Migration paths saturations in meta-epidemic systems

In this paper we consider a simple two-patch model in which a population affected by a disease can freely move. We assume that the capacity of the interconnected paths is limited, and thereby influencing the migration rates. Possible habitat disruptions due to human activities or natural events are accounted for. The demographic assumptions prevent the ecosystem to be wiped out, and the disease remains endemic in both populated patches at a stable equilibrium, but possibly also with an oscillatory behavior in the case of unidirectional migrations. Interestingly, if infected cannot migrate, it is possible that one patch becomes disease-free. This fact could be exploited to keep disease-free at least part of the population

S100P enhances the motility and invasion of human trophoblast cell lines

S100P has been shown to be a marker for carcinogenesis where its expression in solid tumours correlates with metastasis and a poor patient prognosis. This protein’s role in any physiological process is, however, unknown. Here we first show that S100P is expressed both in trophoblasts in vivo as well as in some corresponding cell lines in culture. We demonstrate that S100P is predominantly expressed during the early stage of placental formation with its highest expression levels occurring during the first trimester of gestation, particularly in the invading columns and anchoring villi. Using gain or loss of function studies through overexpression or knockdown of S100P expression respectively, our work shows that S100P stimulates both cell motility and cellular invasion in different trophoblastic and first trimester EVT cell lines. Interestingly, cell invasion was seen to be more dramatically affected than cell migration. Our results suggest that S100P may be acting as an important regulator of trophoblast invasion during placentation. This finding sheds new light on a hitherto uncharacterized molecular mechanism which may, in turn, lead to the identification of novel targets that may explain why significant numbers of confirmed human pregnancies suffer complications through poor placental implantation

Background and Objective: Coronavirus disease 2019 (COVID-19) characterized by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has created serious concerns about its potential adverse effects. There are limited data on clinical, radiological, and neonatal outcomes of pregnant women with COVID-19 pneumonia. This study aimed to assess clinical manifestations and neonatal outcomes of pregnant women with COVID-19. Methods: We conducted a systematic article search of PubMed, EMBASE, Scopus, Google Scholar, and Web of Science for studies that discussed pregnant patients with confirmed COVID-19 between January 1, 2020, and April 20, 2020, with no restriction on language. Articles were independently evaluated by two expert authors. We included all retrospective studies that reported the clinical features and outcomes of pregnant patients with COVID-19. Results: Forty-seven articles were assessed for eligibility; 13 articles met the inclusion criteria for the systematic review. Data is reported for 235 pregnant women with COVID-19. The age range of patients was 2540 years, and the gestational age ranged from 8 to 40 weeks plus 6 days. Clinical characteristics were fever 138/235 (58.72%), cough 111/235 (47.23%), and sore throat 21/235 (8.93%). One hundred fifty six out of 235 (66.38%) pregnant women had cesarean section, and 79 (33.62%) had a vaginal delivery. All the patients showed lung abnormalities in CT scan images, and none of the patients died. Neutrophil cell count, C-reactive protein (CRP) concentration, ALT, and AST were increased but lymphocyte count and albumin levels were decreased. Amniotic fluid, neonatal throat swab, and breastmilk samples were taken to test for SARS-CoV-2 but all found negativ results. Recent published evidence showed the possibility of vertical transmission up to 30%, and neonatal death up to 2.5%. Pre-eclampsia, fetal distress, PROM, pre-mature delivery were the major complications of pregnant women with COVID-19. Conclusions: Our study findings show that the clinical, laboratory and radiological characteristics of pregnant women with COVID-19 were similar to those of the general populations. The possibility of vertical transmission cannot be ignored but C-section should not be routinely recommended anymore according to latest evidences and, in any case, decisions should be taken after proper discussion with the family. Future studies are needed to confirm or refute these findings with a larger number of sample sizes and a long-term follow-up period. © Copyright © 2020 Islam, Poly, Walther, Yang, Wang, Hsieh, Atique, Salmani, Alsinglawi, Lin, Jian and Jack Li

