    Reducing secondary losses by blowing cold air in a turbine

    Local blowing on the profile suction side of the turbine guide wheel blades can be effective in preventing the propagation of secondary flows that is, the transport of casing and hub boundary layers by pressure gradients. Some preliminary results on how the blowing should be accomplished in order to influence the secondary flows in the desired manner are given. The effectiveness of blowing is demonstrated. Blowing is also seen to be more effective than using boundary layer slots as far as diminishing losses in the rim zones is concerned

    Examining the Impact of a University-driven Exercise Programming Event on End-of-semester Stress in Students

    International Journal of Exercise Science 10(5): 754-763, 2017. Stress levels in university students peak during the final exam period. An inverse association exists between Physical Activity (PA) and poor mental health. UNLV has created Fitness4Finals (F4F), an event novel in its approach to academic stress reduction by incorporating both physical activity and mental relaxation. To our knowledge, a university-driven programming event aimed at reducing physiological and psychological stress among students approaching final exams had never been studied. Therefore, the aims of this research were to 1) examine the influence of F4F on physiological stress and perceived psychological stress (PPS) and 2) to examine the relationship between physiological stress and PPS. Fifteen full-time university students were recruited to participate in their choice of one of two groups: F4F or control (NonF4F). Pre-F4F and post-F4F measures of physiological stress, measured by salivary cortisol, and perceived psychological stress, measured by survey were collected. The F4F event was held the week prior to final examinations. Participants in the F4F group engaged in one F4F activity per day for the duration of the 3-day event. Results of the repeated measures MANOVA indicated nonsignificant interaction (p = .864) between F4F participation, physiological stress and PPS. PPS and cortisol were not correlated at the onset of the study (r = -0.18, p = 0.48) or at the last sampling period (r = 0.097, p = 0.73). Preemptive elevated levels of PA in the F4F group may have influenced results. Qualitative data indicates a unanimous perceived reduction in stress from F4F participation. While the physiological measures of stress in the present study were not significantly different, the perceived stress reduction reported by F4F participants is influential. Further investigation with improvements in timing and measurement tools is warranted

    Spatial and temporal pattern of development and decline of the spring diatom populations in Lake Stechlin in 1999

    Integrated samples from the euphotic layer (mostly 0-25 m) were taken between January 1994 and December 2000 in the open water of Lake Stechlin. Monthly sampling was carried out in January-April 1999 and bi-weekly in May-June 1999 at depths of 0, 10, 20, 30, 40, 50 and 60 m. Phytoplankton, with special attention to diatoms, was qualitatively and quantitatively investigated. These data, together with water chemistry and zooplankton monitoring information, were used to describe spring development and decline of the phytoplankton. Results were compared to predictions of the PEG-model. i) The large variety in timing and species composition of early-season diatom growth allows one to conclude that in lakes, which can be either mono- or dimictic, it is primaliry not the trophic state that determines spring growth/composition, rather it is the presence, absence or duration of ice-cover and weather conditions during the onset of the thermocline. ii) Physical stresses (low temperatures, low light, complete turbulent mixing) upon phytoplankton in such lakes has been less severe than growth limitation set by scarcity of nutrients. Diatom growth starts soon after the complete autumnal overturn. Thermocline development, instead if initation of the spring bloom as stated in the PEG model, terminates growth since diatoms, cells sink rapidly into the hypolimnion. iii) „Life time” of unicellular planktonic centric diatoms was the longest int he stage when they turn to decaying or dormant cells. Empty frustules dissolve rather quickly. Frustules of planktonic pennatae diatoms were apparently more persistent than that if centrics. iv) Because of their low abundance, metazooplankton cannot play a significant role in the elimination of spring the phytoplankton crop. v) After the onset of thermocline, rejuvenating diatoms can be found in small numbers throughout the hypolimnion. This study demonstrates that one should be careful with using the term „vegetation period” since the bulk of growth of planktonic primary producers might occur during the isothermal period prior to onset of the thermocline. Moreover, driving forces during this period are largely determined by climatic factors or actual weather, such as the duration of the ice-cover or the onset time of the thermocline

    Systematic Study on the Fluid Dynamical Behaviour of Streamwise and Laterally Inclined Jets in Crossflow

