    Identification of QTLs and candidate genes for high grain Fe and Zn concentration in sorghum [Sorghum bicolor (L.)Moench]

    Sorghum is a major food crop in the semi-arid tropics of Africa and Asia. Enhancing the grain iron (Fe) and zinc (Zn) concentration in sorghum using genetic approaches would help alleviate micronutrient malnutrition in millions of poor people consuming sorghum as a staple food. To localize genomic regions associated with grain Fe and Zn, a sorghum F6 recombinant inbred line (RIL) population (342 lines derived from cross 296B PVK 801) was phenotyped in six environments, and genotyped with simple sequence repeat (SSR), DArT (Diversity Array Technology) and DArTSeq (Diversity Array Technology) markers. Highly significant genotype environment interactions were observed for both micronutrients. Grain Fe showed greater variation than Zn. A sorghum genetic map was constructed with 2088 markers (1148 DArTs, 927 DArTSeqs and 13 SSRs) covering 1355.52 cM with an average marker interval of 0.6 cM. Eleven QTLs (individual) and 3 QTLs (across) environments for Fe and Zn were identified. We identified putative candidate genes from the QTL interval of qfe7.1, qzn7.1, and qzn7.2 (across environments) located on SBI-07 involved in Fe and Zn metabolism. These were CYP71B34, and ZFP 8 (ZINC FINGER PROTEIN 8). After validation, the linked markers identified in this study can help in developing high grain Fe and Zn sorghum cultivars in sorghum improvement programs globally

    Fatigue behaviour of SiC p -reinforced aluminium composites in the very high cycle regime using ultrasonic fatigue

    The fatigue behaviour of a 2009/SiC/15p-T4 DRA composite has been examined in the very high cycle fatigue (VHCF) regime where 10 7 ≤ N f ≤ 10 9 cycles. Ultrasonic fatigue was used to achieve the very high cycle counts. Careful processing yielded a composite with a very homogeneous particle distribution with minimal clustering. Fatigue crack initiation was observed almost exclusively at AlCuFe inclusions with no crack initiation observed at SiC particle clusters. Fatigue lives at a given stress level exhibited minimal scatter and subsurface crack initiation was observed in all cases.     Seroprevalence of brucellosis in buffaloes in North India

    Abstract Brucellosis is an infectious disease characterized by abortion and infertility in several mammalian species including buffaloes, and is one of the most important zoonosis worldwide. The objective of present study was to establish the seroprevalence of brucellosis in buffaloes in North India and to know the exact causal species of Brucella organism. A total of 160 serum samples were analyzed. The seroprevalence of Brucellosis was 4.38% and 7.50% by Rose Bengal Plate Test (RBPT) and Standard Tube Agglutination Test (STAT), respectively. Seroprevalence was zero by both RBPT and STAT in organized dairy farm while in unorganized sector, 5.04 and 8.63% by RBPT and STAT, respectively. Age-wise seroprevalence was higher in 4.5 to 6 years old buffaloes than younger. Area-wise seroprevalence was higher in Satwari block of study area. On screening of 20 milk samples from serological positive and doubtful buffaloes by milk ring test, five samples were positive (25%). On isolation of from samples, only one sample yielded Brucella abortus. The results showed that younger buffaloes were less infected than adults. Prevalence of Brucellosis was higher in Satwari block of Jammu province compared to other study area. The presence of Brucellosis in bovine may pose a significant economic loss to the farmer and a public health hazard to the general population.     Time-Dependent Fatigue Crack Propagation Behavior of Two Solid-Solution-Strengthened Ni-Based Superalloys—INCONEL 617 and HAYNES 230

