    The Effects of Professional Development in Formative Assessment on Mathematics Teaching Performance and Student Achievement

    This article describes a year-long professional development program on formative assessment for middle school mathematics teachers and its impact on teachers’ use of formative assessment during instruction. The focus of the program was to support teachers in (a) embedding formative assessments into their instruction, (b) interpreting student responses and performance on formative assessments, (c) using effective questioning strategies and peer assessment, and (d) identifying student errors and misconceptions and implementing instructional strategies to address them. Teacher and student performance data were collected on participating teachers, control teachers, and their students during the year-long professional development. The professional development had an impact on teachers’ cognitive level of questioning, use of peer assessment, and types of questioning strategies. Results of student performance data revealed differences in performance on some types of items among students of participating teachers and control teachers

    Investigating the relationship between mitochondrial genetic variation and cardiovascular-related traits to develop a framework for mitochondrial phenome-wide association studies

    BACKGROUND: Mitochondria play a critical role in the cell and have DNA independent of the nuclear genome. There is much evidence that mitochondrial DNA (mtDNA) variation plays a role in human health and disease, however, this area of investigation has lagged behind research into the role of nuclear genetic variation on complex traits and phenotypic outcomes. Phenome-wide association studies (PheWAS) investigate the association between a wide range of traits and genetic variation. To date, this approach has not been used to investigate the relationship between mtDNA variants and phenotypic variation. Herein, we describe the development of a PheWAS framework for mtDNA variants (mt-PheWAS). Using the Metabochip custom genotyping array, nuclear and mitochondrial DNA variants were genotyped in 11,519 African Americans from the Vanderbilt University biorepository, BioVU. We employed both polygenic modeling and association testing with mitochondrial single nucleotide polymorphisms (mtSNPs) to explore the relationship between mtDNA variants and a group of eight cardiovascular-related traits obtained from de-identified electronic medical records within BioVU. RESULTS: Using polygenic modeling we found evidence for an effect of mtDNA variation on total cholesterol and type 2 diabetes (T2D). After performing comprehensive mitochondrial single SNP associations, we identified an increased number of single mtSNP associations with total cholesterol and T2D compared to the other phenotypes examined, which did not have more significantly associated SNPs than would be expected by chance. Among the mtSNPs significantly associated with T2D we identified variant mt16189, an association previously reported only in Asian and European-descent populations. CONCLUSIONS: Our replication of previous findings and identification of novel associations from this initial study suggest that our mt-PheWAS approach is robust for investigating the relationship between mitochondrial genetic variation and a range of phenotypes, providing a framework for future mt-PheWAS

    Sensitivity to Sulfited Foods among Sulfite-Sensitive Subjects with Asthma

    Eight individuals with asthma who had been diagnosed as sulfite sensitive on the basis of double-blind capsule-beverage challenges were subjected to challenges with various sulfited foods, including lettuce, shrimp, dried apricots, white grape juice, dehydrated potatoes (as mashed potatoes), and mushrooms. Four of these patients failed to respond to challenges with any of the sulfited foods. The other four patients experienced a decrease in pulmonary function on double-blind challenges with sulfited lettuce. Two of three of these patients reacted to challenges with dried apricots and white grape juice; the fourth patient has not yet been challenged with these products. Only one of these four patients reacted to challenges with dehydrated potatoes and mushrooms, and, in this case, the response to double-blind challenges with dehydrated potatoes was not consistent. None of the sulfite-sensitive subjects with asthma responded to challenges with sulfited shrimp. It is concluded that sulfite-sensitive subjects with asthma will not necessarily react after ingestion of sulfited foods. The likelihood of a reaction is dependent on the nature of the food, the level of residual sulfite, the sensitivity of the patient, and perhaps on the form of residual sulfite and the mechanism of the sulfite-induced reaction

    Development and Use of Engineering Standards for Computational Fluid Dynamics for Complex Aerospace Systems