    ABSTRACT The design of discrete film cooling holes for gas turbine airfoil applications is governed by a number of parameters influencing both their aerodynamic and thermal behaviour. This numerical and experimental study focuses on the marked differences between film cooling holes with combined streamwise and lateral inclination and film cooling holes with streamwise inclination only. The variation in the blowing angle was chosen on a newly dermed and physically motivated basis. High resolution low speed experiments on a large scale turbine airfoil gave insights particularly into the intensified mixing process with lateral ejection. The extensive computational study is performed with the aid of a 3D blockstructured Navier-Stokes solver incorporating a low-Reynolds-number k-e turbulence model. Special attention is paid to mesh generation as a precondition for accurate highresolution results. The downstream temperature fields of the jets show reduced spanwise variations with increasing lateral blowing angle; these variations are quantified for a comprehensive variety of configurations in terms of adiabatic film cooling effectiveness

    Combinations of the azaquinazoline anti-Wolbachia agent, AWZ1066S, with benzimidazole anthelmintics synergise to mediate sub-seven-day sterilising and curative efficacies in experimental models of filariasis

    Lymphatic filariasis and onchocerciasis are two major neglected tropical diseases that are responsible for causing severe disability in 50 million people worldwide, whilst veterinary filariasis (heartworm) is a potentially lethal parasitic infection of companion animals. There is an urgent need for safe, short-course curative (macrofilaricidal) drugs to eliminate these debilitating parasite infections. We investigated combination treatments of the novel anti-Wolbachia azaquinazoline small molecule, AWZ1066S, with benzimidazole drugs (albendazole or oxfendazole) in up to four different rodent filariasis infection models: Brugia malayi—CB.17 SCID mice, B. malayi—Mongolian gerbils, B. pahangi—Mongolian gerbils, and Litomosoides sigmodontis—Mongolian gerbils. Combination treatments synergised to elicit threshold (>90%) Wolbachia depletion from female worms in 5 days of treatment, using 2-fold lower dose-exposures of AWZ1066S than monotherapy. Short-course lowered dose AWZ1066S-albendazole combination treatments also delivered partial adulticidal activities and/or long-lasting inhibition of embryogenesis, resulting in complete transmission blockade in B. pahangi and L. sigmodontis gerbil models. We determined that short-course AWZ1066S-albendazole co-treatment significantly augmented the depletion of Wolbachia populations within both germline and hypodermal tissues of B. malayi female worms and in hypodermal tissues in male worms, indicating that anti-Wolbachia synergy is not limited to targeting female embryonic tissues. Our data provides pre-clinical proof-of-concept that sub-seven-day combinations of rapid-acting novel anti-Wolbachia agents with benzimidazole anthelmintics are a promising curative and transmission-blocking drug treatment strategy for filarial diseases of medical and veterinary importance

    Effectiveness and safety of opicapone in Parkinson's disease patients with motor fluctuations: The OPTIPARK open-label study

    BACKGROUND: The efficacy and safety of opicapone, a once-daily catechol-O-methyltransferase inhibitor, have been established in two large randomized, placebo-controlled, multinational pivotal trials. Still, clinical evidence from routine practice is needed to complement the data from the pivotal trials. METHODS: OPTIPARK (NCT02847442) was a prospective, open-label, single-arm trial conducted in Germany and the UK under clinical practice conditions. Patients with Parkinson’s disease and motor fluctuations were treated with opicapone 50 mg for 3 (Germany) or 6 (UK) months in addition to their current levodopa and other antiparkinsonian treatments. The primary endpoint was the Clinician’s Global Impression of Change (CGI-C) after 3 months. Secondary assessments included Patient Global Impressions of Change (PGI-C), the Unified Parkinson’s Disease Rating Scale (UPDRS), Parkinson’s Disease Questionnaire (PDQ-8), and the Non-Motor Symptoms Scale (NMSS). Safety assessments included evaluation of treatment-emergent adverse events (TEAEs) and serious adverse events (SAEs). RESULTS: Of the 506 patients enrolled, 495 (97.8%) took at least one dose of opicapone. Of these, 393 (79.4%) patients completed 3 months of treatment. Overall, 71.3 and 76.9% of patients experienced any improvement on CGI-C and PGI-C after 3 months, respectively (full analysis set). At 6 months, for UK subgroup only (n = 95), 85.3% of patients were judged by investigators as improved since commencing treatment. UPDRS scores at 3 months showed statistically significant improvements in activities of daily living during OFF (mean ± SD change from baseline: − 3.0 ± 4.6, p < 0.0001) and motor scores during ON (− 4.6 ± 8.1, p < 0.0001). The mean ± SD improvements of − 3.4 ± 12.8 points for PDQ-8 and -6.8 ± 19.7 points for NMSS were statistically significant versus baseline (both p < 0.0001). Most of TEAEs (94.8% of events) were of mild or moderate intensity. TEAEs considered to be at least possibly related to opicapone were reported for 45.1% of patients, with dyskinesia (11.5%) and dry mouth (6.5%) being the most frequently reported. Serious TEAEs considered at least possibly related to opicapone were reported for 1.4% of patients. CONCLUSIONS: Opicapone 50 mg was effective and generally well-tolerated in PD patients with motor fluctuations treated in clinical practice.     Short-course quinazoline drug treatments are effective in the Litomosoides sigmodontis and Brugia pahangi jird models

