    Phase transformation B1 to B2 in TiC, TiN, ZrC and ZrN under pressure

    Phase stability of various phases of MX (M = Ti, Zr; X = C, N) at equilibrium and under pressure is examined based on first-principles calculations of the electronic and phonon structures. The results reveal that all B1 (NaCl-type) MX structures undergo a phase transition to the B2-structures under high pressure in agreement with the previous total-energy calculations. The B1-MX structures are dynamically stable under very high pressure (210-570 GPa). The pressure-induced B2 (CsCl-type) MC phases are dynamically unstable even at high pressures, and TiN and ZrN are found to crystallize with the B2-structure only at pressures above 55 GPa. The first-order B1-to-B2 phase transition in these nitrides is not related to the softening of phonon modes, and the dynamical instability of B2-MX is associated with a high density of states at the Fermi level.Comment: 9 pages, 4 figure

    Relative seismic and tsunami risk assessment for Stromboli Island (Italy)

    An innovative method of estimating the relative risk of buildings exposed to seismic and tsunami hazards in volcanic islands is applied to Stromboli (Italy), a well-known stratovolcano affected by moderate earthquakes and mass-flow-induced tsunamis. The method uses a pre-existing quali-quantitative analysis to assess the relative risk indices of buildings, which provide comparative results useful for prioritisation purposes, in combination with a historical-geographical settlement analysis consistent with the ‘territorialist’ approach to the urban and regional planning and design. The quali-quantitative analysis is based on a new proposed survey-sheet model, useful to collect building information necessary for the relative risk estimation, whereas the historical-geographical investigation is based on the multi-temporal comparison of aerial and satellite images. The proposal to combine two consolidated methods represents an innovation in estimating relative risk. Considering that Stromboli Island had never been subjected to similar analyses, the results of the relative seismic risk assessment are novel and moreover identify buildings with a fairly-low and spatially-uniform relative risk. The results of the relative tsunami risk assessment are consistent with results of similar past studies, identifying buildings with a higher relative risk index on the northern coast of the island. The combined use of a building-by-building survey with a multi-temporal analysis of settlements allows obtaining a higher detail than previously available for the region. If adequately modified, the proposed combination of methods allows assessing relative risk also considering other geo-environmental hazards and their cascading effects, in a multi-hazard risk assessment perspective

    Occurrence of Coxiella burnetii in goat and ewe unpasteurized cheeses: Screening and genotyping

    Q fever is a zoonosis caused by Coxiella burnetii which infects humans as well as several animal species; sheep, goats and cattle are the primary animal reservoir. The main route of human exposure to Coxiella burnetii is inhalation of contaminated aerosols from excreta, especially birth products, while the role of unpasteurized dairy products in the transmission of Q fever to humans remains still controversial. The aim of this work was to evaluate the presence of Coxiella burnetii in unpasteurized cheese samples (n=84) by PCR and to genotype the circulating strains by Multispacer sequence typing (MST) analysis. Coxiella burnetii DNA was detected in 27/84 (32.14%) cheeses and positivity rate of handicraft cheeses reached 17.24%, while positivity rate of non-handicraft cheeses reached 65.38%. In addition, the MST profile of Coxiella burnetii detected in 5 cheese samples have shown the circulation of ST12 and ST32 genotypes in Tuscany

    DNA repair targeted therapy: The past or future of cancer treatment?

    The repair of DNA damage is a complex process that relies on particular pathways to remedy specific types of damage to DNA. The range of insults to DNA includes small, modest changes in structure including mismatched bases and simple methylation events to oxidized bases, intra- and interstrand DNA crosslinks, DNA double strand breaks and protein-DNA adducts. Pathways required for the repair of these lesions include mismatch repair, base excision repair, nucleotide excision repair, and the homology directed repair/Fanconi anemia pathway. Each of these pathways contributes to genetic stability, and mutations in genes encoding proteins involved in these pathways have been demonstrated to promote genetic instability and cancer. In fact, it has been suggested that all cancers display defects in DNA repair. It has also been demonstrated that the ability of cancer cells to repair therapeutically induced DNA damage impacts therapeutic efficacy. This has led to targeting DNA repair pathways and proteins to develop anti-cancer agents that will increase sensitivity to traditional chemotherapeutics. While initial studies languished and were plagued by a lack of specificity and a defined mechanism of action, more recent approaches to exploit synthetic lethal interaction and develop high affinity chemical inhibitors have proven considerably more effective. In this review we will highlight recent advances and discuss previous failures in targeting DNA repair to pave the way for future DNA repair targeted agents and their use in cancer therapy

    Prevalence, virulence and antimicrobial susceptibility of Salmonella spp., Yersinia enterocolitica and Listeria monocytogenes in european wild boar (Sus scrofa) hunted in Tuscany (central Italy)

