    Effects of Expert Testimony and Interrogation Tactics on Perceptions of Confessions

    Evidence obtained through the process of interrogation is frequently undermined by what can be perceived as overzealous interrogation tactics. Although the majority of psychologically oriented tactics are legally permissible, they nonetheless contribute to innocent suspects confessing to crimes they did not commit. The present study examined the effect of expert testimony and interrogation tactics on perceptions of a confession. 182 undergraduates read a transcript of a homicide trial that varied based on interrogation tactic: implicit threat of punishment (maximization) or leniency (minimization) and expert witness testimony (presence or absence of expert testimony). Analysis indicated that the type of interrogation tactic used in obtaining the confession affected participants\u27 perceptions of the coerciveness of the interrogation process

    Characterization of immunoglobulin G antibodies to Plasmodium falciparum sporozoite surface antigen MB2 in malaria exposed individuals

    <p>Abstract</p> <p>Background</p> <p>MB2 protein is a sporozoite surface antigen on the human malaria parasite <it>Plasmodium falciparum</it>. MB2 was identified by screening a <it>P. falciparum </it>sporozoite cDNA expression library using immune sera from a protected donor immunized via the bites of <it>P. falciparum</it>-infected irradiated mosquitoes. It is not known whether natural exposure to <it>P. falciparum </it>also induces the anti-MB2 response and if this response differs from that in protected individuals immunized via the bites of <it>P. falciparum </it>infected irradiated mosquitoes. The anti-MB2 antibody response may be part of a robust protective response against the sporozoite.</p> <p>Methods</p> <p>Fragments of polypeptide regions of MB2 were constructed as recombinant fusions sandwiched between glutathione S-transferase and a hexa histidine tag for bacterial expression. The hexa histidine tag affinity purified proteins were used to immunize rabbits and the polyclonal sera evaluated in an <it>in vitro </it>inhibition of sporozoite invasion assay. The proteins were also used in immunoblots with sera from a limited number of donors immunized via the bites of <it>P. falciparum </it>infected irradiated mosquitoes and plasma and serum obtained from naturally exposed individuals in Kenya.</p> <p>Results</p> <p>Rabbit polyclonal antibodies targeting the non-repeat region of the basic domain of MB2 inhibited sporozoites entry into HepG2-A16 cells <it>in vitro</it>. Analysis of serum from five human volunteers that were immunized via the bites of <it>P. falciparum </it>infected irradiated mosquitoes that developed immunity and were completely protected against subsequent challenge with non-irradiated parasite also had detectable levels of antibody against MB2 basic domain. In contrast, in three volunteers not protected, anti-MB2 antibodies were below the level of detection. Sera from protected volunteers preferentially recognized a non-repeat region of the basic domain of MB2, whereas plasma from naturally-infected individuals also had antibodies that recognize regions of MB2 that contain a repeat motif in immunoblots. Sequence analysis of eleven field isolates and four laboratory strains showed that these antigenic regions of the basic domain of the <it>MB2 </it>gene are highly conserved in parasites obtained from different parts of the world. Moreover, anti-MB2 antibodies also were detected in the plasma of 83% of the individuals living in a malaria endemic area of Kenya (n = 41).</p> <p>Conclusion</p> <p>A preliminary analysis of the human humoral response against MB2 indicates that it may be an additional highly conserved target for immune intervention at the pre-erythrocytic stage of <it>P. falciparum </it>life cycle.</p

    Collective Impact Partnership and Backbone Organizations as Enablers of Children’s Well-Being

    In this article, the question of partnership is approached from a perspective centred around the creation of a common agenda based on trust and from the children´s point of view. Partnership and collaboration have traditionally been viewed as mechanisms to create bridges between organisations and institutions from the private, public and non-governmental sectors in order to enhance funder collaboratives, public-private partnerships, multi-stakeholder initiatives, social sector networks and collective impact initiatives. It was not however until Kania and Kramer´s (2011) seminal work on collective impact when this subject came to be viewed as a developmental process aiming at the creation of a common agenda and mutually agreed activities and consisting of five integral parts: a common agenda, shared measurement systems, mutually reinforcing activities, continuous communication and backbone support organisations. This article, based on a systematic review of the topic, maintains that partnership – approached from the point of view of children and through the lens of collective impact – constitutes a crucial mechanism in the creation of safe and comprehensive wellbeing for children. Thus, this article – using Kania and Kramer´s (2011) definition of collective impact and focusing on the structure of partnerships and the nature of trust in organisations as the prerequisite for partnership – advocates the importance of the UN SDG17 principle as the bringer of inclusive society built upon principles and values, a shared vision, and shared goals that place people at the centre of human endeavour.© Springer Nature Switzerland AG 2020.     Impaired Innate Immunity in Tlr4−/− Mice but Preserved CD8+ T Cell Responses against Trypanosoma cruzi in Tlr4-, Tlr2-, Tlr9- or Myd88-Deficient Mice

    The murine model of T. cruzi infection has provided compelling evidence that development of host resistance against intracellular protozoans critically depends on the activation of members of the Toll-like receptor (TLR) family via the MyD88 adaptor molecule. However, the possibility that TLR/MyD88 signaling pathways also control the induction of immunoprotective CD8+ T cell-mediated effector functions has not been investigated to date. We addressed this question by measuring the frequencies of IFN-γ secreting CD8+ T cells specific for H-2Kb-restricted immunodominant peptides as well as the in vivo Ag-specific cytotoxic response in infected animals that are deficient either in TLR2, TLR4, TLR9 or MyD88 signaling pathways. Strikingly, we found that T. cruzi-infected Tlr2−/−, Tlr4−/−, Tlr9−/− or Myd88−/− mice generated both specific cytotoxic responses and IFN-γ secreting CD8+ T cells at levels comparable to WT mice, although the frequency of IFN-γ+CD4+ cells was diminished in infected Myd88−/− mice. We also analyzed the efficiency of TLR4-driven immune responses against T. cruzi using TLR4-deficient mice on the C57BL genetic background (B6 and B10). Our studies demonstrated that TLR4 signaling is required for optimal production of IFN-γ, TNF-α and nitric oxide (NO) in the spleen of infected animals and, as a consequence, Tlr4−/− mice display higher parasitemia levels. Collectively, our results indicate that TLR4, as well as previously shown for TLR2, TLR9 and MyD88, contributes to the innate immune response and, consequently, resistance in the acute phase of infection, although each of these pathways is not individually essential for the generation of class I-restricted responses against T. cruzi

    The Evolution of the Two Largest Tropical Ice Masses since the 1980s

    As tropical glaciers continue to retreat, we need accurate knowledge about where they are located, how large they are, and their retreat rates. Remote sensing data are invaluable for tracking these hard-to-reach glaciers. However, remotely identifying tropical glaciers is prone to misclassification errors due to ephemeral snow cover. We reevaluate the size and retreat rates of the two largest tropical ice masses, the Quelccaya Ice Cap (Peru) and Nevado Coropuna (Peru), using remote sensing data from the Landsat missions. To quantify their glacial extents more accurately, we expand the time window for our analysis beyond the dry season (austral winter), processing in total 529 Landsat scenes. We find that Landsat scenes from October, November, and December, which are after the dry season, better capture the glacial extent since ephemeral snow cover is minimized. We compare our findings to past studies of tropical glaciers, which have mainly analyzed scenes from the dry season. Our reevaluation finds that both tropical ice masses are smaller but retreating less rapidly than commonly reported. These findings have implications for these ice masses as sustained water resources for downstream communities
