    Early Career Academic Productivity Among Emergency Physicians With R01 Grant Funding

    Objectives:  The objective was to describe the early academic career activities of emergency physician (EP) scientists with recent Research Project Grant Program (R01) grant funding from the National Institutes of Health (NIH). Methods:  The curricula vitae of all EP scientists in the United States currently funded by the NIH were analyzed for evidence of advanced research training and frequency and type of publication and grant writing. Each investigator was surveyed for demographic features and estimation of protected time during their early career development. Results:  Eighteen investigators were identified. The median length of time from completion of residency to receipt of their first R01 grant was 11 years (interquartile range [IQR] = 11 to 15 years), and the median age of investigators at the time of this award was 43 years (IQR = 39 to 47 years). At the time of their award, researchers were publishing five peer‐reviewed manuscripts a year (IQR = 1 to 8 manuscripts) and had already received considerable external funding. Ninety‐four percent of those studied had pursued a research fellowship, an advanced degree, or an NIH K‐award following residency. Conclusions:  For EPs, receipt of an R01 from the NIH requires more than a decade of work following the completion of training.     Articulating the Value of Our Daily Work: An Initial Discussion of the Assessment Challenges of Engineering Librarians

    Engineering librarians need to assess the effectiveness of our library instruction and outreach for many reasons, including communicating library value to institutional stakeholders and making impactful contributions to the scholarly literature. However, as practitioners, most librarians have not been formally educated in research design, data collection, and data analysis. To increase our skills and knowledge and to better align with various publication expectations and guidelines (e.g., ELD Author Guidelines), this panel will lead a discussion on library assessment needs with regard to research design, data collection, data analysis, and dissemination and discovery. The goal of the panel is to facilitate a conversation regarding librarian assessment challenges and needs to design a future ASEE workshop. The case studies here show how Knovel Corporation, a sci-tech e-book vendor, has worked with librarians at large and small institutions and with faculty members to promote awareness and usage of Knovel’s interactive library of electronic books for science and engineering education. Librarians and Knovel partner at a large institution like Drexel University to give on-site workshops for faculty and students. At these workshops important features of Knovel’s content and software are discussed and demonstrated in a live format. Questions are encouraged to ensure participants understand the technology, and short assignments reinforce learning. Knovel works with librarians to promote these sessions and continues the dialogue with attendees afterward. On-site demonstrations are one of the most effective ways to educate users about new technologies and new content. Librarians at smaller institutions often do not have the time and resources to devote to a full-scale on-site workshop. They need an "out-of-the-box" solution that is quick to deploy with an interface that is self-evident to users. Complete integration of e-book titles into the institution’s information toolbox is the key here. Knovel provides resources to promote ready access to meet the needs of smaller institutions, including MARC records for the library's catalog. Finally, librarians and Knovel work directly with faculty to integrate Knovel's content into their courses. An example is Purdue University’s "treasure hunt" assignment given in the Mechanical Engineering Technology program which includes questions which leverage Knovel titles to answer. By supporting outreach and access strategies appropriate for institution size and need, librarians and Knovel illustrate how to create an effective partnership with engineering libraries to increase awareness, usage and, ultimately, cost-effectiveness of electronic resources

    Aspergillus fumigatus Triggers Inflammatory Responses by Stage-Specific β-Glucan Display

    Inhalation of fungal spores (conidia) occurs commonly and, in specific circumstances, can result in invasive disease. We investigated the murine inflammatory response to conidia of Aspergillus fumigatus, the most common invasive mold in immunocompromised hosts. In contrast to dormant spores, germinating conidia induce neutrophil recruitment to the airways and TNF-α/MIP-2 secretion by alveolar macrophages. Fungal β-glucans act as a trigger for the induction of these inflammatory responses through their time-dependent exposure on the surface of germinating conidia. Dectin-1, an innate immune receptor that recognizes fungal β-glucans, is recruited in vivo to alveolar macrophage phagosomes that have internalized conidia with exposed β-glucans. Antibody-mediated blockade of Dectin-1 partially inhibits TNF-α/MIP-2 induction by metabolically active conidia. TLR-2- and MyD88-mediated signals provide an additive contribution to macrophage activation by germinating conidia. Selective responsiveness to germinating conidia provides the innate immune system with a mechanism to restrict inflammatory responses to metabolically active, potentially invasive fungal spores

    Opportunities for improving animal welfare in rodent models of epilepsy and seizures

