    GIBAGADINAMAAGOOM: AN OJIBWE DIGITAL ARCHIVE

    Gibagadinamaagoom, (To Bring to Life, to Sanction, to Give Permission), will bring together a diverse array of partners to discuss how state-of-the-art digital technology can be employed to translate ancient and sophisticated systems of traditional Ojibwe knowledge. Discussions will address the challenging, vitally important questions concerning how to design a digital archive that can be accessed using multiple interfaces. Partners include educational (including tribal colleges) and cultural institutions, native Ojibwe scholars, digital media experts, and a distinguished advisory board. These collaborative partners are well positioned to develop one of the most culturally sensitive Native American archives ever built. This project will contribute significantly to how digital technology can be used to revise academic, museum, and library protocols regarding cultural sensitivity, and enhance teaching and learning with a commitment to honor the cultural sovereignty of the Ojibwe

    SnTox3 Acts in Effector Triggered Susceptibility to Induce Disease on Wheat Carrying the Snn3 Gene

    The necrotrophic fungus Stagonospora nodorum produces multiple proteinaceous host-selective toxins (HSTs) which act in effector triggered susceptibility. Here, we report the molecular cloning and functional characterization of the SnTox3-encoding gene, designated SnTox3, as well as the initial characterization of the SnTox3 protein. SnTox3 is a 693 bp intron-free gene with little obvious homology to other known genes. The predicted immature SnTox3 protein is 25.8 kDa in size. A 20 amino acid signal sequence as well as a possible pro sequence are predicted. Six cysteine residues are predicted to form disulfide bonds and are shown to be important for SnTox3 activity. Using heterologous expression in Pichia pastoris and transformation into an avirulent S. nodorum isolate, we show that SnTox3 encodes the SnTox3 protein and that SnTox3 interacts with the wheat susceptibility gene Snn3. In addition, the avirulent S. nodorum isolate transformed with SnTox3 was virulent on host lines expressing the Snn3 gene. SnTox3-disrupted mutants were deficient in the production of SnTox3 and avirulent on the Snn3 differential wheat line BG220. An analysis of genetic diversity revealed that SnTox3 is present in 60.1% of a worldwide collection of 923 isolates and occurs as eleven nucleotide haplotypes resulting in four amino acid haplotypes. The cloning of SnTox3 provides a fundamental tool for the investigation of the S. nodorum-wheat interaction, as well as vital information for the general characterization of necrotroph-plant interactions.This work was supported by USDA-ARS CRIS projects 5442-22000-043-00D and 5442-22000-030-00D

    Reply to Swartz et al.: Challenges and opportunities for identifying forced labor using satellite-based fishing vessel monitoring

    We appreciate Swartz et al. (1) for highlighting several key considerations for interpreting our results (2). While we discuss many of these in our paper, we are grateful to further highlight our work’s strengths, limitations, and future opportunities. A major challenge with understanding fisheries labor abuses is a lack of data. Automatic identification system (AIS) is only used by a subset of the global fishing fleet. However, AIS is valuable for monitoring certain types of fishing vessels, especially those that are large (∼52 to 85% carry AIS) (3) and those fishing on the high seas (∼80% carry AIS) (4). Mandating AIS and unique identifiers on fishing vessels and publishing vessel registries would facilitate more inclusive AIS-based analyses (5)

    Microfilariae of Brugia malayi Inhibit the mTOR Pathway and Induce Autophagy in Human Dendritic Cells

    Immune modulation is a hallmark of patent filarial infection, including suppression of antigen-presenting cell function and downmodulation of filarial antigen-specific T cell responses. The mammalian target of rapamycin (mTOR) signaling pathway has been implicated in immune regulation, not only by suppressing T cell responses but also by regulating autophagy (through mTOR sensing amino acid availability). Global proteomic analysis (liquid chromatography-tandem mass spectrometry) of microfilaria (mf)-exposed monocyte-derived dendritic cells (DC) indicated that multiple components of the mTOR signaling pathway, including mTOR, eIF4A, and eIF4E, are downregulated by mf, suggesting that mf target this pathway for immune modulation in DC. Utilizing Western blot analysis, we demonstrate that similar to rapamycin (a known mTOR inhibitor), mf downregulate the phosphorylation of mTOR and its regulatory proteins, p70S6K1 and 4E-BP1, a process essential for DC protein synthesis. As active mTOR signaling regulates autophagy, we examined whether mf exposure alters autophagy-associated processes. mf-induced autophagy was reflected in marked upregulation of phosphorylated Beclin 1, known to play an important role in both autophagosome formation and autolysosome fusion, in induction of LC3II, a marker of autophagosome formation, and in induced degradation of p62, a ubiquitin-binding protein that aggregates protein in autophagosomes and is degraded upon autophagy that was reduced significantly by mf exposure and by rapamycin. Together, these results suggest that Brugia malayi mf employ mechanisms of metabolic modulation in DC to influence the regulation of the host immune response by downregulating mTOR signaling, resulting in increased autophagy. Whether this is a result of the parasite-secreted rapamycin homolog is currently under study

    Hippocampus specific iron deficiency alters competition and cooperation between developing memory systems

