PEQUENAS CASAS DE CULTIVO PARA FLORES E HORTALIÇAS
O presente trabalho foi possibilitado através da parceria entre a APP (Associação de Pais e Professores) do Instituto Federal Catarinense - campus Rio do Sul e o Instituto Souza Cruz. Desta parceria o projeto de ensino e extensão, intitulado Novos Rurais IFC/Souza Cruz - Fixação de Jovens no Meio Rural, foi possibilitado. O trabalho contou com uma primeira etapa de formação em empreendedorismo seguido pela implantação dos projetos empreendedores através de subsídios financeiros doados pelo Instituto Souza Cruz. Após a formação em empreendedorismo, identificamos a viabilidade de produzir e comercializar pequenas casas de vegetação (estufas) de baixo custo para suprir demandas de produção de hortaliças, flores e demais mudas necessárias para autossuficiência. O projeto visa a demanda de horticultura e jardinagem em meio urbano, com objetivo de sanar dificuldades de cultivo de espécies vegetais em temperaturas baixas, plantas suscetíveis a pragas e doenças com reduzido uso de agrotóxicos. As casas de vegetação foram construídas com tubos galvanizados e com paredes produzidas em filme plástico de 100 micra, com proteção ultravioleta. As junções foram soldadas por meio de máquina de solda industrial. As dimensões aplicadas foram de 1,90 metros de largura, 2,50 metros de comprimento e 2,00 metros de altura. Pequenas alterações poderiam ser realizadas, adequando a estufa a demanda do consumidor. O aporte financeiro do Instituto Souza Cruz viabilizou a aquisição de máquinas e ferramentas (serra, soldador, furadeira, jogo de chaves e insumos) necessárias para a fabricação das casas de vegetação. A formação (curso de empreendedorismo e orientação individualizadas e coletivas) possibilitou uma visão geral sobre gestão de negócios, inovação e empreendedorismo, o que trouxe mais segurança para a abertura do negócio. Com esse suporte, foi criada a micro empresa denominada Green House, que iniciou suas atividades em 2019, inicialmente com o único produto, casa de vegetação (estufa). As vendas entre 2019 e 2020 foram significativas até que, com a chegada da pandemia de Covid 19, realizamos a diversificação de produtos. A empresa agora também fabrica itens decorativos em metal (principalmente aço), tais como suporte para mangueira, cantinho do café, grades decorativas, entre outros. Estes itens foram vendidos pela internet (plataforma E-Commerce) e levaram a empresa novamente a um bom faturamento, o que possibilitou inclusive a construção de um galpão e aquisição de novos equipamentos, a exemplo de um moderno aparelho de solada MIG (Metal Inerte Gás). A crise decorrente da prolongada pandemia e o aumento dos preços dos insumos (principalmente o ferro) levou o autor buscar uma colocação no mercado de trabalho. Hoje atua como colaborador em empresa na área metalmecânica e fabrica os produtos oportunizados pelo projeto Novos Rurais nos fins de semana, complementando a renda familiar. Assim conclui-se que, mesmo com todo o cenário da pandemia, que atingiu fortemente a economia brasileira, o projeto atingiu seus objetivos uma vez que oportunizou a geração da renda esperada, aprimorou o potencial empreendedor e fixou o autor em ambiente rural, pois neste caso o autor mora e construiu a micro empresa em área rural de Rio do Sul.
Colonic Patch and colonic SILT development are independent and differentially-regulated events
Intestinal lymphoid tissues have to simultaneously ensure protection against pathogens and tolerance towards commensals. Despite such vital functions, their development in the colon is poorly understood. Here, we show that the two distinct lymphoid tissues of the colon–colonic patches and colonic SILTs–can easily be distinguished based on anatomical location, developmental timeframe and cellular organization. Furthermore, whereas colonic patch development depended on CXCL13-mediated LTi cell clustering followed by LTα-mediated consolidation, early LTi clustering at SILT anlagen did not require CXCL13, CCR6 or CXCR3. Subsequent dendritic cell recruitment to and gp38+VCAM-1+ lymphoid stromal cell differentiation within SILTs required LTα; B cell recruitment and follicular dendritic cell differentiation depended on MyD88-mediated signalling, but not the microflora. In conclusion, our data demonstrate that different mechanisms, mediated mainly by programmed stimuli, induce the formation of distinct colonic lymphoid tissues, therefore suggesting that these tissues may have different functions
Short-term antigen presentation and single clonal burst limit the magnitude of the CD8(+) T cell responses to malaria liver stages.
