    ACTIVE SUPPRESSION AS A POSSIBLE MECHANISM OF TOLERANCE IN TETRAPARENTAL MICE

    Previous work has indicated that tetraparental mice, chimeric since the eight-cell stage because of embryo fusion using histoincompatible strain combinations, possess autospecific immune cells and blocking antibodies. Although this phenomenon has been demonstrated in vitro, it may have relevance to the self-tolerance shown by these mice in vivo. The experiments described here indicate that spleen cells from tetraparental mice can block mixed lymphocyte reactions between the two parental cell types, but not between unrelated strains. Furthermore, this suppressive ability is not affected by an otherwise effective treatment of the tetraparental spleen cells with anti-θ antibody and complement. The in vitro experimental system elaborated here should help to characterize the cell type responsible for the suppression

    Evaluation of the QBC-vet autoread haematology system for domestic and pet animal species

    A comprehensive evaluation was initiated to determine the suitability of the QBC-Vet Autoread haematology system for veterinary purposes in domestic and pet animal species. The system determines haematocrit (HCT), haemoglobin (HGB), white blood cell (WBC) count, granulocyte count, combined lymphocyte and monocyte count (L/M), platelet count, as well as eosinophil and neutrophil counts (canine samples only), and reticulocyte count (canine and feline samples only). Linearity assessed for a canine sample usually surpassed the physiological range. Within-batch precision was very good for the majority of the parameters in feline and canine samples: Coefficients of variation (CV) were below 5.5% for HCT, HGB and WBC. In order to test the accuracy of the system with respect to reference methods, a total of 300 blood samples from the Clinics of Internal Veterinary Medicine and Surgery (101 canine, 99 feline and 100 equine) were analysed. Strong linear correlation was demonstrated for HCT, HGB (rτ;/0.96) and WBC (rτ;/0.93) based on high correlation coefficients and narrow confidence intervals. A somewhat higher degree of variation from the estimated regression lines was found in differential blood cell counts, especially for eosinophil counts of the dog where the automated reader erroneously attributed some lymphocytes or monocytes to eosinophil counts. Accuracy of the system was also assessed with respect to clinical relevance of results. The majority of leukocytosis (50 of 53), neutrophilias (3 of 4), or eosinophilias (4 of 5) was detected properly by the QBC-Vet Autoread haematology system, but only 20 out of 35 leukopenic samples were identified correctly. The system detected the presence of reticulocytes in the majority of feline (9 of 10) and canine (6 of 7) samples with a regenerative anaemia. Unexpectedly, platelets of cats were measured with high within-batch precision (mean CV=4.64%). No ‘streaming' effect (no discrimination between erythrocytes and granulocytes) was observed with this advanced QBC system. The system was found to be easy both in handling and interpretation of results. The buffy coat profile appeared particularly useful and informative. In conclusion, the QBC-Vet Autoread-System has excellent analytical properties and is well suited for veterinary purpose

    No detection of brodifacoum residues in the marine and terrestrial food web three years after rat eradication at Palmyra Atoll, Central Pacific

    Invasive alien species represent one of the greatest threats to native plants and animals on islands. Rats (Rattus spp.) have invaded most of the world’s oceanic islands, causing lasting or irreversible damage to ecosystems and biodiversity. To counter this threat, techniques to eradicate invasive rats from islands have been developed and applied across the globe. Eradication of alien rats from large or complex island ecosystems has only been successful with the use of bait containing a rodenticide. While effective at eradicating rats from islands, rodenticide can persist in the ecosystem longer than the time required to eradicate the target rat population and can potentially harm non-target species. However, the persistence of rodenticides in ecosystems following rat eradication campaigns is poorly understood, though predictions can be made based on the chemical properties of the rodenticide and the environment it is applied in. Brodifacoum, a relatively persistent second-generation anticoagulant, was used to successfully eradicate rats from Palmyra Atoll. With this study, we evaluated the persistence of brodifacoum residues in terrestrial and marine species at Palmyra Atoll (Northern Line Islands) three years after rat eradication. We collected 44 pooled samples containing 121 individuals of the following: mullet (Moolgarda engeli), cockroaches (Periplaneta sp.), geckos (Lepidodactylus lugubris), hermit crabs (Coenobita perlatus), and fiddler crabs (Uca tetragonon). Despite detection of brodifacoum residue in all five of the species sampled in this study 60 days after the application of bait to Palmyra Atoll in 2011, brodifacoum residue was not found in any of the pooled samples collected three years after bait application.     Fifty-kDa Hyaluronic Acid Upregulates Some Epidermal Genes without Changing TNF-α Expression in Reconstituted Epidermis

