    01-03 "Community Control in a Global Economy: Lessons from Mexico's Economic Integration Process"

    The North American Free Trade Agreement appeared to promise economic growth for Mexico and improved living conditions for its people. While the Mexican economy has recovered significantly from its post-NAFTA collapse, there is mounting evidence that many of the pre-NAFTA warnings of worsening poverty and deteriorating environmental conditions were true, if exaggerated. However one interprets the statistics, there is little doubt that the economic integration process, which began a full decade before NAFTA took effect, has created a significant restructuring of the Mexican economy, with some of the country's most vulnerable residents facing the harshest conditions. How have those most affected by the economic integration process responded to the challenges and opportunities posed by globalization? Based on a collaborative research project between U.S. and Mexican researchers, the authors provide an overview of the existing English-language research on the subject and suggest a research agenda to assess adaptive strategies and to draw from those experiences lessons for the construction of future trade agreements.

    A globalised practice of local peace agreements

    In recent years, the difficulties and failures of national level peace processes have prompted increased attention on ‘the local’ as a space of conflict resolution. The papers presented here suggest both the existence of local peacemaking, and the impossibility of delimiting what is merely ‘local’ about it. This in turn points to a need for a new political imaginary for peace processes, which would go beyond the idea that it is about brokering elite pacts reached in a comprehensive peace agreement. The contributions to this Collection all conclude that local agreements deserve greater attention as a peacemaking tool. They also demonstrate variation in how local agreements are used within and across contexts, and indicate significant involvement of international actors in many contexts

    Functional analysis of a human A1 adenosine receptor/green fluorescent protein/Gi1α fusion protein following stable expression in CHO cells

    AbstractFusion proteins between the human A1 adenosine receptor and the pertussis toxin resistant (Cys351Gly) mutant of the G-protein α subunit Gi1α (A1/Gi), and between the human A1 adenosine receptor, the Aequorea victoria green fluorescent protein (GFP) and Cys351Gly Gi1α (A1/GFP/Gi), were expressed in CHO cells. The agonist NECA caused a stimulation of [35S]GTPγS binding at both fusion proteins with similar concentration dependence as at the native receptor. However in the presence of pertussis toxin NECA stimulation of [35S]GTPγS binding was only seen at the A1/GFP/Gi fusion protein. The regulation of the adenylyl cyclase and MAP kinase effector systems by both fusion proteins was attenuated following pertussis toxin treatment. These studies demonstrate for the first time the characterisation of a fusion protein between a G-protein coupled receptor, GFP and a G-protein α subunit

    Inclusive Chemical Characterization of Tourmaline: Mossbauer Study of Fe Valence and Site Occupancy

    We report here the results of a series of inclusive chemical characterizations, including all elements except oxygen, for a suite of 54 tourmaline samples. A combination of analytical techniques was used to analyze for major and light elements (electron microprobe), Fe3+ and Fe2+ (Mossbauer spectroscopy), H (U extraction), and B, Li, and F (ion microprobe, or SIMS). The B content of the tourmalines studied ranges from 2.86 to 3.26 B per formula unit (pfu) with 31 anions; excess boron is believed to reside in the Si site. Li ranges from 0.0 to 1.44 Li pfu and F contents are 0.0-0.91 pfu. H contents range from nearly anhydrous up to 3.76 H pfu and do not correlate simply with Fe3+ content. Mossbauer results show that tourmaline exhibits the entire range of Fe3+/Sigma Fe from 0.0-1.0. Fe2+ is represented in the spectra by three doublets, with occupancy in at least three distinct types of Y sites (with different types of nearest and next nearest neighbors). Fe3+ was found in 26 of the 54 samples studied. Although Mossbauer data do not allow the distinction between Fe-[Y](3+) and Fe-[Z](3+) site occupancies to be made, XRD data on these samples suggest that the majority of Fe3+ is also in Y. Of the samples studied, Fe-[4](3+) occurs in nine; five of those were either olenite or uvite with extensive Na substitution. A mixed valence doublet corresponding to delocalized electrons shared between adjacent octahedra was observed in 14 of the samples studied. Projection pursuit regression analysis shows that distribution of Fe among doublets is a function (albeit a complex one) of bulk composition of the tourmaline and supports the interpretation of doublets representing different populations of neighbors. Variations in Fe3+/Fe2+ ratio cannot be directly related to variations in charge in any single site of the structure. Fe3+/Fe2+ ratio is probably controlled by the prevailing oxidation state in the bull; rock assemblage, rather than by any particular crystal chemical substitution