Malaria sporozoites induce swift activation of antigen-specific CD8(+) T cells that inhibit the intracellular development of liver-stage parasites. The length of time of functional in vivo antigen presentation, estimated by monitoring the activation of antigen-specific CD8(+) T cells, is of short duration, with maximum T cell activation occurring within the first 8 h after immunization and lasting approximately 48 h. Although the magnitude of the CD8(+) T cell response closely correlates with the number of parasites used for immunization, increasing the time of antigen presentation by daily immunizations does not enhance the magnitude of this response. Thus, once a primary clonal burst is established, the CD8(+) T cell response becomes refractory or unresponsive to further antigenic stimulation. These findings strongly suggest that the most efficient strategy for the induction of primary CD8(+) T cell responses is the delivery of a maximal amount of antigen in a single dose, thereby ensuring a clonal burst that involves the largest number of precursors to become memory cells
Using argument notation to engineer biological simulations with increased confidence
The application of computational and mathematical modelling to explore the mechanics of biological systems is becoming prevalent. To significantly impact biological research, notably in developing novel therapeutics, it is critical that the model adequately represents the captured system. Confidence in adopting in silico approaches can be improved by applying a structured argumentation approach, alongside model development and results analysis. We propose an approach based on argumentation from safety-critical systems engineering, where a system is subjected to a stringent analysis of compliance against identified criteria. We show its use in examining the biological information upon which a model is based, identifying model strengths, highlighting areas requiring additional biological experimentation and providing documentation to support model publication. We demonstrate our use of structured argumentation in the development of a model of lymphoid tissue formation, specifically Peyer's Patches. The argumentation structure is captured using Artoo (www.york.ac.uk/ycil/software/artoo), our Web-based tool for constructing fitness-for-purpose arguments, using a notation based on the safety-critical goal structuring notation. We show how argumentation helps in making the design and structured analysis of a model transparent, capturing the reasoning behind the inclusion or exclusion of each biological feature and recording assumptions, as well as pointing to evidence supporting model-derived conclusions
Caracterização do vírus da raiva isolado de uma colônia de morcegos Eptesicus furinalis, do Brasil
Some bat species have adapted to the expanding human population by acquiring the ability to roost in urban buildings, increasing the exposure risk for people and domestic animals, and consequently, the likelihood of transmitting rabies. Three dead bats were found in the yard of a house in an urban area of Jundiaí city in the state of São Paulo in southeast Brazil. Two of the three bats tested positive for rabies, using Fluorescent Antibody and Mouse Inoculation techniques. A large colony of Eptesicus furinalis was found in the house's attic, and of the 119 bats captured, four more tested positive for rabies. The objectives of this study were to report the rabies diagnosis, characterize the isolated virus antigenically and genetically, and study the epidemiology of the colony.Algumas espécies de morcegos têm se adaptado ao uso de abrigos em construções urbanas, aumentando a possibilidade de contato desses morcegos com pessoas e animais domésticos e conseqüentemente, o potencial risco de transmissão de raiva. Três morcegos foram encontrados no jardim de uma casa na área urbana da cidade de Jundiaí, Estado de São Paulo, Sudeste do Brasil, dois deles foram positivos para raiva pelas técnicas de imunofluorescência e inoculação em camundongos. Uma grande colônia de E. furinalis foi identificada, vivendo no sótão da casa e 119 morcegos foram encaminhados para diagnóstico de raiva, com mais quatro morcegos positivos. O objetivo desse estudo é apresentar a caracterização genética e antigênica do vírus da raiva isolado desses morcegos e o estudo epidemiológico da colônia
Therapeutic limitations in tumor-specific CD8+ memory T cell engraftment
