Spinning straw into milk: Can a 95% byproduct diet support milk production?

One way the livestock industry has addressed this is by using human inedible feedstuffs, including industrial byproducts. Many dairy and feedlot diets incorporate 20–40% byproduct feeds, but few studies have evaluated responses of lactating dairy cattle to diets composed almost entirely of byproducts. Our objective was to evaluate such a diet in comparison to a more traditional lactation diet. The control diet was primarily composed of alfalfa hay, corn silage, corn gluten feed, and corn grain. The by-product diet included wheat straw, corn hominy, post-extraction algae residue, and corn gluten feed; in addition, 4% molasses was included to improve palatability. The control and by-product diets had similar concentrations of DM (50.6%) and CP (17.2%), whereas the byproduct diet included slightly more NDF (32.9 vs. 30.5%) and less fat (4.7 vs. 5.2%). Twelve Holstein cows (154 ± 20 DIM) were blocked by parity (primiparous vs. multiparous) and randomly assigned to treatment sequence in a crossover design. Diets were fed for 20 d, with data and sample collections over the final 3 d of each period. One cow was removed from byproduct diet after refusing to consume it, and data from this period were not included in the analysis. Data were analyzed with mixed models to assess fixed effects of diet, parity, and their interaction as well as the random effects of cow and period, and significance was declared at P < 0.05. The one selective cow notwithstanding, DMI was not affected by treatment. Milk yield of multiparous cows was decreased by the byproduct diet (38.7 vs. 42.3 ± 2.2 kg/d) but there was no treatment effect in primiparous cows (39.3 vs. 39.4 ± 2.2 kg/d). The byproduct diet decreased milk fat content (3.3 vs. 3.6 ± 0.12%) and tended to decrease protein content (2.94 vs. 2.99 ± 0.05%), and energy-corrected milk yield was decreased by 5.4 kg/d in multiparous cows and 1.5 kg/d in primiparous cows. No effects on BW or BCS were detected. Despite negative productivity responses, calculated recoveries of human-edible protein and energy in the diet were increased by approximately 50% with the byproduct diet, changing from a net loss to a net gain in human-edible energy and protein. A diet composed of 95% byproduct feeds supported milk yield of 39 kg/d and increased the efficiency of production from a human-edible input perspective
Educational attainment trajectories among children and adolescents with depression, and the role of sociodemographic characteristics: longitudinal data-linkage study
Background
Depression is associated with lower educational attainment, but there has been little investigation of long-term educational trajectories in large cohorts with diagnosed depression.
Aims:
To describe the educational attainment trajectories of children with a depression diagnosis in secondary care, and to investigate whether these trajectories vary by sociodemographic characteristics. /
Method:
We identified new referrals to South London and Maudsley's NHS Foundation Trust between 2007 and 2013 who received a depression diagnosis at under 18 years old. Linking their health records to the National Pupil Database, we standardised their performance on three assessments (typically undertaken at ages 6–7 years (school Year 2), 10–11 (Year 6) and 15–16 (Year 11)) relative to the local reference population in each academic year. We used mixed models for repeated measures to estimate attainment trajectories. /
Results:
In our sample of 1492 children, the median age at depression diagnosis was 15 years (interquartile range = 14–16). Their attainment showed a decline between school Years 6 and 11. Attainment was consistently lower among males and those eligible for free school meals. Black ethnic groups also showed lower attainment than White ethnic groups between Years 2 and 6, but showed a less pronounced drop in attainment at Year 11. /
Conclusions:
Those who receive a depression diagnosis during their school career show a drop in attainment in Year 11. Although this pattern was seen among multiple sociodemographic groups, gender, ethnicity and socioeconomic status predict more vulnerable subgroups within this clinical population who might benefit from additional educational support or more intensive treatment
Systematic review and meta-analysis: The association between child and adolescent depression and later educational attainment

is unclear. This systematic review and meta-analysis examines the longitudinal association
between depression and subsequent attainment, and its potential effect modifiers and
mediators.
Method: We searched Embase, PsycINFO, PubMed, ERIC and British Education Index from
inception to 23 October 2019, conducted citation searching, and contacted authors for
articles. Eligible studies reported on the longitudinal association between depression in
children and adolescents age 4-18 years and later educational attainment. Two reviewers
independently conducted screening, data extraction, and risk of bias assessment. Correlation
coefficients were pooled in meta-analysis, and effect modifiers were explored using metaregression and stratification. Other evidence on confounders, modifiers and mediators was
narratively synthesized. PROSPERO record: CRD42019123068.
Results: Thirty-one studies were included, of which 22 were pooled in meta-analysis. There
was a small but statistically significant association between depression and lower subsequent
attainment (pooled Fisher’s z=-0.19, 95% CI=-0.22 to -0.16, I
2=62.9%). Fifteen studies also
reported an enduring effect after adjusting for various confounders. No statistically
significant effect modifiers were identified. Social and school problems may mediate between
depression and low attainment.
Conclusion: Depression was associated with lower educational attainment, but further
research is needed to establish mechanisms. Nonetheless, there is a clear need for mental

The association between depression and later educational attainment in children and adolescents: a systematic review protocol.

