    Lysine and methionine supplementation in high soy protein content diets for silvery-black porgy (Sparidentex hasta) juveniles

    A two-month feeding trial was conducted to evaluate the effect of partial replacement of fish meal (FM) by soybean protein (SP) alone or in combination with lysine (Lys) and methionine (Met) supplementation in practical diets for silvery-black porgy juveniles (16.7±0.1 g). Seven isoproteic (ca. 50% crude protein) and isoenergetic (ca. 22.4 MJ kg-1) diets were formulated in which 45% (SP45), 60% (SP60) and 75% (SP75) of FM protein were replaced by SP and the control diet (FM) was prepared with FM as the major source of protein. In SP45+, SP60+ and SP75+ diets, 45 to 75% of FM was replaced by SP with supplementing blends of Lys and Met. Growth performance, feed utilization, and protein and lipid digestibility decreased with increasing dietary SP levels (p<0.05). Fish fed SP75 had the highest whole body lipid content, but with the lowest whole body arginine, lysine, histidine, phenylalanine and taurine concentrations (p<0.05). Supplementing Lys and Met in SP based diets did not improve growth performance, suggesting that the anti-nutritional factors in soybean protein products rather than Met and Lys deïŹciency might limit FM substitution with SP in silvery-black porgy juveniles.info:eu-repo/semantics/publishedVersio

    A rare IL33 loss-of-function mutation reduces blood eosinophil counts and protects from asthma.

Common variants at IL33 and IL1RL1, encoding the IL-33 receptor ST2, associate with eosinophil counts and asthma. Through whole-genome sequencing and imputation into the Icelandic population, we found a rare variant in IL33 (NM_001199640:exon7:c.487-1G>C (rs146597587-C), allele frequency = 0.65%) that disrupts a canonical splice acceptor site before the last coding exon. It is also found at low frequency in European populations. rs146597587-C associates with lower eosinophil counts (ÎČ = -0.21 SD, P = 2.5×10-16, N = 103,104), and reduced risk of asthma in Europeans (OR = 0.47; 95%CI: 0.32, 0.70, P = 1.8×10-4, N cases = 6,465, N controls = 302,977). Heterozygotes have about 40% lower total IL33 mRNA expression than non-carriers and allele-specific analysis based on RNA sequencing and phased genotypes shows that only 20% of the total expression is from the mutated chromosome. In half of those transcripts the mutation causes retention of the last intron, predicted to result in a premature stop codon that leads to truncation of 66 amino acids. The truncated IL-33 has normal intracellular localization but neither binds IL-33R/ST2 nor activates ST2-expressing cells.     Sequence variant affects GCSAML splicing, mast cell specific proteins, and risk of urticaria

In order to identify sequence variants associated with urticaria, we performed a meta-analysis of genome-wide association studies for urticaria with a total of 40,694 cases and 1,230,001 controls from Iceland, the UK, Finland, and Japan. We also performed transcriptome- and proteome-wide analyses in Iceland and the UK. We found nine sequence variants at nine loci associating with urticaria. The variants are at genes participating in type 2 immune responses and/or mast cell biology (CBLB, FCER1A, GCSAML, STAT6, TPSD1, ZFPM1), the innate immunity (C4), and NF-ÎșB signaling. The most significant association was observed for the splice-donor variant rs56043070[A] (hg38: chr1:247556467) in GCSAML (MAF = 6.6%, OR = 1.24 (95%CI: 1.20–1.28), P-value = 3.6 × 10-44). We assessed the effects of the variants on transcripts, and levels of proteins relevant to urticaria pathophysiology. Our results emphasize the role of type 2 immune response and mast cell activation in the pathogenesis of urticaria. Our findings may point to an IgE-independent urticaria pathway that could help address unmet clinical need.Peer reviewe

    A mixed-data evaluation in group TOPSIS with differentiated decision power

    [[abstract]]This main objective of this paper is to provide decision support for mixed data in group Technique for Order Preference by Similarity to Idea Solution (TOPSIS) with differentiated decision power. We use a signum function to compare the ordinal performance of alternatives on any qualitative criterion, or the partial information provided by decision makers. The proposed process for ordinal information is uniformly coherent with the traditional TOPSIS steps, preserving the characteristic of distance-based utilities. Ordinal weights are also considered herein, and the decision power of the group members is formulated by their weights under an agreement in the group. Two examples demonstrate that the proposed approach has some benefits and achieves robustness with two types of sensitivity analyses. Nordiska ministerrÄdet inrÀttade dÀrför i september 2018 en arbetsgrupp för att systematisera de nordiska lÀndernas erfarenheter frÄn torkan 2018 samt föreslÄ konkreta nordiska samarbetsomrÄden som kan bidra till ett mer motstÄndskraftigt nordiskt jordbruk.