Oxygen tension regulates the miRNA profile and bioactivity of exosomes released from extravillous trophoblast cells - liquid biopsies for monitoring complications of pregnancy

Our understanding of how cells communicate has undergone a paradigm shift since the recent recognition of the role of exosomes in intercellular signaling. In this study, we investigated whether oxygen tension alters the exosome release and miRNA profile from extravillous trophoblast (EVT) cells, modifying their bioactivity on endothelial cells (EC). Furthermore, we have established the exosomal miRNA profile at early gestation in women who develop pre-eclampsia (PE) and spontaneous preterm birth (SPTB). HTR-8/SVneo cells were used as an EVT model. The effect of oxygen tension (i.e. 8% and 1% oxygen) on exosome release was quantified using nanocrystals (Qdot®) coupled to CD63 by fluorescence NTA. A real-time, live-cell imaging system (Incucyte™) was used to establish the effect of exosomes on EC. Plasma samples were obtained at early gestation (<18 weeks) and classified according to pregnancy outcomes. An Illumina TrueSeq Small RNA kit was used to construct a small RNA library from exosomal RNA obtained from EVT and plasma samples. The number of exosomes was significantly higher in EVT cultured under 1% compared to 8% oxygen. In total, 741 miRNA were identified in exosomes from EVT. Bioinformatic analysis revealed that these miRNA were associated with cell migration and cytokine production. Interestingly, exosomes isolated from EVT cultured at 8% oxygen increased EC migration, whilst exosomes cultured at 1% oxygen decreased EC migration. These changes were inversely proportional to TNF-α released from EC. Finally, we have identified a set of unique miRNAs in exosomes from EVT cultured at 1% oxygen and exosomes isolated from the circulation of mothers at early gestation, who later developed PE and SPTB. We suggest that aberrant exosomal signalling by placental cells is a common aetiological factor in pregnancy complications characterised by incomplete SpA remodeling and is therefore a clinically relevant biomarker of pregnancy complications.Grace Truong, Dominic Guanzon, Vyjayanthi Kinhal, Omar Elfeky, Andrew Lai, Sherri Longo, Zarin Nuzhat, Carlos Palma, Katherin Scholz-Romero, Ramkumar Menon, Ben W. Regulated Expression of ADAMTS-12 in Human Trophoblastic Cells: A Role for ADAMTS-12 in Epithelial Cell Invasion?

Metastatic carcinoma cells exploit the same molecular machinery that allows human placental cytotrophoblasts to develop an invasive phenotype. As altered expression levels of ADAMTS (A Disintegrin And Metalloproteinase with ThromboSpondin repeats) subtypes have been associated with cancer progression, we have examined the function and regulation of members of this gene family in epithelial cell invasion using cultures of highly invasive extravillous cytotrophoblasts and the poorly invasive JEG-3 cytotrophoblast cell line as model systems. Of the multiple ADAMTS subtypes identified in first trimester human placenta and these two trophoblastic cell types, only ADAMTS-12 was preferentially expressed by extravillous cytotrophoblasts. Transforming growth factor-β1 and interleukin-1β, two cytokines that promote and restrain cytotrophoblast invasion in vitro, were also found to differentially regulate trophoblastic ADAMTS-12 mRNA levels. Loss- or gain-of-function studies confirmed that ADAMTS-12, independent of its proteolytic activity, plays a specific, non-redundant role in trophoblast invasion. Furthermore, we demonstrated that ADAMTS-12 regulated cell-extracellular matrix adhesion and invasion through a mechanism involving the αvβ3 integrin heterodimer. This study identifies a novel biological role for ADAMTS-12, and highlights the importance and complexity of its non-proteolytic domain(s) pertaining to its function