Available from BMJ Open via the DOI in this record. INTRODUCTION: Depression represents a major public health concern for children and adolescents, and is thought to negatively impact subsequent educational attainment. However, the extent to which depression and educational attainment are directly associated, and whether other factors play a role, is uncertain. Therefore, we aim to systematically review the literature to provide an up-to-date estimate on the strength of this association, and to summarise potential mediators and moderators on the pathway between the two. METHODS AND ANALYSIS: To identify relevant studies, we will systematically search Embase, PsycINFO, PubMed, Education Resources Information Centre and British Education Index, manually search reference lists and contact experts in the field. Studies will be included if they investigate and report on the association between major depression diagnosis or depressive symptoms in children and adolescents aged 4-18 years (exposure) and later educational attainment (outcome). Two independent reviewers will screen titles, abstracts and full texts according to eligibility criteria, perform data extraction and assess study quality according to a modified version of the Newcastle-Ottawa Scale. If sufficiently homogeneous studies are identified, summary effect estimates will be pooled in meta-analysis, with further tests for study heterogeneity, publication bias and the effects of moderators using meta-regression. ETHICS AND DISSEMINATION: Because this review will make use of already published data, ethical approval will not be sought. The review will be submitted for publication in a peer-reviewed journal, presented at practitioner-facing conferences, and a lay summary will be written for non-scientific audiences such as parents, young people and teachers. Vesicular stomatitis virus enables gene transfer and transsynaptic tracing in a wide range of organisms
Current limitations in technology have prevented an extensive analysis of the connections among neurons, particularly within nonmammalian organisms. We developed a transsynaptic viral tracer originally for use in mice, and then tested its utility in a broader range of organisms. By engineering the vesicular stomatitis virus (VSV) to encode a fluorophore and either the rabies virus glycoprotein (RABV-G) or its own glycoprotein (VSV-G), we created viruses that can transsynaptically label neuronal circuits in either the retrograde or anterograde direction, respectively. The vectors were investigated for their utility as polysynaptic tracers of chicken and zebrafish visual pathways. They showed patterns of connectivity consistent with previously characterized visual system connections, and revealed several potentially novel connections. Further, these vectors were shown to infect neurons in several other vertebrates, including Old and New World monkeys, seahorses, axolotls, and Xenopus. They were also shown to infect two invertebrates, Drosophila melanogaster, and the box jellyfish, Tripedalia cystophora, a species previously intractable for gene transfer, although no clear evidence of transsynaptic spread was observed in these species. These vectors provide a starting point for transsynaptic tracing in most vertebrates, and are also excellent candidates for gene transfer in organisms that have been refractory to other methods
Image informatics strategies for deciphering neuronal network connectivity
Brain function relies on an intricate network of highly dynamic neuronal connections that rewires dramatically under the impulse of various external cues and pathological conditions. Among the neuronal structures that show morphologi- cal plasticity are neurites, synapses, dendritic spines and even nuclei. This structural remodelling is directly connected with functional changes such as intercellular com- munication and the associated calcium-bursting behaviour. In vitro cultured neu- ronal networks are valuable models for studying these morpho-functional changes. Owing to the automation and standardisation of both image acquisition and image analysis, it has become possible to extract statistically relevant readout from such networks. Here, we focus on the current state-of-the-art in image informatics that enables quantitative microscopic interrogation of neuronal networks. We describe the major correlates of neuronal connectivity and present workflows for analysing them. Finally, we provide an outlook on the challenges that remain to be addressed, and discuss how imaging algorithms can be extended beyond in vitro imaging studies
A proposal for a coordinated effort for the determination of brainwide neuroanatomical connectivity in model organisms at a mesoscopic scale
In this era of complete genomes, our knowledge of neuroanatomical circuitry
remains surprisingly sparse. Such knowledge is however critical both for basic
and clinical research into brain function. Here we advocate for a concerted
effort to fill this gap, through systematic, experimental mapping of neural
circuits at a mesoscopic scale of resolution suitable for comprehensive,
brain-wide coverage, using injections of tracers or viral vectors. We detail
the scientific and medical rationale and briefly review existing knowledge and
experimental techniques. We define a set of desiderata, including brain-wide
coverage; validated and extensible experimental techniques suitable for
standardization and automation; centralized, open access data repository;
compatibility with existing resources, and tractability with current
informatics technology. We discuss a hypothetical but tractable plan for mouse,
additional efforts for the macaque, and technique development for human. We
estimate that the mouse connectivity project could be completed within five
years with a comparatively modest budget.Comment: 41 page
Input-specific control of reward and aversion in the ventral tegmental area
Ventral tegmental area (VTA) dopamine neurons have important roles in adaptive and pathological brain functions related to reward and motivation. However, it is unknown whether subpopulations of VTA dopamine neurons participate in distinct circuits that encode different motivational signatures, and whether inputs to the VTA differentially modulate such circuits. Here we show that, because of differences in synaptic connectivity, activation of inputs to the VTA from the laterodorsal tegmentum and the lateral habenula elicit reward and aversion in mice, respectively. Laterodorsal tegmentum neurons preferentially synapse on dopamine neurons projecting to the nucleus accumbens lateral shell, whereas lateral habenula neurons synapse primarily on dopamine neurons projecting to the medial prefrontal cortex as well as on GABAergic (γ-aminobutyric-acid-containing) neurons in the rostromedial tegmental nucleus.
