    Sustainability-focused international PBL project:Rethinking digital education for individuals of low socioeconomic status

    Providing access to education for individuals of lower socio-economic status is a significant way to reduce poverty, as it empowers them to grow as professionals and as individuals. Although there is an increasing sense of urgency to promote these changes, notably motivated by the Sustainable Development Goals (SDGs) set by the UN 2030 Agenda, there are still few successful ways to solve this problem on a large-scale. As digital technology develops and affordability increases, new ways to share quality educational content are created. In an exploratory case study, with a qualitative approach, this paper presents the development of a digital application focused on providing quality educational content directed to vulnerable groups that lack access to formal learning experiences, specifically focused on waste pickers in Brasilia, Brazil. The main data collection methods used to make the decisions through the development process were observation, surveys, and interviews. Within the framework of Problem Based Learning (PBL) an international cross-disciplinary collaboration among different universities, the project, denominated “Mobile Education”, involved: (i) the design and implementation of a system consisting of both a web and mobile application; (ii) the research of a viable business model to provide long-term sustainability for the project; (iii) and the creation of a pilot course of financial education for the preliminary target group, i.e., waste pickers from Brasilia, Brazil. Conclusively, the project aims to positively impact social transformation for individuals who work at the Waste Sorting facilities and lack financial knowledge. The Mobile Education project resulted in a functional version of the app (Web and Mobile) as well as the delimitation of a viable business model to keep it providing digital equality in Brazilian education

    Comparação do equilíbrio postural de idosos sedentários, praticantes de diferentes modalidades de exercícios e adultos jovens

    O objetivo deste estudo foi comparar o equilíbrio postural de idosos praticantes de duas modalidades de exercícios físicos, idosos não praticantes e adultos jovens, na presença e ausência de informação visual. Participaram 51 indivíduos (n = 31 do sexo feminino e n = 20 do sexo masculino), subdivididos em quatro grupos: idosos praticantes de karatê (GIKA, n = 11), idosos praticantes de ginástica generalizada (GIGG, n = 16), idosos não praticantes de exercícios físicos (GINP, n = 12), e um grupo controle de adultos jovens (GAJ, n = 12). A tarefa constituiu em permanecer em apoio estático, posição semi tandem, sobre a plataforma de força por 40 segundos. Duas condições experimentais foram realizadas: com visão e sem visão. As variáveis de controle postural obtidas a partir do centro de pressão (Cop) foram: área de oscilação do Cop, velocidade e amplitude média de oscilação do Cop nas direções ântero-posterior e médio-lateral. Os resultados indicaram que os idosos praticantes de exercícios físicos GIKA e GIGG obtiveram desempenhos semelhantes, não diferenciando a modalidade praticada, tanto na condição com visão quanto sem visão, aproximando seu desempenho de adultos jovens, mais especificamente o grupo GIGG. Na condição sem visão, todos os grupos mostraram-se mais suscetíveis à instabilidade postural. O grupo GINP apresentou maior oscilação postural em todas as variáveis analisadas quando comparado aos outros grupos, apresentando maiores diferenças com o grupo GIGG e GAJ. Sendo assim, os grupos de idosos praticantes não se diferenciaram, se aproximando ao desempenho dos adultos jovens. Compreende-se que a prática de exercícios físicos tem papel importante e eficaz na manutenção do equilíbrio postural durante o envelhecimento e pode evitar possíveis quedas

    Desenvolvimento de software para produção de biodiesel a partir do óleo residual

O biodiesel existe como uma alternativa ao diesel derivado do petróleo. Biodiesel é obtido por uma reação de transesterificação entre um óleo vegetal ou gordura animal e um álcool de cadeia curta. O óleo de soja residual é uma alternativa de utilização de um resíduo para produção desse biocombustível. O presente estudo traz o desenvolvimento de um software que vem a ser apoio na produção de biodiesel a partir do óleo residual. Foram feitas reações para produção de biodiesel a partir do óleo de soja residual alterando as variáveis: tipo de catalisador (KOH e NaOH), razão molar óleo:metanol (1:5 e 1:7) e tempo de reação de reação (40 min e 1h). O maior rendimento de óleo em biodiesel de 97,30% foi obtido na reação utilizando a razão molar óleo:metanol de 1:7, KOH como catalisador na temperatura de 50ºC por 1h. O software coleta do usuário a massa do óleo, dispondo com isto o catalisador a ser utilizado, quantidade de massa do catalisador e álcool, tempo reacional, temperatura e porção necessária ou não de HCl para posterior purificação.ABSTRACT: The demand for energy grows more each year and with it increases pollution. Biodiesel exists as an alternative to diesel derived from petroleum. Biodiesel is obtained by a transesterification reaction between a vegetable oil or animal fat and a short-chain alcohol. Residual soybean oil is an alternative to using a residue to produce this biofuel. The present study brings the development of software that comes to support the production of biodiesel from residual oil. Reactions were made for the production of biodiesel from residual soy oil by changing the variables: type of catalyst (KOH and NaOH), molar ratio oil: methanol (1: 5 and 1: 7) and reaction time of reaction (40 min and 1h). The highest oil yield in biodiesel of 97.30% was obtained in the reaction using the oil: methanol molar ratio of 1: 7, KOH as a catalyst at a temperature of 50ºC for 1h. The software collects the mass of the oil from the user, thereby providing the catalyst to be used, the amount of mass of the catalyst and alcohol, reaction time, temperature and the necessary portion or not of HCl for further purification.info:eu-repo/semantics/publishedVersio