In 1999 Mnemiopsis leidyi was introduced to the Caspian Sea from the Black Sea with ballast waters from the ships. The comprehensive study on probability of controlling Caspian Sea invasive Ctenophora planned after a remarkable of decreeing in Kilka fish catches stocks and fisheris community problems. This study focus on reproduction experiments of Beroe ovata as the best candidate for control of Mnemiopsis population size in the Caspian Sea that was performed in Turkey and Iran during 2002-2003. At 2002, 87 specimens of B. ovata, 10-50 mm transferred to Caspian sea ecology research center from Marmareh sea where acclimated with Caspian sea water gradually. At 2003, experiments were performed near to Black sea (at Sinop) with freshly collected Beroe ovata, 40-65 mm size in three salinity level treatment, the Black sea water 18 , Mixed water 15 and Caspian water 12 . 130 individuals of Beroe ovata were brought from Sinop (Turkey) to Iran during 2003. A number of Beroe specimens were sent to Guilan province for reproduction studies and another part were sent to Mazandaran province for both reproduction and mesocosem studies. For control we had 1 Beroe, length 30 mm in the Black Sea water that was alive during of study in Iran. The Jars were examined each day for ova and larvae and they were collected and put into glass container of Caspian water for hatching and developing survey, some of them were left without any handling for larvae developing. Also in another experiment the eggs collected from jars were placed in the same three treatments for studying of growth and survival. The results were unsuccessful on propagation experiments at 2002 since the spawning and hatching rates were very low (20 ova) and, none of the larvae developed into adults in Caspian Sea water. The spawning was more in Marmareh sea water with 138 ova where only 7 larvae was hatched. Results showed that Beroe specimens is able to survive and reproduce in Caspian water but was not as well as Black Sea also the Beroe larvae growth rate is low in the Caspian Sea water. Maximum fecundity of Beroe individual was 2212 and 235 ovae in Caspian Sea water in site Sinop and Iran respectively. Results showed 34-100% eggs in Caspian Sea water were destroyed and did not develop. In Iran we obtained only one larvae with 5 mm length, other larvae were at different stages of development but most of them were 1.2 2 mm. The results of mesocosm survey showed most of ova and larvae have been obtained from the tanks where individuals B. ovata were with Mnemiopsis. Fecundity of Beroe in the control with Black Sea water were between 17 to 1879 with average of 828 ± 112 ova. The poor results of B. ovata reproduction obtained in this survey in Caspian Sea may be due to transportation and acclimation stress and low salinity of Caspian Sea water

