    Co-operative learning and adaptive instruction in a mathematics curriculum

    The AGO 12 to 16 Project (the acronym AGO stands for the Dutch equivalent of 'Adaptive Instruction and Co-operative Learning') seeks to develop and evaluate a mathematics curriculum which is suitable for mixed-ability groups in secondary education. The research questions we will address here are, first, whether this curriculum is feasible and effective, and, second, what effects, if any, the context variables time and mean cognitive level of the class have on learning. Many mathematics programmes make insufficient allowance for the differences in intellectual ability that exist in mixed-ability classes. In order to change this situation we developed a mathematics curriculum with adaptive qualities. The evaluation of the experimental curriculum was carried out in two stages. During the first stage the curriculum was used at two schools with the aim of investigating the feasibility of the programme. Experience with the implementation of the programme led to some improvements in the experimental materials. By and large the AGO model appeared to be feasible in secondary classrooms. In the second stage, which was on a large scale, the focus was on the effectiveness of the programme. Six hundred students, 13 teachers and six schools were involved in the research. Teachers in the experimental group were trained in AGO methods and in implementing the new AGO curriculum. Teachers in the control groups worked with the existing programme following their usual methods of teaching. The main conclusion of the study is positive. The AGO model as a whole proved to be practical and effective in learning mathematics. The AGO model has a positive effect on the intercept, which means that the mean scores of AGO classes are higher than the mean scores of non-AGO classes. It may be concluded that, on the average, students benefit from learning in AGO classes as compared with non-AGO classes. AGO does not increase or decrease the differences between students in the same class. As expected, positive effects of two context variables were found: (1) the total amount of time spent in class covering the mathematical content and (2) class composition as indicated by the mean pretest score (aptitude) of the class

    The merger of vertically offset quasi-geostrophic vortices

    We examine the critical merging distance between two equal-volume, equal-potential-vorticity quasi-geostrophic vortices. We focus on how this distance depends on the vertical offset between the two vortices, each having a unit mean height-to-width aspect ratio. The vertical direction is special in the quasi-geostrophic model (used to capture the leading-order dynamical features of stably stratified and rapidly rotating geophysical flows) since vertical advection is absent. Nevertheless vortex merger may still occur by horizontal advection. In this paper, we first investigate the equilibrium states for the two vortices as a function of their vertical and horizontal separation. We examine their basic properties together with their linear stability. These findings are next compared to numerical simulations of the nonlinear evolution of two spheres of potential vorticity. Three different regimes of interaction are identified, depending on the vertical offset. For a small offset, the interaction differs little from the case when the two vortices are horizontally aligned. On the other hand, when the vertical offset is comparable to the mean vortex radius, strong interaction occurs for greater horizontal gaps than in the horizontally aligned case, and therefore at significantly greater full separation distances. This perhaps surprising result is consistent with the linear stability analysis and appears to be a consequence of the anisotropy of the quasi-geostrophic equations. Finally, for large vertical offsets, vortex merger results in the formation of a metastable tilted dumbbell vortex.Publisher PDFPeer reviewe

    Selective loss of noradrenaline exacerbates early cognitive dysfunction and synaptic deficits in APP/PS1 mice.

