    Arbuscular Mycorrhizas Reduce Nitrogen Loss via Leaching

    The capacity of mycorrhizal and non-mycorrhizal root systems to reduce nitrate (NO3−) and ammonium (NH4+) loss from soils via leaching was investigated in a microcosm-based study. A mycorrhiza defective tomato mutant and its mycorrhizal wildtype progenitor were used in this experiment in order to avoid the indirect effects of establishing non-mycorrhizal control treatments on soil nitrogen cycling and the wider soil biota. Mycorrhizal root systems dramatically reduced nitrate loss (almost 40 times less) via leaching, compared to their non-mycorrhizal counterparts, following a pulse application of ammonium nitrate to experimental microcosms. The capacity of AM to reduce nutrient loss via leaching has received relatively little attention, but as demonstrated here, can be significant. Taken together, these data highlight the need to consider the potential benefits of AM beyond improvements in plant nutrition alone

    Local and distal effects of arbuscular mycorrhizal colonization on direct pathway Pi uptake and root growth in <i>Medicago truncatula</i>

    Two pathways exist for plant Pi uptake from soil: via root epidermal cells (direct pathway) or via associations with arbuscular mycorrhizal (AM) fungi, and the two pathways interact in a complex manner. This study investigated dis - tal and local effects of AM colonization on direct root Pi uptake and root growth, at different soil P levels. Medicago truncatula was grown at three soil P levels in split-pots with or without AM fungal inoculation and where one root half grew into soil labelled with 33 P. Plant genotypes included the A17 wild type and the mtpt4 mutant. The mtpt4 mutant, colonized by AM fungi, but with no functional mycorrhizal pathway for Pi uptake, was included to better understand effects of AM colonization per se . Colonization by AM fungi decreased expression of direct Pi transporter genes locally, but not distally in the wild type. In mtpt4 mutant plants, direct Pi transporter genes and the Pi starvation- induced gene Mt4 were more highly expressed than in wild-type roots. In wild-type plants, less Pi was taken up via the direct pathway by non-colonized roots when the other root half was colonized by AM fungi, compared with non- mycorrhizal plants. Colonization by AM fungi strongly influenced root growth locally and distally, and direct root Pi uptake activity locally, but had only a weak influence on distal direct pathway activity. The responses to AM coloniza - tion in the mtpt4 mutant suggested that in the wild type, the increased P concentration of colonized roots was a major factor driving the effects of AM colonization on direct root Pi uptake.Stephanie J.     Wine Terroir and the Soil Bacteria: An Amplicon Sequencing-Based Assessment of the Barossa Valley and Its Sub-Regions

    A wines’ terroir, represented as wine traits with regional distinctiveness, is a reflection of both the biophysical and human-driven conditions in which the grapes were grown and wine made. Soil is an important factor contributing to the uniqueness of a wine produced by vines grown in specific conditions. Here, we evaluated the impact of environmental variables on the soil bacteria of 22 Barossa Valley vineyard sites based on the 16S rRNA gene hypervariable region 4. In this study, we report that both dispersal isolation by geographic distance and environmental heterogeneity (soil plant-available P content, elevation, rainfall, temperature, spacing between row and spacing between vine) contribute to microbial community dissimilarity between vineyards. Vineyards located in cooler and wetter regions showed lower beta diversity and a higher ratio of dominant taxa. Differences in soil bacterial community composition were significantly associated with differences in fruit and wine composition. Our results suggest that environmental factors affecting wine terroir, may be mediated by changes in microbial structure, thus providing a basic understanding of how growing conditions affect interactions between plants and their soil bacteria

