    Oxidized flavors in strawberry ice cream

    At the time this project was initiated only a small amount of material had been published concerning this defect and even that was subject to considerable controversy. That the defect under consideration is important has been emphasized by other workers and needs no reiteration here. It was the opinion of the authors that the study should be made with ice cream manufactured in a commercial manner and from commercial products. This was done wherever possible. The objectives of the project were to attempt to determine which single factor or group of the following possible factors-oxidases in the fruit, copper and iron in the mix, strawberries and types of added solids - was responsible for the occurrence of the defect. It was likewise considered advisable to see whether or not such changes as occur in the fat, if the defect were a fat oxidation, were sufficient to cause detectable variations in the iodine, acetyl and Reichert- Meissl numbers of the fat of the ice cream

    Expression of the Bcl-2 Protein BAD Promotes Prostate Cancer Growth

    BAD, a pro-apoptotic protein of the Bcl-2 family, has recently been identified as an integrator of several anti-apoptotic signaling pathways in prostate cancer cells. Thus, activation of EGFR, GPCRs or PI3K pathway leads to BAD phosphorylation and inhibition of apoptosis. Increased levels of BAD in prostate carcinomas have also been reported. It appears contradictory that instead of limiting expression of pro-apoptotic protein, prostate cancer cells choose to increase BAD levels while keeping it under tight phosphorylation control. Analysis of the effect of BAD on prostate cancer xenografts has shown that increased BAD expression enhances tumor growth, while knockdown of BAD expression by shRNA inhibits tumor growth. Tissue culture experiments demonstrated that increased BAD expression stimulates proliferation of prostate cancer cells. These results suggest that increased expression of BAD provides a proliferative advantage to prostate tumors, while BAD dephosphorylation increases sensitivity of prostate cancer cells to apoptosis. Combination of proliferative and apoptotic properties prompts prostate cancer cells to be “addicted” to increased levels of phosphorylated BAD. Thus, kinases that phosphorylate BAD are plausible therapeutic targets; while monitoring BAD phosphorylation could be used to predict tumor response to treatments

    Determination of Point and Nonpoint Source Toxicity in the Clark Fork River Basin Using the Daphnid, Ceriodaphnia Dubia

    Ceriodaphnia dubia, a small planktonic daphnid was used to biomonitor point sources of toxicity in wastewater and nonpoint source toxicity in stream samples obtained from the Clark Fork River Basin, MT. Brief descriptions, results and discussions are presented for studies of wastewater from a kraft mill near Frenchtown, MT and potential toxicity of water samples from 19 sites along the Clark Fork River in 1985. In 1987, dilutions of Missoula, MT municipal wastewater fortified with ammonia were tested, as was the wastewater before and after chlorination. Potential toxicity of water samples from eight sites along the upper Clark Fork River were also tested. All studies were cooperative efforts with the Montana Departments of Health and Environmental Sciences and Fish, Wildlife and Parks. Ceriodaphnia appear to be indicators of toxicity in a variety of test conditions such as ammonia in wastewater and metals from past mining activities. The daphnids indicated toxicity from other substances in the wastewater or perhaps the influence of characteristics of the wastewater that increased ammonia toxicity. All example of nonpoint source effects was toxicity in samples from Silver Bow Creek MT, where impaired conditions to aquatic life resulting from the presence of metals have been reported for years. During some of the tests with wastewater, toxicological endpoints were observed using the actual number of daphnids that reproduced in a test, not the average number of young. There was circumstantial evidence in 1985 that copper alone was responsible for the toxicity in Silver Bow Creek. However, the later studies performed under different hydrological conditions found toxicity was probably due to a combination of metals, some of which had not been measurable earlier. For well-defined control of standard conditions during testing, there are indications that waters to be used as reference media for Ceriodaphnia need further research. Nevertheless, the use of daphnids to test the ambient conditions described in this paper should encourage environmental managers to consider approaches with this or similar species in the future

    Detecting Mutations in the Mycobacterium tuberculosis Pyrazinamidase Gene pncA to Improve Infection Control and Decrease Drug Resistance Rates in Human Immunodeficiency Virus Coinfection.

