    Concentração de chumbo, defeitos de esmalte e cárie em dentes decíduos

    OBJECTIVE: To verify the relationship between lead concentration in the enamel of deciduous teeth and the presence of enamel defects and, consequently, with dental caries among preschool children. METHODS: The sample consisted of 329 preschool children in Piracicaba, State of São Paulo: 132 attending municipal kindergartens close to industrial plants and 197 attending kindergartens in non-industrial areas. This sample belonged to an initial study made between 2000 and 2001, in which the lead concentration was obtained by means of enamel biopsy. Oral clinical examination of the children from both regions was performed to verify the prevalence of enamel defects, using the Developmental Defects of Enamel (DDE) Index of the World Dental Federation (FDI), and of dental caries, using the decayed, missing and filled surfaces (dmfs) index of the World Health Organization. The chi-squared test and relative risk calculation were utilized in relation to a significance level of 5%, considering each region separately. RESULTS: Among the children from the non-industrial area, there was a higher proportion with dental caries among those with higher lead concentrations in deciduous teeth (p=0.02). This was not, however, observed among the children from the industrial area (p=0.89). There was an increased relative risk (RR) of caries among the children from the non-industrial area, but this was not seen among the children from the industrial area. No relationship was observed between the presence of lead and enamel defects. CONCLUSIONS: No data was found that would give evidence of a relationship between lead concentration and enamel defects in either of the areas studied. No relationship was found between lead and dental caries in the industrial area, thus emphasizing that more studies of such relationships are needed.OBJETIVO: Verificar a relação da concentração de chumbo no esmalte de dentes decíduos com a presença de defeitos de esmalte e sua relação com cárie dental em pré-escolares. MÉTODOS: A amostra foi de 329 crianças de pré-escolas municipais de uma área próxima de indústrias (N=132) e outra não industrial (N=197) da cidade de Piracicaba, Estado de São Paulo. Essa amostra pertencente a um estudo inicial realizado entre 2000 e 2001 no qual o chumbo foi analisado por meio de biópsia de esmalte. Foram realizados exames clínicos bucais para verificação da prevalência de defeitos de esmalte (Developmental Defects of Enamel Index - DDE, da Federação Dentária Internacional - FDI) e cárie (Índice ceos, Organização Mundial da Saúde), em ambas regiões. Foram utilizados teste de qui-quadrado e cálculo do risco relativo ao nível de significância de 5%, considerando cada região separadamente. RESULTADOS: Houve maior proporção de crianças com cárie entre aquelas com maiores concentrações de chumbo nos decíduos na região não industrial (p=0,02), o que não se observou na região industrial (p=0,89). Houve risco relativo (RR) aumentado de cárie nas crianças da região não industrial o que não foi verificado nas crianças da região industrial. Não se observou relação entre a presença de chumbo e os defeitos de esmalte. CONCLUSÕES: Não foram encontados dados que evidenciassem a relação entre concentração de chumbo e defeitos no esmalte em nenhuma das regiões pesquisadas. Não foi encontrada relação entre chumbo e cárie na região industrial, ressaltando a necessidade de mais estudos dessas relações

    Annona coriacea Mart. fractions promote cell cycle arrest and inhibit autophagic flux in human cervical cancer cell lines

    Plant-based compounds are an option to explore and perhaps overcome the limitations of current antitumor treatments. Annona coriacea Mart. is a plant with a broad spectrum of biological activities, but its antitumor activity is still unclear. The purpose of our study was to determine the effects of A. coriacea fractions on a panel of cervical cancer cell lines and a normal keratinocyte cell line. The antitumor effect was investigated in vitro by viability assays, cell cycle, apoptosis, migration, and invasion assays. Intracellular signaling was assessed by Western blot, and major compounds were identified by mass spectrometry. All fractions exhibited a cytotoxic effect on cisplatin-resistant cell lines, SiHa and HeLa. C3 and C5 were significantly more cytotoxic and selective than cisplatin in SiHa and Hela cells. However, in CaSki, a cisplatin-sensitive cell line, the compounds did not demonstrate higher cytotoxicity when compared with cisplatin. Alkaloids and acetogenins were the main compounds identified in the fractions. These fractions also markedly decreased cell proliferation with p21 increase and cell cycle arrest in G2/M. These effects were accompanied by an increase of H2AX phosphorylation levels and DNA damage index. In addition, fractions C3 and C5 promoted p62 accumulation and decrease of LC3II, as well as acid vesicle levels, indicating the inhibition of autophagic flow.     Leptin Prevents Lipopolysaccharide-Induced Depressive-Like Behaviors in Mice: Involvement of Dopamine Receptors

