    Random coefficient modeling research on short-term forecast of passenger flow into an urban rail transit station

    Taking a representative metro station in Beijing as example, this research has newly developed a random coefficient model to predict the short-term passenger flows with sudden increases sometimes into an urban rail transit station. The hierarchical Bayesian approach is iteratively applied in this work to estimate the new model and the estimation outcomes in each of the iterative calibrations are improved by sequential Bayesian updating. It has been proved that the estimation procedure is able to effectively converge to rational results with satisfying accuracies. In addition, the model application study reveals that besides sufficient preparations in manpower, devices, etc.; the information of the factors affecting the passenger flows into an urban rail transit station should be timely transferred in advance from important buildings, road intersections, squares and so on in neighborhood to this station. In this way, this station is able to cope with the unexpectedly sharp increases of the passenger flows into the station to ensure its operation safety

    [Purpose/significance] This paper aims to discover the factors affecting user behavior in the derivative situation of e-commerce, social commerce, and explore the sustainable development and related marketing advice of it. [Method/process]This paper put forward a theoretical model of factors affecting user behavior in social commerce by integrating emotional state impact into the Stimulus-Organism-Response (S-O-R) framework. 277 valid samples were collected by questionnaires and PLS. [Result/conclusion]The results show that information quality and tie strength significantly affect user emotional states, while emotional states positively affect user behavior. In addition, graphic features of business information have indirect effects on user emotional states, while it has direct effect on purchase intention

    Quantitative Proteomics and Functional Characterization Reveal That Glutathione Peroxidases Act as Important Antioxidant Regulators in Mulberry Response to Drought Stress

    Mulberry (Morus alba L.) has been an economically important food crop for the domesticated silkworm, Bombyx mori, in China for more than 5000 years. However, little is known about the mechanism underlying mulberry response to environmental stress. In this study, quantitative proteomics was applied to elucidate the molecular mechanism of drought response in mulberry. A total of 604 differentially expressed proteins (DEPs) were identified via LC-MS/MS. The proteomic profiles associated with antioxidant enzymes, especially five glutathione peroxidase (GPX) isoforms, as a scavenger of reactive oxygen species (ROS), were systematically increased in the drought-stressed mulberry. This was further confirmed by gene expression and enzymatic activity. Furthermore, overexpression of the GPX isoforms led to enhancements in both antioxidant system and ROS-scavenging capacity, and greater tolerance to drought stress in transgenic plants. Taken together, these results indicated that GPX-based antioxidant enzymes play an important role in modulating mulberry response to drought stress, and higher levels of GPX can improve drought tolerance through enhancing the capacity of the antioxidant system for ROS scavenging

    Excess total organic carbon in the intermediate water of the South China Sea and its export to the North Pacific

    Depth profiles of total organic carbon (TOC) were measured in spring (2005) and winter (2006) in the South China Sea (SCS), the largest marginal sea adjacent to the North Western Pacific (NWP). Compared to TOC profiles in the NWP, excess TOC (3.2 +/- 1.1 mu mol kg(-1)) was revealed in the intermediate layer of the SCS at sigma(theta) similar to 27.2-27.6 (similar to 1000-1500 m). Below the depth of 2000 m, TOC concentrations were identical between the SCS and the NWP. Based on a one-dimensional steady state diffusion advection model constrained by potential temperature, we estimated a net TOC production rate of 0.12 +/- 0.04 mu mol kg(-1) yr(-1) to maintain this excess. A positive relationship between TOC and apparent oxygen utilization in the SCS deep water lent support to such a model-derived TOC production. This excess TOC in the out-flowing intermediate water may carry 3.1 +/- 2.1 Tg C yr(-1) of organic carbon out from the SCS and potentially into the deep open ocean. In light of the short residence time of the SCS deep water, the exported TOC was likely from the recently fixed organic carbon within the SCS. The export of such organic carbon, thereby less likely to return to the atmosphere may therefore contribute significantly to the carbon sequestration in the SCS

    G-quadruplex inducer/stabilizer pyridostatin targets SUB1 to promote cytotoxicity of a transplatinum complex

    Pyridostatin (PDS) is a well-known G-quadruplex (G4) inducer and stabilizer, yet its target genes have remained unclear. Herein, combining mass spectrometry based proteomics strategy with bioinformatics analysis, we revealed that PDS significantly downregulated 22 proteins, of which the genes contain rich G4 potential sequences, in HeLa cancer cells, consequently upregulating 16 proteins remarkably. The PDS-regulated proteins appeared to work synergistically to activate cyclin and cell cycle regulation, and to restrain the inhibition of ARE-mediated mRNA decay pathway, suggesting that PDS itself is not a potential anticancer agent, at least towards HeLa cancer. Importantly, among the PDS targeted genes, SUB1, which expresses the human positive cofactor and DNA lesion sensor PC4, was down-regulated by 4.76-fold. Further studies demonstrated that the downregulation of PC4 dramatically promoted the cytotoxicity of trans-[PtCl2(NH3)(thiazole)] towards HeLa cells to a similar level to that of cisplatin, contributable to retarding the repair of 1,3-trans-platinated DNA lesion mediated by PC4. These findings not only provide new insights into better understanding on the biological functions of PDS, but also implicate a strategy for the rational design of novel multi-targeting platinum anticancer drugs via conjugation of PDS as a ligand to the coordination scaffold of transplatin for battling drug resistance to cisplatin

    Post-lockdown SARS-CoV-2 nucleic acid screening in nearly ten million residents of Wuhan, China

    Stringent COVID-19 control measures were imposed in Wuhan between January 23 and April 8, 2020. Estimates of the prevalence of infection following the release of restrictions could inform post-lockdown pandemic management. Here, we describe a city-wide SARS-CoV-2 nucleic acid screening programme between May 14 and June 1, 2020 in Wuhan. All city residents aged six years or older were eligible and 9,899,828 (92.9%) participated. No new symptomatic cases and 300 asymptomatic cases (detection rate 0.303/10,000, 95% CI 0.270–0.339/10,000) were identified. There were no positive tests amongst 1,174 close contacts of asymptomatic cases. 107 of 34,424 previously recovered COVID-19 patients tested positive again (re-positive rate 0.31%, 95% CI 0.423–0.574%). The prevalence of SARS-CoV-2 infection in Wuhan was therefore very low five to eight weeks after the end of lockdown
