    Constructing a man-made c-type cytochrome maquette in vivo:electron transfer, oxygen transport and conversion to a photoactive light harvesting maquette

    The successful use of man-made proteins to advance synthetic biology requires both the fabrication of functional artificial proteins in a living environment, and the ability of these proteins to interact productively with other proteins and substrates in that environment. Proteins made by the maquette method integrate sophisticated oxidoreductase function into evolutionarily naive, non-computationally designed protein constructs with sequences that are entirely unrelated to any natural protein. Nevertheless, we show here that we can efficiently interface with the natural cellular machinery that covalently incorporates heme into natural cytochromes c to produce in vivo an artificial c-type cytochrome maquette. Furthermore, this c-type cytochrome maquette is designed with a displaceable histidine heme ligand that opens to allow functional oxygen binding, the primary event in more sophisticated functions ranging from oxygen storage and transport to catalytic hydroxylation. To exploit the range of functions that comes from the freedom to bind a variety of redox cofactors within a single maquette framework, this c-type cytochrome maquette is designed with a second, non-heme C, tetrapyrrole binding site, enabling the construction of an elementary electron transport chain, and when the heme C iron is replaced with zinc to create a Zn porphyrin, a light-activatable artificial redox protein. The work we describe here represents a major advance in de novo protein design, offering a robust platform for new c-type heme based oxidoreductase designs and an equally important proof-of-principle that cofactor-equipped man-made proteins can be expressed in living cells, paving the way for constructing functionally useful man-made proteins in vivo

    Use of Mixed CH_3−/HC(O)CH_2CH_2−Si(111) Functionality to Control Interfacial Chemical and Electronic Properties During the Atomic-Layer Deposition of Ultrathin Oxides on Si(111)

    Silicon surfaces terminated with a mixed monolayer containing both a propyl aldehyde functionality and methyl groups were prepared and used to control the interfacial chemical and electronic properties of Si(111) surfaces during atomiclayer deposition (ALD) of Al_2O_3 or MnO. Si(111) surfaces functionalized only with the aldehyde moiety exhibited surface recombination velocities, S, of 2500 ± 600 cm s^(−1) whereas the mixed CH_3−/HC(O)CH_2CH_2−Si(111) surfaces displayed S = 25 ± 7 cm s^(−1). During the ALD growth of either Al_2O_3 or MnO, both the HC(O)CH_2CH_2−Si(111) and CH_3−/HC(O)CH_2CH_2−Si(111) surfaces produced increased metal oxide deposition at low cycle number, relative to H−Si(111) or CH_3−Si(111) surfaces. As detected by X-ray photoelectron spectroscopy after the ALD process, the CH_3− and mixed CH_3−/HC(O)CH_2CH_2− functionalized Si(111) surfaces exhibited less interfacial SiO_x than was observed for ALD of metal oxides on H−Si(111) substrates

    Very Low Mass Stellar and Substellar Companions to Solar-Like Stars From MARVELS V: A Low Eccentricity Brown Dwarf from the Driest Part of the Desert, MARVELS-6b

    We describe the discovery of a likely brown dwarf (BD) companion with a minimum mass of 31.7 +/- 2.0 M_Jup to GSC 03546-01452 from the MARVELS radial velocity survey, which we designate as MARVELS-6b. For reasonable priors, our analysis gives a probability of 72% that MARVELS-6b has a mass below the hydrogen-burning limit of 0.072 M_Sun, and thus it is a high-confidence BD companion. It has a moderately long orbital period of 47.8929 +0.0063/-0.0062 days with a low eccentricty of 0.1442 +0.0078/-0.0073, and a semi-amplitude of 1644 +12/-13 m/s. Moderate resolution spectroscopy of the host star has determined the following parameters: T_eff = 5598 +/- 63, log g = 4.44 +/- 0.17, and [Fe/H] = +0.40 +/- 0.09. Based upon these measurements, GSC 03546-01452 has a probable mass and radius of M_star = 1.11 +/- 0.11 M_Sun and R_star = 1.06 +/- 0.23 R_Sun with an age consistent with less than ~6 Gyr at a distance of 219 +/- 21 pc from the Sun. Although MARVELS-6b is not observed to transit, we cannot definitively rule out a transiting configuration based on our observations. There is a visual companion detected with Lucky Imaging at 7.7 arcsec from the host star, but our analysis shows that it is not bound to this system. The minimum mass of MARVELS-6b exists at the minimum of the mass functions for both stars and planets, making this a rare object even compared to other BDs.Comment: 15 pages, 15 figures, 5 tables.     People of the British Isles: preliminary analysis of genotypes and surnames in a UK control population

    There is a great deal of interest in fine scale population structure in the UK, both as a signature of historical immigration events and because of the effect population structure may have on disease association studies. Although population structure appears to have a minor impact on the current generation of genome-wide association studies, it is likely to play a significant part in the next generation of studies designed to search for rare variants. A powerful way of detecting such structure is to control and document carefully the provenance of the samples involved. Here we describe the collection of a cohort of rural UK samples (The People of the British Isles), aimed at providing a well-characterised UK control population that can be used as a resource by the research community as well as providing fine scale genetic information on the British population. So far, some 4,000 samples have been collected, the majority of which fit the criteria of coming from a rural area and having all four grandparents from approximately the same area. Analysis of the first 3,865 samples that have been geocoded indicates that 75% have a mean distance between grandparental places of birth of 37.3km, and that about 70% of grandparental places of birth can be classed as rural. Preliminary genotyping of 1,057 samples demonstrates the value of these samples for investigating fine scale population structure within the UK, and shows how this can be enhanced by the use of surnames

    Using [C II] 158 μm Emission from Isolated ISM Phases as a Star Formation Rate Indicator

    The brightest observed emission line in many star-forming galaxies is the [C II] 158 μm line, making it detectable up to z ~ 7. In order to better understand and quantify the [C II] emission as a tracer of star formation, the theoretical ratio between the [N II] 205 μm emission and the [C II] 158 μm emission has been employed to empirically determine the fraction of [C II] emission that originates from the ionized and neutral phases of the interstellar medium (ISM). Sub-kiloparsec measurements of the [C II] 158 μm and [N II] 205 μm lines in nearby galaxies have recently become available as part of the Key Insights in Nearby Galaxies: a Far Infrared Survey with Herschel (KINGFISH) and Beyond the Peak programs. With the information from these two far-infrared lines along with the multi-wavelength suite of KINGFISH data, a calibration of the [C II] emission line as a star formation rate (SFR) indicator and a better understanding of the [C II] deficit are pursued. [C II] emission is also compared to polycyclic aromatic hydrocarbon (PAH) emission in these regions to compare photoelectric heating from PAH molecules to cooling by [C II] in the neutral and ionized phases of the ISM. We find that the [C II] emission originating in the neutral phase of the ISM does not exhibit a deficit with respect to the infrared luminosity and is therefore preferred over the [C II] emission originating in the ionized phase of the ISM as an SFR indicator for the normal star-forming galaxies included in this sample
