DHHC Proteins and the Role of Protein S-palmitoylation in the Encystation Process

    Protein S-palmitoylation, a hydrophobic post-translational modification, is performed by protein acyltransferases that have a common DHHC Cys-rich domain (DHHC proteins), and provides a regulatory switch for protein membrane association. In this work, we analyzed the presence of DHHC proteins in the protozoa parasite Giardia lamblia and the function of the reversible S-palmitoylation of proteins during parasite differentiation into cyst. Two specific events were observed: encysting cells displayed a larger amount of palmitoylated proteins, and parasites treated with palmitoylation inhibitors produced a reduced number of mature cysts. With bioinformatics tools, we found nine DHHC proteins, potential protein acyltransferases, in the Giardia proteome. These proteins displayed a conserved structure when compared to different organisms and are distributed in different monophyletic clades. Although all Giardia DHHC proteins were found to be present in trophozoites and encysting cells, these proteins showed a different intracellular localization in trophozoites and seemed to be differently involved in the encystation process when they were overexpressed. dhhc transgenic parasites showed a different pattern of cyst wall protein expression and yielded different amounts of mature cysts when they were induced to encyst.     Tumor Organoids for Primary Liver Cancers: A Systematic Review of Current Applications in Diagnostics, Disease Modeling, and Drug Screening

    Background/AimsLiver cancer ranks third in cancer-related deaths globally, projected to exceed one millionannually by 2030. Existing therapies have significant limitations, including severe sideeffects and inconsistent efficacy. Innovative therapeutic approaches to address primary livercancer (PLC) have led to the ongoing development of tumor-derived organoids. These aresophisticated three-dimensional structures capable of mimicking native tissue architectureand function in vitro, improving our ability to model in vivo homeostasis and disease.MethodsThis systematic review consolidates known literature on human and mouse liver organoidsacross all PLC subtypes, emphasizing diagnostic precision, disease modeling, and drugscreening capabilities.ResultsAcross all 39 included studies, organoids were frequently patient derived organoids (PDO),closely followed by cancer cell line derived organoids (CCO). The literature concentrated onHepatocellular Carcinoma (HCC) and Intrahepatic Cholangiocarcinoma (ICC), whileexploration of other subtypes was limited. These studies demonstrate a valuable role for PLCorganoid cultures in biomarker discovery, disease modeling, and therapeutic exploration.ConclusionsEncouraging advancements such as organoid-on-a-chip and co-culturing systems presentpromising prospects in advancing treatment regimens for PLC. Standardizing in vitroprotocols is crucial to integrate research breakthroughs into practical treatment strategies forPLC.Impact and ImplicationsThis review underscores the expanding utility of PLC organoids across therapeutic discovery,diagnostics, and disease modeling. PDOs replicate many tumor characteristics. Novel genesfrom HCC organoids offer promising biomarkers for personalized treatments. Innovativemethodologies, like microfluidic chips, enhance organoid culture reproducibility. Despitelimitations, co-culturing, and organ-on-a-chip show potential in better mimicking the in vivo tumor microenvironment. These advancements position PLC organoids as crucial tools forpersonalized cancer therapy, biomarker discovery, and disease modeling, with ongoingprotocol standardization efforts essential for clinical applications.<br/

    OGLE-2009-BLG-092/MOA-2009-BLG-137: A Dramatic Repeating Event With the Second Perturbation Predicted by Real-Time Analysis

    We report the result of the analysis of a dramatic repeating gravitational microlensing event OGLE-2009-BLG-092/MOA-2009-BLG-137, for which the light curve is characterized by two distinct peaks with perturbations near both peaks. We find that the event is produced by the passage of the source trajectory over the central perturbation regions associated with the individual components of a wide-separation binary. The event is special in the sense that the second perturbation, occurring 100\sim 100 days after the first, was predicted by the real-time analysis conducted after the first peak, demonstrating that real-time modeling can be routinely done for binary and planetary events. With the data obtained from follow-up observations covering the second peak, we are able to uniquely determine the physical parameters of the lens system. We find that the event occurred on a bulge clump giant and it was produced by a binary lens composed of a K and M-type main-sequence stars. The estimated masses of the binary components are M1=0.69±0.11 MM_1=0.69 \pm 0.11\ M_\odot and M2=0.36±0.06 MM_2=0.36\pm 0.06\ M_\odot, respectively, and they are separated in projection by r=10.9±1.3 AUr_\perp=10.9\pm 1.3\ {\rm AU}. The measured distance to the lens is DL=5.6±0.7 kpcD_{\rm L}=5.6 \pm 0.7\ {\rm kpc}. We also detect the orbital motion of the lens system.Comment: 18 pages, 5 figures, 1 tabl