    Trajectories of exposure and vulnerability of small islands to climate change

It uses the trajectory of exposure and vulnerability (TEV) approach that it defines as ‘storylines of driving factors and processes that have influenced past and present territorial system exposure and vulnerability to impacts associated with climate variability and change.’ The study is based on the analysis of six peer-reviewed Pacific island case studies covering various geographical settings (high islands vs low-lying reef islands, urban vs rural) and hazards associated with climate variability and change; that addressed the interactions between natural and anthropogenic driving factors; and adopted multidecadal past-to-present approaches. The findings emphasize that most urban and rural reef and high islands have undergone increasing exposure and vulnerability as a result of major changes in settlement and demographic patterns, lifestyles and economies, natural resources availability, and environmental conditions. The article highlights three generic and successive periods of change in the studied islands’ TEV: from geopolitical and political over the colonization-to-political independence period; to demographic, socio-economic, and cultural from the 1960s to the 1980s; culminating in the dominance of demographic, socio-economic, cultural, and environmental drivers since the 1980s. Based on these empirical insights, the article emphasizes the existence of anthropogenic-driven path-dependency effects in TEV, thus arguing for the analysis of the temporal dimensions of exposure and vulnerability to be a prerequisite for science to be able to inform policy- and decision-making processes toward robust adaptation pathways.PostprintPeer reviewe

    Comparative transcriptomics in the Triticeae

    <p>Abstract</p> <p>Background</p> <p>Barley and particularly wheat are two grass species of immense agricultural importance. In spite of polyploidization events within the latter, studies have shown that genotypically and phenotypically these species are very closely related and, indeed, fertile hybrids can be created by interbreeding. The advent of two genome-scale Affymetrix GeneChips now allows studies of the comparison of their transcriptomes.</p> <p>Results</p> <p>We have used the Wheat GeneChip to create a "gene expression atlas" for the wheat transcriptome (cv. Chinese Spring). For this, we chose mRNA from a range of tissues and developmental stages closely mirroring a comparable study carried out for barley (cv. Morex) using the Barley1 GeneChip. This, together with large-scale clustering of the probesets from the two GeneChips into "homologous groups", has allowed us to perform a genomic-scale comparative study of expression patterns in these two species. We explore the influence of the polyploidy of wheat on the results obtained with the Wheat GeneChip and quantify the correlation between conservation in gene sequence and gene expression in wheat and barley. In addition, we show how the conservation of expression patterns can be used to elucidate, probeset by probeset, the reliability of the Wheat GeneChip.</p> <p>Conclusion</p> <p>While there are many differences in expression on the level of individual genes and tissues, we demonstrate that the wheat and barley transcriptomes appear highly correlated. This finding is significant not only because given small evolutionary distance between the two species it is widely expected, but also because it demonstrates that it is possible to use the two GeneChips for comparative studies. This is the case even though their probeset composition reflects rather different design principles as well as, of course, the present incomplete knowledge of the gene content of the two species. We also show that, in general, the Wheat GeneChip is not able to distinguish contributions from individual homoeologs. Furthermore, the comparison between the two species leads us to conclude that the conservation of both gene sequence as well as gene expression is positively correlated with absolute expression levels, presumably reflecting increased selection pressure on genes coding for proteins present at high levels. In addition, the results indicate the presence of a correlation between sequence and expression conservation within the Triticeae.</p

    The LOFAR Transients Pipeline

    Current and future astronomical survey facilities provide a remarkably rich opportunity for transient astronomy, combining unprecedented fields of view with high sensitivity and the ability to access previously unexplored wavelength regimes. This is particularly true of LOFAR, a recently-commissioned, low-frequency radio interferometer, based in the Netherlands and with stations across Europe. The identification of and response to transients is one of LOFAR's key science goals. However, the large data volumes which LOFAR produces, combined with the scientific requirement for rapid response, make automation essential. To support this, we have developed the LOFAR Transients Pipeline, or TraP. The TraP ingests multi-frequency image data from LOFAR or other instruments and searches it for transients and variables, providing automatic alerts of significant detections and populating a lightcurve database for further analysis by astronomers. Here, we discuss the scientific goals of the TraP and how it has been designed to meet them. We describe its implementation, including both the algorithms adopted to maximize performance as well as the development methodology used to ensure it is robust and reliable, particularly in the presence of artefacts typical of radio astronomy imaging.     Social trends and new geographies