Limnological study of Sabalan run off Damfor aquaculture cold water fish was conducted in sixtimesat four stations from July to December 2009. Average minimum and maximum parameters measured in different stations were included water temperature, dissolved Oxygen, total hardness, electrical conductivity, pH, BOD5, COD, TDS, NH4, NO2, NO3, PO4, TN, TP, SiO2, respectively (10/3±9/5-14±8/7°C), (9±1/2-11/4±2/1 mg/liter), (367±19/8-381±17/8mg/liter), (0/95±0/07-1/05±0/07 mz/cm), (8/3±0/1-8/4±0/1), (3/3±1/14-3/88 ± 0/71mg/ liter), (15/5 ± 3/1-16/3 ± 3 mg/liter), (0/48 ± 0/04-0/54 ± 0/06mg/ liter), (0/08 ± 0/02-0/13 ± 0/07 mg/liter), (0/004 ± 0/003- 0/05 ± 0/1 mg/ liter), (0/6 ± 0/2-0/64 ± 0/29 mg/ liter), (0/19 ± 0/07-0/22 ± 0/07 mg/ liter), (10/48 ± 0/6-10/98 ± 0/54mg/liter), (0/39 ± 0/11-0/44 ± 0/18 mg/liter), and(9/9 ±0/6-11/1 ± 1/4 mg/liter). Average minimum and maximum of water heavy metalswas included of Cd, Pb, Zn, Fe, Cuand Ni respectively was measured as ND(No Deliver), ND, ND- 0/242, ND- 1/075, ND- 0/018 and ND-0/014inmg/ liter. Also among the 16 organochlorinecom pounds measured the highest amount of was belonged totoxin Hexachlorine Benzene (1/5 mg/ liter). The results of the analysis of water quality parameters showed that parameters except ofCOD, TDS, PO4, TP, Zn, Fe and water temperature, other parameters do not haveany restrictions foraquaculturecold water fish in therunoffDam. Based on data results it was noticed that there was temperature restriction up to November at downstream and station 2 showed less temperature fluctuation and more suitable condition compared to other stations. BOD5 value was within the allowable range for growing trout, in which station 2 somewhat with lower volatility of value had the most suitable BOD5 condition. In the present study, the amount of TDS in the study area was somewhat higher than the limitation and since TDS values increased with distance from the dam, therefore station 2 had somewhat better conditions for fish farming. In addition the amount of EC in the study area, indicating the inappropriateness of it for various activities in the fisheries. Although none of the stations in terms of the amount of phosphorus didn't show favorable conditions for rainbow trout fish propagation, it appears between the researches stations, station 2 had the most favorable terms. The results of this study (regardless of the unseen), it seems that in August and September; Zn had overestimated value for cold fish propagation. Recent study showed that among heavy metals, Fe in water, sediments and fish was over limitation value. Also, Fe value was measured of 1.08 at station 4, which showed over normal value for cold fish propagation. Therefore, station 2 at downstream seemed to have suitable cold fish propagation due to lower Fe value. On the based on the recent study, planktonic biological index did not endorse water quality for fish propagation. In this study, aquatic plant such as Potamogeton pectinatus and Pheragmitie saustralis was found at water of higher and somehow polluted organic matter. Probably, based on the results it was noticed that downstream water of dam was polluted to organic matter. At the recent study, resisted maroinvertebrates to organic matter (Chironomidae) was consisted noticeable frequency compared to other sensitive invertebrates to organic matter (EPT).Statistically, it can be expressed that probably organic pollution resulted to increase of resistance invertebrates and decreased of sensitive groups. In recent study, EPT/CHIR index was measured 0.6 in station 2 and 1.74 in station 4. This ration was less to many other mountain rivers. As ETP sensitive invertebrates has higher sensitive to other groups with the regards of environmental condition and pollution, unpredicted increase of Chironomidae compare to sensitive groups resulted of EPT/CHIR value which showed environmental stress. In this study, HFBI index was measured as 7.49 at station 2 and 7 in station 4. Based on HFBI index, station 2 was classified as a high organic pollution and station 4 with noticeable organic pollution. At the recent study, station 2 was catch more fish to station4. For instance, Squaliuscephalus and Alburnoides pinctatus were catch in station 2 and were not found in station 4. As a whole, Biological indicators whichwas included Plankton Bioindicator, EPT / CHIR. ,Hilsenh off Biological Indicator, diversity and distribution ofaquatic plants and fish, showed water situation in run off Damrich inorganic matter. Finally, the study area was approved for fish farm about 50 tons and needs management arrangements with the increased production rate

    Get PDF
Extreme disruptive events, such as the volcano eruption in Iceland, the Japanese tsunami, and the COVID-19 pandemic, as well as constant changes in customers’ needs and expectations, have forced supply chains to continuously adapt to new environments. Consequently, it is paramount to understand the supply chain characteristics for possible future scenarios, in order to know how to respond to threats and take advantage of the opportunities that the next years will bring. This chapter focuses on describing the characteristics of the supply chain in each of the six macro-scenarios presented in Sardesai et al. (2020b), as final stage of the scenario building methodology. Supply chains for each scenario are characterized in eight dimensions: Products and Services, Supply Chain Paradigm, Sourcing and Distribution, Technology Level, Supply Chain Configuration, Manufacturing Systems, Sales Channel, and Sustainability