    Elevated plasma adiponectin levels in patients with chronic obstructive pulmonary disease

    BACKGROUND: Adiponectin is an anti-inflammatory adipokine that may play a role in chronic obstructive pulmonary disease (COPD) pathogenesis. OBJECTIVE: To investigate the relationship between adiponectin, interleukin (IL) 6, IL-8 and C-reactive protein (CRP) and COPD by evaluating these biomarkers in ever-smokers with or without the disease. METHOD: Plasma levels of adiponectin, IL-6, IL-8 and CRP were measured using commercially available kits in COPD patients (n = 71), healthy ever-smokers (n = 62) and non-smokers (n = 51). RESULTS: There were significant increases in plasma adiponectin, IL-6 and CRP in COPD patients (median [IQR] 4.39 μg/ml [2.68-6.98], 4.19 pg/ml [<2.40-6.40], 8.75 mg/l [4.26-40.63], respectively) compared to healthy ever-smokers (1.90 μg/ml [0.86-2.86], <2.40 pg/ml [<2.40-2.77], 3.71 mg/l [1.97-10.37 mg/l], respectively, P < 0.001) and non-smokers (1.76 μg/ml [1.34-2.52], <2.40 pg/ml [<2.40-2.78], 3.12 mg/l [2.11-5.71], respectively, P < 0.001). COPD patients had lower plasma IL-8 levels than healthy ever-smokers. Among ever-smokers with or without COPD, plasma adiponectin, IL-6 and CRP levels were inversely correlated with forced expiratory volume in 1 second (% predicted) after adjustment for age, body mass index, smoking status and pack-years.     Functional Redundancy of Two Pax-Like Proteins in Transcriptional Activation of Cyst Wall Protein Genes in Giardia lamblia

    The protozoan Giardia lamblia differentiates from a pathogenic trophozoite into an infectious cyst to survive outside of the host. During encystation, genes encoding cyst wall proteins (CWPs) are coordinately induced. Pax family transcription factors are involved in a variety of developmental processes in animals. Nine Pax proteins have been found to play an important role in tissue and organ development in humans. To understand the progression from primitive to more complex eukaryotic cells, we tried to identify putative pax genes in the G. lamblia genome and found two genes, pax1 and pax2, with limited similarity. We found that Pax1 may transactivate the encystation-induced cwp genes and interact with AT-rich initiatior elements that are essential for promoter activity and transcription start site selection. In this study, we further characterized Pax2 and found that, like Pax1, Pax2 was present in Giardia nuclei and it may specifically bind to the AT-rich initiator elements of the encystation-induced cwp1-3 and myb2 genes. Interestingly, overexpression of Pax2 increased the cwp1-3 and myb2 gene expression and cyst formation. Deletion of the C-terminal paired domain or mutation of the basic amino acids of the paired domain resulted in a decrease of nuclear localization, DNA-binding activity, and transactivation activity of Pax2. These results are similar to those found in the previous Pax1 study. In addition, the profiles of gene expression in the Pax2 and Pax1 overexpressing cells significantly overlap in the same direction and ERK1 associated complexes may phosphorylate Pax2 and Pax1, suggesting that Pax2 and Pax1 may be downstream components of a MAPK/ERK1 signaling pathway. Our results reveal functional redundancy between Pax2 and Pax1 in up-regulation of the key encystation-induced genes. These results illustrate functional redundancy of a gene family can occur in order to increase maintenance of important gene function in the protozoan organism G. lamblia

    Binary microlensing event OGLE-2009-BLG-020 gives a verifiable mass, distance and orbit predictions

    We present the first example of binary microlensing for which the parameter measurements can be verified (or contradicted) by future Doppler observations. This test is made possible by a confluence of two relatively unusual circumstances. First, the binary lens is bright enough (I=15.6) to permit Doppler measurements. Second, we measure not only the usual 7 binary-lens parameters, but also the 'microlens parallax' (which yields the binary mass) and two components of the instantaneous orbital velocity. Thus we measure, effectively, 6 'Kepler+1' parameters (two instantaneous positions, two instantaneous velocities, the binary total mass, and the mass ratio). Since Doppler observations of the brighter binary component determine 5 Kepler parameters (period, velocity amplitude, eccentricity, phase, and position of periapsis), while the same spectroscopy yields the mass of the primary, the combined Doppler + microlensing observations would be overconstrained by 6 + (5 + 1) - (7 + 1) = 4 degrees of freedom. This makes possible an extremely strong test of the microlensing solution. We also introduce a uniform microlensing notation for single and binary lenses, we define conventions, summarize all known microlensing degeneracies and extend a set of parameters to describe full Keplerian motion of the binary lenses.Comment: 51 pages, 8 figures, 2 appendices.