    Background: Degeneration of the locus coeruleus (LC), the major noradrenergic nucleus in the brain, occurs early and is ubiquitous in Alzheimer’s disease (AD). Experimental lesions to the LC exacerbate AD-like neuropathology and cognitive deficits in several transgenic mouse models of AD. Because the LC contains multiple neuromodulators known to affect amyloid toxicity and cognitive function, the specific role of noradrenaline (NA) in AD is not well understood. Methods: To determine the consequences of selective NA deficiency in an AD mouse model, we crossed dopamine -hydroxylase (DBH) knockout mice with amyloid precursor protein (APP)/presenilin-1 (PS1) mice overexpressing mutant APP and PS1. Dopamine -hydroxylase (/) mice are unable to synthesize NA but otherwise have normal LC neurons and co-transmitters. Spatial memory, hippocampal long-term potentiation, and synaptic protein levels were assessed. Results: The modest impairments in spatial memory and hippocampal long-term potentiation displayed by young APP/PS1 or DBH (/) single mutant mice were augmented in DBH (/)/APP/PS1 double mutant mice. Deficits were associated with reduced levels of total calcium/calmodulin-dependent protein kinase II and N-methyl-D-aspartate receptor 2A and increased N-methyl-D-aspartate receptor 2B levels and were independent of amyloid accumulation. Spatial memory performance was partly improved by treatment with the NA precursor drug L-threo-dihydroxyphenylserine. Conclusions: These results indicate that early LC degeneration and subsequentNAdeficiency inADmay contribute to cognitive deficits via altered levels of calcium/calmodulin-dependent protein kinase II and N-methyl-D-aspartate receptors and suggest that NA supplementation could be beneficial in early AD

    Towards a knowledge-rich learning environment in preparatory secondary education

    In this case study a novel educational programme for students in preparatory vocational education was studied. The research questions were: (1) Which teaching/learning processes occur in a simulated workplace using the concept of a knowledge-rich workplace? (2) What is the role of models and modelling in the teaching/learning processes? The curriculum project consisted of design and construction tasks. The students were collaboratively involved in the process of designing a tricycle for a real customer. This real-life activity creates opportunities for students to develop and use models, which can be used in more than in one context. The case study explored how the teachers deal with the students' explicit and implicit need for knowledge and skills. The main findings are that teachers more often provide this knowledge, rather than guide the students in reconstructing it, and towards the end of the project, knowledge tended to remain situated

    Photometric study of the late-time near-infrared plateau in Type Ia supernovae

    We present an in-depth study of the late-time near-infrared plateau in Type Ia supernovae (SNe Ia), which occurs between 70-500 d. We double the existing sample of SNe Ia observed during the late-time near-infrared plateau with new observations taken with the Hubble Space Telescope, Gemini, New Technology Telescope, the 3.5m Calar Alto Telescope, and the Nordic Optical Telescope. Our sample consists of 24 nearby SNe Ia at redshift < 0.025. We are able to confirm that no plateau exists in the Ks band for most normal SNe Ia. SNe Ia with broader optical light curves at peak tend to have a higher average brightness on the plateau in J and H, most likely due to a shallower decline in the preceding 100 d. SNe Ia that are more luminous at peak also show a steeper decline during the plateau phase in H. We compare our data to state-of-the-art radiative transfer models of nebular SNe Ia in the near-infrared. We find good agreement with the sub-Mch model that has reduced non-thermal ionisation rates, but no physical justification for reducing these rates has yet been proposed. An analysis of the spectral evolution during the plateau demonstrates that the ratio of [Fe II] to [Fe III] contribution in a near-infrared filter determines the light curve evolution in said filter.     Generation and characterization of antibodies specific for caspase-cleaved neo-epitopes: a novel approach

    Apoptosis research has been significantly aided by the generation of antibodies against caspase-cleaved peptide neo-epitopes. However, most of these antibodies recognize the N-terminal fragment and are specific for the protein in question. The aim of this project was to create antibodies, which could identify caspase-cleaved proteins without a priori knowledge of the cleavage sites or even the proteins themselves. We hypothesized that many caspase-cleavage products might have a common antigenic shape, given that they must all fit into the same active site of caspases. Rabbits were immunized with the eight most prevalent exposed C-terminal tetrapeptide sequences following caspase cleavage. After purification of the antibodies we demonstrated (1) their specificity for exposed C-terminal (but not internal) peptides, (2) their ability to detect known caspase-cleaved proteins from apoptotic cell lysates or supernatants from apoptotic cell culture and (3) their ability to detect a caspase-cleaved protein whose tetrapeptide sequence differs from the eight tetrapeptides used to generate the antibodies. These antibodies have the potential to identify novel neo-epitopes produced by caspase cleavage and so can be used to identify pathway-specific caspase cleavage events in a specific cell type. Additionally this methodology may be applied to generate antibodies against products of other proteases, which have a well-defined and non-promiscuous cleavage activity

