    Elimination of Potential Pathogenic Microorganisms in Sewage Sludge Using Electron Beam Irradiation

    Microbiological analyses on municipal sewage sludge sample treated in a pilot plant process utilizing an electron accelerator with a beam energy of 3 MeV were conducted as a way to show the potential of this technology to decontaminate sludge containing 15% solids. Bacterial counts including total heterotrophic bacterial, total coliform, and fecal coliform counts were performed on sewage sludge samples pre- and postirradiation with the electron beam at doses ranging between 2.7 and 30.7 kGy. At each irradiation dose, bacterial and Ascaris ova counts and survival were measured in triplicate as colony forming units (CFUs) per milliliter (ml) of sewage sludge. Experimental results obtained revealed that a dose of 6.7 kGy is enough to reduce bacterial load to consider the treated sewage sludge safe for both the environment and human according to the Environmental Protection Agency standards. However, a dose of 25.7 kGy was needed to reduce the concentration of Ascaris ova at levels deemed safe for land applications. This study also showed that electron beam treatment is less energy consuming with shorter processing times than conventional techniques used to decontaminate sludge. Taken altogether, these observations open new avenues for large urban agglomerations to save money on sewage sludge treatment

    Fluence Uniformity Measurements in an Electron Accelerator Used for Irradiation of Extended Area Solar Cells and Electronic Circuits for Space Applications

    It is common to have liquid crystal displays and electronic circuit boards with area sizes of the order of 20x20 sq cm on board of satellites and space vehicles. Usually irradiating them at different fluence values assesses the radiation damage in these types of devices. As a result, there is a need for a radiation source with large spatial fluence uniformity for the study of the damage by radiation from space in those devices. Kent State University s Program on Electron Beam Technology has access to an electron accelerator used for both research and industrial applications. The electron accelerator produces electrons with energies in the interval from 1 to 5 MeV and a maximum beam power of 150 kW. At such high power levels, the electron beam is continuously scanned back and forth in one dimension in order to provide uniform irradiation and to prevent damage to the sample. This allows for the uniform irradiation of samples with an area of up to 1.32 sq m. This accelerator has been used in the past for the study of radiation damage in solar cells (1). However in order to irradiate extended area solar cells there was a need to measure the uniformity of the irradiation zone in terms of fluence. In this paper the methodology to measure the fluence uniformity on a sample handling system (linear motion system), used for the irradiation of research samples, along the irradiation zone of the above-mentioned facility is described and the results presented. We also illustrate the use of the electron accelerator for the irradiation of large area solar cells (of the order of 156 sq cm) and include in this paper the electrical characterization of these types of solar cells irradiated with 5 MeV electrons to a total fluence of 2.6 x 10(exp 15) e/sq cm

    Variación latitudinal del ciclo reproductivo de dos bivalvos con orígenes biogeográficos contrastantes a lo largo del ecosistema de afloramiento de la Corriente de Humboldt