    The quinazolines CBR417 and CBR490 were previously shown to be potent anti-wolbachials that deplete Wolbachia endosymbionts of filarial nematodes and present promising pre-clinical candidates for human filarial diseases such as onchocerciasis. In the present study we tested both candidates in two models of chronic filarial infection, namely the Litomosoides sigmodontis and Brugia pahangi jird model and assessed their long-term effect on Wolbachia depletion, microfilariae counts and filarial embryogenesis 16-18 weeks after treatment initiation (wpt). Once per day (QD) oral treatment with CBR417 (50 mg/kg) for 4 days or twice per day (BID) with CBR490 (25 mg/kg) for 7 days during patent L. sigmodontis infection reduced the Wolbachia load by >99% and completely cleared peripheral microfilaremia from 10-14 wpt. Similarly, 7 days of QD treatments (40 mg/kg) with CBR417 or CBR490 cleared >99% of Wolbachia from B. pahangi and reduced peritoneal microfilariae counts by 93% in the case of CBR417 treatment. Transmission electron microscopy analysis indicated intensive damage to the B. pahangi ovaries following CBR417 treatment and in accordance filarial embryogenesis was inhibited in both models after CBR417 or CBR490 treatment. Suboptimal treatment regimens of CBR417 or CBR490 did not lead to a maintained reduction of the microfilariae and Wolbachia load. In conclusion, CBR417 or CBR490 are pre-clinical candidates for filarial diseases, which achieve long-term clearance of Wolbachia endosymbionts of filarial nematodes, inhibit filarial embryogenesis and clear microfilaremia with treatments as short as 7 days

    Factors associated with spontaneous stone passage in a contemporary cohort of patients presenting with acute ureteric colic. Results from the MIMIC Study (A Multi-centre cohort study evaluating the role of Inflammatory Markers in patients presenting with acute ureteric Colic)

    Objectives There is conflicting data on the role of white blood cell count (WBC) and other inflammatory markers in spontaneous stone passage in patients with acute ureteric colic. The aim of the study was to assess the relationship of WBC and other routinely collected inflammatory and clinical markers including stone size, stone position and Medically Expulsive Therapy use (MET) with spontaneous stone passage (SSP) in a large contemporary cohort of patients with acute ureteric colic. Subjects and Methods Multi‐centre retrospective cohort study coordinated by the British Urology Researchers in Surgical Training (BURST) Research Collaborative at 71 secondary care hospitals across 4 countries (United Kingdom, Republic of Ireland, Australia and New Zealand). 4170 patients presented with acute ureteric colic and a computer tomography confirmed single ureteric stone. Our primary outcome measure was SSP as defined by the absence of need for intervention to assist stone passage. Multivariable mixed effects logistic regression was used to explore the relationship between key patient factors and SSP. Results 2518 patients were discharged with conservative management and had further follow up with a SSP rate of 74% (n = 1874/2518). Sepsis after discharge with conservative management was reported in 0.6% (n = 16/2518). On multivariable analysis neither WBC, Neutrophils or CRP were seen to predict SSP, with an adjusted OR of 0.97 [95% CI 0.91 to 1.04, p = 0.38], 1.06 [95% CI 0.99 to 1.13, p = 0.1] and 1.00 [95% CI 0.99 to 1.00, p = 0.17], respectively. Medical expulsive therapy (MET) also did not predict SSP [adjusted OR 1.11 [95% CI 0.76 to 1.61]). However, stone size and stone position were significant predictors. SSP for stones 7mm. For stones in the upper ureter the SSP rate was 52% [95% CI 48 to 56], middle ureter was 70% [95% CI 64 to 76], and lower ureter was 83% [95% CI 81 to 85]. Conclusion In contrast to the previously published literature, we found that in patients with acute ureteric colic who are discharged with initial conservative management, neither WBC, Neutrophil count or CRP help determine the likelihood of spontaneous stone passage. We also found no overall benefit from the use of MET. Stone size and position are important predictors and our findings represent the most comprehensive stone passage rates for each mm increase in stone size from a large contemporary cohort adjusting for key potential confounders. We anticipate that these data will aid clinicians managing patients with acute ureteric colic and help guide management decisions and the need for intervention
