    A Mixed Methods Approach to Identifying Administration Issues Pertinent in Interscholastic Sports

    The purpose of this study was to investigate three propositions: a) What are the administration issues most pertinent to interscholastic sport today, as well as the next five years?, b) How important are those administration issues to athletic administrators?, and c) What are the potential implications of those pertinent administration issues to practicing athletic administrators? The literature provides a general overview of relevant issues surrounding interscholastic athletics. However, the importance and implications of relevant issues to practicing high school athletic administrators are difficult to discern. To answer the first proposition, the Minnesota State High School League (MSHSL) provided 10 contemporary administration issues that were most pertinent to interscholastic sport today, as well as the next five years. To answer the second proposition, a Likert-Scale was created so that practicing athletic administrators could rate each issue on a scale of 5 = extremely important to 1 = very little importance. A national study was conducted with athletic directors from the National Interscholastic Athletic Administrators Association (N = 170) annual conference. A one-tailed ANOVA was executed to determine significant differences among the 10 administration issues identified by the MSHSL. Four issues were found to be significant; Athletic Facilities, Athletic Training, Health Issues and Travel Teams. A Games-Howell post hoc was executed to determine significant differences across geographical regions of the United States. For the third proposition, semi-structured interviews were completed to provide insight on the implications for practicing athletic administrators. The results offer insight from which further investigations could be conducted to continue building on policies that influence interscholastic athletic administrators’ day-to-day accountability when overseeing their athletic programs

    Nematode community development early in ecological restoration: The role of organic amendments

    a b s t r a c t Soil food web structure is an integral component of ecosystem function, but there are few strategies orientated towards managing its development in restoration projects. The objective of this study was to direct nematode community structure and function through the application of organic amendments to the soil of an urban landfill remediation project using native grassland vegetation. We used a 2 Â 3 factorial design in which an organic amendment was added to the soil at different locations (incorporated versus surface-applied) and amounts (none, light, heavy). Nematode and plant community structure were monitored over three growing seasons to determine the rate and direction of change. Surface application of organic amendments supported greater grass and total plant densities compared to incorporated amendment treatments, but plant density did not vary with amendment amount. Total nematode density, family diversity and family richness were not affected by the amendment treatments, but both family richness and seasonal nematode density increased over the duration of the experiment. Other descriptors of nematode community development (Structure, Maturity, and Plant Parasite Indexes) were not influenced by either amendment amount or location, but varied significantly over time. Contrary to expectations, the surface amendment treatments significantly increased bacterivorous, plant parasitic, omnivorous and predator nematode densities, but had no influence on fungi/root-tip feeding nematodes. Also contrary to our hypotheses, the surface treatments had smaller Channel Index and greater Enrichment Index values relative to the incorporated treatments during the first 15 month of the experiment. We hypothesize that the surface amendments are indirectly affecting the structure of the nematode community by promoting greater plant density, thus increasing the concentration of highquality organic matter (such as root exudates) in the soil. This promotes the development of a nematode community dominated by opportunistic groups that respond rapidly to increased resource availability. Future studies should aim to distinguish between the organic amendment's direct function as a potential food source for the soil biota versus their indirect role as an environmental variable, including their capability to alter the availability of plant-derived resources

    Neoadjuvant pazopanib and molecular analysis of tissue response in renal cell carcinoma

    BACKGROUND. Surgery remains the frontline therapy for patients with localized clear cell renal cell carcinoma (ccRCC); however, 20%–40% recur. Angiogenesis inhibitors have improved survival in metastatic patients and may result in responses in the neoadjuvant setting. The impact of these agents on the tumor genetic heterogeneity or the immune milieu is largely unknown. This phase II study was designed to evaluate safety, response, and effect on tumor tissue of neoadjuvant pazopanib. METHODS. ccRCC patients with localized disease received pazopanib (800 mg daily; median 8 weeks), followed by nephrectomy. Five tumors were examined for mutations by whole exome sequencing from samples collected before therapy and at nephrectomy. These samples underwent RNA sequencing; 17 samples were available for posttreatment assessment. RESULTS. Twenty-one patients were enrolled. The overall response rate was 8 of 21 (38%). No patients with progressive disease. At 1-year, response-free survival and overall survival was 83% and 89%, respectively. The most frequent grade 3 toxicity was hypertension (33%, 7 of 21). Sequencing revealed strong concordance between pre- and posttreatment samples within individual tumors, suggesting tumors harbor stable core profiles. However, a reduction in private mutations followed treatment, suggesting a selective process favoring enrichment of driver mutations. CONCLUSION. Neoadjuvant pazopanib is safe and active in ccRCC. Future genomic analyses may enable the segregation of driver and passenger mutations. Furthermore, tumor infiltrating immune cells persist during therapy, suggesting that pazopanib can be combined with immune checkpoint inhibitors without dampening the immune response. FUNDING.     Prospective Study of TMVR Using Balloon-Expandable Aortic Transcatheter Valves in MAC: MITRAL Trial 1-Year Outcomes

