Effect of fatigue loading on impact response of rat ulna
Stress fracture is a common injury among athletes, such as basketball players. The occurrence of stress fracture is a consequence of both fatigue and impact loading of the bone that will potentially threat athletes’ careers. Scientists and engineers have studied the fatigue properties of many engineering materials, and more recently biological materials. Investigating the role of fatigue on impact properties has received much less attention in bone. In this study, cyclic axial compressive loading was applied in vivo on the right ulnae of sixteen rats (Sprague-Dawley, Charles River), and the left served as contralateral control. The animals were divided into two groups: one day of rest before they were sacrificed and the other seven days. Afterwards, the ulnae were harvested and potted in epoxy and then scanned using micro-Computed Tomography (CT). Impact tests were performed using a customized figure where the impact energy was normalized for all specimens and following impact the specimens were re-scanned using micro-CT scans. There was no significant change in bone volume between the control (mean = 7.01 0.61mm3) and loaded (mean = 6.63 0.19mm3) ulnae in the group with one day rest (p = 0.28). However, after seven days of rest, the average bone volume increased by 4.35% among the control ulnae (mean = 7.32 0.49mm3) , and 15.10% among the loaded (mean = 7.63 0.47mm3). The increase in volume was attributed to woven bone formation and was visually confirmed from the micro CT images. The peak impact force was 37.5% higher in the control (mean = 174.96 33.25N) specimens than the loaded (mean = 130.34 22.37N ).
Our data is limited to some degree by the sample size and two specimens fractured after the cyclic loading which further decease the sample size. Future work should investigate the effect of different rest times. This study indicated that cyclic fatigue loading had a negative impact on bone’s impact response. Bones that experienced fatigue loading became less stiff and resulted in lower peak forces, and an increased fracture rate when subjected to impact. Rest time was crucial to the recovery of fatigue damage. Seven days rest decreased the fracture rate by 66.67%. If more rest time was given, the peak force could return to the same level as the control or even higher as new bone would possibly mineralize. This study can provide a baseline guidance of the training, competition and rest arrangement to minimize the risk of stress fracture and prolong athletes’ careers

PeerJ 9:e12436 DOI 10.7717/peerj.12436In this study, size selectivity and exploitation pattern of six diamond-mesh codends with different mesh sizes, ranging from 25 to 54 mm, for Southern velvet shrimp (Metapenaeopsis palmensis) were tested and compared in a shrimp trawl fishery of the South China Sea (SCS). We used a codend with a mesh size of 25 mm (D25) as a starting point, which is the minimum mesh size (MMS) currently regulated in the studied area. Four different fishing population scenarios were applied to quantify and compare how mesh sizes of codends used would impact the size selectivity and exploitation pattern for the target shrimp species. The results demonstrated that the D25 codend was not proper for protecting juvenile shrimp at the studied area. By applying this legal codend, L50 (50% retention length) of the target shrimp species was below its minimum conservation reference size (MCRS, 7.0 cm total length), the retention probability of shrimp with a length of MCRS was above 95% CI [91–99] and more than 43% of undersized shrimp was retained. To mitigate the bycatch issue of undersized shrimp, increasing the mesh size in the diamond mesh codend is a simple and effective option. However, the loss of catch efficiency for marketable shrimp is a major concern while increasing the mesh size. A good compromise between releasing undersized shrimp and maintaining the legal individuals is manifested by using the codend with 35 mm mesh size (D35). CoRide: Joint Order Dispatching and Fleet Management for Multi-Scale Ride-Hailing Platforms
How to optimally dispatch orders to vehicles and how to tradeoff between
immediate and future returns are fundamental questions for a typical
ride-hailing platform. We model ride-hailing as a large-scale parallel ranking
problem and study the joint decision-making task of order dispatching and fleet
management in online ride-hailing platforms. This task brings unique challenges
in the following four aspects. First, to facilitate a huge number of vehicles
to act and learn efficiently and robustly, we treat each region cell as an
agent and build a multi-agent reinforcement learning framework. Second, to
coordinate the agents from different regions to achieve long-term benefits, we