    The fatigue crack propagation (FCP) as well as the sustained loading crack growth (SLCG) behavior of two solid-solution-strengthened Ni-based superalloys, INCONEL 617 (Special Metals Corporation Family of Companies) and HAYNES 230 (Haynes International, Inc., Kokomo, IN), were studied at increased temperatures in laboratory air under a constant stress-intensity- factor (K) condition. The crack propagation tests were conducted using a baseline cyclic triangular waveform with a frequency of 1 3 Hz. Various hold times were imposed at the maximum load of a fatigue cycle to study the hold time effect. The results show that a linear elastic fracture mechanics (LEFM) parameter, stress intensity factor (K), is sufficient to describe the FCP and SLCG behavior at the testing temperatures ranging from 873 K to 1073 K (600 C to 800 C). As observed in the precipitation-strengthened superalloys, both INCONEL 617 and HAYNES 230 exhibited the time-dependent FCP, steady SLCG behavior, and existence of a damage zone ahead of crack tip. A thermodynamic equation was adapted to correlate the SLCG rates to determine thermal activation energy. The fracture modes associated with crack propagation behavior were discussed, and the mechanism of time-dependent FCP as well as SLCG was identified. Compared with INCONEL 617, the lower crack propagation rates of HAYNES 230 under the time-dependent condition were ascribed to the different fracture mode and the presence of numerous W-rich M6C-type and Cr-rich M23C6-type carbides. Toward the end, a phenomenological model was employed to correlate the FCP rates at cycle/time-dependent FCP domain. All the results suggest that an environmental factor, the stress assisted grain boundary oxygen embrittlement (SAGBOE) mechanism, is mainly responsible for the accelerated time dependent FCP rates of INCONEL 617 and HAYNES 230

    Taxane-Platin-Resistant Lung Cancers Co-develop Hypersensitivity to JumonjiC Demethylase Inhibitors

    Although non-small cell lung cancer (NSCLC) patients benefit from standard taxane-platin chemotherapy, many relapse, developing drug resistance. We established preclinical taxane-platin-chemoresistance models and identified a 35-gene resistance signature, which was associated with poor recurrence-free survival in neoadjuvant-treated NSCLC patients and included upregulation of the JumonjiC lysine demethylase KDM3B. In fact, multi-drug-resistant cells progressively increased the expression of many JumonjiC demethylases, had altered histone methylation, and, importantly, showed hypersensitivity to JumonjiC inhibitors in vitro and in vivo. Increasing taxane-platin resistance in progressive cell line series was accompanied by progressive sensitization to JIB-04 and GSK-J4. These JumonjiC inhibitors partly reversed deregulated transcriptional programs, prevented the emergence of drug-tolerant colonies from chemo-naive cells, and synergized with standard chemotherapy in vitro and in vivo. Our findings reveal JumonjiC inhibitors as promising therapies for targeting taxane-platin-chemoresistant NSCLCs.Fil: Dalvi, Maithili P.. University of Texas.     Effect of AGM and Fetal Liver-Derived Stromal Cell Lines on Globin Expression in Adult Baboon (P. anubis) Bone Marrow-Derived Erythroid Progenitors

    This study was performed to investigate the hypothesis that the erythroid micro-environment plays a role in regulation of globin gene expression during adult erythroid differentiation. Adult baboon bone marrow and human cord blood CD34+ progenitors were grown in methylcellulose, liquid media, and in co-culture with stromal cell lines derived from different developmental stages in identical media supporting erythroid differentiation to examine the effect of the micro-environment on globin gene expression. Adult progenitors express high levels of γ-globin in liquid and methylcellulose media but low, physiological levels in stromal cell co-cultures. In contrast, γ-globin expression remained high in cord blood progenitors in stromal cell line co-cultures. Differences in γ-globin gene expression between adult progenitors in stromal cell line co-cultures and liquid media required cell-cell contact and were associated with differences in rate of differentiation and γ-globin promoter DNA methylation. We conclude that γ-globin expression in adult-derived erythroid cells can be influenced by the micro-environment, suggesting new potential targets for HbF induction

    Emergence and Evolution of Cooperation Under Resource Pressure

    We study the influence that resource availability has on cooperation in the context of hunter-gatherer societies. This paper proposes a model based on archaeological and ethnographic research on resource stress episodes, which exposes three different cooperative regimes according to the relationship between resource availability in the environment and population size. The most interesting regime represents moderate survival stress in which individuals coordinate in an evolutionary way to increase the probabilities of survival and reduce the risk of failing to meet the minimum needs for survival. Populations self-organise in an indirect reciprocity system in which the norm that emerges is to share the part of the resource that is not strictly necessary for survival, thereby collectively lowering the chances of starving.
