    Metabotropic glutamate receptors (mGluRs) include receptors sensitive to L-2-amino-4-phosphonobutyrate (L-AP4) and 1S,3R-1-aminocyclopentane-1,3-dicarboxylic acid (1S,3R-ACPD). To determine whether 1S,3R-ACPD is an agonist at retinal L-AP4 receptors, whole cell voltage clamp recordings were obtained from mudpuppy ON bipolar cells in a superfused retinal slice and L-AP4 and 1S,3R-ACPD were bath applied. Both compounds evoked similar outward currents which reversed near 0 mV and were accompanied by an increased input resistance. Responses to both agonists washed out in parallel suggesting they act through the same second messenger pathway(s). Inhibitors of cGMP-PDE activity suppressed responses to both L-AP4 and 1SR,3RS-ACPD, suggesting that both compounds activate cGMP-PDE. Responses to 1S,3R-ACPD were occluded by prior activation of L-AP4 receptors, but not blocked by the non-AP4, mGluR antagonists, L-aminophosphonopropionic acid (L-AP3) or 4-carboxy-3-hydroxyphenylglycine (4C3H-PG). These results indicate that 1S,3R-ACPD is an agonist at L-AP4 receptors. 1S,3S-ACPD and 4C3H-PG evoked outward currents similar to L-AP4 suggesting they may also be L-AP4 receptor agonists. Using the b-wave of the ERG as an assay for ON bipolar cell responses, concentration/response curves were obtained for ACPD enantiomers. The rank-order potency of ACPD enantiomers at L-AP4 receptors in ON bipolar cells is similar to their rank-order potency at non-AP4, mGluRs in brain which suggests that the receptors possess similar binding sites and may be members of a common receptor family

    Removal of extracellular chloride suppresses transmitter release from photoreceptor terminals in the mudpuppy retina.

    Removal of extracellular Cl- has been shown to suppress light-evoked voltage responses of ON bipolar and horizontal cells, but not photoreceptors or OFF bipolar cells, in the amphibian retina. A substantial amount of experimental evidence has demonstrated that the photoreceptor transmitter, L-glutamate, activates cation, not Cl-, channels in these cells. The mechanism for Cl-free effects was therefore reexamined in a superfused retinal slice preparation from the mudpuppy (Necturus maculosus) using whole-cell voltage and current clamp techniques. In a Cl-free medium, light-evoked currents were maintained in rod and cone photoreceptors but suppressed in horizontal, ON bipolar, and OFF bipolar cells. Changes in input resistance and dark current in bipolar and horizontal cells were consistent with the hypothesis that removal of Cl- suppresses tonic glutamate release from photoreceptors. The persistence of light-evoked voltage responses in OFF bipolar cells, despite the suppression of light-evoked currents, is due to a compensatory increase in input resistance. Focal application of hyperosmotic sucrose to photoreceptor terminals produced currents in bipolar and horizontal cells arising from two sources: (a) evoked glutamate release and (b) direct actions of the hyperosmotic solution on postsynaptic neurons. The inward currents resulting from osmotically evoked release of glutamate in OFF bipolar and horizontal cells were suppressed in a Cl-free medium. For ON bipolar cells, both the direct and evoked components of the hyperosmotic response resulted in outward currents and were thus difficult to separate. However, in some cells, removal of extracellular Cl- suppressed the outward current consistent with a suppression of presynaptic glutamate release. The results of this study suggest that removal of extracellular Cl- suppresses glutamate release from photoreceptor terminals. Thus, it is possible that control of [Cl-] in and around photoreceptors may regulate glutamate release from these cells

    A spatial rainfall model was applied to radar data of air mass thunderstorms to yield a rainstorm representation as a set of convective rain cells. The modeled rainfall was used as input into hydrological model, instead of the standard radar-grid data. This approach allows a comprehensive linkage between runoff responses and rainfall structures.     Our knowledge of the function of the auditory nervous system is based upon a wealth of data obtained, for the most part, in anaesthetised animals. More recently, it has been generally acknowledged that factors such as attention profoundly modulate the activity of sensory systems and this can take place at many levels of processing. Imaging studies, in particular, have revealed the greater activation of auditory areas and areas outside of sensory processing areas when attending to a stimulus. We present here a brief review of the consequences of such non-passive listening and go on to describe some of the experiments we are conducting to investigate them. In imaging studies, using fMRI, we can demonstrate the activation of attention networks that are non-specific to the sensory modality as well as greater and different activation of the areas of the supra-temporal plane that includes primary and secondary auditory areas. The profuse descending connections of the auditory system seem likely to be part of the mechanisms subserving attention to sound. These are generally thought to be largely inactivated by anaesthesia. However, we have been able to demonstrate that even in an anaesthetised preparation, removing the descending control from the cortex leads to quite profound changes in the temporal patterns of activation by sounds in thalamus and inferior colliculus. Some of these effects seem to be specific to the ear of stimulation and affect interaural processing. To bridge these observations we are developing an awake behaving preparation involving freely moving animals in which it will be possible to investigate the effects of consciousness (by contrasting awake and anaesthetized), passive and active listening

    Teacher learning communities and educational change in Scotland: the Highland experience

