Measurement And Evolution Of The Thickness Distribution And Morphology Of Deformed Features Of Antarctic Sea Ice
Thesis (Ph.D.) University of Alaska Fairbanks, 2003Antarctic sea ice thickness data obtained from drilling on sea ice floes were examined with the goal of enhancing our capability to estimate ice thickness remotely, especially from air- or space-borne altimetry and shipboard visual observations. The state of hydrostatic equilibrium of deformed ice features and the statistical relationships between ice thickness and top surface roughness were examined. Results indicate that ice thickness may be estimated fairly reliably from surface measurements of snow elevation on length scales of ?100 m. Examination of the morphology of deformed ice features show that Antarctic pressure ridges are flatter and less massive than Arctic pressure ridges and that not all surface features (ridge sails) are associated with features underwater (ridge keels). I propose that the differences in morphology are due to differences in sampling strategies, parent ice characteristics and the magnitude and duration of driving forces. As a result of these findings, the existing methodology used to estimate ice thickness from shipboard visual observations was modified to incorporate the probability that a sail is associated with a keel underwater, and the probability that keels may be found under level surfaces. Using the improved methodology, ice thickness was estimated from ship observations data obtained during two cruises in the Ross Sea, Antarctica. The dynamic and thermodynamic processes involved in the development of the ice prior to their observation were examined employing a regional sea ice-mixed layer-pycnocline model. Both our model results and previously published ice core data indicate that thermodynamic thickening is the dominant process that determines the thickness of first year ice in the central Ross Sea, although dynamic thickening also plays a significant role. Ice core data also indicate that snow ice forms a significant proportion of the total ice mass. For ice in the northeast Ross Sea in the summer, model results and evidence from ice core and oceanographic data indicate that dynamic thickening, snow ice formation and bottom melting compete to determine the ice thickness during mid and late winter
Polymer Immobilised Ionic Liquid Supported Catalysts: Synthesis, Characterisation and Applications
PhD ThesisDue to increasing demand for synthetic commodities, to reduce the overall
environmental impact of existing chemical processes, a sustainable approach is
required. In order to accomplish this, it will be necessary to use highly efficient, durable,
and scalable catalyst technologies which are low in energy consumption and waste
production. In addition, bio-renewable feedstock will also be required due its
abundancy and availability. In this regard, chapter 1 provides an overview of the unique
physicochemical properties and concepts of ionic liquids (ILs). The incorporation of ILs
can produce functional materials, which in turn, can be used for the sustainable
conversion of bio-renewable feedstock.
Chapter 2 discusses the rational design and synthesis of a range of ruthenium
nanoparticle (RuNP) catalysts, in which, the catalysts were tested in the hydrogenation
of the benchmark substrate acetophenone. The overall aim of this work was to
investigate the effect of the interactions between the ruthenium and the polymer
support on the catalyst performance and selectivity. The resulting catalysts were highly
active and selective in the hydrogenation of a range of , -unsaturated ketones,
aldehydes, and the study was extended to include bio-derived substrates. In total four
reactions were tested, namely the hydrogenation of acetophenone, furfural, levulinic
acid and ethyl levulinate. The low ruthenium loadings and the reusability of these
heterogenous catalysts, in addition to the use of water as the co-solvent/solvent, has
provided a sustainable hydrogenation method, which is among one of the highest
reported on comparison with the literature. Chapter 3 covers the application of the
RuNPs for the hydrazine-mediated reduction of a broad range of nitroaromatics to their
corresponding aromatic amines under mild conditions with low catalyst loadings.
Chapter 4 details the synthesis of a set of imidazolium polystyrene-based
Brønsted acid catalysts immobilised with peroxophosphotungstate anions. These
efficient and selective catalysts were tested, as catalysts for the alcoholysis of furfuryl
alcohol to produce alkyl levulinates, under batch as well as continuous flow regimes.
