    HISTOLOGICAL AND IMMUNOHISTOCHEMICAL PHENOTYPE CORRELATION WITH TUMOR MARKERS IN PATIENTS WITH TESTICULAR NEOPLASIA

Cancerul testicular cuprinde diverse neoplazii, în funcție de celula de origine și de vârsta tipică de prezentare. Deși mai multe tipuri de celule din testicule pot suferi transformări neoplazice, tumorile derivate din celulele germinale constituie marea majoritate a cazurilor de neoplasme testiculare. Gonadotropina corionică umană, alfa fetoproteina și lactat dehidrogenaza joacă un rol crucial în diagnosticarea, stadializarea, prognosticul, monitorizarea răspunsului la tratament și supravegherea tumorilor germinale seminomatoase și neseminomatoase. Scopul lucrării. Corelarea fenotipului histologic cu valorile markerilor tumorali. Material și metode. Studiul retrospectiv a fost realizat pentru perioada 2018 – 2023, prin identificarea a 40 de bărbați din cadrul Cancer Registru IO, cu vârsta medie de 27,64±7,76, supuși intervențiilor de orhectomie pentru tumori testiculare. Valorile markerilor tumorali beta HCG, AFP și LDH au fost analizate în raport cu rezultatul histologic și imunohistochimic (SALL-4, OCT-4, PLAP, CD30, PanCK, CD117). Rezultate. Afectarea testicolului stâng a predominat în lotul studiat 21 (52,5%) vs 19 (47,5%). Toți pacienții au prezentat neoplazii unilaterale, efectuându-se orhectomia cu excizia funiculului spermatic. Din variantele histologice, 13 (32,5%) au fost diagnosticați cu tumori mixte, 9 (22,5%) Seminoame, 6 (15%) Carcinom Embrionar și 12 (30%) pacienți au fost diagnosticați cu alte tipuri de tumori (Leiomiosarcom, Teratom, Coriocarcinom, etc.). Valorile medii ale beta HCG au constituit 1749,71 și AFP – 828,44. Valoarea maximă a AFP si beta HCG s-a determinat în cazul unei tumori mixte (carcinom embrionar 70%, teratom 20% și tumora de sac vitelin 10%). 12 (46,15%) au prezentat valori normale ale AFP și 7 (24,13%) pentru beta HCG. Per ansamblu, cele mai înalte valori ale beta HCG au fost caracteristice tumorilor cu conținut histologic de carcinom embrionar. Concluzii. Valorile markerilor oncologici au o valorea predictivă redusă pentru fenotipul histologic.Background. Testicular cancer encompasses various neoplasms, depending on the cell of origin and the typical age of presentation. Although several types of testicular cells can undergo neoplastic mutation, tumors derived from germ cells represent the vast majority of testicular neoplasms. Human chorionic gonadotropin, alpha-fetoprotein and lactate dehydrogenase play a crucial role in the diagnosis, staging, prognosis, monitoring of treatment response and surveillance of seminomatous and non-seminomatous germ tumors. Objective of the study. Histological phenotype correlation with tumor marker values. Material and methods. The retrospective study was carried out for the period 2018 - 2023, by identifying 40 men from the PMHI Oncology Institute Cancer Registry, with an average age of 27.64±7.76, undergoing orchectomy interventions for testicular tumors. The values of tumor markers beta HCG, AFP and LDH were analyzed in relation to the histological and immunohistochemical result (SALL-4, OCT-4, PLAP, CD30, PanCK, CD117). Results. Damage of the left testicle prevails in the studied group 21 (52.5%) vs 19 (47.5%). All patients presented with unilateral neoplasia, were performed orchectomy with excision of the spermatic cord. Of all the histological variants, 13 (32.5%) were diagnosed with mixed tumors, 9 (22.5%) Seminomas, 6 (15%) Embryonal Carcinoma and 12 (30%) patients were diagnosed with other types of tumors (Leiomyosarcoma, Teratoma, Choriocarcinoma, etc.). The mean values of beta HCG were 1749.71 and AFP – 828.44. The highest value of AFP and beta HCG was determined in the case of a mixed tumor (embryonic carcinoma 70%, teratoma 20% and yolk sac tumor 10%). 12 (46.15%) showed normal values for AFP and 7 (24.13%) for beta HCG. Overall, the highest values of beta HCG were characteristic of tumors with histological variant of embryonal carcinoma. Conclusions. The values of oncological markers have a low predictive value for the histological phenotype

El capítulo 3 describe la planificación del proyecto, la metodología usada, la estimación de recursos y costes y seguimiento del proyecto.     Elaboration of biodegradable whey isolate film incorporated with green synthesized silver nanoparticles

    Finding a feasible alternative to reduce the use of conventional polymers in the plastic sector has become a top priority, since industrially generated plastic waste, mostly traditional food packaging, has turned into a global environmental disaster. The aim of the study was the preparation and characterization of biodegradable films based on whey protein isolate, functionalized with green synthesized nanoparticles. Green silver nanoparticles (AgNPs) were synthesized using cinnamon extract as both capping and reducing agent. Biodegradable nanocomposite blend films based on silver nanoparticles and whey protein isolate (WPI) were made by casting denatured WPI film solutions incorporated with green synthesized silver nanoparticles. The film thickness and water vapor permeability (WVP) of both control and AgNPs were determined. The biodegradable films based on whey and (1%) green synthesized nanoparticles have potential application as active food packaging in the cheese industry