    OBJECTIVES: The aim of this study was to evaluate 1-year outcomes of valve-in-mitral annular calcification (ViMAC) in the MITRAL (Mitral Implantation of Transcatheter Valves) trial. BACKGROUND: The MITRAL trial is the first prospective study evaluating the feasibility of ViMAC using balloon-expandable aortic transcatheter heart valves. METHODS: A multicenter prospective study was conducted, enrolling high-risk surgical patients with severe mitral annular calcification and symptomatic severe mitral valve dysfunction at 13 U.S. sites. RESULTS: Between February 2015 and December 2017, 31 patients were enrolled (median age 74.5 years [interquartile range (IQR): 71.3 to 81.0 years], 71% women, median Society of Thoracic Surgeons score 6.3% [IQR: 5.0% to 8.8%], 87.1% in New York Heart Association functional class III or IV). Access was transatrial (48.4%), transseptal (48.4%), or transapical (3.2%). Technical success was 74.2%. Left ventricular outflow tract obstruction (LVOTO) with hemodynamic compromise occurred in 3 patients (transatrial, n = 1; transseptal, n = 1; transapical, n = 1). After LVOTO occurred in the first 2 patients, pre-emptive alcohol septal ablation was implemented to decrease risk in high-risk patients. No intraprocedural deaths or conversions to open heart surgery occurred during the index procedures. All-cause mortality at 30 days was 16.7% (transatrial, 21.4%; transseptal, 6.7%; transapical, 100% [n = 1]; p = 0.33) and at 1 year was 34.5% (transatrial, 38.5%; transseptal, 26.7%; p = 0.69). At 1-year follow-up, 83.3% of patients were in New York Heart Association functional class I or II, the median mean mitral valve gradient was 6.1 mm Hg (IQR: 5.6 to 7.1 mm Hg), and all patients had ≤1+ mitral regurgitation. CONCLUSIONS: At 1 year, ViMAC was associated with symptom improvement and stable transcatheter heart valve performance. Pre-emptive alcohol septal ablation may prevent transcatheter mitral valve replacement-induced LVOTO in patients at risk. Thirty-day mortality of patients treated via transseptal access was lower than predicted by the Society of Thoracic Surgeons score. Further studies are needed to evaluate safety and efficacy of ViMAC

    Transcatheter or surgical aortic-valve replacement in intermediate-risk patients

    BACKGROUND: Previous trials have shown that among high-risk patients with aortic stenosis, survival rates are similar with transcatheter aortic-valve replacement (TAVR) and surgical aorticvalve replacement. We evaluated the two procedures in a randomized trial involving intermediate-risk patients. METHODS: We randomly assigned 2032 intermediate-risk patients with severe aortic stenosis, at 57 centers, to undergo either TAVR or surgical replacement. The primary end point was death from any cause or disabling stroke at 2 years. The primary hypothesis was that TAVR would not be inferior to surgical replacement. Before randomization, patients were entered into one of two cohorts on the basis of clinical and imaging findings; 76.3% of the patients were included in the transfemoral-access cohort and 23.7% in the transthoracic-access cohort. RESULTS: The rate of death from any cause or disabling stroke was similar in the TAVR group and the surgery group (P=0.001 for noninferiority). At 2 years, the Kaplan–Meier event rates were 19.3% in the TAVR group and 21.1% in the surgery group (hazard ratio in the TAVR group, 0.89; 95% confidence interval [CI], 0.73 to 1.09; P=0.25). In the transfemoralaccess cohort, TAVR resulted in a lower rate of death or disabling stroke than surgery (hazard ratio, 0.79; 95% CI, 0.62 to 1.00; P=0.05), whereas in the transthoracic-access cohort, outcomes were similar in the two groups. TAVR resulted in larger aortic-valve areas than did surgery and also resulted in lower rates of acute kidney injury, severe bleeding, and new-onset atrial fibrillation; surgery resulted in fewer major vascular complications and less paravalvular aortic regurgitation. CONCLUSIONS: In intermediate-risk patients, TAVR was similar to surgical aortic-valve replacement with respect to the primary end point of death or disabling stroke.     Scholarship on Gender and Sport in Sex Roles and Beyond

    In this paper we critically review how research on girls or women and sport has developed over the last 35 years. We use a post-positivist lens to explore the content of the papers published in Sex Roles in the area of women, gender and sport and examine the shifts in how gender and sport have been conceptualized in these accounts. In order to initiate a broader dialogue about the scholarly analysis of gender and sport, we subsequently explore ideas inspired by feminist theorizing that have dominated/guided related research in other outlets over this time period but have received relatively little attention in papers published in Sex Roles. We conclude by briefly making suggestions for further research in this area

    Computational Prediction and Experimental Verification of New MAP Kinase Docking Sites and Substrates Including Gli Transcription Factors

    In order to fully understand protein kinase networks, new methods are needed to identify regulators and substrates of kinases, especially for weakly expressed proteins. Here we have developed a hybrid computational search algorithm that combines machine learning and expert knowledge to identify kinase docking sites, and used this algorithm to search the human genome for novel MAP kinase substrates and regulators focused on the JNK family of MAP kinases. Predictions were tested by peptide array followed by rigorous biochemical verification with in vitro binding and kinase assays on wild-type and mutant proteins. Using this procedure, we found new ‘D-site’ class docking sites in previously known JNK substrates (hnRNP-K, PPM1J/PP2Czeta), as well as new JNK-interacting proteins (MLL4, NEIL1). Finally, we identified new D-site-dependent MAPK substrates, including the hedgehog-regulated transcription factors Gli1 and Gli3, suggesting that a direct connection between MAP kinase and hedgehog signaling may occur at the level of these key regulators. These results demonstrate that a genome-wide search for MAP kinase docking sites can be used to find new docking sites and substrates
