Intrinsic Electrostatic Potential in the BK Channel Pore: Role in Determining Single Channel Conductance and Block
The internal vestibule of large-conductance Ca2+ voltage-activated K+ (BK) channels contains a ring of eight negative charges not present in K+ channels of lower conductance (Glu386 and Glu389 in hSlo) that modulates channel conductance through an electrostatic mechanism (Brelidze, T.I., X. Niu, and K.L. Magleby. 2003. Proc. Natl. Acad. Sci. USA. 100:9017–9022). In BK channels there are also two acidic amino acid residues in an extracellular loop (Asp326 and Glu329 in hSlo). To determine the electrostatic influence of these charges on channel conductance, we expressed wild-type BK channels and mutants E386N/E389N, D326N, E329Q, and D326N/E329Q channels on Xenopus laevis oocytes, and measured the expressed currents under patch clamp. Contribution of E329 to the conductance is negligible and single channel conductance of D326N/E329Q channels measured at 0 mV in symmetrical 110 mM K+ was 18% lower than the control. Current–voltage curves displayed weak outward rectification for D326N and the double mutant. The conductance differences between the mutants and wild-type BK were caused by an electrostatic effect since they were enhanced at low K+ (30 mM) and vanished at high K+ (1 M K+). We determine the electrostatic potential change, Δφ, caused by the charge neutralization using TEA+ block for the extracellular charges and Ba2+ for intracellular charges. We measured 13 ± 2 mV for Δφ at the TEA+ site when turning off the extracellular charges, and 17 ± 2 mV for the Δφ at the Ba2+ site when the intracellular charges were turned off. To understand the electrostatic effect of charge neutralizations, we determined Δφ using a BK channel molecular model embedded in a lipid bilayer and solving the Poisson-Boltzmann equation. The model explains the experimental results adequately and, in particular, gives an economical explanation to the differential effect on the conductance of the neutralization of charges D326 and E329
Baryon Resonances
In this talk I show recent results on how many excited baryon resonances
appear as systems of one meson and one baryon, or two mesons and one baryon,
with the mesons being either pseudoscalar or vectors. Connection with
experiment is made including a discussion on old predictions and recent results
for the photoproduction of the resonance, as well as the
prediction of one baryon state around 1920 MeV which might have been

Prevalence of Intestinal Parasites in Endangered Ashy Red Colobus Monkeys (Piliocolobus tephrosceles) in Tanzania
Intestinal parasites constitute one of the most frequent causes of gastrointestinal diseases in primates, directly affecting their health. We sampled 3 populations of the Endangered ashy red colobus monkey (Piliocolobus tephrosceles) with different levels of anthropogenic disturbance in Tanzania. We collected faecal samples (n=157) soon after defecation and fixed them in situ in 70% ethanol. We then re-fixed half of each sample in MIF (merthiolate iodine formaline) for microscopic study and saved the rest for molecular analysis. We examined helminth eggs, larvae and protozoan cysts using a light microscope after faecal sedimentation. We analysed samples positive for Giardia using Polymerase Chain Reaction (PCR) to determine genotypes. The overall prevalence of protozoan and helminth infection was 94.3% (148/157), with 64.9% (96/148) being infected by 1 species, 25.7% (38) by 2 species, and 9.5% (14) by 3 or more species. We detected 8 species of intestinal parasites: Ancylostoma sp. (13.4%), Trichuris trichiura (3.8%), Strongyloides stercoralis rhabditoid larvae (2.5%), Entamoeba chatonni (82.8%), Iodamoeba butschlii (14%), Endolimax nana (4.5%), Blastocystis hominis (2.5%) and Giardia duodenalis (14%). These species were detected in different combinations in the 3 areas, while Giardia was detected in only 1 area. The molecular analysis of positive Giardia samples showed that all of them belonged to assemblage B, which could also infect humans. However, we could not identify an exclusively anthropogenic origin of the parasitic species found. Our study contributes to our knowledge of parasitic infections in ashy monkeys in Tanzania, allowing us to assess their health status and disease risk, which in turn will help us design more successful conservation strategies for this Endangered primate in Tanzani
The Chandra Galactic Bulge Survey: optical catalogue and point-source counterparts to X-ray sources
As part of the Chandra Galactic Bulge Survey (GBS), we present a catalogue of
optical sources in the GBS footprint. This consists of two regions centered at
Galactic latitude b = 1.5 degrees above and below the Galactic Centre, spanning
(l x b) = (6x1) degrees. The catalogue consists of 2 or more epochs of
observations for each line of sight in r', i' and H{\alpha} filters. It is
complete down to r' = 20.2 and i' = 19.2 mag; the mean 5{\sigma} depth is r' =
22.5 and i' = 21.1 mag. The mean root-mean-square residuals of the astrometric
solutions is 0.04 arcsec. We cross-correlate this optical catalogue with the
1640 unique X-ray sources detected in Chandra observations of the GBS area, and
find candidate optical counterparts to 1480 X-ray sources. We use a false alarm
probability analysis to estimate the contamination by interlopers, and expect ~
10 per cent of optical counterparts to be chance alignments. To determine the
most likely counterpart for each X-ray source, we compute the likelihood ratio
for all optical sources within the 4{\sigma} X-ray error circle. This analysis
yields 1480 potential counterparts (~ 90 per cent of the sample). 584
counterparts have saturated photometry (r'<17, i'<16), indicating these objects
are likely foreground sources and the real counterparts. 171 candidate
counterparts are detected only in the i'-band. These sources are good qLMXB and

The Respiratory Exchange Ratio is Associated with Fitness Indicators Both in Trained and Untrained Men: A Possible Application for People with Reduced Exercise Tolerance
Background The respiratory exchange ratio (RER) indirectly shows the muscle's oxidative capacity to get energy. Sedentarism, exercise and physically active lifestyles modify it. For that reason, this study evaluates the associations between RER during sub-maximum exercise and other well established fitness indicators (body fat, maximum heart rate, maximum O 2 uptake, workload, and lactate threshold), in physically active trained and untrained men. Methods The RER, O 2 uptake and blood lactate were measured in eight endurance trained and eight untrained men (age, 22.9 ± 4.5 vs. 21.9 ± 2.8 years; body mass, 67.1 ± 5.4 vs. 72.2 ± 7.7 kg; body fat, 10.6 ± 2.4% vs. 16.6 ± 3.8% and maximum O2 uptake, 68.9 ± 6.3 vs. 51.6 ± 5.8 ml · kg −1 · min −1 ), during maximum exercise test and during three different sub-maximum exercises at fixed workload: below, within or above the lactate threshold. Results Endurance trained men presented higher O 2 uptake, lower blood lactate concentrations and lower RER values than those in untrained men at the three similar relative workloads. Even though with these differences in RER, a strong association (p < 0.05) of RER during sub-maximum exercise with the other well established fitness indicators was observed, and both maximum O 2 uptake and lactate threshold determined more than 57% of its variance (p < 0.05). Conclusions These data demonstrate that RER measurement under sub-maximum exercise conditions was well correlated with other established physical fitness indicators, despite training condition. Furthermore, the results suggest that RER could help obtain an easy approach of fitness status under low exercise intensity and could be utilized in subjects with reduced exercise tolerance