    Iron deficiency (ID) is the most common gestational micronutrient deficiency in the world, targets the fetal hippocampus and striatum and results in long-term behavioral abnormalities. These structures primarily mediate spatial and procedural memory, respectively, in the rodent but have interconnections that result in competition or cooperation during cognitive tasks. We determined whether ID-induced impairment of one alters the function of the other by genetically inducing a 40% reduction of hippocampus iron content in late fetal life in mice and measuring dorsal striatal gene expression and metabolism and the behavioral balance between the two memory systems in adulthood. Slc11a2hipp/hipp mice had similar striatum iron content, but 18% lower glucose and 44% lower lactate levels, a 30% higher phosphocreatine:creatine ratio, and reduced iron transporter gene expression compared to wild type (WT) littermates, implying reduced striatal metabolic function. Slc11a2hipp/hipp mice had longer mean escape times on a cued task paradigm implying impaired procedural memory. Nevertheless, when hippocampal and striatal memory systems were placed in competition using a Morris Water Maze task that alternates spatial navigation and visual cued responses during training, and forces a choice between hippocampal and striatal strategies during probe trials, Slc11a2hipp/hipp mice used the hippocampus-dependent response less often (25%) and the visual cued response more often (75%) compared to WT littermates that used both strategies approximately equally. Hippocampal ID not only reduces spatial recognition memory performance but also affects systems that support procedural memory, suggesting an altered balance between memory systems

    Faster decline of pitch memory over time in congenital amusia

    Congenital amusia (amusia, hereafter) is a developmental disorder that impacts negatively on the perception of music. Psychophysical testing suggests that individuals with amusia have above average thresholds for detection of pitch change and pitch direction discrimination; however, a low-level auditory perceptual problem cannot completely explain the disorder, since discrimination of melodies is also impaired when the constituent intervals are suprathreshold for perception. The aim of the present study was to test pitch memory as a function of (a) time and (b) tonal interference, in order to determine whether pitch traces are inherently weaker in amusic individuals. Memory for the pitch of single tones was compared using two versions of a paradigm developed by Deutsch (1970a). In both tasks, participants compared the pitch of a standard (S) versus a comparison (C) tone. In the time task, the S and C tones were presented, separated in time by 0, 1, 5, 10, and 15 s (blocked presentation). In the interference task, the S and C tones were presented with a fixed time interval (5 s) but with a variable number of irrelevant tones in between: 0, 2, 4, 6, and 8 tones (blocked presentation). In the time task, control performance remained high for all time in tervals, but amusics showed a performance decrement over time. In the interference task, controls and amusics showed a similar performance decrement with increasing number of irrelevant tones. Overall, the results suggest that the pitch representations of amusic individuals are less stable and more prone to decay than those of matched non-amusic individuals

    Adapted conservation agriculture practices can increase energy productivity and lower yield-scaled greenhouse gas emissions in coastal Bangladesh

    While numerous studies have documented the benefits of conservation agriculture (CA) in South Asia, most focus on favorable environments where farmers have reliable access to energy supporting irrigation and inputs. The performance of CA in South Asia’s under-developed coastal environments is comparatively understudied. In these environments, farmers are increasingly interested in growing a second crop to meet food security and income generation objectives in rotation following the predominant monsoon season rice crop, though labor, energy costs, and investment constraints limit their ability to do so. We hypothesized that rotating rice (Oryza sativa) with maize (Zea mays) using conservation agriculture, or CA (i.e., strip-tilled maize followed by unpuddled transplanted rice), or seasonally alternating tillage (SAT, i.e., strip-tilled maize followed by fully-tilled, puddled rice with residues retained across rotations) would reduce costs and energy use, increase energy-use efficiency, and reduce yield-scaled CO2-eq emissions (YSE) and total global warming potential (GWP), compared to farmers’ own practices (FP) and conventional full-tillage (CT) under the same rotation in Bangladesh’s coastal region. Starting with winter maize followed by summer rice, we evaluated four tillage and crop establishment treatments in farmer-managed experiments in partially irrigated and rainfed environments over three years in 35 farmer’s fields across Bangladesh’s coastal districts. Treatments included FP, CT, complete CA, and SAT under a rice-maize rotation. Across years, the full suite of CA practices and SAT were significantly more energy-efficient and energy-productive than FP or CT. The order of YSE in rice was CA< CT or FP < SAT while in maize, it was CA or SAT < FP < CT. Across environments, CA and SAT resulted in 15-18% higher yield at the cropping systems level (maize and rice yields combined) and 26-40% less manual labor than CT or FP. CA and SAT also reduced by 1-12% and 33-35% total production costs respective to CT and FP. This was associated with 13-17% greater grain energy output in CA and SAT, and 2-18% lower YSE, compared to CT or FP. While our data suggest that both CA and SAT can result in a range of positive agronomic, economic, and environmental outcomes compared to FP or CT, post-trial surveys and discussions with farmers revealed a strong practical aversion to use of the full suite of CA practices and preference for adapted practices due to logistical constraints in negotiating the hire of laborers for unpuddled manual transplanting
