    Gestational age acceleration is associated with epigenetic biomarkers of prenatal physiologic stress exposure

    BACKGROUND: Physiological maternal stress response, such as imbalance in the glucocorticoid pathway and immune system seems to be mediated by DNA methylation (DNAm) and might translate intrauterine stress exposures into phenotypic changes in a sex-specific manner. DNAm in specific sites can also predict newborn gestational age and gestational age acceleration (GAA). GAA occurs when the predicted biological age is higher than the chronological age. In adults, poor health outcomes related to this deviance are well documented and raise questions for the interpretation and prediction in early stages of life. Boys seem to be more vulnerable to intrauterine stress exposure than girls; however, the mechanisms of adaptive sex-specific responses are still unclear. We hypothesize that intrauterine stress exposure is associated with GAA and could be different in boys and girls if inflammatory or glucocorticoid pathways exposure is considered. RESULTS: Using the Western Region Birth Cohort (ROC-Sao Paulo, Brazil) (n = 83), we calculated DNAm age and GAA from cord blood samples. Two epigenetic risk scores were calculated as an indirect proxy for low-grade inflammation (i-ePGS) and for glucocorticoid exposure (GES). Multivariate linear regression models were applied to investigate associations of GAA with prenatal exposures. The i-ePGS and GES were included in different models with the same co-variates considering sex interactions. The first multivariate model investigating inflammatory exposure (adj. R(2) = 0.31, p = < 0.001) showed that GAA was positively associated with i-ePGS (CI, 0.26-113.87, p = 0.049) and negative pregnancy-related feelings (CI, 0.04-0.48 p = 0.019). No sex interaction was observed. The second model investigating glucocorticoid exposure (adj. R(2) = 0.32, p = < 0.001) showed that the higher was the GAA was associated with a lower the lower was the GES in girls (CI, 0.04-2.55, p = 0.044). In both models, maternal self-reported mental disorder was negatively associated with GAA. CONCLUSION: Prenatal epigenetic score of exposure to low-grade inflammatory was a predictor of GAA for both sexes. Glucocorticoid epigenetic score seems to be more important to GAA in girls. This study supports the evidence of sex-specificity in stress response, suggesting the glucocorticoid as a possible pathway adopted by girls to accelerate the maturation in an adverse condition

    Gene expression profiling of Trypanosoma cruzi in the presence of heme points to glycosomal metabolic adaptation of epimastigotes inside the vector

    Chagas disease, also known as American trypanosomiasis, is a potentially life-threatening illness caused by the protozoan parasite, Trypanosoma cruzi, and is transmitted by triatomine insects during its blood meal. Proliferative epimastigotes forms thrive inside the insects in the presence of heme (iron protoporphyrin IX), an abundant product of blood digestion, however little is known about the metabolic outcome of this signaling molecule in the parasite. Trypanosomatids exhibit unusual gene transcription employing a polycistronic transcription mechanism through trans-splicing that regulates its life cycle. Using the Deep Seq transcriptome sequencing we characterized the heme induced transcriptome of epimastigotes and determined that most of the upregulated genes were related to glucose metabolism inside the glycosomes. These results were supported by the upregulation of glycosomal isoforms of PEPCK and fumarate reductase of heme-treated parasites, implying that the fermentation process was favored. Moreover, the downregulation of mitochondrial gene enzymes in the presence of heme also supported the hypothesis that heme shifts the parasite glycosomal glucose metabolism towards aerobic fermentation. These results are examples of the environmental metabolic plasticity inside the vector supporting ATP production, promoting epimastigotes proliferation and survival

    The first observed stellar occultations by the irregular satellite Phoebe (Saturn IX) and improved rotational period

    peer reviewedWe report six stellar occultations by Phoebe (Saturn IX), an irregular satellite of Saturn, obtained between mid-2017 and mid-2019. The 2017 July 6 event was the first stellar occultation by an irregular satellite ever observed. The occultation chords were compared to a 3D shape model of the satellite obtained from Cassini observations. The rotation period available in the literature led to a sub-observer point at the moment of the observed occultations where the chords could not fit the 3D model. A procedure was developed to identify the correct sub-observer longitude. It allowed us to obtain the rotation period with improved precision compared to the currently known value from literature. We show that the difference between the observed and the predicted sub-observer longitude suggests two possible solutions for the rotation period. By comparing these values with recently observed rotational light curves and single- chord stellar occultations, we can identify the best solution for Phoebe's rotational period as 9.27365 ± 0.00002 h. From the stellar occultations, we also obtained six geocentric astrometric positions in the ICRS as realized by the Gaia DR2 with uncertainties at the 1-mas level

    A big data approach to identify the loss of coastal cultural ecosystem services caused by the 2019 Brazilian oil spill disaster

    In August 2019, the Northeast coast of Brazil was impacted by an extensive oil spill, with immediate effects on marine and coastal ecosystems and significant impacts on tourism and food security. The human dimension of those impacts also includes the loss of cultural ecosystem services (CES); the non-material benefits stemming from strongly rooted cultural practices and relationships with nature. CES are of great importance for local residents and visitors that flock to Brazilian iconic beaches, however, they are difficult to measure using traditional assessment methods due to their subjective and non-tangible nature. Here, we use a big data approach to assess and map the loss of CES in the Northeast coast of Brazil caused by the recent oil spill. We analysed 2,880 digital images (published on the image sharing platform Flickr) taken before and during the disaster in affected locations, using a combination of automated techniques. Results showed a sharp decline in the number of users posting photos of locations affected by oil spill, and a decline in photos representing landscape and cultural appreciation. Our big data approach provides a fast and automated way to assess CES at large spatial scales that can be used to monitor the social impacts of environmental disasters.Peer reviewe