BACKGROUND: Adoptive immunotherapy with cytotoxic T lymphocytes (CTL) represents an alternative approach to treating solid tumors. Ideally, this would confer long-term protection against tumor. We previously demonstrated that in vitro-generated tumor-specific CTL from the ovalbumin (OVA)-specific OT-I T cell receptor transgenic mouse persisted long after adoptive transfer as memory T cells. When recipient mice were challenged with the OVA-expressing E.G7 thymoma, tumor growth was delayed and sometimes prevented. The reasons for therapeutic failures were not clear. METHODS: OT-I CTL were adoptively transferred to C57BL/6 mice 21 – 28 days prior to tumor challenge. At this time, the donor cells had the phenotypical and functional characteristics of memory CD8+ T cells. Recipients which developed tumor despite adoptive immunotherapy were analyzed to evaluate the reason(s) for therapeutic failure. RESULTS: Dose-response studies demonstrated that the degree of tumor protection was directly proportional to the number of OT-I CTL adoptively transferred. At a low dose of OT-I CTL, therapeutic failure was attributed to insufficient numbers of OT-I T cells that persisted in vivo, rather than mechanisms that actively suppressed or anergized the OT-I T cells. In recipients of high numbers of OT-I CTL, the E.G7 tumor that developed was shown to be resistant to fresh OT-I CTL when examined ex vivo. Furthermore, these same tumor cells no longer secreted a detectable level of OVA. In this case, resistance to immunotherapy was secondary to selection of clones of E.G7 that expressed a lower level of tumor antigen. CONCLUSIONS: Memory engraftment with tumor-specific CTL provides long-term protection against tumor. However, there are several limitations to this immunotherapeutic strategy, especially when targeting a single antigen. This study illustrates the importance of administering large numbers of effectors to engraft sufficiently efficacious immunologic memory. It also demonstrates the importance of targeting several antigens when developing vaccine strategies for cancer

Câmara inalatória para ensaios boilógicos in vivo.
Câmara inalatória para ensaios biológicos in vivo, particularmente de uma câmara que visa viabilizar estudos farmacológicos e toxicológicos in vivo de substâncias vaporizadas e/ou nebulizadas. Para tal, não se exige o controle de pressão, pelo contrário, a câmara objeto da invenção apresenta orifícios para saída do ar interiorizado na mesma, possibilitando renovação desse ar sem que haja alteração de pressão em seu interior. De maneira mais específica, a presente câmara objetiva avaliar a atividade terapêutica e toxicidade de formulações para administração pela via pulmonar, em modelo in vivo para roedores. Além disso, também poderá ser empregada para avaliar os malefícios causados ao organismo por vapores, gases e fumaças. A invenção contempla uma câmara de inalação (1) com pelo menos cinco juntas esmerilhadas fêmeas (2) receptoras dos tubos de vidro (3) de acondicionamento dos animais, referidos tubos de vidro (3) recebendo, cada qual, uma tampa (4), sendo que, na extremidade da câmara inalatória propriamente dita (1), se acopla a cabeça (5), enquanto que, esta câmara inalatória propriamente dita (1) possui uma área de liberação do ar (6) próximo ao fundo da mesma; a referida cabeça (5) se acopla a uma mangueira de silicone (7) e esta a uma junção de vidro cilíndrica (8) que se acopla, na extremidade oposta à mangueira de silicone (7), ao copo de nebulização (9) e este ao aparelho de nebulização (10).Dados atualizados em 31. jan. 2023. The Cell Cycle Time of CD8+ T Cells Responding In Vivo Is Controlled by the Type of Antigenic Stimulus
A hallmark of cells comprising the mammalian adaptive immune system is the requirement for these rare naïve T (and B) lymphocytes directed to a specific microorganism to undergo proliferative expansion upon first encounter with this antigen. In the case of naïve CD8+ T cells the ability of these rare quiescent lymphocytes to rapidly activate and expand into effector T cells in numbers sufficient to control viral and certain bacterial infections can be essential for survival. In this report we examined the activation, cell cycle time and initial proliferative response of naïve murine CD8+ T cells responding in vivo to Influenza and Vaccinia virus infection or vaccination with viral antigens. Remarkably, we observed that CD8+ T cells could divide and proliferate with an initial cell division time of as short as 2 hours. The initial cell cycle time of responding CD8+ T cells is not fixed but is controlled by the antigenic stimulus provided by the APC in vivo. Initial cell cycle time influences the rate of T cell expansion and the numbers of effector T cells subsequently accumulating at the site of infection. The T cell cycle time varies with duration of the G1 phase of the cell cycle. The duration of G1 is inversely correlated with the phosphorylation state of the retinoblastoma (Rb) protein in the responding T cells. The implication of these findings for the development of adaptive immune responses and the regulation of cell cycle in higher eukaryotic cells is discussed