    This article discusses the issue of the sustainability of educational change, in the light of findings from research undertaken in tandem with a development project initiated by a Scottish Education Authority, The Highland Council. The project aimed to promote self and peer assessment practices, as well as other participative pedagogies associated with Scotland’s new Curriculum for Excellence, in secondary schools. The article reviews some of the key themes that have emerged from recent literature on educational change, before drawing on the project data to address two key issues: the factors that have helped to promote and sustain changes within the schools; and the barriers to innovation experienced in these schools. We conclude the article by identifying a range of considerations that should be taken into account by those seeking to innovate, and we suggest that, while the Highland model for change has enjoyed a degree of success in inculcating change, more needs to be done to address systemic issues, such as the pervasive influence of a narrow attainment agenda in shaping classroom practice

    Limit-feeding a high-concentrate diet may alter nutrient absorption

    Feeding newly arrived cattle is commonly characterized by a few days of feeding longstemmed hay followed by a series of step-up diets, wherein concentrate levels are increased to promote ruminal adaptation to a highconcentrate finishing diet. This is done to give the rumen microbes time to adjust to larger amounts of readily fermentable starches in cereal grains. Rumen epithelial adaptation may be achievable by limit-feeding a finishing diet, with gradual increases in feed intake, until the cattle are on full feed. If this can be achieved without causing ruminal disorders and days off feed, then the cost of feeding cattle can be reduced. By limit-feeding, the higher roughage step-up diets are replaced with a single high-concentrate diet. The cost of grain is less than that of roughage, and there are decreased costs in terms of storage space, waste disposal (due to decreased manure production), and mixing and hauling of rations. The purpose of this experiment was to examine the effects of a traditional step-up program versus a limit-fed finishing diet in terms of dry matter intake, acetate to propionate ratio, and ruminal dilution rate. Diet effects on volatile fatty acid concentration and absorption were also examined by using valerate as a marker for volatile fatty acid absorption

    Geologic context of geodetic data across a Basin and Range normal fault, Crescent Valley, Nevada

    Geodetic strain and late Quaternary faulting in the Basin and Range province is distributed over a region much wider than historic seismicity, which is localized near the margins of the province. In the relatively aseismic interior, both the magnitude and direction of geodetic strain may be inconsistent with the Holocene faulting record. We document the best example of such a disagreement across the NE striking, ~55Β° NW dipping Crescent normal fault, where a NW oriented, 70 km geodetic baseline records contemporary shortening of ~2 mm/yr orthogonal to the fault trace. In contrast, our geomorphic, paleoseismic, and geochronologic analyses of the Crescent fault suggest that a large extensional rupture occurred during the late Holocene epoch. An excavation across the fault at Fourmile Canyon reveals that the most recent event occurred at 2.8 Β± 0.1 ka, with net vertical tectonic displacement of 4.6 Β± 0.4 m at this location, corresponding to the release of ~3 m of accumulated NW-SE extension. Measured alluvial scarp profiles suggest a minimum rupture length of 30 km along the range front for the event, implying a moment magnitude M_w of at least 6.6. No prior event occurred between ~2.8 ka and ~6.4 Β± 0.1 ka, the ^(14)C calender age of strata near the base of the exposed section. Assuming typical slip rates for Basin and Range faults (~0.3 mm/yr), these results imply that up to one third, or ~1 m, of the extensional strain released in the previous earthquake could have reaccumulated across the fault since ~2.8 ka. However, the contemporary shortening implies that the fault is unloading due to a transient process, whose duration is limited to between 6 years (geodetic recording time) and 2.8 ka (the age of the most recent event). These results emphasize the importance of providing accurate geologic data on the timescale of the earthquake cycle in order to evaluate geodetic measurements

    Sleep duration and incidence of obesity in infants, children and adolescents : a systematic review and meta-analysis of prospective studies

    Study Objective: To assess the prospective relationship between sleep and obesity in a paediatric population. Methods: We performed a systematic search using PubMed, Embase, Web of Science and Cochrane (up to 25th September 2017). Included studies were prospective, had follow-up >1 year, had duration of sleep at baseline, and measures of incidence of overweight or obesity and/or changes in body mass index (BMI) z-score and BMI during follow-up. We extracted relative risks or changes in BMI z-score or BMI and 95% confidence intervals (CI) and pooled them using a random effect model. Results: Forty-two studies were included but, as there was significant heterogeneity, results are presented by age strata. Short sleep was associated with a greater risk of developing overweight or obesity in infancy (7 Studies, 14 738 participants, RR: 1.40; 95% CI 1.19 to 1.65; p<0.001), early childhood (8 Studies, 31 104 participants, RR: 1.57; 1.40 to 1.76; p<0.001), middle childhood (3 studies, 3 005 participants, RR: 2.23; 2.18 to 2.27; p<0.001) and adolescence (3 studies, 26 652 participants, RR: 1.30; 1.11 to 1.53; p<0.002). Sleep duration was also associated with a significant change in BMI z-score (14 studies, 18 cohorts, 31 665 participants) (mean difference -0.03; -0.04 to -0.01 per h sleep; P=0.001) and in BMI (16 studies, 24 cohorts, 24 894 participants) (mean difference -0.03 kg/m2; -0.04 to -0.01 for every h of increase in sleep; P=0.001). Conclusions: Short sleep duration is a risk factor or marker of the development of obesity in infants, children and adolescents