    Argopecten purpuratus (Lamarck, 1819) and Mesodesma donacium (Lamarck, 1818) are bivalves that inhabit the Humboldt Current Upwelling Ecosystem. They have contrasting biogeographical origins, suggesting that their responses to exogenous factors should differ. Using circular statistics, we examine synchrony/asynchrony in the reproductive cycle between populations of each species. The results indicate that there is reproductive asynchrony in both species along their distributional range. However, there was synchrony for A. purpuratus in several location-pairs, including Paita-Chimbote, Chimbote-Callao, Callao-Pisco and Pisco-Antofagasta. For M. donacium, there were only two synchronic groups: Camaná-Capellanía-Mehuín and Hornitos-Peñuelas-Longotoma-La Ligua-Cucao-Quilanlar. A. purpuratus showed gametogenenic activity throughout the year. In contrast, M. donacium showed strong seasonality, with gametogenesis in winter and spawning in spring/summer. In conclusion, the patterns observed for these sympatric species suggest that on a large scale the reproductive cycles follow the expected patterns for the contrasting biogeographic origin of each species, so it could be argued that they are modulated by endogenous factors. However, at a local scale, the reproductive cycles of these species show variation, likely determined by local oceanographic or hydrographic processes.Argopecten purpuratus (Lamarck, 1819) y Mesodesma donacium (Lamarck, 1818) son moluscos bivalvos endémicos del sistema de afloramiento de la Corriente de Humboldt pero tienen orígenes biogeográficos opuestos. Por tanto, se esperaría que los factores exógenos afectaran sus ritmos biológicos diferencialmente, entre ellos sus ciclos reproductivos. Mediante análisis de distribución circular se evaluó el sincronismo/asincronismo en ciclos reproductivos entre poblaciones de cada especie. Los resultados indican que existe asincronía reproductiva en ambas especies a lo largo de su distribución. Sin embargo para A. purpuratus se encontró varios pares de localidades sincrónicas: Paita-Chimbote; Chimbote-Callao; Callao-Pisco y Pisco-Antofagasta, y para M. donacium se dieron dos grupos sincrónicos, el primero entre las poblaciones de Camaná, Capellanía y Mehuín; y el segundo grupo entre Hornitos, Peñuelas, Longotoma, La Ligua, Cucao y Quilanlar. A. purpuratus presentó actividad gametogénica continua. En contraste, M. donacium presentó una marcada estacionalidad, con gametogénesis durante invierno y desoves durante primavera y/o verano. En conclusión, los patrones observados para ambas especies simpátricas sugieren que, a gran escala el ciclo reproductivo sigue los patrones esperados para el origen biogeográfico contrastante de cada especie, por lo tanto se podría argumentar que es modulada por factores endógenos. Sin embargo, a escala local el ciclo reproductivo de ambas especies muestra variaciones, aparentemente influenciados por los factores oceanográficos o hidrográficos locales

Publisher Copyright: © 2022 by the authors.Myotonic dystrophy type 1 (DM1) is an autosomal dominant disease caused by a CTG repeat expansion in the 3′ untranslated region of the dystrophia myotonica protein kinase gene. AKT dephosphorylation and autophagy are associated with DM1. Autophagy has been widely studied in DM1, although the endocytic pathway has not. AKT has a critical role in endocytosis, and its phosphorylation is mediated by the activation of tyrosine kinase receptors, such as epidermal growth factor receptor (EGFR). EGF-activated EGFR triggers the internalization and degradation of ligand–receptor complexes that serve as a PI3K/AKT signaling platform. Here, we used primary fibroblasts from healthy subjects and DM1 patients. DM1-derived fibroblasts showed increased autophagy flux, with enlarged endosomes and lysosomes. Thereafter, cells were stimulated with a high concentration of EGF to promote EGFR internalization and degradation. Interestingly, EGF binding to EGFR was reduced in DM1 cells and EGFR internalization was also slowed during the early steps of endocytosis. However, EGF-activated EGFR enhanced AKT and ERK1/2 phosphorylation levels in the DM1-derived fibroblasts. Therefore, there was a delay in EGF-stimulated EGFR endocytosis in DM1 cells; this alteration might be due to the decrease in the binding of EGF to EGFR, and not to a decrease in AKT phosphorylation.publishersversionpublishe

    Incident Use of Hydroxychloroquine for the Treatment of Rheumatoid Arthritis and Systemic Lupus Erythematosus During the COVID‐19 Pandemic

    Objective: We studied whether the use of hydroxychloroquine (HCQ) for COVID‐19 resulted in supply shortages for patients with rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE). Methods: We used US claims data (IQVIA PHARMETRICS® Plus for Academics [PHARMETRICS]) and hospital electronic records from Spain (Institut Municipal d'Assistència Sanitària Information System [IMASIS]) to estimate monthly rates of HCQ use between January 2019 and March 2022, in the general population and in patients with RA and SLE. Methotrexate (MTX) use was estimated as a control. Results: More than 13.5 million individuals (13,311,811 PHARMETRICS, 207,646 IMASIS) were included in the general population cohort. RA and SLE cohorts enrolled 135,259 and 39,295 patients, respectively, in PHARMETRICS. Incidence of MTX and HCQ were stable before March 2020. On March 2020, the incidence of HCQ increased by 9‐ and 67‐fold in PHARMETRICS and IMASIS, respectively, and decreased in May 2020. Usage rates of HCQ went back to prepandemic trends in Spain but remained high in the United States, mimicking waves of COVID‐19. No significant changes in HCQ use were noted among patients with RA and SLE. MTX use rates decreased during HCQ approval period for COVID‐19 treatment. Conclusion: Use of HCQ increased dramatically in the general population in both Spain and the United States during March and April 2020. Whereas Spain returned to prepandemic rates after the first wave, use of HCQ remained high and followed waves of COVID‐19 in the United States. However, we found no evidence of general shortages in the use of HCQ for both RA and SLE in the United States