    This opening chapter sets the scene for subsequent more detailed analysis of many of the issues raised here. We start by discussing in Section 1 the tension in the current era between humanity’s simultaneously standing at “the peak of possibilities” while also, possibly, facing an abyss due to growing inequalities, political conflict and the ever-present danger of climate catastrophe. We turn in Sections 2 and 3 to the main social and spatial transformations that have characterised the last twenty five years. Again we see advances and regressions, above all uneven and fragile development. These sections set the scene for a consideration of three specific challenges: the tension between capitalism and democracy (Section 4); that between production and reproduction with an emphasis on gender relations (Section 5); and that between demographic change and sustainability (Section 6). We then conclude with a sober appraisal of the prospects for the emergence of viable agents for social transformation (Section 7) before making some general remarks on the challenges and possibilities for social progress (Section 8). The underlying hypothesis for social progress is that development is, and always has been, contradictory. Poverty amongst plenty, individual advancement versus collective regression and repression intertwined with liberty. If the industrial era emerged through what Karl Polanyi called a “great transformation” are we headed towards, or do we need a ‘new’ great transformation? We posit a general need for the market to be re-embedded in society if social progress is not to be halted or even reversed. In terms of the political order we find that the recent transformations of democracy and capitalism have had hugely ambiguous features. It is not wrong to say that the planet is currently both more democratic and more affluent than it was three decades ago. But the ways in which such progress has come about endangers not only future progress, it even puts past progress at risk. In political terms, the increasing diffusion of democracy means that more people across the globe have a say on the collective matters that concern them. But under current circumstances, their participation may not be able to reach the kind of decisions that one would understand as collective self-determination. In economic terms, material affluence is being created in unprecedented forms and volume. But, first, this affluence is so unevenly generated and distributed that poverty and hardship do not disappear and are even reproduced in new and possibly more enduring forms. And second, the continuing production of this material affluence may/will endanger the inhabitability of the planet, or large parts of it, even in the short- or medium-term. We have seen our task as one of offering a complex assessment of the current situation that has not been over-determined by our own political preferences. The positive and negative components of the picture we offer are constitutive of the ambivalent nature of the social progress. We are acutely aware that the world looks very different according to our standpoint geographically, socially and by our social and cultural identity. So we have not posed a false unity in terms of outlook. We consider it useful to pose the key questions as clearly as possible from a collective perspective that includes many diverse disciplinary and subject stand-points. We also seek to avoid an analysis determined by either a depressed Weltanschauung that sees only catastrophe ahead given recent political developments or what some have called a Polanyian Pollyanna tendency that is emotionally committed to positive social transformation regardless of the evidence. Quite simply, neither pessimism nor optimism are adequate diagnostic tools. This is particularly the case when we turn to the possible agents of the ‘new’ social transformation we advocate. While we show the decline of 20th-century agents of social change we also try to bring to life the new potential actors for redistribution, social justice and recognition

    Desmopressin for prevention of bleeding for thrombocytopenic, critically ill patients undergoing invasive procedures: A randomised, double‐blind, placebo‐controlled feasibility trial

    Thrombocytopenic patients have an increased risk of bleeding when undergoing invasive procedures. In a multicentre, phase II, blinded, randomised, controlled feasibility trial, critically ill patients with platelet count 100 × 109/L or less were randomised 1:1 to intravenous desmopressin (0.3 ”g/kg) or placebo before an invasive procedure. Forty‐three participants (18.8% of those eligible) were recruited, with 41 eligible for analysis. Post‐procedure bleeding occurred in one of 22 (4.5%) in the placebo arm and zero of 19 in the desmopressin arm. Despite liberal inclusion criteria, there were significant feasibility challenges recruiting patients in the critical care setting prior to invasive procedures
