Succinate Dehydrogenase Is a Direct Target of Sirtuin 3 Deacetylase Activity
BACKGROUND: Sirtuins (SIRT1-7) are a family of NAD-dependent deacetylases and/or ADP-ribosyltransferases that are involved in metabolism, stress responses and longevity. SIRT3 is localized to mitochondria, where it deacetylates and activates a number of enzymes involved in fuel oxidation and energy production. METHODOLOGY/PRINCIPAL FINDINGS: In this study, we performed a proteomic screen to identify SIRT3 interacting proteins and identified several subunits of complex II and V of the electron transport chain. Two subunits of complex II (also known as succinate dehydrogenase, or SDH), SDHA and SDHB, interacted specifically with SIRT3. Using mass spectrometry, we identified 13 acetylation sites on SDHA, including six novel acetylated residues. SDHA is hyperacetylated in SIRT3 KO mice and SIRT3 directly deacetylates SDHA in a NAD-dependent manner. Finally, we found that SIRT3 regulates SDH activity both in cells and in murine brown adipose tissue. CONCLUSIONS/SIGNIFICANCE: Our study identifies SDHA as a binding partner and substrate for SIRT3 deacetylase activity. SIRT3 loss results in decreased SDH enzyme activity, suggesting that SIRT3 may be an important physiological regulator of SDH activity

Language support for immigrant children: a study of state schools in the UK and US
In recent decades, immigrants, refugees and asylum seekers have sought a new way of life in large numbers, often leaving their countries of origin behind in search of places that offer a better way of life. The purpose of this study was to investigate how elementary and middle school students in state schools in Reading, England (primarily speakers of Asian languages), and Richmond, Virginia (primarily speakers of Spanish), were supported academically, when most children’s first language was not English. The authors were interested in exploring whether or not there were cultural or structural differences in the way each country helped or hindered these students as they progressed through the school systems. Three UK schools in a district of approximately 100,000 and three US schools in a district of approximately 250,000 were the focus of this exploration from 2000 to 2003. Findings indicated that there were cultural and legislative differences and similarities. Teachers and administrators in both countries attempted to provide services with limited and sometimes diminishing resources. Community support varied based on resources, attitudes toward various ethnic groups, and the coping strategies adopted by these groups in their new environments. Marked differences appeared with regard to the manner in which assessments took place and how the results were made available to the public
The Relativistic N-body Problem in a Separable Two-Body Basis
We use Dirac's constraint dynamics to obtain a Hamiltonian formulation of the
relativistic N-body problem in a separable two-body basis in which the
particles interact pair-wise through scalar and vector interactions. The
resultant N-body Hamiltonian is relativistically covariant. It can be easily
separated in terms of the center-of-mass and the relative motion of any
two-body subsystem. It can also be separated into an unperturbed Hamiltonian
with a residual interaction. In a system of two-body composite particles, the
solutions of the unperturbed Hamiltonian are relativistic two-body internal
states, each of which can be obtained by solving a relativistic
Schr\"odinger-like equation. The resultant two-body wave functions can be used
as basis states to evaluate reaction matrix elements in the general N-body
problem. We prove a relativistic version of the post-prior equivalence which
guarantees a unique evaluation of the reaction matrix element, independent of
the ways of separating the Hamiltonian into unperturbed and residual
interactions. Since an arbitrary reaction matrix element involves composite
particles in motion, we show explicitly how such matrix elements can be
evaluated in terms of the wave functions of the composite particles and the