leverage the geographical hierarchy of the region grids to perform hierarchical
reinforcement learning. Third, to deal with the heterogeneous and variant
action space for joint order dispatching and fleet management, we design the
action as the ranking weight vector to rank and select the specific order or
the fleet management destination in a unified formulation. Fourth, to achieve
the multi-scale ride-hailing platform, we conduct the decision-making process
in a hierarchical way where a multi-head attention mechanism is utilized to
incorporate the impacts of neighbor agents and capture the key agent in each
scale. The whole novel framework is named as CoRide. Extensive experiments
based on multiple cities real-world data as well as analytic synthetic data
demonstrate that CoRide provides superior performance in terms of platform
revenue and user experience in the task of city-wide hybrid order dispatching
and fleet management over strong baselines.Comment: CIKM 201
Short-Term Photovoltaic Power Generation Forecasting Based on Multivariable Grey Theory Model with Parameter Optimization
Owing to the environment, temperature, and so forth, photovoltaic power generation volume is always fluctuating and subsequently impacts power grid planning and operation seriously. Therefore, it is of great importance to make accurate prediction of the power generation of photovoltaic (PV) system in advance. In order to improve the prediction accuracy, in this paper, a novel particle swarm optimization algorithm based multivariable grey theory model is proposed for short-term photovoltaic power generation volume forecasting. It is highlighted that, by integrating particle swarm optimization algorithm, the prediction accuracy of grey theory model is expected to be highly improved. In addition, large amounts of real data from two separate power stations in China are being employed for model verification. The experimental results indicate that, compared with the conventional grey model, the mean relative error in the proposed model has been reduced from 7.14% to 3.53%. The real practice demonstrates that the proposed optimization model outperforms the conventional grey model from both theoretical and practical perspectives
Homogeneous environmental selection mainly determines the denitrifying bacterial community in intensive aquaculture water
Nitrate reduction by napA (encodes periplasmic nitrate reductase) bacteria and nitrous oxide reduction by nosZ (encodes nitrous oxide reductase) bacteria play important roles in nitrogen cycling and removal in intensive aquaculture systems. This study investigated the diversity, dynamics, drivers, and assembly mechanisms of total bacteria as well as napA and nosZ denitrifiers in intensive shrimp aquaculture ponds over a 100-day period. Alpha diversity of the total bacterial community increased significantly over time. In contrast, the alpha diversity of napA and nosZ bacteria remained relatively stable throughout the aquaculture process. The community structure changed markedly across all groups over the culture period. Total nitrogen, phosphate, total phosphorus, and silicate were identified as significant drivers of the denitrifying bacterial communities. Network analysis revealed complex co-occurrence patterns between total, napA, and nosZ bacteria which fluctuated over time. A null model approach showed that, unlike the total community dominated by stochastic factors, napA and nosZ bacteria were primarily governed by deterministic processes. The level of determinism increased with nutrient loading, suggesting the denitrifying community can be manipulated by bioaugmentation. The dominant genus Ruegeria may be a promising candidate for introducing targeted denitrifiers into aquaculture systems to improve nitrogen removal. Overall, this study provides important ecological insights into aerobic and nitrous oxide-reducing denitrifiers in intensive aquaculture, supporting strategies to optimize microbial community structure and function
SIAD: Self-supervised Image Anomaly Detection System
Recent trends in AIGC effectively boosted the application of visual
inspection. However, most of the available systems work in a human-in-the-loop
manner and can not provide long-term support to the online application. To make
a step forward, this paper outlines an automatic annotation system called SsaA,
working in a self-supervised learning manner, for continuously making the
online visual inspection in the manufacturing automation scenarios. Benefit
from the self-supervised learning, SsaA is effective to establish a visual
inspection application for the whole life-cycle of manufacturing. In the early
stage, with only the anomaly-free data, the unsupervised algorithms are adopted
to process the pretext task and generate coarse labels for the following data.