    Animal models of epilepsy and seizures, mostly involving mice and rats, are used to understand the pathophysiology of the different forms of epilepsy and their comorbidities, to identify biomarkers, and to discover new antiepileptic drugs and treatments for comorbidities. Such models represent an important area for application of the 3Rs (replacement, reduction and refinement of animal use). This report provides background information and recommendations aimed at minimising pain, suffering and distress in rodent models of epilepsy and seizures in order to improve animal welfare and optimise the quality of studies in this area. The report includes practical guidance on principles of choosing a model, induction procedures, in vivo recordings, perioperative care, welfare assessment, humane endpoints, social housing, environmental enrichment, reporting of studies and data sharing. In addition, some model-specific welfare considerations are discussed, and data gaps and areas for further research are identified. The guidance is based upon a systematic review of the scientific literature, survey of the international epilepsy research community, consultation with veterinarians and animal care and welfare officers, and the expert opinion and practical experience of the members of a Working Group convened by the United Kingdom's National Centre for the Replacement, Refinement and Reduction of Animals in Research (NC3Rs)

    The Palomar Testbed Interferometer Calibrator Catalog

    The Palomar Testbed Interferometer (PTI) archive of observations between 1998 and 2005 is examined for objects appropriate for calibration of optical long-baseline interferometer observations - stars that are predictably point-like and single. Approximately 1,400 nights of data on 1,800 objects were examined for this investigation. We compare those observations to an intensively studied object that is a suitable calibrator, HD217014, and statistically compare each candidate calibrator to that object by computing both a Mahalanobis distance and a Principal Component Analysis. Our hypothesis is that the frequency distribution of visibility data associated with calibrator stars differs from non-calibrator stars such as binary stars. Spectroscopic binaries resolved by PTI, objects known to be unsuitable for calibrator use, are similarly tested to establish detection limits of this approach. From this investigation, we find more than 350 observed stars suitable for use as calibrators (with an additional 140\approx 140 being rejected), corresponding to 95\gtrsim 95% sky coverage for PTI. This approach is noteworthy in that it rigorously establishes calibration sources through a traceable, empirical methodology, leveraging the predictions of spectral energy distribution modeling but also verifying it with the rich body of PTI's on-sky observations.Comment: 100 pages, 7 figures, 7 tables; to appear in the May 2008ApJS, v176n

    synaptojanin1 Is Required for Temporal Fidelity of Synaptic Transmission in Hair Cells

    To faithfully encode mechanosensory information, auditory/vestibular hair cells utilize graded synaptic vesicle (SV) release at specialized ribbon synapses. The molecular basis of SV release and consequent recycling of membrane in hair cells has not been fully explored. Here, we report that comet, a gene identified in an ENU mutagenesis screen for zebrafish larvae with vestibular defects, encodes the lipid phosphatase Synaptojanin 1 (Synj1). Examination of mutant synj1 hair cells revealed basal blebbing near ribbons that was dependent on Cav1.3 calcium channel activity but not mechanotransduction. Synaptojanin has been previously implicated in SV recycling; therefore, we tested synaptic transmission at hair-cell synapses. Recordings of post-synaptic activity in synj1 mutants showed relatively normal spike rates when hair cells were mechanically stimulated for a short period of time at 20 Hz. In contrast, a sharp decline in the rate of firing occurred during prolonged stimulation at 20 Hz or stimulation at a higher frequency of 60 Hz. The decline in spike rate suggested that fewer vesicles were available for release. Consistent with this result, we observed that stimulated mutant hair cells had decreased numbers of tethered and reserve-pool vesicles in comparison to wild-type hair cells. Furthermore, stimulation at 60 Hz impaired phase locking of the postsynaptic activity to the mechanical stimulus. Following prolonged stimulation at 60 Hz, we also found that mutant synj1 hair cells displayed a striking delay in the recovery of spontaneous activity. Collectively, the data suggest that Synj1 is critical for retrieval of membrane in order to maintain the quantity, timing of fusion, and spontaneous release properties of SVs at hair-cell ribbon synapses

    Impaired Release of Antimicrobial Peptides into Nasal Fluid of Hyper-IgE and CVID Patients

    Patients with primary immunodeficiency (PID) often suffer from frequent respiratory tract infections. Despite standard treatment with IgG-substitution and antibiotics many patients do not improve significantly. Therefore, we hypothesized that additional immune deficits may be present among these patients.To investigate if PID patients exhibit impaired production of antimicrobial peptides (AMPs) in nasal fluid and a possible link between AMP-expression and Th17-cells.Nasal fluid, nasopharyngeal swabs and peripheral blood mononuclear cells (PBMCs) were collected from patients and healthy controls. AMP levels were measured in nasal fluid by Western blotting. Nasal swabs were cultured for bacteria. PBMCs were stimulated with antigen and the supernatants were assessed for IL-17A release by ELISA.In healthy controls and most patients, AMP levels in nasal fluid were increased in response to pathogenic bacteria. However, this increase was absent in patients with common variable immunodeficiency (CVID) and Hyper-IgE syndrome (HIES), despite the presence of pathogenic bacteria. Furthermore, stimulation of PBMCs revealed that both HIES and CVID patients exhibited an impaired production of IL-17A.CVID and HIES patients appear to have a dysregulated AMP response to pathogenic bacteria in the upper respiratory tract, which could be linked to an aberrant Th17 cell response
