    Experimental Pulmonary Granuloma Mimicking Sarcoidosis Induced by Propionibacterium acnes in Mice

    Propionibacterium acnes has been implicated as an etiologic agent of sarcoidosis since the isolation of this bacterium from sarcoid lesions. We experimentally produced a murine pulmonary granuloma model using P. acnes with several features that simulate sarcoidosis. Mice were sensitized with heat-killed P. acnes and complete Freund's adjuvant and were subsequently challenged with heat-killed P. acnes at 2-week intervals. P. acnes-challenged mice developed epitheloid cell granulomas in the lungs. These mice showed a pulmonary immune response characterized by an increased number of T-lymphocytes, especially CD4 cells, and the ratio of CD4/CD8 in bronchoalveolar lavage (BAL) fluid also increased. Furthermore, significant elevations in both angiotensin-converting enzyme (ACE) serum levels and antibody titers against P. acnes were observed. Mice sensitized with P. acnes without complete Freund's adjuvant were capable of forming pulmonary granulomas, which appeared to be caused by indigenous P. acnes. The genome of P. acnes was found in the lungs, BAL cells, hilar lymph nodes, liver, and spleen in non-sensitized mice, which were thought to be germ-free. These results suggest that the immune response against indigenous P. acnes may play an important role in the pathogenesis of granuloma formation in a murine model.</p

    Stress relaxation arrested the mainshock rupture of the 2016 Central Tottori earthquake

Additional large earthquakes typically do not occur, despite the fact that the large static stress near the edges of the fault is expected to trigger further large earthquakes at these locations. Here we analyse ~10, 000 highly accurate focal mechanism solutions of aftershocks of the 2016 Mw 6.2 Central Tottori earthquake in Japan. We determine the location of the horizontal edges of the mainshock fault relative to the aftershock hypocentres, with an accuracy of approximately 200 m. We find that aftershocks rarely occur near the horizontal edges and extensions of the fault. We propose that the mainshock rupture was arrested within areas characterised by substantial stress relaxation prior to the main earthquake. This stress relaxation along fault edges could explain why mainshocks are rarely followed by further large earthquakes

    Lavender Essential Oil and Its Main Constituents Inhibit the Expression of TNF-α-induced Cell Adhesion Molecules in Endothelial Cells

    Lavender essential oil (Lvn) has anti-inflammatory effects in an ovalbumin-sensitized murine model of asthma, and inhibits inflammatory cell infiltration into the lungs. The anti-inflammatory effects of Lvn on cell adhesion molecules are not clear. Here we evaluated the effects of Lvn and its main constituents, linalyl acetate (LA) and linalool (LO), on the expression of tumor necrosis factor-alpha (TNF-α)-induced cell adhesion molecules in murine brain endothelial bEnd.3 cells and human umbilical vein endothelial cells (HUVECs). The bEnd.3 cells were treated with Lvn, LA, or LO and subsequently stimulated with TNF-α. The mRNA expression levels of cell adhesion molecules were detected using RT-PCR. E-selectin and P-selectin protein and phosphorylated-NF-κB p65 were detected by western blotting. The effects of Lvn on HUVECs were measured by RT-PCR. In bEnd.3 cells, Lvn and LA suppressed TNF-α-induced E-selectin, P-selectin, vascular cell adhesion molecule-1, intercellular adhesion molecule-1, and phosphorylated-NF-κB p65 in the nucleus; LO did not suppress P-selectin or phosphorylated-NF-κB p65. Lvn inhibited TNF-α-induced E-selectin mRNA in HUVECs. These results indicate that Lvn and LA inhibit TNF-α-induced cell adhesion molecules in endothelial cells through the suppression of NF-κB activation. Consequently, Lvn or other essential oils including LA may be useful as alternative anti-inflammatory medicines

    Alginate–lavender nanofibers with antibacterial and anti-inflammatory activity to effectively promote burn healing

Here, we show the combined use of two naturally derived compounds, sodium alginate and lavender essential oil, for the production of bioactive nanofibrous dressings by electrospinning, and their efficacy for the treatment of skin burns induced by midrange ultraviolet radiation (UVB). We demonstrate that the engineered dressings reduce the risk of microbial infection of the burn, since they stop the growth of Staphylococcus aureus. Furthermore, they are able to control and reduce the inflammatory response that is induced in human foreskin fibroblasts by lipopolysaccharides, and in rodents by UVB exposure. In particular, we report a remarkable reduction of pro-inflammatory cytokines when fibroblasts or animals are treated with the alginate-based nanofibers. The down-regulation of cytokines production and the absence of erythema on the skin of the treated animals confirm that the here described dressings are promising as advanced biomedical devices for burn management

    The 2018 Hokkaido Eastern Iburi earthquake (M-JMA=6.7) was triggered by a strike-slip faulting in a stepover segment: insights from the aftershock distribution and the focal mechanism solution of the main shock

    The Hokkaido Eastern Iburi earthquake (MJMA = 6.7) occurred on September 6, 2018, in the Hokkaido corner region where the Kurile and northeastern Japan island arcs meet. We relocated aftershocks of this intraplate earthquake immediately after the main shock by using data from a permanent local seismic network and found that aftershock depths were concentrated from 20 to 40 km, which is extraordinarily deep compared with other shallow intraplate earthquakes in the inland area of Honshu and Kyushu, Japan. Further, we found that the aftershock area consists of three segments. The first segment is located in the northern part of the aftershock area, the second segment lies in the southern part, and the third segment forms a stepover between the other two segments. The hypocenter of the main shock, from which the rupture initiated, is located on the stepover segment. The centroid moment tensor solution for the main shock indicates a reverse faulting, whereas the focal mechanism solution determined by using the first-motion polarity of the P wave indicates strike-slip faulting. To explain this discrepancy qualitatively, we present a model in which the rupture started as a small strike-slip fault in the stepover segment of the aftershock area, followed by two large reverse faulting ruptures in the northern and southern segments