Then supervised algorithms are trained for the downstream task. With
user-friendly web-based interfaces, SsaA is very convenient to integrate and
deploy both of the unsupervised and supervised algorithms. So far, the SsaA

Phase I dose-escalation trial of the oral AKT inhibitor uprosertib in combination with the oral MEK1/MEK2 inhibitor trametinib in patients with solid tumors.
PurposeThis study aimed to determine the safety, tolerability, and recommended phase II doses of trametinib plus uprosertib (GSK2141795) in patients with solid tumors likely to be sensitive to MEK and/or AKT inhibition.MethodsThis was a phase I, open-label, dose-escalation, and dose-expansion study in patients with triple-negative breast cancer or BRAF-wild type advanced melanoma. The primary outcome of the expansion study was investigator-assessed response. Among 126 enrolled patients, 63 received continuous oral daily dosing of trametinib and uprosertib, 29 received various alternative dosing schedules, and 34 were enrolled into expansion cohorts. Doses tested in the expansion cohort were trametinib 1.5 mg once daily (QD) + uprosertib 50 mg QD.ResultsAdverse events (AEs) were consistent with those reported in monotherapy studies but occurred at lower doses and with greater severity. Diarrhea was the most common dose-limiting toxicity; diarrhea and rash were particularly difficult to tolerate. Overall, 59% and 6% of patients reported AEs with a maximum severity of grade 3 and 4, respectively. Poor tolerability prevented adequate delivery of uprosertib with trametinib at a concentration predicted to have clinical activity. The study was terminated early based on futility in the continuous-dosing expansion cohorts and a lack of pharmacological or therapeutic advantage with intermittent dosing. The objective response rate was < 5% (1 complete response, 5 partial responses).ConclusionsContinuous and intermittent dosing of trametinib in combination with uprosertib was not tolerated, and minimal clinical activity was observed in all schedules tested
Impacts of coagulation on the appearance time method for new particle growth rate evaluation and their corrections
The growth rate of atmospheric new particles is a key parameter that determines their survival probability of becoming cloud condensation nuclei and hence their impact on the climate. There have been several methods to estimate the new particle growth rate. However, due to the impact of coagulation and measurement uncertainties, it is still challenging to estimate the initial growth rate of new particles, especially in polluted environments with high background aerosol concentrations. In this study, we explore the influences of coagulation on the appearance time method to estimate the growth rate of sub-3 nm particles. The principle of the appearance time method and the impacts of coagulation on the retrieved growth rate are clarified via derivations. New formulae in both discrete and continuous spaces are proposed to correct for the impacts of coagulation. Aerosol dynamic models are used to test the new formulae. New particle formation in urban Beijing is used to illustrate the importance of considering the impacts of coagulation on the sub-3 nm particle growth rate and its calculation. We show that the conventional appearance time method needs to be corrected when the impacts of coagulation sink, coagulation source, and particle coagulation growth are non-negligible compared to the condensation growth. Under the simulation conditions with a constant concentration of non-volatile vapors, the corrected growth rate agrees with the theoretical growth rates. However, the uncorrected parameters, e.g., vapor evaporation and the variation in vapor concentration, may impact the growth rate obtained with the appearance time method. Under the simulation conditions with a varying vapor concentration, the average bias in the corrected 1.5-3 nm particle growth rate ranges from 6 %-44 %, and the maximum bias in the size-dependent growth rate is 150 %. During the test new particle formation event in urban Beijing, the corrected condensation growth rate of sub-3 nm particles was in accordance with the growth rate contributed by sulfuric acid condensation, whereas the conventional appearance time method overestimated the condensation growth rate of 1.5 nm particles by 80 %.Peer reviewe
