    Early Career ESOL Teachers’ Practical Knowledge of Teaching Speaking

    This thesis presents the findings of qualitative multiple-case study research investigating ESOL teachers’ practical knowledge of teaching speaking. Although there has been increased recognition of the value of practical knowledge research in recent years, such research remains extremely limited and the practical knowledge and teachers in an ESOL context and in the curricular domain of teaching speaking skills were previously unexplored areas. The four research participants were all early career ESOL teachers in the United Kingdom. Classroom observation data and interview data were generated at multiple points over the course of an academic year. This methodological approach introduced a longitudinal dimension to the research enabling any possible practical knowledge growth to be investigated. The research identified the largely contemporary nature of the ESOL teachers’ practices in teaching speaking. However, the teachers’ practical knowledge was identified as being atheoretical: teachers did not refer to public theory in the explanations of their practices. Instead, the findings suggest that teachers may experience a process of socialisation (both institutional and sectorial) through which many practices are adopted without a theoretical basis. A significant degree of commonality was identified amongst the teachers’ practical knowledge. Individual differences appeared to be significant, however, and were identifiable both in teachers’ practices and the beliefs underlying them. Teachers’ exercising of significant agency in their practices meant that these differences were evident despite certain sectorial pressure on teachers, particularly through exam washback. There was very limited evidence of growth in the teachers’ practical knowledge of teaching speaking. The research indicated a number of factors which appeared to inhibit such growth. The study discusses the implications of these findings for ESOL teacher development programmes. Recommendations for teacher development programmes include constructivist approaches to teacher engagement with public theory and institutional mechanisms for a sharing of practices amongst teachers

    Biofilm-specific extracellular matrix proteins of nontypeable Haemophilus influenzae

    Nontypeable Haemophilus influenzae (NTHi), a human respiratory tract pathogen, can form colony biofilms in vitro. Bacterial cells and the amorphous extracellular matrix (ECM) constituting the biofilm can be separated using sonication. The ECM from 24- and 96-h NTHi biofilms contained polysaccharides and proteinaceous components as detected by nuclear magnetic resonance (NMR) and Fourier transform infrared spectroscopy (FTIR) spectroscopy. More conventional chemical assays on the biofilm ECM confirmed the presence of these components and also DNA. Proteomics revealed eighteen proteins present in biofilm ECM that were not detected in planktonic bacteria. One ECM protein was unique to 24-h biofilms, two were found only in 96-h biofilms, and fifteen were present in the ECM of both 24- and 96-h NTHi biofilms. All proteins identified were either associated with bacterial membranes or cytoplasmic proteins. Immunocytochemistry showed two of the identified proteins, a DNA-directed RNA polymerase and the outer membrane protein OMP P2, associated with bacteria and biofilm ECM. Identification of biofilm-specific proteins present in immature biofilms is an important step in understanding the in vitro process of NTHi biofilm formation. The presence of a cytoplasmic protein and a membrane protein in the biofilm ECM of immature NTHi biofilms suggests that bacterial cell lysis may be a feature of early biofilm formation

    Understanding power, social capital and trust alongside near real-time water quality monitoring and technological development collaboration

    We report on qualitative social research conducted with stakeholders in a local agricultural knowledge and advice network associated with a collaborative water quality monitoring project. These farmers, advisors and researchers allude to existing social dynamics, technological developments, and (more general) social evolution which is analysed against a novel analytical framework. This framework considers notions of power, social capital, and trust as related and dynamic, forming the basis of our contribution to knowledge. We then probe the data to understand perceived impacts of the collaborative project and social interaction associated with this research project, which involved cutting edge automated and frequent water quality monitoring that allowed for near real-time access to data visualisation displayed via a bespoke mobile or web ‘app’ (1622WQ). Our findings indicate that a multi-faceted approach to assessing and intervening based on consideration of multiple social dimensions holds promise in terms of creating conditions that allow for individual and group learning to encourage changes in thinking required to result in improved land management practice

    Metabolic molecular markers of the tidal clock in the marine crustacean Eurydice pulchra

    SummaryIn contrast to the well mapped molecular orchestration of circadian timekeeping in terrestrial organisms, the mechanisms that direct tidal and lunar rhythms in marine species are entirely unknown. Using a combination of biochemical and molecular approaches we have identified a series of metabolic markers of the tidal clock of the intertidal isopod Eurydice pulchra. Specifically, we show that the overoxidation of peroxiredoxin (PRX), a conserved marker of circadian timekeeping in terrestrial eukaryotes [1], follows a circatidal (approximately 12.4 hours) pattern in E. pulchra, in register with the tidal pattern of swimming. In parallel, we show that mitochondrially encoded genes are expressed with a circatidal rhythm. Together, these findings demonstrate that PRX overoxidation rhythms are not intrinsically circadian; rather they appear to resonate with the dominant metabolic cycle of an organism, regardless of its frequency. Moreover, they provide the first molecular leads for dissecting the tidal clockwork

    Climate change alterations to ecosystem dominance: how might sponge-dominated reefs function?