    Why farmers should manage the arbuscular mycorrhizal symbiosis

    The Tansley review by Ryan & Graham (2018) provided a welcome critical perspective on the role of arbuscular mycorrhizal (AM) fungi in large‐scale industrial agriculture, with a focus on cereals (wheat, Triticum aestivum). They conclude that there is little evidence that farmers should consider the abundance or diversity of AM fungi when managing crops. We welcome many of the points made in the paper, as they give an opportunity for self‐reflection, considering that the importance of AM fungi in agroecosystems is often taken for granted. However, we suggest that it is too early to draw the overall conclusion that the management of AM fungi by farmers is currently not warranted. We offer the following points to contribute to the discussion. The first point pertains to the overall focus of Ryan & Graham (2018), which strongly determines the recommendations at which the authors arrive. This scope is limited to yield, at the expense of neglecting aspects of sustainability. We then argue that AM fungal communities do respond negatively to aspects of agricultural management, and list evidence for their positive effects to agronomically important traits, including yield in cereals. In our final argument, we advocate for transitioning to agroecosystems that are more AM compatible in order to increasingly take advantage of all the potential services these ancient symbionts, and other soil biota, can provide

    Frequency Versus Quantity: Phenotypic Response of Two Wheat Varieties to Water and Nitrogen Variability

    Due to climate change, water availability will become increasingly variable, affecting nitrogen (N) availability. Therefore, we hypothesised watering frequency would have a greater impact on plant growth than quantity, affecting N availability, uptake and carbon allocation. We used a gravimetric platform, which measures the unit of volume per unit of time, to control soil moisture and precisely compare the impact of quantity and frequency of water under variable N levels. Two wheat genotypes (Kukri and Gladius) were used in a factorial glasshouse pot experiment, each with three N application rates (25, 75 and 150mgNkg−1 soil) and five soil moisture regimes (changing water frequency or quantity). Previously documented drought tolerance, but high N use efficiency, of Gladius as compared to Kukri provides for potentially different responses to N and soil moisture content. Water use, biomass and soil N were measured. Both cultivars showed potential to adapt to variable watering, producing higher specific root lengths under low N coupled with reduced water and reduced watering frequency (48h watering intervals), or wet/dry cycling. This affected mineral N uptake, with less soil N remaining under constant watering × high moisture, or 48h watering intervals × high moisture. Soil N availability affected carbon allocation, demonstrated by both cultivars producing longer, deeper roots under low N. Reduced watering frequency decreased biomass more than reduced quantity for both cultivars. Less frequent watering had a more negative effect on plant growth compared to decreasing the quantity of water. Water variability resulted in differences in C allocation, with changes to root thickness even when root biomass remained the same across N treatments. The preferences identified in wheat for water consistency highlights an undeveloped opportunity for identifying root and shoot traits that may improve plant adaptability to moderate to extreme resource limitation, whilst potentially encouraging less water and nitrogen use

    A slow release nitrogen fertiliser produced by simultaneous granulation and superheated steam drying of urea with brown coal

    Background: The inefficient use of nitrogen (N) fertiliser as a consequence of N losses from soil is a pressing issue in broad-acre agricultural systems. The research reported here tested the hypothesis that granulation of synthetic N fertiliser (urea) with a natural organic C resource (brown coal) would reduce fertiliser N loss from the soil system. Results: Urea-enriched brown coal granules were simultaneously formed and dried within a pilot-scale superheated steam dryer. After application to unplanted soil columns, the urea-brown coal granules reduced nitrous oxide emission by up to 40&nbsp;%, reduced mineral nitrogen leaching and maintained higher levels of N in topsoil when compared to conventional urea alone. Reduced gaseous N losses without a reduction in plant N uptake were also observed in planted soil columns treated with urea-brown coal granules. Conclusions: Brown coal-urea blended fertiliser showed potential for more efficient use of N in the long term and has environmental benefits in retaining more N in the soil

    The state of the world’s urban ecosystems: What can we learn from trees, fungi, and bees?