    Computational fluid dynamics (CFD) and other advanced modeling and simulation (M&S) methods are increasingly relied on for predictive performance, reliability and safety of engineering systems. Analysts, designers, decision makers, and project managers, who must depend on simulation, need practical techniques and methods for assessing simulation credibility. The AIAA Guide for Verification and Validation of Computational Fluid Dynamics Simulations (AIAA G-077-1998 (2002)), originally published in 1998, was the first engineering standards document available to the engineering community for verification and validation (V&V) of simulations. Much progress has been made in these areas since 1998. The AIAA Committee on Standards for CFD is currently updating this Guide to incorporate in it the important developments that have taken place in V&V concepts, methods, and practices, particularly with regard to the broader context of predictive capability and uncertainty quantification (UQ) methods and approaches. This paper will provide an overview of the changes and extensions currently underway to update the AIAA Guide. Specifically, a framework for predictive capability will be described for incorporating a wide range of error and uncertainty sources identified during the modeling, verification, and validation processes, with the goal of estimating the total prediction uncertainty of the simulation. The Guide's goal is to provide a foundation for understanding and addressing major issues and concepts in predictive CFD. However, this Guide will not recommend specific approaches in these areas as the field is rapidly evolving. It is hoped that the guidelines provided in this paper, and explained in more detail in the Guide, will aid in the research, development, and use of CFD in engineering decision-making

    The role of taxonomic expertise in interpretation of metabarcoding studies

    Abstract The performance of DNA metabarcoding approaches for characterizing biodiversity can be influenced by multiple factors. Here, we used morphological assessment of taxa in zooplankton samples to develop a large barcode database and to assess the congruence of taxonomic identification with metabarcoding under different conditions. We analysed taxonomic assignment of metabarcoded samples using two genetic markers (COI, 18S V1–2), two types of clustering into molecular operational taxonomic units (OTUs, ZOTUs), and three methods for taxonomic assignment (RDP Classifier, BLASTn to GenBank, BLASTn to a local barcode database). The local database includes 1042 COI and 1108 18S (SSU) barcode sequences, and we added new high-quality sequences to GenBank for both markers, including 109 contributions at the species level. The number of phyla detected and the number of taxa identified to phylum varied between a genetic marker and among the three methods used for taxonomic assignments. Blasting the metabarcodes to the local database generated multiple unique contributions to identify OTUs and ZOTUs. We argue that a multi-marker approach combined with taxonomic expertise to develop a curated, vouchered, local barcode database increases taxon detection with metabarcoding, and its potential as a tool for zooplankton biodiversity surveys

    Updated site compilation of the Latin American Pollen Database

    Flantua, Suzette G A et al.The updated inventory of the Latin American Pollen Database (LAPD) offers a wide range of new insights. This paper presents a systematic compilation of palynological research in Latin America. A comprehensive inventory of publications in peer-reviewed and grey literature shows a major expansion of studies over the last decades. The inventory includes 1379 cores and sections with paleoecological data and more than 4800 modern samples from throughout the continent. Through the years, pollen datasets extend over increasing spans of time and show improved taxonomic and temporal resolution. Currently, these datasets are from 12 modern biomes and 30 countries, covering an altitudinal range of 0 to 6300. m asl. The most densely sampled regions are the Colombian Andes, the southeast coast of Brazil, and Patagonia. Underrepresented biomes are the warm temperate mixed forest (3%), dry forests (3%), and warm temperate rainforest (1%); whereas steppe, tropical rainforest, and cool grass shrublands, such as the páramos, are generally well represented (all >. 17%). There are 126 records that span the late Pleistocene to the Last Glacial Maximum transition (21,000. cal. yr BP), and >. 20% of the records cover the Younger Dryas interval and the Pleistocene/Holocene transition. Reanalysis of numerous sites using multiproxy tools emphasize the informative value of this approach in paleoenvironmental reconstruction. We make suggestions for several pollen sites and regions to be visited again; similarly we identify some key research questions that have yet to be answered. The updated LAPD now provides the platform to support an exciting new phase of global palynological research in which multi-site data are being integrated to address current cutting-edge research questions.