    Anthropogenic stressors are impacting ecological systems across the world. Of particular concern are the recent rapid changes occurring in coral reef systems. With ongoing degradation from both local and global stressors, future reefs are likely to function differently to current coral-dominated ecosystems. Determining key attributes of future reef states is critical to reliably predict outcomes for ecosystem service provision. Here we explore the impacts of changing sponge dominance on coral reefs. Qualitative modelling of reef futures suggests that changing sponge dominance due to increased sponge abundance will have different outcomes for other trophic levels compared with increased sponge dominance as a result of declining coral abundance. By exploring uncertainty in the model outcomes we identify the need to: i) quantify changes in carbon flow through sponges, ii) determine the importance of food limitation for sponges, iii) assess the ubiquity of the recently described 'sponge loop', iv) determine the competitive relationships between sponges and other benthic taxa, particularly algae, and v) understand how changing dominance of other organisms alters trophic pathways and energy flows through ecosystems. Addressing these knowledge gaps will facilitate development of more complex models that assess functional attributes of sponge-dominated reef ecosystems. This article is protected by copyright.     Sandcastle: software for revealing latent information in multiple experimental ChIP-chip datasets via a novel normalisation procedure

    ChIP-chip is a microarray based technology for determining the genomic locations of chromatin bound factors of interest, such as proteins. Standard ChIP-chip analyses employ peak detection methodologies to generate lists of genomic binding sites. No previously published method exists to enable comparative analyses of enrichment levels derived from datasets examining different experimental conditions. This restricts the use of the technology to binary comparisons of presence or absence of features between datasets. Here we present the R package Sandcastle — Software for the Analysis and Normalisation of Data from ChIP-chip AssayS of Two or more Linked Experiments — which allows for comparative analyses of data from multiple experiments by normalising all datasets to a common background. Relative changes in binding levels between experimental datasets can thus be determined, enabling the extraction of latent information from ChIP-chip experiments. Novel enrichment detection and peak calling algorithms are also presented, with a range of graphical tools, which facilitate these analyses.     3D-DIP-Chip: a microarray-based method to measure genomic DNA damage

    Genotoxins cause DNA damage, which can result in genomic instability. The genetic changes induced have far-reaching consequences, often leading to diseases such as cancer. A wide range of genotoxins exists, including radiations and chemicals found naturally in the environment, and in man-made forms created by human activity across a variety of industries. Genomic technologies offer the possibility of unravelling the mechanisms of genotoxicity, including the repair of genetic damage, enhancing our ability to develop, test and safely use existing and novel materials. We have developed 3D-DIP-Chip, a microarray-based method to measure the prevalence of genomic genotoxin-induced DNA damage. We demonstrate the measurement of both physical and chemical induced DNA damage spectra, integrating the analysis of these with the associated changes in histone acetylation induced in the epigenome.     DREAMS-START (Dementia RElAted Manual for Sleep; STrAtegies for RelaTives) for people with dementia and sleep disturbances: a single-blind feasibility and acceptability randomized controlled trial

    Background: 40% of people with dementia have disturbed sleep but there are currently no known effective treatments. Studies of sleep hygiene and light therapy have not been powered to indicate feasibility and acceptability and have shown 40–50% retention. We tested the feasibility and acceptability of a six-session manualized evidence-based non-pharmacological therapy; Dementia RElAted Manual for Sleep; STrAtegies for RelaTives (DREAMS-START) for sleep disturbance in people with dementia. Methods: We conducted a parallel, two-armed, single-blind randomized trial and randomized 2:1 to intervention: Treatment as Usual. Eligible participants had dementia and sleep disturbances (scoring ≥4 on one Sleep Disorders Inventory item) and a family carer and were recruited from two London memory services and Join Dementia Research. Participants wore an actiwatch for two weeks pre-randomization. Trained, clinically supervised psychology graduates delivered DREAMS-START to carers randomized to intervention; covering Understanding sleep and dementia; Making a plan (incorporating actiwatch information, light exposure using a light box); Daytime activity and routine; Difficult night-time behaviors; Taking care of your own (carer's) sleep; and What works? Strategies for the future. Carers kept their manual, light box, and relaxation recordings post-intervention. Outcome assessment was masked to allocation. The co-primary outcomes were feasibility (≥50% eligible people consenting to the study) and acceptability (≥75% of intervention group attending ≥4 intervention sessions). Results: In total, 63out of 95 (66%; 95% CI: 56–76%) eligible referrals consented between 04/08/2016 and 24/03/2017; 62 (65%; 95% CI: 55–75%) were randomized, and 37 out of 42 (88%; 95% CI: 75–96%) adhered to the intervention. Conclusions: DREAM-START for sleep disorders in dementia is feasible and acceptable

    Four regioisomeric azulenes bearing pentafluorosulfanylphenyl substituents have been prepared and characterised by various spectroscopic techniques.
