The classic theory of mammalian brain and behavioral sexual differentiation holds that an organizational action of testosterone, secreted perinatally by the male’s testes, controls male-typical aspects of brain and behavioral development, whereas no active perinatal sex hormone signaling is required for female-typical differentiation. However, it has recently been shown using aromatase knock out (ArKO) mice, which are deprived of estradiol due to a targeted mutation in the aromatase gene, that estradiol may also actively contribute to the differentiation of female-typical aspects of behavior over a specific pre-pubertal period. Sexual behavior is a well known sexually dimorphic behavior, and thus provides a good model for studying the sexual differentiation of the brain. Female ArKO mice showed significantly lower levels of lordosis behavior than wild type controls following adult ovariectomy and treatment with ovarian hormones. This deficit was restored by treating female ArKO mice with estradiol benzoate between postnatal days 15 and 25. However, the mechanism by which estradiol acts prepubertally in female mice to feminize lordosis behavior remains to be elucidated. Therefore, in a first attempt, we will determine which neuropeptides are in particularly sensitive to estradiol action over the prepubertal period. We will focus on neuropeptides which have been shown to be implicated in lordosis behavior. Studies by the group of Micevych have proposed an important role for NPY and beta-endorphin (B-END) neurons in the arcuate nucleus (ARC) in the estradiol-mediated activation of lordosis behavior in female rats. An in vitro study recently showed that kisspeptin directly activates arcuate (ARC) B-END neurons. This is particularly interesting since unpublished results from our laboratory showed that Kp neurons are directly involved in the expression of lordosis behavior. We are thus currently determining the expression of NPY, B-END, and kisspeptin in the ARC as well as μ-opioid receptors MORs in the MPN over the prepubertal period and their possible modulation by prepubertal estradiol treatment. Interestingly, we also observed that our treatment may have an organizational effect on the regulation of energy balance. Indeed, administration of estradiol over the prepubertal period restores a normal weight in ArKO mice, which are normally obese in adulthood. It has been shown that ArKO mice present a lower spontaneous physical activity which can be one cause of their obesity. However, our treated mice did not show an increase in their locomotion behavior. Rodents utilize chemical cues to recognize and avoid other conspecifics infected with pathogens. Infection with pathogens and acute inflammation alter the repertoire and signature of olfactory stimuli emitted by a sick individual. These cues are recognized by healthy conspecifics via the vomeronasal or accessory olfactory system, triggering an innate form of avoidance behavior. National audiencePlus qu'une méthode, la prospective est une démarche de réflexion collective sur le changement dans le temps et dans le cas précis de la géoprospective, dans l'espace.L'intérêt d'un exercice de géoprospective appliquée aux systèmes (ou socio-écosystèmes) marins, dans notre cas la pêcherie benthique du golfe de Gascogne, repose sur deux principaux constats. D'une part, la pêcherie est soumise à changements environnementaux, politiques ou encore socio-économiques. Ces changements modifient l'état du système et multiplient les incertitudes quant à son devenir. D'autre part, la gestion centralisée et sectorielle qui a longtemps prévalu est aujourd'hui remise en question au profit de la gouvernance et d'un modèle intégré qui nécessite d'adapter, sinon d'élargir, les outils de l'halieutique opérationnelle.La géoprospective se positionne alors comme un complément utile aux démarches classiques d'anticipation quantitative et d'évaluation bioéconomique de scénarios de gestion. En effet, elle entend répondre à de nouveaux standards politiques et de nouveaux défis de l'exercice de la recherche sur les pêches maritimes qui en découlent : régionalisation de la gestion, intégration des parties-prenantes, transversalité.L'ambition de cette présentation est alors de développer le concept de géoprospective, d'expliquer le dispositif méthodologique qui sera employé dans le cadre d'ateliers participatifs et de discuter sa pertinence pour anticiper l'état d'une pêcherie et les actions de gestion possibles

Blood Signature of Pre-Heart Failure: A Microarrays Study

International audienceBACKGROUND: The preclinical stage of systolic heart failure (HF), known as asymptomatic left ventricular dysfunction (ALVD), is diagnosed only by echocardiography, frequent in the general population and leads to a high risk of developing severe HF. Large scale screening for ALVD is a difficult task and represents a major unmet clinical challenge that requires the determination of ALVD biomarkers. METHODOLOGY/PRINCIPAL FINDINGS: 294 individuals were screened by echocardiography. We identified 9 ALVD cases out of 128 subjects with cardiovascular risk factors. White blood cell gene expression profiling was performed using pangenomic microarrays. Data were analyzed using principal component analysis (PCA) and Significant Analysis of Microarrays (SAM). To build an ALVD classifier model, we used the nearest centroid classification method (NCCM) with the ClaNC software package. Classification performance was determined using the leave-one-out cross-validation method. Blood transcriptome analysis provided a specific molecular signature for ALVD which defined a model based on 7 genes capable of discriminating ALVD cases. Analysis of an ALVD patients validation group demonstrated that these genes are accurate diagnostic predictors for ALVD with 87% accuracy and 100% precision. Furthermore, Receiver Operating Characteristic curves of expression levels confirmed that 6 out of 7 genes discriminate for left ventricular dysfunction classification. CONCLUSIONS/SIGNIFICANCE: These targets could serve to enhance the ability to efficiently detect ALVD by general care practitioners to facilitate preemptive initiation of medical treatment preventing the development of HF