Such generated levulinates are highly desirable as bio-renewable fuel additives

SPARC Deficiency Results in Improved Surgical Survival in a Novel Mouse Model of Glaucoma Filtration Surgery
Glaucoma is a disease frequently associated with elevated intraocular pressure that can be alleviated by filtration surgery. However, the post-operative subconjunctival scarring response which blocks filtration efficiency is a major hurdle to the achievement of long-term surgical success. Current application of anti-proliferatives to modulate the scarring response is not ideal as these often give rise to sight-threatening complications. SPARC (secreted protein, acidic and rich in cysteine) is a matricellular protein involved in extracellular matrix (ECM) production and organization. In this study, we investigated post-operative surgical wound survival in an experimental glaucoma filtration model in SPARC-null mice. Loss of SPARC resulted in a marked (87.5%) surgical wound survival rate compared to 0% in wild-type (WT) counterparts. The larger SPARC-null wounds implied that aqueous filtration through the subconjunctival space was more efficient in comparison to WT wounds. The pronounced increase in both surgical survival and filtration efficiency was associated with a less collagenous ECM, smaller collagen fibril diameter, and a loosely-organized subconjunctival matrix in the SPARC-null wounds. In contrast, WT wounds exhibited a densely packed collagenous ECM with no evidence of filtration capacity. Immunolocalization assays confirmed the accumulation of ECM proteins in the WT but not in the SPARC-null wounds. The observations in vivo were corroborated by complementary data performed on WT and SPARC-null conjunctival fibroblasts in vitro. These findings indicate that depletion of SPARC bestows an inherent change in post-operative ECM remodeling to favor wound maintenance. The evidence presented in this report is strongly supportive for the targeting of SPARC to increase the success of glaucoma filtration surgery
Integrated Omics Reveals the Orchestrating Role of Calycosin in Danggui Buxue Tang, a Herbal Formula Containing Angelicae Sinensis Radix and Astragali Radix, in Inducing Osteoblastic Differentiation and Proliferation
Systems biology unravels the black box of signaling pathway of cells; but which has not been extensively applied to reveal the mechanistic synergy of a herbal formula. The therapeutic efficacies of a herbal formula having multi-target, multi-function and multi-pathway are the niches of traditional Chinese medicine (TCM). Here, we reported an integrated omics approach, coupled with the knockout of an active compound, to measure the regulation of cellular signaling, as to reveal the landscape in cultured rat osteoblasts having synergistic pharmacological efficacy of Danggui Buxue Tang (DBT), a Chinese herbal formula containing Angelicae Sinensis Radix and Astragali Radix. The changes in signaling pathways responsible for energy metabolism, RNA metabolism and protein metabolism showed distinct features between DBT and calycosin-depleted DBT. Here, our results show that calycosin within DBT can orchestrate the osteoblastic functions and signaling pathways of the entire herbal formula. This finding reveals the harmony of herbal medicine in pharmacological functions, as well as the design of drug/herbal medicine formulation. The integration of systems biology can provide novel and essential insights into the synergistic property of a herbal formula, which is a key in modernizing TCM
The Magnitude of Global Marine Species Diversity
Background: The question of how many marine species exist is important because it provides a metric for how much we do and do not know about life in the oceans. We have compiled the first register of the marine species of the world and used this baseline to estimate how many more species, partitioned among all major eukaryotic groups, may be discovered.
Results: There are ∼226,000 eukaryotic marine species described. More species were described in the past decade (∼20,000) than in any previous one. The number of authors describing new species has been increasing at a faster rate than the number of new species described in the past six decades. We report that there are ∼170,000 synonyms, that 58,000–72,000 species are collected but not yet described, and that 482,000–741,000 more species have yet to be sampled. Molecular methods may add tens of thousands of cryptic species. Thus, there may be 0.7–1.0 million marine species. Past rates of description of new species indicate there may be 0.5 ± 0.2 million marine species. On average 37% (median 31%) of species in over 100 recent field studies around the world might be new to science.
Conclusions: Currently, between one-third and two-thirds of marine species may be undescribed, and previous estimates of there being well over one million marine species appear highly unlikely. More species than ever before are being described annually by an increasing number of authors. If the current trend continues, most species will be discovered this century
A common variant near TGFBR3 is associated with primary open angle glaucoma
Primary open angle glaucoma (POAG), a major cause of blindness worldwide, is a complex disease with a significant genetic contribution. We performed Exome Array (Illumina) analysis on 3504 POAG cases and 9746 controls with replication of the most significant findings in 9173 POAG cases and 26 780 controls across 18 collections of Asian, African and European descent. Apart from confirming strong evidence of association at CDKN2B-AS1 (rs2157719 [G], odds ratio [OR] = 0.71, P = 2.81 × 10−33), we observed one SNP showing significant association to POAG (CDC7–TGFBR3 rs1192415, ORG-allele = 1.13, Pmeta = 1.60 × 10−8). This particular SNP has previously been shown to be strongly associated with optic disc area and vertical cup-to-disc ratio, which are regarded as glaucoma-related quantitative traits. Our study now extends this by directly implicating it in POAG disease pathogenesis