    SN 2022joj: A Peculiar Type Ia Supernova Possibly Driven by an Asymmetric Helium-shell Double Detonation

    We present observations of SN 2022joj, a peculiar Type Ia supernova (SN Ia) discovered by the Zwicky Transient Facility (ZTF). SN 2022joj exhibits an unusually red gZTFrZTFg_\mathrm{ZTF}-r_\mathrm{ZTF} color at early times and a rapid blueward evolution afterwards. Around maximum brightness, SN 2022joj shows a high luminosity (MgZTF,max19.7M_{g_\mathrm{ZTF},\mathrm{max}}\simeq-19.7 mag), a blue broadband color (gZTFrZTF0.2g_\mathrm{ZTF}-r_\mathrm{ZTF}\simeq-0.2 mag), and shallow Si II absorption lines, consistent with those of overluminous, SN 1991T-like events. The maximum-light spectrum also shows prominent absorption around 4200 \r{A}, which resembles the Ti II features in subluminous, SN 1991bg-like events. Despite the blue optical-band colors, SN 2022joj exhibits extremely red ultraviolet - optical colors at maximum luminosity (uv1.6u-v\simeq1.6 mag and uvw1v4.0uvw1 - v\simeq4.0 mag), suggesting a suppression of flux between \sim2500--4000 \r{A}. Strong C II lines are also detected at peak. We show that these unusual spectroscopic properties are broadly consistent with the helium-shell double detonation of a sub-Chandrasekhar mass (M1MM\simeq1\mathrm{M_\odot}) carbon/oxygen (C/O) white dwarf (WD) from a relatively massive helium shell (Ms0.04M_s\simeq0.04--0.1M0.1\mathrm{M_\odot}), if observed along a line of sight roughly opposite to where the shell initially detonates. None of the existing models could quantitatively explain all the peculiarities observed in SN 2022joj. The low flux ratio of [Ni II] λ\lambda7378 to [Fe II] λ\lambda7155 emission in the late-time nebular spectra indicates a low yield of stable Ni isotopes, favoring a sub-Chandrasekhar mass progenitor. The significant blueshift measured in the [Fe II] λ\lambda7155 line is also consistent with an asymmetric chemical distribution in the ejecta, as is predicted in double-detonation models.Comment: 24 pages, 11 figures, 6 tables.     Age-Related Impairment of Ultrasonic Vocalization in Tau.P301L Mice: Possible Implication for Progressive Language Disorders

    Tauopathies, including Alzheimer's Disease, are the most frequent neurodegenerative diseases in elderly people and cause various cognitive, behavioural and motor defects, but also progressive language disorders. For communication and social interactions, mice produce ultrasonic vocalization (USV) via expiratory airflow through the larynx. We examined USV of Tau.P301L mice, a mouse model for tauopathy expressing human mutant tau protein and developing cognitive, motor and upper airway defects.At age 4-5 months, Tau.P301L mice had normal USV, normal expiratory airflow and no brainstem tauopathy. At age 8-10 months, Tau.P301L mice presented impaired USV, reduced expiratory airflow and severe tauopathy in the periaqueductal gray, Kolliker-Fuse and retroambiguus nuclei. Tauopathy in these nuclei that control upper airway function and vocalization correlates well with the USV impairment of old Tau.P301L mice.In a mouse model for tauopathy, we report for the first time an age-related impairment of USV that correlates with tauopathy in midbrain and brainstem areas controlling vocalization. The vocalization disorder of old Tau.P301L mice could be, at least in part, reminiscent of language disorders of elderly suffering tauopathy