    Can scenario-planning support community-based natural resource management? Experiences from three countries in latin america

    Community Based Natural Resource Management (CBNRM) is a concept critical to managing socio-ecological systems, but whose implementation needs strengthening. Scenario-planning is one approach that may offer benefits relevant to CBNRM, but whose potential is not yet well understood. We therefore designed, trialled and evaluated a scenario-planning method intended to support CBNRM at three cases, located in Colombia, Mexico and Argentina. Implementing scenario-planning was judged as worthwhile in all three cases, although aspects of it could be challenging to facilitate. The benefits generated were relevant to strengthening CBNRM: encouraging the participation of local people and using their knowledge; enhanced consideration and adaption of future change; and supporting the development of systems thinking. Tracing exactly when and how these benefits arise is challenging, but two elements of the method seemed particularly useful. Firstly, using a systematic approach to discuss how drivers of change may affect local socio-ecological systems helped to foster systems thinking and identify connections between issues. Secondly, explicitly focusing on how to use and respond to scenarios helped identify specific practical activities ('response options') that would support CBNRM despite the pressures of future change. Discussions about response options also highlighted the need for support by other actors (e.g. policy groups): this raises the question of when and how other actors and other sources of knowledge should be involved in scenario-planning, so as to encourage their buy-in to actions identified by the process. We suggest other CBNRM initiatives may benefit from adapting and applying scenario-planning. However, these initiatives should be carefully monitored since further research is required to understand how and when scenario-planning methods may produce benefits, and their strengths and weaknesses versus other methods

    The effects of essential oil from Lippia origanoides and herbal betaine on performance, intestinal integrity, bone mineralization and meat quality in broiler chickens subjected to cyclic heat stress

    Essential oils (EO) affect performance, intestinal integrity, bone mineralization, and meat quality in broiler chickens subjected to cyclic heat stress (HS). Day-of-hatch Cobb 500 male broiler chicks (n = 475) were randomly divided into four groups. Group 1: No heat stress (Thermoneutral) + control diets with no antibiotics; Group 2: heat stress control + control diets; Group 3: heat stress + control diets supplemented with thymol chemotype (45 ppm) and herbal betaine (150 ppm) formulation EO1; Group 4: heat stress + control diets supplemented with phellandrene (45 ppm) and herbal betaine (150 ppm) formulation EO2. From day 10–42, the heat stress groups were exposed to cyclic HS at 35°C for 12 h (8:00–20:00). BW, BWG, FI, and FCRc were measured at d 0, 10, 28, and 42. Chickens were orally gavaged with FITC-d on days 10 (before heat stress) and 42. Morphometric analysis of duodenum and ileum samples and bone mineralization of tibias were done. Meat quality was assessed on day 43 with ten chickens per pen per treatment. Heat stress reduced BW by day 28 (p &lt; 0.05) compared to thermoneutral chickens. At the end of the trial, chickens that received both formulations of EO1 and EO2 had significantly higher BW than HS control chickens. A similar trend was observed for BWG. FCRc was impaired by EO2 supplementation. There was a significant increase in total mortality in EO2 compared with EO1 EO1 chickens had lower FITC-d concentrations at day 42 than the HS control. In addition, EO1 treatment is not statistically different if compared to EO2 and thermoneutral. Control HS broilers had significantly lower tibia breaking strength and total ash at day 42 than heat-stressed chickens supplemented with EO1 and EO2. Heat stress affected intestinal morphology more than thermoneutral chickens. EO1 and EO2 improved intestinal morphology in heat-stressed chickens. Woody breast and white striping were more common in thermoneutral chickens than heat stress chickens. In conclusion, the EO-containing diet could improve broiler chicken growth during cyclic heat stress, becoming increasingly relevant in antibiotic-free production in harsh climates