    Schistosoma mansoni Tegument Protein Sm29 Is Able to Induce a Th1-Type of Immune Response and Protection against Parasite Infection

    Schistosomiasis is the most important human helminth infection in terms of morbidity and mortality. Although the efforts to develop a vaccine against this disease have experienced failures, a new generation of surface antigens revealed by proteomic studies changed this scenario. Our group has characterized the protein Sm29 described previously as one of the most exposed and expressed antigens in the outer tegument of Schistosoma mansoni. Studies in patients living in endemic areas for schistosomiasis revealed high levels of IgG1 and IgG3 anti-Sm29 in resistant individuals. In this study, confocal microscope analysis showed Sm29 present in the surface of lung-stage schistosoluma and adult worms. Recombinant Sm29, when used as vaccine candidate, induced high levels of protection in mice. This protection was associated with a typical Th1 immune response and reduction of worm burden, liver granulomas and in intestinal eggs. Further, microarray analysis of worms recovered from vaccinated mice showed significant down-regulation of several genes encoding previously characterized vaccine candidates and/or molecules exposed on the surface, suggesting an immune evasion strategy of schistosomes under immune attack.     Identification of Piecemeal Degranulation and Vesicular Transport of MBP-1 in Liver-Infiltrating Mouse Eosinophils During Acute Experimental Schistosoma mansoni Infection

    Eosinophils have been long associated with helminthic infections, although their functions in these diseases remain unclear. During schistosomiasis caused by the trematode Schistosoma mansoni, eosinophils are specifically recruited and migrate to sites of granulomatous responses where they degranulate. However, little is known about the mechanisms of eosinophil secretion during this disease. Here, we investigated the degranulation patterns, including the cellular mechanisms of major basic protein-1 (MBP-1) release, from inflammatory eosinophils in a mouse model of S. mansoni infection (acute phase). Fragments of the liver, a major target organ of this disease, were processed for histologic analyses (whole slide imaging), conventional transmission electron microscopy (TEM), and immunonanogold EM using a pre-embedding approach for precise localization of major basic protein 1 (MBP-1), a typical cationic protein stored pre-synthesized in eosinophil secretory (specific) granules. A well-characterized granulomatous inflammatory response with a high number of infiltrating eosinophils surrounding S. mansoni eggs was observed in the livers of infected mice. Moreover, significant elevations in the levels of plasma Th2 cytokines (IL-4, IL-13, and IL-10) and serum enzymes (alanine aminotransferase and aspartate aminotransferase) reflecting altered liver function were detected in response to the infection. TEM quantitative analyses revealed that while 19.1% of eosinophils were intact, most of them showed distinct degranulation processes: cytolysis (13.0%), classical and/or compound exocytosis identified by granule fusions (1.5%), and mainly piecemeal degranulation (PMD) (66.4%), which is mediated by vesicular trafficking. Immunonanogold EM showed a consistent labeling for MBP-1 associated with secretory granules. Most MBP-1-positive granules had PMD features (79.0 ± 4.8%). MBP-1 was also present extracellularly and on vesicles distributed in the cytoplasm and attached to/surrounding the surface of emptying granules. Our data demonstrated that liver-infiltrating mouse eosinophils are able to degranulate through different secretory processes during acute experimental S. mansoni infections with PMD being the predominant mechanism of eosinophil secretion. This means that a selective secretion of MBP-1 is occurring. Moreover, our study demonstrates, for the first time, a vesicular trafficking of MBP-1 within mouse eosinophils elicited by a helminth infection. Vesicle-mediated secretion of MBP-1 may be relevant for the rapid release of small concentrations of MBP-1 under cell activation