    Depression is a chronic and recurrent disorder, associated with high morbidity and risk of suicide. Leptin was firstly described as an anti-obesity hormone, but several actions of leptin in CNS have been reported. In fact, leptin regulates dopaminergic neurotransmission in mesolimbic areas and has antidepressant-like properties in stress-based models. In the present study, we investigated, for the first time, putative antidepressant-like effects of leptin in an animal model of depressive-like behaviors induced by lipopolysaccharide (LPS), and the potential involvement of dopamine receptors as mediators of those behavioral effects. Mice were injected leptin (1.5 mg/kg, IP) or imipramine prior to LPS administration. To evaluate the involvement of dopamine receptors, different experimental groups were pretreated with either the dopaminergic antagonist SCH23390, for D1 receptors or raclopride, for D2/D3 receptors, prior to leptin injection. Twenty-four hours post-LPS, mice were submitted to the forced swimming and sucrose preference tests. In addition, IL-1β levels were determined in the prefrontal cortex (PFC), hippocampus and striatum. BDNF levels were measured in the hippocampus. Our results showed that leptin, similarly to imipramine, prevented the core behavioral alterations induced by LPS (despair-like behavior and anhedonia), without altering locomotion. In neurochemical analysis, leptin restored LPS-induced changes in IL-1β levels in the PFC and striatum, and increased BDNF levels in the hippocampus. The blockade of dopamine D1 and D2/D3 receptors inhibited leptin's antidepressant-like effects, whilst only the blockade of D1-like receptors blunted leptin-induced increments in prefrontal IL-1β levels. Our results indicate that leptin has antidepressant-like effects in an inflammatory model of depression with the contribution, at least partial, of dopamine receptors

    Genome of the Avirulent Human-Infective Trypanosome—Trypanosoma rangeli

    Background: Trypanosoma rangeli is a hemoflagellate protozoan parasite infecting humans and other wild and domestic mammals across Central and South America. It does not cause human disease, but it can be mistaken for the etiologic agent of Chagas disease, Trypanosoma cruzi. We have sequenced the T. rangeli genome to provide new tools for elucidating the distinct and intriguing biology of this species and the key pathways related to interaction with its arthropod and mammalian hosts.  Methodology/Principal Findings: The T. rangeli haploid genome is ,24 Mb in length, and is the smallest and least repetitive trypanosomatid genome sequenced thus far. This parasite genome has shorter subtelomeric sequences compared to those of T. cruzi and T. brucei; displays intraspecific karyotype variability and lacks minichromosomes. Of the predicted 7,613 protein coding sequences, functional annotations could be determined for 2,415, while 5,043 are hypothetical proteins, some with evidence of protein expression. 7,101 genes (93%) are shared with other trypanosomatids that infect humans. An ortholog of the dcl2 gene involved in the T. brucei RNAi pathway was found in T. rangeli, but the RNAi machinery is non-functional since the other genes in this pathway are pseudogenized. T. rangeli is highly susceptible to oxidative stress, a phenotype that may be explained by a smaller number of anti-oxidant defense enzymes and heatshock proteins.  Conclusions/Significance: Phylogenetic comparison of nuclear and mitochondrial genes indicates that T. rangeli and T. cruzi are equidistant from T. brucei. In addition to revealing new aspects of trypanosome co-evolution within the vertebrate and invertebrate hosts, comparative genomic analysis with pathogenic trypanosomatids provides valuable new information that can be further explored with the aim of developing better diagnostic tools and/or therapeutic targets

