    Synthetic biology for evolutionary engineering: from perturbation of genotype to acquisition of desired phenotype

    With the increased attention on bio-based industry, demands for techniques that enable fast and effective strain improvement have been dramatically increased. Evolutionary engineering, which is less dependent on biological information, has been applied to strain improvement. Currently, synthetic biology has made great innovations in evolutionary engineering, particularly in the development of synthetic tools for phenotypic perturbation. Furthermore, discovering biological parts with regulatory roles and devising novel genetic circuits have promoted high-throughput screening and selection. In this review, we first briefly explain basics of synthetic biology tools for mutagenesis and screening of improved variants, and then describe how these strategies have been improved and applied to phenotypic engineering. Evolutionary engineering using advanced synthetic biology tools will enable further innovation in phenotypic engineering through the development of novel genetic parts and assembly into well-designed logic circuits that perform complex tasks.11Ysciescopu

    Novel fungal genera and species associated with the sooty blotch and flyspeck complex on apple in China and the USA

    Fungi in the sooty blotch and flyspeck (SBFS) complex cause blemishes on apple and pear fruit that result in economic losses for growers. The SBFS fungi colonise the epicuticular wax layer of pomaceous fruit but do not invade the cuticle. Fungi causing fuliginous and punctate mycelial types on apple are particularly difficult to identify based on morphological criteria because many species in the SBFS complex share the same mycelial phenotypes. We compared the morphology and nuclear ribosomal DNA phylogeny (ITS, LSU) of 11 fungal strains isolated from SBFS blemishes on apple obtained from two provinces in China and five states in the USA. Parsimony analysis, supported by cultural characteristics and morphology in vitro, provided support to delimit the isolates into three novel genera, representing five new species. Phaeothecoidiella, with two species, P. missouriensis and P. illinoisensis, is introduced as a new genus with pigmented endoconidia in the Dothideomycetes. Houjia (Capnodiales) is introduced for H. pomigena and H. yanglingensis. Although morphologically similar to Stanjehughesia (Chaetosphaeriaceae), Houjia is distinct in having solitary conidiogenous cells. Sporidesmajora (Capnodiales), based on S. pennsylvaniensis, is distinguished from Sporidesmium (Sordariomycetes) in having long, multiseptate conidiophores that frequently have a subconical, darkly pigmented apical cell, and very long, multi-euseptate conidia

    Energy Spectrum of Bloch Electrons Under Checkerboard Field Modulations

    Two-dimensional Bloch electrons in a uniform magnetic field exhibit complex energy spectrum. When static electric and magnetic modulations with a checkerboard pattern are superimposed on the uniform magnetic field, more structures and symmetries of the spectra are found, due to the additional adjustable parameters from the modulations. We give a comprehensive report on these new symmetries. We have also found an electric-modulation induced energy gap, whose magnitude is independent of the strength of either the uniform or the modulated magnetic field. This study is applicable to experimentally accessible systems and is related to the investigations on frustrated antiferromagnetism.Comment: 8 pages, 6 figures (reduced in sizes), submitted to Phys.     Is traditional Chinese medicine recommended in Western medicine clinical practice guidelines in China? A systematic analysis

    BackgroundEvidence-based medicine promotes and relies on the use of evidence in developing clinical practice guidelines (CPGs). The Chinese healthcare system includes both traditional Chinese medicine (TCM) and Western medicine, which are expected to be equally reflected in Chinese CPGs.ObjectiveTo evaluate the inclusion of TCM-related information in Western medicine CPGs developed in China and the adoption of high level evidence.MethodsAll CPGs were identified from the China Guideline Clearinghouse (CGC), which is the main Chinese organisation maintaining the guidelines issued by the Ministry of Health of China, the Chinese Medical Association and the Chinese Medical Doctors’ Association.TCM-related contents were extracted from all the CPGs identified. Extracted information comprised the institution issuing the guideline, date of issue, disease, recommendations relating to TCM, evidence level of the recommended content and references supporting the recommendations.ResultsA total of 604 CPGs were identified, only a small number of which (74/604; 12%) recommended TCM therapy and only five guidelines (7%) had applied evidence grading. The 74 CPGs involved 13 disease systems according to the International Classification of Diseases 10th edition. TCM was mainly recommended in the treatment part of the guidelines (73/74, 99%), and more than half of the recommendations (43/74, 58%) were related to Chinese herbal medicine (single herbs or herbal treatment based on syndrome differentiation).ConclusionsFew Chinese Western medicine CPGs recommend TCM therapies and very few provide evidence grading for the TCM recommendation. We suggest that future guideline development should be based on systematic searches for evidence to support CPG recommendations and involve a multidisciplinary approach including TCM expertise