Instances and connectors : issues for a second generation process language
This work is supported by UK EPSRC grants GR/L34433 and GR/L32699Over the past decade a variety of process languages have been defined, used and evaluated. It is now possible to consider second generation languages based on this experience. Rather than develop a second generation wish list this position paper explores two issues: instances and connectors. Instances relate to the relationship between a process model as a description and the, possibly multiple, enacting instances which are created from it. Connectors refers to the issue of concurrency control and achieving a higher level of abstraction in how parts of a model interact. We believe that these issues are key to developing systems which can effectively support business processes, and that they have not received sufficient attention within the process modelling community. Through exploring these issues we also illustrate our approach to designing a second generation process language.Postprin
A small world of citations? The influence of collaboration networks on citation practices
This paper examines the proximity of authors to those they cite using degrees
of separation in a co-author network, essentially using collaboration networks
to expand on the notion of self-citations. While the proportion of direct
self-citations (including co-authors of both citing and cited papers) is
relatively constant in time and across specialties in the natural sciences (10%
of citations) and the social sciences (20%), the same cannot be said for
citations to authors who are members of the co-author network. Differences
between fields and trends over time lie not only in the degree of co-authorship
which defines the large-scale topology of the collaboration network, but also
in the referencing practices within a given discipline, computed by defining a
propensity to cite at a given distance within the collaboration network.
Overall, there is little tendency to cite those nearby in the collaboration
network, excluding direct self-citations. By analyzing these social references,
we characterize the social capital of local collaboration networks in terms of
the knowledge production within scientific fields. These results have
implications for the long-standing debate over biases common to most types of
citation analysis, and for understanding citation practices across scientific
disciplines over the past 50 years. In addition, our findings have important
practical implications for the availability of 'arm's length' expert reviewers
of grant applications and manuscripts

Individual common variants exert weak effects on the risk for autism spectrum disorders.
While it is apparent that rare variation can play an important role in the genetic architecture of autism spectrum disorders (ASDs), the contribution of common variation to the risk of developing ASD is less clear. To produce a more comprehensive picture, we report Stage 2 of the Autism Genome Project genome-wide association study, adding 1301 ASD families and bringing the total to 2705 families analysed (Stages 1 and 2). In addition to evaluating the association of individual single nucleotide polymorphisms (SNPs), we also sought evidence that common variants, en masse, might affect the risk. Despite genotyping over a million SNPs covering the genome, no single SNP shows significant association with ASD or selected phenotypes at a genome-wide level. The SNP that achieves the smallest P-value from secondary analyses is rs1718101. It falls in CNTNAP2, a gene previously implicated in susceptibility for ASD. This SNP also shows modest association with age of word/phrase acquisition in ASD subjects, of interest because features of language development are also associated with other variation in CNTNAP2. In contrast, allele scores derived from the transmission of common alleles to Stage 1 cases significantly predict case status in the independent Stage 2 sample. Despite being significant, the variance explained by these allele scores was small (Vm< 1%). Based on results from individual SNPs and their en masse effect on risk, as inferred from the allele score results, it is reasonable to conclude that common variants affect the risk for ASD but their individual effects are modest
An assessment of multimodal imaging of subsurface text in mummy cartonnage using surrogate papyrus phantoms
Ancient Egyptian mummies were often covered with an outer casing, panels and masks made from cartonnage: a lightweight material made from linen, plaster, and recycled papyrus held together with adhesive. Egyptologists, papyrologists, and historians aim to recover and read extant text on the papyrus contained within cartonnage layers, but some methods, such as dissolving mummy casings, are destructive. The use of an advanced range of different imaging modalities was investigated to test the feasibility of non-destructive approaches applied to multi-layered papyrus found in ancient Egyptian mummy cartonnage. Eight different techniques were compared by imaging four synthetic phantoms designed to provide robust, well-understood, yet relevant sample standards using modern papyrus and replica inks. The techniques include optical (multispectral imaging with reflection and transillumination, and optical coherence tomography), X-ray (X-ray fluorescence imaging, X-ray fluorescence spectroscopy, X-ray micro computed tomography and phase contrast X-ray) and terahertz-based approaches. Optical imaging techniques were able to detect inks on all four phantoms, but were unable to significantly penetrate papyrus. X-ray-based techniques were sensitive to iron-based inks with excellent penetration but were not able to detect carbon-based inks. However, using terahertz imaging, it was possible to detect carbon-based inks with good penetration but with less sensitivity to iron-based inks. The phantoms allowed reliable and repeatable tests to be made at multiple sites on three continents. The tests demonstrated that each imaging modality needs to be optimised for this particular application: it is, in general, not sufficient to repurpose an existing device without modification. Furthermore, it is likely that no single imaging technique will to be able to robustly detect and enable the reading of text within ancient Egyptian mummy cartonnage. However, by carefully selecting, optimising and combining techniques, text contained within these fragile and rare artefacts may eventually be open to non-destructive imaging, identification, and interpretation