    Avaliação de genótipos de arroz sob efeito do ácido butírico

    Solos do tipo hidromórfico apresentam uma reduzida capacidade de drenagem, sendo utilizados principalmente para cultivo de arroz irrigado. Esta condição favorece o desenvolvimento de microrganismos anaeróbios que produzem substâncias fitotóxicas. O objetivo do trabalho foi avaliar a resposta de 25 genótipos de arroz ao ácido butírico, um composto produzido em solos de deficiente drenagem e alto teor de matéria orgânica. O trabalho foi executado em sistema de hidroponia com 4 doses do ácido e o delineamento utilizado foi blocos casualizados com 3 repetições em um esquema fatorial. As variáveis mensuradas foram comprimento de raízes (CR) e de parte aérea (CPA), número de raízes (NR) e matéria seca de raízes (MSR) e parte aérea (MSPA). Foram procedidas análise de variância, desempenho relativo e ajuste de regressões. Os efeitos para genótipos e doses de ácido butírico foram todos significativos. Apenas os efeitos de interação entre doses x genótipos para as variáveis CR e MSR revelaram significância. A variável CR foi a mais afetada pelo ácido e as regressões estabelecidas para essa variável revelaram 9 genótipos tolerantes e 16 sensíveis ao efeito fitototoxico do ácido butírico.     Pervasive gaps in Amazonian ecological research

    Biodiversity loss is one of the main challenges of our time,1,2 and attempts to address it require a clear un derstanding of how ecological communities respond to environmental change across time and space.3,4 While the increasing availability of global databases on ecological communities has advanced our knowledge of biodiversity sensitivity to environmental changes,5–7 vast areas of the tropics remain understudied.8–11 In the American tropics, Amazonia stands out as the world’s most diverse rainforest and the primary source of Neotropical biodiversity,12 but it remains among the least known forests in America and is often underrepre sented in biodiversity databases.13–15 To worsen this situation, human-induced modifications16,17 may elim inate pieces of the Amazon’s biodiversity puzzle before we can use them to understand how ecological com munities are responding. To increase generalization and applicability of biodiversity knowledge,18,19 it is thus crucial to reduce biases in ecological research, particularly in regions projected to face the most pronounced environmental changes. We integrate ecological community metadata of 7,694 sampling sites for multiple or ganism groups in a machine learning model framework to map the research probability across the Brazilian Amazonia, while identifying the region’s vulnerability to environmental change. 15%–18% of the most ne glected areas in ecological research are expected to experience severe climate or land use changes by 2050.     Influence of socioeconomic factors on pregnancy outcome in women with structural heart disease

    OBJECTIVE: Cardiac disease is the leading cause of indirect maternal mortality. The aim of this study was to analyse to what extent socioeconomic factors influence the outcome of pregnancy in women with heart disease.  METHODS: The Registry of Pregnancy and Cardiac disease is a global prospective registry. For this analysis, countries that enrolled ≥10 patients were included. A combined cardiac endpoint included maternal cardiac death, arrhythmia requiring treatment, heart failure, thromboembolic event, aortic dissection, endocarditis, acute coronary syndrome, hospitalisation for cardiac reason or intervention. Associations between patient characteristics, country characteristics (income inequality expressed as Gini coefficient, health expenditure, schooling, gross domestic product, birth rate and hospital beds) and cardiac endpoints were checked in a three-level model (patient-centre-country).  RESULTS: A total of 30 countries enrolled 2924 patients from 89 centres. At least one endpoint occurred in 645 women (22.1%). Maternal age, New York Heart Association classification and modified WHO risk classification were associated with the combined endpoint and explained 37% of variance in outcome. Gini coefficient and country-specific birth rate explained an additional 4%. There were large differences between the individual countries, but the need for multilevel modelling to account for these differences disappeared after adjustment for patient characteristics, Gini and country-specific birth rate.  CONCLUSION: While there are definite interregional differences in pregnancy outcome in women with cardiac disease, these differences seem to be mainly driven by individual patient characteristics. Adjustment for country characteristics refined the results to a limited extent, but maternal condition seems to be the main determinant of outcome