    Partial Wave Analysis of J/Ïˆâ†’Îł(K+K−π+π−)J/\psi \to \gamma (K^+K^-\pi^+\pi^-)

    BES data on J/Ïˆâ†’Îł(K+K−π+π−)J/\psi \to \gamma (K^+K^-\pi^+\pi^-) are presented. The K∗Kˉ∗K^*\bar K^* contribution peaks strongly near threshold. It is fitted with a broad 0−+0^{-+} resonance with mass M=1800±100M = 1800 \pm 100 MeV, width Γ=500±200\Gamma = 500 \pm 200 MeV. A broad 2++2^{++} resonance peaking at 2020 MeV is also required with width ∌500\sim 500 MeV. There is further evidence for a 2−+2^{-+} component peaking at 2.55 GeV. The non-K∗Kˉ∗K^*\bar K^* contribution is close to phase space; it peaks at 2.6 GeV and is very different from K∗K∗ˉK^{*}\bar{K^{*}}.Comment: 15 pages, 6 figures, 1 table, Submitted to PL

    P-wave excited baryons from pion- and photo-induced hyperon production

    We report evidence for N(1710)P11N(1710)P_{11}, N(1875)P11N(1875)P_{11}, N(1900)P13N(1900)P_{13}, Δ(1600)P33\Delta(1600)P_{33}, Δ(1910)P31\Delta(1910)P_{31}, and Δ(1920)P33\Delta(1920)P_{33}, and find indications that N(1900)P13N(1900)P_{13} might have a companion state at 1970\,MeV. The controversial Δ(1750)P31\Delta(1750)P_{31} is not seen. The evidence is derived from a study of data on pion- and photo-induced hyperon production, but other data are included as well. Most of the resonances reported here were found in the Karlsruhe-Helsinki (KH84) and the Carnegie-Mellon (CM) analyses but were challenged recently by the Data Analysis Center at GWU. Our analysis is constrained by the energy independent πN\pi N scattering amplitudes from either KH84 or GWU. The two πN\pi N amplitudes from KH84 or GWU, respectively, lead to slightly different πN\pi N branching ratios of contributing resonances but the debated resonances are required in both series of fits.Comment: 22 pages, 28 figures.     A Study of Cosmic Ray Secondaries Induced by the Mir Space Station Using AMS-01

    The Alpha Magnetic Spectrometer (AMS-02) is a high energy particle physics experiment that will study cosmic rays in the ∌100MeV\sim 100 \mathrm{MeV} to 1TeV1 \mathrm{TeV} range and will be installed on the International Space Station (ISS) for at least 3 years. A first version of AMS-02, AMS-01, flew aboard the space shuttle \emph{Discovery} from June 2 to June 12, 1998, and collected 10810^8 cosmic ray triggers. Part of the \emph{Mir} space station was within the AMS-01 field of view during the four day \emph{Mir} docking phase of this flight. We have reconstructed an image of this part of the \emph{Mir} space station using secondary π−\pi^- and Ό−\mu^- emissions from primary cosmic rays interacting with \emph{Mir}. This is the first time this reconstruction was performed in AMS-01, and it is important for understanding potential backgrounds during the 3 year AMS-02 mission.Comment: To be submitted to NIM B Added material requested by referee.     The proton spectrum in the kinetic energy range 0.1 to 200 GeV was measured by the Alpha Magnetic Spectrometer (AMS) during space shuttle flight STS-91 at an altitude of 380 km. Above the geomagnetic cutoff the observed spectrum is parameterized by a power law. Below the geomagnetic cutoff a substantial second spectrum was observed concentrated at equatorial latitudes with a flux ~ 70 m^-2 sec^-1 sr^-1. Most of these second spectrum protons follow a complicated trajectory and originate from a restricted geographic region.Comment: 19 pages, Latex, 7 .eps figure

    Search for antihelium in cosmic rays

    The Alpha Magnetic Spectrometer (AMS) was flown on the space shuttle Discovery during flight STS-91 in a 51.7 degree orbit at altitudes between 320 and 390 km. A total of 2.86 * 10^6 helium nuclei were observed in the rigidity range 1 to 140 GV. No antihelium nuclei were detected at any rigidity. An upper limit on the flux ratio of antihelium to helium of < 1.1 * 10^-6 is obtained.Comment: 18 pages, Latex, 9 .eps figure