    Wild boar is an animal the population of which constantly increases in Europe. This animal plays an important role as a reservoir for several pathogens, including three of the most important zoonoses: salmonellosis, yersiniosis and listeriosis. The aim of this investigation was to eval-uate the occurrence of antimicrobial-resistant and virulence factor genes of Salmonella spp., Yersinia enterocolitica and Listeria monocytogenes isolated from wild boar in Tuscany (Central Italy). During two consequent hunting seasons (2018/2019 and 2019/2020), rectal swabs, spleens and livers were collected from 287 hunted wild boar to isolate strains. Each isolate was tested to investigate its an-timicrobial resistance and to detect virulence factor genes by PCR. Eighteen Salmonella strains (6.27%) were isolated. Of these, 66.7% were resistant to streptomycin, 13.4% to cephalothin, 6.67% to imipenem and one isolate (6.67%) was resistant simultaneously to five antimicrobials. Moreover, the most detected genes were sopE (73.4%), pipB (66.7%), sodCI (53.3%), spvR and spvC (46.7%). In total, 54 (17.8%) Yersinia enterocolitica were isolated; of them, 26 (48.1%), 9 (16.7%), 17 (31.5%), 1 (1.85%) and 1 (1.85%) belonged to biotypes 1, 2, 3, 4 and 5, respectively. All strains (100%) demon-strated resistance to cephalothin and 70.4% to amoxicillin-clavulanic acid, 55.6% to ampicillin, and 37.0% to cefoxitin. Additionally, the most detected genes were ystA (25.9%), inv (24.1%), ail (22.2%), ystB (18.5%) and virF (14.8%). Finally, only one Listeria monocytogenes isolate (0.35%) was obtained, belonging to serogroup IVb, serovar 4b, and it was found to be resistant to cefoxitin, cefotaxime and nalidixic acid. The results highlighted the role of wild boar as a carrier for pathogenic and antimi-crobial-resistant Salmonella spp., Yersinia enterocolitica and Listeria monocytogens, representing a possible reservoir for domestic animals and human pathogens

    A tight-binding potential for atomistic simulations of carbon interacting with transition metals: Application to the Ni-C system

    We present a tight-binding potential for transition metals, carbon, and transition metal carbides, which has been optimized through a systematic fitting procedure. A minimal basis, including the s, p electrons of carbon and the d electrons of the transition metal, is used to obtain a transferable tight-binding model of the carbon-carbon, metal-metal and metal-carbon interactions applicable to binary systems. The Ni-C system is more specifically discussed. The successful validation of the potential for different atomic configurations indicates a good transferability of the model and makes it a good choice for atomistic simulations sampling a large configuration space. This approach appears to be very efficient to describe interactions in systems containing carbon and transition metal elements

    Exploring a structural protein-drug interactome for new therapeutics in lung cancer

    The pharmacology of drugs is often defined by more than one protein target. This property can be exploited to use approved drugs to uncover new targets and signaling pathways in cancer. Towards enabling a rational approach to uncover new targets, we expand a structural protein-ligand interactome () by scoring the interaction among 1000 FDA-approved drugs docked to 2500 pockets on protein structures of the human genome. This afforded a drug-target network whose properties compared favorably with previous networks constructed using experimental data. Among drugs with the highest degree and betweenness two are cancer drugs and one is currently used for treatment of lung cancer. Comparison of predicted cancer and non-cancer targets reveals that the most cancer-specific compounds were also the most selective compounds. Analysis of compound flexibility, hydrophobicity, and size showed that the most selective compounds were low molecular weight fragment-like heterocycles. We use a previously-developed screening approach using the cancer drug erlotinib as a template to screen other approved drugs that mimic its properties. Among the top 12 ranking candidates, four are cancer drugs, two of them kinase inhibitors (like erlotinib). Cellular studies using non-small cell lung cancer (NSCLC) cells revealed that several drugs inhibited lung cancer cell proliferation. We mined patient records at the Regenstrief Medical Record System to explore the possible association of exposure to three of these drugs with occurrence of lung cancer. Preliminary in vivo studies using the non-small cell lung cancer (NCLSC) xenograft model showed that losartan- and astemizole-treated mice had tumors that weighed 50 (p < 0.01) and 15 (p < 0.01) percent less than the treated controls. These results set the stage for further exploration of these drugs and to uncover new drugs for lung cancer therapy

    Factors Associated with SARS-CoV-2 Infection Evaluated by Antibody Response in a Sample of Workers from the Emilia-Romagna Region, Northern Italy

    Factors associated with SARS-CoV-2 infection risk are still debated. This case–control study aims to investigate the possible relationship between SARS-CoV-2 infection, evaluated through antibody response, and the main sociodemographic, occupational, clinical-anamnestic, and biochemical factors in a population of Modena province (Northern Italy), mainly workers. Both workers who voluntarily joined the screening campaign proposed by companies and self-referred individuals who underwent serological testing were enrolled. Subjects with antibody positivity were recruited as cases (n = 166) and subjects tested negative (n = 239) as controls. A questionnaire on sociodemographic, occupational, and clinical data was administered through telephone interviews. Serum zinc/iron/copper/chromium/nickel, vitamins D/B12, folates, triglycerides, and LDL/HDL/total cholesterol were measured. Cases lived more often in urban areas (61.8% vs. 57%). Cases and controls did not differ significantly by working macrocategories, but the percentage of workers in the ceramic sector was higher among cases. Low adherence to preventive measures in the workplace was more frequent among seropositives. Folate concentration was significantly lower among cases. Therefore, adequate folate levels, living in rural areas, and good adherence to preventive strategies seem protective against infection. Workers in the ceramic sector seem to be at greater risk; specific factors involved are not defined, but preventive interventions are needed
