Pharmakokinetik und Metabolismus des 20-Hyroxyecdysons im Menschen
Ecdysteroide sind Steroidhormone, die in
Arthropoden und im Pflanzenreich vorkommen. Sie werden vom
Menschen in relevanter Menge ĂŒber die Nahrung aufgenommen. In
der Literatur gibt es zahlreiche Hinweise auf Wirkungen dieser
Hormone auf den menschlichen Organismus. Ecdysteroide finden in
der Landwirtschaft (als Insektizide), als Medikamente und auch
zur Leistungssteigerung bei Sportlern Verwendung. Sie wurden
jedoch bisher nicht in die Dopingliste des Internationalen
Olympischen Komitees aufgenommen. Ziel dieser Arbeit war es,
grundlegende pharmakologische Eigenschaften der Ecdysteroide zu
untersuchen. Dies erfolgte in einem Versuch, in dem ein Proband
20 mg 20-Hydroxyecdyson als Kapsel einnahm und in den folgenden
zwei Tagen Urinproben zur spÀteren Analyse gesammelt wurden.
FĂŒr die Analysen wurde vor allem die HPLC verwandt, zum
weitgehend selektiven Nachweis von Ecdysteroiden diente ein
Peroxidase-gekoppelter EIA. Die Strukturbestimmung der
Metabolite erfolgte mittels LC/MS. In Rahmen dieser Arbeit
gelang es, die Probenvorbereitung von Urinproben fĂŒr
verschiedene analytische Methoden zu optimieren,
pharmakokinetische Daten von 20E im Menschen zu bestimmen und
zumindest einen Metaboliten dieser Substanz sicher zu
bestimmen. BezĂŒglich der Probenvorbereitung hat sich fĂŒr
chromatographische Verfahren wie DC, HPTLC, HPLC aber auch fĂŒr
die LC/MS die SchĂŒttelextraktion mit nachfolgender
Festphasenextraktion als herausragend dargestellt. Die
Pharmakokinetik zeigte einen zweigipfligen Verlauf der
Ausscheidung mit Maxima bei vier bis fĂŒnf Stunden nach Einnahme
und bei 21 Stunden nach Einnahme. Insgesamt konnten ca. 6% der
eingenommenen Dosis 20E im Urin als immunoreaktive Steroide
wiedergefunden werden. BezĂŒglich des Metabolismus zeigte sich,
dass in den Urinproben, die sich mit dem ersten
Exkretionsmaximum nach vier bis fĂŒnf Stunden deckten, sowohl
die Ausgangssubstanz 20-Hydroxyecdyson als auch das Ecdysteroid
14-Desoxy-20-Hydroxyecdyson vorlag. Nach 21 Stunden war
lediglich 20-Hydroxyecdyson nachweisbar und eine um 16
Masseneinheiten schwerere Substanz, die gut vereinbar mit einer
Hydroxylierung von 20E wÀre
Monitoring of Daily Training Load and Training Load Responses in Endurance Sports: What do Coaches Want?
Accurate assessment of training load (TL) and training load responses (TLR) might be useful for an optimized training regulation and prevention of overtraining. No consensus on a gold-standard for measuring TL or intensity in endurance sports has been reported in the available literature so far. The aim of the present article is i) to identify feasible parameters to measure TL and TLR in daily training and ii) to compare these scientific approaches with the needs of elite endurance coaches. Therefore, the first part provides a systematic review of the current literature and the second part concentrates on the results of a questionnaire that assessed the coachesâ requirements for monitoring daily endurance training. The systematic review revealed that the combination of both quantitative and qualitative data seems most promising to evaluate TL and TLR. Thus, validated questionnaires or rating of perceived exertion (RPE), combined with physiological parameters, such as heart rate, are often used and seem to provide the most reliable results. From the coachesâ perspective, duration and kind of training, RPE, as well as personal remarks in the athletesâ training diaries are considered to be essential information. Further, the coaches favor a feasible system that collects large amounts of directly measurable and perceived data and that is able to learn from previous events in order to present the most important information in a short individual overview. When comparing both parts of the present study, it becomes clear that the scientific research cannot yet fully respond to the coachesâ requests, however, based on the coachesâ propositions, scientific research might be stimulated to tackle this challenge in the near future

Analysis of a Streptomyces coelicolor A3(2) locus containing the nucleoside diphosphate kinase (ndk) and folylpolyglutamate synthetase (folC) genes
A 3.6-kb DNA fragment from Streptomyces coelicolor A3(2) with the genes valS probably encoding a valyl-tRNA synthetase, folC encoding folylpolyglutamate synthetase, and ndk encoding a nucleoside diphosphate kinase was analysed. folC and ndk are separated by a small open reading frame of unknown function, orfX. The deduced folC gene product is a protein of 46â677 Da whose sequence is similar to other folylpolyglutamate synthetases and folylpolyglutamate synthetase-dihydrofolate synthetases from both Gram-positive and Gram-negative bacteria. After cloning folC behind the lacZ promoter, the Streptomyces folC complemented a folC mutant of Escherichia coli. Enzymatic CâC-Coupling Prenylation: Bioinformatics â Modelling â Mechanism â Protein-Redesign â Biocatalytic Application
The functional role of isoprenoids and especially enzymatic prenylation in nature and human application is briefly covered, with the focus on bioinformatical, mechanistical and structural aspects of prenyltransferases and terpene synthases. These enzymes are as yet underrepresented
but perspectively useful biocatalysts for CâC couplings of aromatic and isoprenoid substrates. Some examples of the successful use in chemoenzymatic synthesis are given including an application for the otherwise difficult synthesis of Kuhistanol A. Computational structure-based site-directed
mutagenesis can be used for rational enzyme redesign to obtain altered substrate and product specificities, which is demonstrated for terpene cyclases
Differences in Hypercholesterolemia and Atherogenesis Induced by Common Androgen Deprivation Therapies in Male Mice
Background-Treatment of prostate cancer often involves androgen deprivation therapy (ADT) by gonadotropin-releasing hormone (GnRH) receptor agonists, GnRH receptor antagonists, or orchiectomy. ADT may increase the rate of cardiovascular disease events, but recent clinical studies suggested that not all means of ADT carry the same risk, raising the possibility of non-testosterone-mediated effects of different forms of ADT on atherosclerosis. Here we compared effects of ADT on atherosclerosis in intact and orchiectomized Apoe-deficient mice. Methods and Results-Chow-fed Apoe-deficient mice were allocated to orchiectomy and/or monthly injections with the GnRH receptor agonist leuprolide or the GnRH receptor antagonist degarelix. Atherosclerosis was quantified at 26 weeks of age in the aortic arch by en face examination and in the aortic root by histology. In intact Apoe-deficient mice, all types of ADT reduced testosterone production to castration levels. Although hypercholesterolemia was accentuated in leuprolide-treated mice, the amount and composition of atherosclerosis was not different between the different types of ADT. In orchiectomized Apoe-deficient mice, leuprolide, but not degarelix, augmented hypercholesterolemia, changed body, thymus, and spleen weights, and increased atherosclerosis in the aortic root. No direct effects of the drugs were detectable on cytokine secretion from murine bone marrow-derived macrophages or on splenocyte proliferation. Conclusions-No differences in the development of atherosclerosis were detected among groups of intact Apoe-deficient mice treated with different types of ADT. A pro-atherogenic, possibly cholesterol-mediated, effect of leuprolide was seen in orchiectomized mice that might be relevant for understanding the potential cardiovascular risk associated with GnRH agonist-based ADT.The study was investigator-initiated and sponsored by Ferring Pharmaceuticals.S