    Background: Due to its strong water binding potential, hyaluronic acid (HA) is a well-known active ingredient for cosmetic applications. However, based on its varying molecular size, skin penetration of HA may be limited. Recent studies have demonstrated that low-molecular-weight HA (LMW HA) may show a certain proinflammatory activity. We thus aimed to characterize an LMW-sized HA molecule that combines strong anti-aging abilities with efficient skin penetration but lacks potential proinflammatory effects. Methods: Total RNA and total protein were isolated from reconstituted human epidermis following incubation with HAs of various molecular weights (20, 50, 130, 300, 800 and 1,500 kDa). Tumor necrosis factor-alpha expression was determined using quantitative PCR. Genonnic and proteomic expression of various junctional proteins was determined using Affymetrix and common Western blotting techniques. Results: LMW HA of approximately 50 kDa did not significantly alter tumor necrosis factor-alpha expression compared to 20-kDa HA, but revealed significantly higher skin penetration rates than larger sized HA associated with increased expression of genes and proteins known to be involved in tight junction formation and keratinocyte cohesion. Conclusion: LMW HA of approximately 50 kDa shows better penetration abilities than larger-sized HA. In addition, LMW HA influences the expression of various genes including those contributing to keratinocyte differentiation and formation of intercellular tight junction complexes without showing proinflammatory activity. These observations contribute to current knowledge on the effects of LMW HA on keratinocyte biology and cutaneous physiology. Copyright (C) 2011 S. Karger AG, Base

    Spectral Analysis of the Chandra Comet Survey

    We present results of the analysis of cometary X-ray spectra with an extended version of our charge exchange emission model (Bodewits et al. 2006). We have applied this model to the sample of 8 comets thus far observed with the Chandra X-ray observatory and ACIS spectrometer in the 300-1000 eV range. The surveyed comets are C/1999 S4 (LINEAR), C/1999 T1 (McNaught-Hartley), C/2000 WM1 (LINEAR), 153P/2002 (Ikeya-Zhang), 2P/2003 (Encke), C/2001 Q4 (NEAT), 9P/2005 (Tempel 1) and 73P/2006-B (Schwassmann-Wachmann 3) and the observations include a broad variety of comets, solar wind environments and observational conditions. The interaction model is based on state selective, velocity dependent charge exchange cross sections and is used to explore how cometary X-ray emission depend on cometary, observational and solar wind characteristics. It is further demonstrated that cometary X-ray spectra mainly reflect the state of the local solar wind. The current sample of Chandra observations was fit using the constrains of the charge exchange model, and relative solar wind abundances were derived from the X-ray spectra. Our analysis showed that spectral differences can be ascribed to different solar wind states, as such identifying comets interacting with (I) fast, cold wind, (II), slow, warm wind and (III) disturbed, fast, hot winds associated with interplanetary coronal mass ejections.     The absorption properties of HgCdTe-based infrared detectors can be greatly increased in the mid-infrared band, by incorporating nanostructured plasmonic arrays on the illuminated detector face. The array periodicity, combined with the excitation of surface plasmon-polariton stationary modes, enhances the absorption efficiency by a substantial amount, allowing to reduce in turn the HgCdTe absorption thickness

    Effects of daunorubicin, mitomycin C, azathioprine and cyclosporin A on human retinal pigmented epithelial, corneal endothelial and conjunctival cell lines

    BACKGROUND: We wished to investigate the toxicity of four immunosuppressant and antimetabolic drugs, which are known to influence postoperative wound healing, on three different human ocular cell lines. METHODS: Acute toxicity to cyclosporin A, azathioprine, mitomicyn C and daunorubicin was assessed in Chang cells by monitoring their uptake of propidium iodide during a 3-h period. Chronic toxicity was assessed by monitoring the proliferation and viability of subconfluent cultures of Chang cells, human corneal endothelial cells (HCECs) and retinal pigmented epithelial (RPE) cells after continuous exposure to the drugs for 7 days. RESULTS: Acute toxicity testing revealed no obvious effects. However, the chronic toxicity tests disclosed a narrow concentration range over which cell proliferation decreased dramatically but calcein metabolism was sustained. Although the three lines reacted similarly to each agent, HCECs were the most vulnerable to daunorubicin and mitomycin. At a daunorubicin concentration of 0.05 microg/ml, a 75% decrease in calcein metabolism (P or = 95% cell loss (P < 0.001) were observed. At a mitomycin concentration of 0.01 mug/ml, cell density decreased by 61% (P < 0.001) without a change in calcein metabolism, but at 0.1 microg/ml, the latter parameter decreased to 12% (P = 0.00014). At this concentration the proliferation of Chang and RPE cells decreased by more than 50%, whilst calcein metabolism was largely sustained. Cyclosporin inhibited cell proliferation moderately at lower concentrations (< 5 microg/ml; P=0.05) and substantially at higher ones, with a corresponding decline in calcein metabolism. Azathioprine induced a profound decrease in both parameters at concentrations above 5 microg/ml. CONCLUSION: Daunorubicin, cyclosporin and azathioprine could be used to inhibit excessive intraocular scarring after glaucoma and vitreoretinal surgery without overly reducing cell viability. The attributes of immunosuppressants lie in their combined antiproliferative and immunomodulatory effects