    Public awareness and engagement in relation to the coastal oil spill in northeast Brazil

    Social media data is a rich source of information to assess human activities in catastrophic events. Here, we use social media data to understand how the 2019 Brazilian oil spill influenced social attitudes. Data were collected from the globally popular Instagram platform between August 1, 2019 and March 1, 2020. First, we manually identified the 5 most popular (portuguese language) hashtags related to the oil spill #oleonononordeste;#desastreambiental;#ma rsemoleo;#sosnordeste;#ma rsempetroleo. In the sequence, we collected information on captions, post metadata and users associated with posts retrieved using the selected hashtags. We identified a total of 7,413 posts. These posts were grouped in topics: government (47.76%), protest (24.37%), volunteers (24.45%), biodiversity (0.003%), origin (0.006%), tourism (0.008%) and others (0.016%). All topics had the peak of posts in October and November 2019. Nevertheless, interest in the oil spill was temporary, with most posts appearing in the 2-4 months after the beginning of the disaster. Our findings illustrate the enormous potential of using social media data for understanding and monitoring human engagement with environmental disasters, but also suggest that conservationists and environmental groups may only have a limited 'window of opportunity' to engage and mobilize public support for remediation and restoration efforts.Peer reviewe

    WRONG WAY HOME: AN INFANT SOUTHERN ELEPHANT SEAL (MIROUNGA LEONINA) ARRIVAL ON SOUTHEASTERN BRAZILIAN BEACHES

    ABSTRACTRecords of southern elephant seals (Mirounga leonina) as vagrants along the SE Brazilian coast date back the late 50’s and have been relatively common over the last decades. These large marine mammals usually call much attention when they arrive on tropical beaches worldwide and are generally treated as ‘occasional visitors’ and ‘vagrants’. This note reports on sightings of a youngelephant seal along SE Brazil in the summer and autumn of 2020. We also reviewed records in both the literature and open sources, totaling eight records of infant southern elephant seals known since the late 70’s along the Brazilian coast. It was noted that the arrival of an infant in February of 2020is coincident with a previous cyclonic activity off the SE and NE Brazilian coast. The connection of such unlikely records of pinnipeds on tropical beaches and extreme weather events associated to ocean currents should be better evaluated in the context of climatic change RESUMOÉ reportada uma ocorrência recente de um infante de elefante-marinho (Mirounga leonina) na costa do estado do Rio de Janeiro com um intervalo aproximado de um mês entre as avistagens do mesmo indivíduo. A comparação das fotografias obtidas em ambos os registros permitiu a comparação e confirmação. Em adição, se discute a presença de infantes na costa brasileira, que totalizam oito casos, mas que não apontam uma sazonalidade marcada, mas uma tendência aos registros serem reportados em junho, seguido por outubro e novembro. Como o presente registro se deu em janeiro, pico do verão, fatores climáticos de larga escala podem estar atuando para deslocar alguns indivíduos muito ao longe das suas colônias.Palavras-chave: Mirounga leonina; Movimentos; Vagante; Atlântico Sul

    Hypoxia enhances ILC3 responses through HIF-1α-dependent mechanism

    Group 3 innate lymphoid cells (ILC3) have a prominent role in the maintenance of intestine mucosa homeostasis. The hypoxia-inducible factor (HIF) is an important modulator of immune cell activation and a key mechanism for cellular adaptation to oxygen deprivation. However, its role on ILC3 is not well known. In this study, we investigated how a hypoxic environment modulates ILC3 response and the subsequent participation of HIF-1 signaling in this process. We found increased proliferation and activation of intestinal ILC3 at low oxygen levels, a response that was phenocopied when HIF-1α was chemically stabilized and was reversed when HIF-1 was blocked. The increased activation of ILC3 relied on a HIF-1α-dependent transcriptional program, but not on mTOR-signaling or a switch to glycolysis. HIF-1α deficiency in RORyt compartment resulted in impaired IL-17 and IL-22 production by ILC3 in vivo, which reflected in a lower expression of their target genes in the intestinal epithelium and an increased susceptibility to Clostridiodes difficile infection. Taken together, our results show that HIF-1α activation in intestinal ILC3 is relevant for their functions in steady state and infectious conditions

The new Genius Proflex (25/0.04), Vortex Blue (25/0.04), and TruNatomy (26/0.04v) instruments (n = 41 per group) were analyzed regarding design, metallurgy, and mechanical performance, while shaping ability (untouched canal walls, volume of removed dentin and hard tissue debris) was tested in 36 anatomically matched root canals of mandibular molars. The results were compared using one-way ANOVA, post hoc Tukey, and Kruskal–Wallis tests, with a significance level set at 5%. All instruments showed symmetrical cross-sections, with asymmetrical blades, no radial lands, no major defects, and almost equiatomic nickel–titanium ratios. Differences were noted in the number of blades, helical angles, cross-sectional design, and tip geometry. The Genius Proflex and the TruNatomy instruments had the highest and lowest R-phase start and finish temperatures, as well as the highest and lowest time and cycles to fracture (p 0.05). No differences among tested systems were observed regarding the maximum torque, angle of rotation prior to fracture, and shaping ability (p > 0.05). The instruments showed similarities and differences in their design, metallurgy, and mechanical properties. However, their shaping ability was similar, without any clinically significant errors. Understanding these characteristics may help clinicians to make decisions regarding which instrument to choose for a particular clinical situation.publishersversionpublishe
