    Abstract. This thesis studies the possibilities to automate UVM testbench creation in the telecommunications industry. First, the ideas behind UVM are looked at and automatable parts of the testbench coding process are studied. Facilitating the reuse of code is also examined.     Topological descriptors for 3D surface analysis

    We investigate topological descriptors for 3D surface analysis, i.e. the classification of surfaces according to their geometric fine structure. On a dataset of high-resolution 3D surface reconstructions we compute persistence diagrams for a 2D cubical filtration. In the next step we investigate different topological descriptors and measure their ability to discriminate structurally different 3D surface patches. We evaluate their sensitivity to different parameters and compare the performance of the resulting topological descriptors to alternative (non-topological) descriptors. We present a comprehensive evaluation that shows that topological descriptors are (i) robust, (ii) yield state-of-the-art performance for the task of 3D surface analysis and (iii) improve classification performance when combined with non-topological descriptors.Comment: 12 pages, 3 figures, CTIC 201

    Correlating Pedestrian Flows and Search Engine Queries

    An important challenge for ubiquitous computing is the development of techniques that can characterize a location vis-a-vis the richness and diversity of urban settings. In this paper we report our work on correlating urban pedestrian flows with Google search queries. Using longitudinal data we show pedestrian flows at particular locations can be correlated with the frequency of Google search terms that are semantically relevant to those locations.     Biogeochemical cycling and ecological thresholds in a High Arctic lake (Svalbard)

    Lakes are a dominant feature of the Arctic landscape and a focal point of regional and global biogeochemical cycling. We collected a sediment core from a High Arctic Lake in southwestern Svalbard for multiproxy paleolimnological analysis. The aim was to find linkages between the terrestrial and aquatic environments in the context of climate change to understand centennial-long Arctic biogeochemical cycling and environmental dynamics. Two significant thresholds in elemental cycling were found based on sediment physical and biogeochemical proxies that were associated with the end of the cold Little Ice Age and the recent warming. We found major shifts in diatom, chironomid and cladoceran communities and their functionality that coincided with increased summer temperatures since the 1950s. We also discovered paleoecological evidence that point toward expanded bird (Little Auk) colonies in the catchment alongside climate warming. Apparently, climate-driven increase in glacier melt water delivery as well as a prolonged snow- and ice-free period have increased the transport of mineral matter from the catchment, causing significant water turbidity and disappearance of several planktonic diatoms and clear-water chironomids. We also found sedimentary accumulation of microplastic particles following the increase in Little Auk populations suggesting that seabirds potentially act as biovectors for plastic contamination. Our study demonstrates the diverse nature of climate-driven changes in the Arctic lacustrine environment with increased inorganic input from the more exposed catchment, larger nutrient delivery from the increased bird colonies at the surrounding mountain summits and subsequent alterations in aquatic communities.Peer reviewe

    Isoflurane cracks the polycarbonate connector of extra-corporeal circuit -A case report-

    Cardiopulmonary bypass (CPB) is widely used for cardiac surgery by virtue of its proven safety over the course of its use during the past half century. Even though perfusion is safer, incidents still occur. During the repair of a ventricular-septal defect in an 11-month-old infant, we experienced a critical incident related to the potential hazardous effect of volatile anesthetics on the polycarbonate connector of extra-corporeal circuit. The damage to the polycarbonate connector had occurred after spillage of isoflurane during the filling of the vaporizer, causing it to crack and leak. The incident was managed by replacement of the cracked connector during a temporary circulatory arrest. The patient was hypothermic and the time off bypass was less than 1.5 min. There were no neurologic sequelae, the patient made an uneventful recovery. In conclusion, the spillage of volatile anesthetics can cause cracks in the polycarbonate connector of the extra-corporeal circuit, leading to potentially interruption of CPB

    In Vivo Selection of a Computationally Designed SCHEMA AAV Library Yields a Novel Variant for Infection of Adult Neural Stem Cells in the SVZ

    Directed evolution continues to expand the capabilities of complex biomolecules for a range of applications, such as adeno-associated virus vectors for gene therapy; however, advances in library design and selection strategies are key to develop variants that overcome barriers to clinical translation. To address this need, we applied structure-guided SCHEMA recombination of the multimeric adeno-associated virus (AAV) capsid to generate a highly diversified chimeric library with minimal structural disruption. A stringent in vivo Cre-dependent selection strategy was implemented to identify variants that transduce adult neural stem cells (NSCs) in the subventricular zone. A novel variant, SCH9, infected 60% of NSCs and mediated 24-fold higher GFP expression and a 12-fold greater transduction volume than AAV9. SCH9 utilizes both galactose and heparan sulfate as cell surface receptors and exhibits increased resistance to neutralizing antibodies. These results establish the SCHEMA library as a valuable tool for directed evolution and SCH9 as an effective gene delivery vector to investigate subventricular NSCs

    Multifrequency Observations of the Gamma-Ray Blazar 3C 279 in Low-State during Integral AO-1

    We report first results of a multifrequency campaign from radio to hard X-ray energies of the prominent gamma-ray blazar 3C 279 during the first year of the INTEGRAL mission. The variable blazar was found at a low activity level, but was detected by all participating instruments. Subsequently a multifrequency spectrum could be compiled. The individual measurements as well as the compiled multifrequency spectrum are presented.