    In the Brazilian Amazon, two monospecific genera, the Harpy Eagle and Crested Eagle have low densities and are classified by IUCN as Near Threatened due to habitat loss, deforestation, habitat degradation and hunting. In this study, we evaluate occurrence of these large raptors using the environmental surveys database from Belo Monte Hydroelectric Power Plant. Integrating the dataset from two methods, we plotted a distribution map along the Xingu River, including records over a 276-km stretch of river. Terrestrial surveys (RAPELD method) were more efficient for detecting large raptors than standardized aquatic surveys, although the latter were complementary in areas without modules. About 53% of the records were obtained during activities of wildlife rescue/flushing, vegetation suppression or in transit. Between 2012 and 2014, four Harpy Eagles were removed from the wild; two shooting victims, one injured by collision with power lines and one hit by a vehicle. Also, seven nests were mapped. The mean distance between Harpy Eagle records was 15 km along the river channel, with a mean of 20 km between nests near the channel, which allowed us to estimate 20 possible pairs using the alluvial forest, riverine forest and forest fragments. Territories of another ten pairs will probably be affected by inundation of the Volta Grande channel, which is far from the main river. The average distance between Crested Eagle records was 16 km along the river channel. The only nest found was 1.3 km away from a Harpy Eagle nest. The remnant forests are under threat of being replaced by cattle pastures, so we recommend that permanently protected riparian vegetation borders (APP) be guaranteed, and that forest fragments within 5 km of the river be conserved to maintain eagle populations. © 2015, Instituto Internacional de Ecologia.     High anti-SARS-CoV-2 antibody seroconversion rates before the second wave in Manaus, Brazil, and the protective effect of social behaviour measures: results from the prospective DETECTCoV-19 cohort

    Background: The city of Manaus, Brazil, has seen two collapses of the health system due to the COVID-19 pandemic. We report anti-SARS-CoV-2 nucleocapsid IgG antibody seroconversion rates and associated risk factors in Manaus residents before the second wave of the epidemic in Brazil. Methods: A convenience sample of adult (aged ≥18 years) residents of Manaus was recruited through online and university website advertising into the DETECTCoV-19 study cohort. The current analysis of seroconversion included a subgroup of DETECTCoV-19 participants who had at least two serum sample collections separated by at least 4 weeks between Aug 19 and Oct 2, 2020 (visit 1), and Oct 19 and Nov 27, 2020 (visit 2). Those who reported (or had no data on) having a COVID-19 diagnosis before visit 1, and who were positive for anti-SARS-CoV-2 nucleocapsid IgG antibodies at visit 1 were excluded. Using an in-house ELISA, the reactivity index (RI; calculated as the optical density ratio of the sample to the negative control) for serum anti-SARS-CoV-2 nucleocapsid IgG antibodies was measured at both visits. We calculated the incidence of seroconversion (defined as RI values ≤1·5 at visit 1 and ≥1·5 at visit 2, and a ratio >2 between the visit 2 and visit 1 RI values) during the study period, as well as incidence rate ratios (IRRs) through cluster-corrected and adjusted Poisson regression models to analyse associations between seroconversion and variables related to sociodemographic characteristics, health access, comorbidities, COVID-19 exposure, protective behaviours, and symptoms. Findings: 2496 DETECTCoV-19 cohort participants returned for a follow-up visit between Oct 19 and Nov 27, 2020, of whom 204 reported having COVID-19 before the first visit and 24 had no data regarding previous disease status. 559 participants were seropositive for anti-SARS-CoV-2 nucleocapsid IgG antibodies at baseline. Of the remaining 1709 participants who were seronegative at baseline, 71 did not meet the criteria for seroconversion and were excluded from the analyses. Among the remaining 1638 participants who were seronegative at baseline, 214 showed seroconversion at visit 2. The seroconversion incidence was 13·06% (95% CI 11·52–14·79) overall and 6·78% (5·61–8·10) for symptomatic seroconversion, over a median follow-up period of 57 days (IQR 54–61). 48·1% of seroconversion events were estimated to be asymptomatic. The sample had higher proportions of affluent and higher-educated people than those reported for the Manaus city population. In the fully adjusted and corrected model, risk factors for seroconversion before visit 2 were having a COVID-19 case in the household (IRR 1·49 [95% CI 1·21–1·83]), not wearing a mask during contact with a person with COVID-19 (1·25 [1·09–1·45]), relaxation of physical distancing (1·31 [1·05–1·64]), and having flu-like symptoms (1·79 [1·23–2·59]) or a COVID-19 diagnosis (3·57 [2·27–5·63]) between the first and second visits, whereas working remotely was associated with lower incidence (0·74 [0·56–0·97]). Interpretation: An intense infection transmission period preceded the second wave of COVID-19 in Manaus. Several modifiable behaviours increased the risk of seroconversion, including non-compliance with non-pharmaceutical interventions measures such as not wearing a mask during contact, relaxation of protective measures, and non-remote working. Increased testing in high-transmission areas is needed to provide timely information about ongoing transmission and aid appropriate implementation of transmission mitigation measures.