    Publication history: Accepted - 2 July 2020; Published online - 29 September 2020Trees are a foundation for biodiversity in urban ecosystems and therefore must be able to withstand global change and biological challenges over decades and even centuries to prevent urban ecosystems from deteriorating. Tree quality and diversity should be prioritized over simply numbers to optimize resilience to these challenges. Successful establishment and renewal of trees in cities must also consider belowground (e.g., mycorrhizas) and aboveground (e.g., pollinators) interactions to ensure urban ecosystem longevity, biodiversity conservation and continued provision of the full range of ecosystem services provided by trees. Positive interactions with nature inspire people to live more sustainable lifestyles that are consistent with stopping biodiversity loss and to participate in conservation actions such as tree‐planting and supporting pollinators. Interacting with nature simultaneously provides mental and physical health benefits to people. Since most people live in cities, here we argue that urban ecosystems provide important opportunities for increasing engagement with nature and educating people about biodiversity conservation. While advocacy on biodiversity must communicate in language that is relevant to a diverse audience, over‐simplified messaging, may result in unintended negative outcomes. For example, tree planting actions typically focus on numbers rather than diversity while the call to save bees has inspired unsustainable proliferation of urban beekeeping that may damage wild bee conservation through increased competition for limited forage in cities and disease spread. Ultimately multiple ecosystem services must be considered (and measured) to optimize their delivery in urban ecosystems and messaging to promote the value of nature in cities must be made widely available and more clearly defined.Animal Plant Health Agency (UK).     Genome-wide characterization of simple sequence repeats in cucumber (Cucumis sativus L.)

    <p>Abstract</p> <p>Background</p> <p>Cucumber, <it>Cucumis sativus </it>L. is an important vegetable crop worldwide. Until very recently, cucumber genetic and genomic resources, especially molecular markers, have been very limited, impeding progress of cucumber breeding efforts. Microsatellites are short tandemly repeated DNA sequences, which are frequently favored as genetic markers due to their high level of polymorphism and codominant inheritance. Data from previously characterized genomes has shown that these repeats vary in frequency, motif sequence, and genomic location across taxa. During the last year, the genomes of two cucumber genotypes were sequenced including the Chinese fresh market type inbred line '9930' and the North American pickling type inbred line 'Gy14'. These sequences provide a powerful tool for developing markers in a large scale. In this study, we surveyed and characterized the distribution and frequency of perfect microsatellites in 203 Mbp assembled Gy14 DNA sequences, representing 55% of its nuclear genome, and in cucumber EST sequences. Similar analyses were performed in genomic and EST data from seven other plant species, and the results were compared with those of cucumber.</p> <p>Results</p> <p>A total of 112,073 perfect repeats were detected in the Gy14 cucumber genome sequence, accounting for 0.9% of the assembled Gy14 genome, with an overall density of 551.9 SSRs/Mbp. While tetranucleotides were the most frequent microsatellites in genomic DNA sequence, dinucleotide repeats, which had more repeat units than any other SSR type, had the highest cumulative sequence length. Coding regions (ESTs) of the cucumber genome had fewer microsatellites compared to its genomic sequence, with trinucleotides predominating in EST sequences. AAG was the most frequent repeat in cucumber ESTs. Overall, AT-rich motifs prevailed in both genomic and EST data. Compared to the other species examined, cucumber genomic sequence had the highest density of SSRs (although comparable to the density of poplar, grapevine and rice), and was richest in AT dinucleotides. Using an electronic PCR strategy, we investigated the polymorphism between 9930 and Gy14 at 1,006 SSR loci, and found unexpectedly high degree of polymorphism (48.3%) between the two genotypes. The level of polymorphism seems to be positively associated with the number of repeat units in the microsatellite. The <it>in silico </it>PCR results were validated empirically in 660 of the 1,006 SSR loci. In addition, primer sequences for more than 83,000 newly-discovered cucumber microsatellites, and their exact positions in the Gy14 genome assembly were made publicly available.</p> <p>Conclusions</p> <p>The cucumber genome is rich in microsatellites; AT and AAG are the most abundant repeat motifs in genomic and EST sequences of cucumber, respectively. Considering all the species investigated, some commonalities were noted, especially within the monocot and dicot groups, although the distribution of motifs and the frequency of certain repeats were characteristic of the species examined. The large number of SSR markers developed from this study should be a significant contribution to the cucurbit research community.</p
