METHODS: Thirty experimental animals were divided into groups as healthy (HG), desflurane control (DCG), TPP and desflurane group (TDG). 20 mg/kg TPP was injected to intraperitoneally TDG. After one hour of TPP administration, desflurane was applied for two hours. After 24 hours, liver tissues of the animals killed with decapitation were removed. the oxidant/antioxidant levels and ALT, AST and LDH activities were measured. the histopathological examinations were performed in the liver tissues for all rats. RESULTS: Notwithstanding the levels of oxidants and liver enzymes were significantly increased (p<0.0001), antioxidant levels were significantly decreased in DCG (p<0.0001). on contrary to the antioxidant parameters were increased (p<0.05) the oxidant parameters and liver enzymes were decreased in TDG (p<0.0001). Whereas multiple prominent, congestion, hemorrhage and dilatation were observed in sinusoids and lymphocyte-rich inflammation results in the centrilobular and portal areas of liver tissue in DCG, these findings were observed less frequently in TDG. CONCLUSION: Thiamine pyrophosphate prevented liver oxidative damage induced with desflurane and may be useful in prophylaxis of desflurane induced hepatotoxicity.Erzincan University Scientific Research Projects DepartmentErzincan Binali Yildirim University [SAG-B-080715-0173]Erzincan University Scientific Research Projects Department (Project no SAG-B-080715-0173
Phytochemical profile and some biological activities of three Centaurea species from Turkey
Purpose: To characterise the phytochemical profile of whole plants of Centaurea balsamita, C. depressa and C. lycopifolia with LC-ESI-MS/MS, and as well as their antioxidant, anticholinesterase and antimicrobial activities. Methods: Organic and aqueous extracts of the three Centaurea species were evaluated for DPPH free radical, ABTS cation radical scavenging and cupric reducing antioxidant capacity (CUPRAC). Acetyland butyryl-cholinesterase enzyme inhibition abilities of the extracts using petroleum ether, acetone, methanol and water were studied to determine anticholinesterase activity, while antimicrobial activity was determined by disc diffusion method using appropriate antimicrobial standards and organisms. The phytochemical components of the methanol extracts were assessed by LC-MS/MS.Results: The methanol extract of C. balsamita exhibited much higher DPPH free and ABTS cation radicals scavenging activities (with IC50 of 62.65 ± 0.97 and 24.21 ± 0.70 mg/ml, respectively) than the other extracts. The petroleum ether extracts of the plant species exhibited moderate inhibitory activity against butyrylcholinesterase enzymes while the acetone extract of C. balsamita showed good antifungal activity against Candida albicans. Quinic acid (17513 ± 813 μg/g, 63874 ± 3066 μg/g and 108234 ± 5195 μg/g) was the major compound found in the methanol extracts of C. balsamita, C. depressa and C. Lycopifolia, respectively. Conclusion: These results indicate quinic acid is the major compound in the three plant species and that Centaurea balsamita has significant antioxidant, anticholinesterase and antimicrobial properties. Further studies to identify the compounds in the extracts responsible for the activities are required. Keywords: Centaurea, LC-ESI-MS/MS, Anticholinesterase, Antioxidant, Antimicrobia

Postharvest Treatments of Different Doses of Calcium Lactate in Combination with Chitosan Improves Biochemical Characters of Cucumis sativus L. during Storage
This study aimed to determine the effects of the postharvest coating created with different doses of calcium compounds and chitosan on the quality of the cucumbers during storage. For this purpose, cucumber fruits were dipped into solutions containing the doses of 2, 4, and 6% calcium chloride (CaCl2), calcium propionate (C6H10CaO4), and calcium lactate (C6H10CaO6) + 1.0% chitosan separately before storage. After the treatments, the cucumber fruits were dried and placed in boxes with plastic lids and stored at 7±1oC and 90-95% relative humidity for 20 days. Calcium change in fruits, flesh firmness (N), weight loss (%), fruit skin color (L*, a*, b*, hue angle, yellowness index), chlorophyll SPAD amount, total soluble solids (TSS), titratable acid (%), taste quality, electrolyte leakage (%), decay rate (%) observations and analyzes were evaluated at five-day intervals during storage. According to the results, it was found that especially the dose of 2% calcium lactate was more effective in preserving fruit firmness and green color, increasing the amount of TSS, and also delaying the decay, amongst calcium chloride, calcium propionate, and calcium lactate treatments. The KT and CaP4 treatments were successful in reducing weight loss, whereas CaL4 and CaL6 were found to be effective in maintaining chlorophyll content. Also, it observed that CaK4 and CaK6 treatments were increased electrolyte leakage due to causing damage to the cell wall, thus the shortened storage time of cucumber fruits in this study
Chemoradiotherapy-Associated Myelosuppression: A Retrospective Analysis of Risk Factors for Patients with Non-Small Cell Lung Cancer
Objective: Concomitant chemoradiotherapy (CRT) is the standard treatment for patients with stage III non-small cell lung cancer (NSCLC). Myelosuppression can be a significant problem in concomitant CRT. The aim of this study was to assess the parameters obtained before concomitant CRT to define the risk factors for myelosuppression in patients with locally advanced NSCLC.
Methods: We retrospectively analyzed 81 patients with NSCLC who received concomitant platinum-based chemoradiotherapy between January 2008 and December 2012. Seventy-eight patients (96.2%) received etoposide (50 mg/m2, intravenously on days 1–5, 29–33) plus cisplatin (50 mg/m2, on days 1, 8, 29, and 36), and 3 patients (3.8%) received docetaxel (20 mg/m2/w, on weeks 1–8) plus cisplatin (20 mg/m2/w, on weeks 1–8) concurrently with thoracic radiotherapy to a total dose of 40–66.6 Gy. The risk factors were examined for their association with myelosupression (grade 3 or 4 leukopenia, neutropenia, thrombocytopenia, or anemia) by logistic regression analysis.
Results: Grade 3 or higher neutropenia, leukopenia, thrombocytopenia, or anemia occurred in 51.8%, 53%, 8.6%, and 7.4% of the patients, respectively. Multivariate analysis revealed that the risk factors for neutropenia were performance status (p=0.032), white blood cell count (p=0.023), and pretreatment creatinine level (p=0.018). On multivariate analysis, white blood cell count (OR, 3.311; p=0.027; 95% CI, 1.148–9.545) was found as significant risk factor for CRT-induced leukopenia
Conclusion: Patients with a poor pretreatment performance status, low white blood cell count, and high creatinine level are at a risk of myelosupression. These characteristics of the patients should be considered while making treatment decisions