Cardiomyopathie dilatée primitive (mise en évidence de facteurs pronostiques à l'IRM)

Objectif. Evaluer le pronostic global de la cardiomyopathie dilatée primitive (CMD) à l'IRM. Matériel et méthodes. 117 patients ont été inclus prospectivement dans deux centres français. Un critère composite combinant décès, hospitalisation pour décompensation cardiaque, et arythmie ventriculaire, a été étudié sur un suivi de 17 mois. Des paramètres ont été mesurés à l'IRM standard, et après reconstruction de la surface endocardique du VG en 4 dimensions.Résultats. Les patients ayant présenté un évènement ont des volumes VG, un ratio [volume/masse VG], un indice de sphéricité VG, un volume télésystolique VD plus élevés ; des FEVG et FEVD plus altérées ; un ratio [strain global / VTDVG] plus bas. En analyse multivariée, la FEVG à l'IRM et le volume télésystolique VD sont facteurs prédictifs indépendants d'évènement, plus forts à eux seuls que l'ensemble des paramètres cliniques, biologiques, et échocardiographiques réunis. Discussion. Conclusion. L'IRM myocardique est fondamentale dans la CMD: en plus d'informations nécessaires au diagnostic étiologique, elle permet une évaluation pronostique du patient. Leucomalacies kystiques, étude rétrospective de 31 cas (description, devenir et facteurs de risque)

La LPVc est responsable de la majorité des séquelles motrices de la prématurité. L'hypoxie cérébral et l'inflammation sont les 2 mécanismes conduisant à la lésion. Sur 6 ans, nous avons réuni 31 cas de LPVc, 9 anténatales et 21 postnatales pour réaliser une étude rétrospective afin de décrire les populations, leur devenir et les facteurs de risque. L'incidence de LPVc anténatales est de 0,46%, l'âge gestationnel moyen de 31,92SA, le taux d'hypotrophie de 33%. 88% des lésions sont frontales et 56% unilatérales. Ces lésions sont de bon pronostique. L'incidence des LPVc postnatales est de 1,16%, l'âge gestationnel moyen de 28,76SA. L'ETF précoce est pathologique chez 82% des cas. 91% des lésions sont pariétales et 82% sont bilatérales. Le pronostic est sévère avec des séquelles motrices (80% à 2 ans) et des séquelles cognitives plus tardivement (30%). L’avaluació de la creativitat de les produccions escrites en un context d'ensenyament i aprenentatge

[cat] Aquesta recerca qualitativa, en la qual la investigadora també fa de docent, vol oferir un marc teòric i pràctic sobre l’avaluació de la creativitat de les produccions escrites des de la didàctica de la llengua i la literatura. En la part experimental, la interpretació de les dades obtingudes d’una mostra d’estudiants adults, de professors de les diverses etapes educatives i d’experts en escriptura creativa d’ensenyaments reglats i no reglats ofereixen una fotografia de les creences i les actuacions de la comunitat educativa sobre l’avaluació de la creativitat. El treball també proposa, abans de presentar les conclusions, alguns suggeriments didàctics per aplicar tant a les proves certificatives com als productes escrits produïts a l’aula de llengua i literatura.[eng] This qualitative research, in which the investigator is also the teacher, wants to offer a theoretical and practical framework about the evaluation of the creativity of written work from the field of didactics of language and literature. In the experimental part, the interpretation of the data obtained from a sample of adult students, teachers of different educational stages, and experts in creative writing from regulated and non- regulated teaching methods offer a snapshot of the beliefs and behaviour of the educational community with respect to the evaluation of creativity. Les pêches maritimes : un terrain d’expérimentation de la géoprospective

La prospective construit des scénarios d’évolution – pour la planification ou pour la protection de l’environnement – dans le but de susciter la discussion et la mise en perspective des visions d’avenir. Cette démarche orientée vers le futur rencontre un tournant spatial avec l’émergence de la géoprospective qui, en spatialisant les conjectures, permet de mettre en évidence des dynamiques géographiques et d’intégrer les enjeux spatiaux dans le débat. Une application participative de géoprospective aux pêches maritimes présente un double intérêt. Le premier est scientifique : il s’agit de contribuer aux corpus méthodologique et épistémologique de la géoprospective. En effet, la proposition de méthode permet de discuter la fonction des modèles, d’une part, et le rôle des chercheurs et des acteurs, d’autre part. Le second intérêt d’une géoprospective des pêches maritimes est opérationnel : l’amélioration de la connaissance collective sur les dynamiques et les enjeux sociospatiaux des pêches est de nature à faciliter le dialogue entre parties prenantes. À terme, ce dialogue peut être prolongé sous la forme d’une stratégie de gestion coconstruite.A prospective approach can be used to establish management and environmental protection options to stimulate discussion and bring different scenarios for the future into perspective.