    Hospital infection control measures are crucial to tuberculosis (TB) control strategies within settings caring for human immunodeficiency virus (HIV)-positive patients, as these patients are at heightened risk of developing TB. Pyrazinamide (PZA) is a potent drug that effectively sterilizes persistent Mycobacterium tuberculosis bacilli. However, PZA resistance associated with mutations in the nicotinamidase/pyrazinamidase coding gene, pncA, is increasing. A total of 794 patient isolates obtained from four sites in Lima, Peru, underwent spoligotyping and drug resistance testing. In one of these sites, the HIV unit of Hospital Dos de Mayo (HDM), an isolation ward for HIV/TB coinfected patients opened during the study as an infection control intervention: circulating genotypes and drug resistance pre- and postintervention were compared. All other sites cared for HIV-negative outpatients: genotypes and drug resistance rates from these sites were compared with those from HDM. HDM patients showed high concordance between multidrug resistance, PZA resistance according to the Wayne method, the two most common genotypes (spoligotype international type [SIT] 42 of the Latino American-Mediterranean (LAM)-9 clade and SIT 53 of the T1 clade), and the two most common pncA mutations (G145A and A403C). These associations were absent among community isolates. The infection control intervention was associated with 58-92% reductions in TB caused by SIT 42 or SIT 53 genotypes (odds ratio [OR] = 0.420, P = 0.003); multidrug-resistant TB (OR = 0.349, P < 0.001); and PZA-resistant TB (OR = 0.076, P < 0.001). In conclusion, pncA mutation typing, with resistance testing and spoligotyping, was useful in identifying a nosocomial TB outbreak and demonstrating its resolution after implementation of infection control measures

    Transcriptional landscape of the human and fly genomes: Nonlinear and multifunctional modular model of transcriptomes

    Regions of the genome not coding for proteins or not involved in cis-acting regulatory activities are frequently viewed as lacking in functional value. However, a number of recent large-scale studies have revealed significant regulated transcription of unannotated portions of a variety of plant and animal genomes, allowing a new appreciation of the widespread transcription of large portions of the genome. High-resolution mapping of the sites of transcription of the human and fly genomes has provided an alternative picture of the extent and organization of transcription and has offered insights for biological functions of some of the newly identified unannotated transcripts. Considerable portions of the unannotated transcription observed are developmental or cell-type-specific parts of protein-coding transcripts, often serving as novel, alternative 5′ transcriptional start sites. These distal 5′ portions are often situated at significant distances from the annotated gene and alternatively join with or ignore portions of other intervening genes to comprise novel unannotated protein-coding transcripts. These data support an interlaced model of the genome in which many regions serve multifunctional purposes and are highly modular in their utilization. This model illustrates the underappreciated organizational complexity of the genome and one of the functional roles of transcription from unannotated portions of the genome.     Context dependent learning in the serial RT task

    This study investigated the development of contextual dependencies for sequential perceptual-motor learning on static features in the learning environment. In three experiments we assessed the effect of manipulating task irrelevant static context features in a serial reaction-time task. Experiment 1 demonstrated impaired performance after simultaneously changing display color, placeholder shape, and placeholder location. Experiment 2 showed that this effect was mainly caused by changing placeholder shape. Finally, Experiment 3 indicated that changing context affected both the application of sequence knowledge and the selection of individual responses. It is proposed either that incidental stimulus features are integrated with a global sequence representation, or that the changed context causes participants to strategically inhibit sequence skills

    Endemicity of Zoonotic Diseases in Pigs and Humans in Lowland and Upland Lao PDR: Identification of Socio-cultural Risk Factors

    In Lao People's Democratic Republic pigs are kept in close contact with families. Human risk of infection with pig zoonoses arises from direct contact and consumption of unsafe pig products. This cross-sectional study was conducted in Luang Prabang (north) and Savannakhet (central-south) Provinces. A total of 59 villages, 895 humans and 647 pigs were sampled and serologically tested for zoonotic pathogens including: hepatitis E virus (HEV), Japanese encephalitis virus (JEV) and Trichinella spiralis; In addition, human sera were tested for Taenia spp. and cysticercosis. Seroprevalence of zoonotic pathogens in humans was high for HEV (Luang Prabang: 48.6%, Savannakhet: 77.7%) and T. spiralis (Luang Prabang: 59.0%, Savannakhet: 40.5%), and lower for JEV (around 5%), Taenia spp. (around 3%) and cysticercosis (Luang Prabang: 6.1, Savannakhet 1.5%). Multiple correspondence analysis and hierarchical clustering of principal components was performed on descriptive data of human hygiene practices, contact with pigs and consumption of pork products. Three clusters were identified: Cluster 1 had low pig contact and good hygiene practices, but had higher risk of T. spiralis. Most people in cluster 2 were involved in pig slaughter (83.7%), handled raw meat or offal (99.4%) and consumed raw pigs' blood (76.4%). Compared to cluster 1, cluster 2 had increased odds of testing seropositive for HEV and JEV. Cluster 3 had the lowest sanitation access and had the highest risk of HEV, cysticercosis and Taenia spp. Farmers which kept their pigs tethered (as opposed to penned) and disposed of manure in water sources had 0.85 (95% CI: 0.18 to 0.91) and 2.39 (95% CI: 1.07 to 5.34) times the odds of having pigs test seropositive for HEV, respectively. The results have been used to identify entry-points for intervention and management strategies to reduce disease exposure in humans and pigs, informing control activities in a cysticercosis hyper-endemic village
