A rapid, non invasive and simple molecular method for sex determination in pygoscelis penguins
In Adélie penguins (Pygoscelis adeliae) and gentoo penguins (Pygoscelis papua), the conspicuous sexual dimorphism often makes it difficult to determine sex on the basis of external morphology. The information about sex is important in many ecology and conservation studies. In this paper we evaluated the use of an established primer pair (2550F/2718R) to identify sex in sexually monomorphic birds. In both penguin species, it resulted in two distinct CHD1Z and CHD1W PCR bands, allowing sex identification. This is a simple, rapid and cheap system for molecular sexing of gentoo and Adélie penguins.Instituto Multidisciplinario de Biología CelularFacultad de Ciencias Naturales y MuseoConsejo Nacional de Investigaciones Científicas y Técnica
Physiological and proteomic response of Escherichia coli O157:H7 to a bioprotective lactic acid bacterium in a meat environment
The enterohemorrhagic Escherichia (E.) coli (EHEC) is a pathogen of great concern for public health and the meat industry all over the world. The high economic losses in meat industry and the high costs of the illness highlight the necessity of additional efforts to control this pathogen. Previous studies have demonstrated the inhibitory activity of Enterococcus mundtii CRL35 towards EHEC, showing a specific proteomic response during the co-culture. In the present work, additional studies of the EHEC-Ent. mundtii interaction were carried out: i) differential protein expression of E. coli O157:H7 NCTC12900 growing in co-culture with Ent. mundtii in a meat environment, ii) the reciprocal influence between these two microorganisms in the adhesion to extracellular matrix (ECM) proteins and iii) the possible induction of the phage W933, coding for Shiga toxin (Stx1), by Ent. mundtii CRL35. Proteomic analysis showed a significant repression of a number of E. coli NCTC12900 proteins in co-culture respect to its single culture, these mostly related to the metabolism and transport of amino acids and nucleotides. On the other hand, statistically significant overexpression of EHEC proteins involved in stress, energy production, amino acid metabolism and transcription was observed at 30 h respect to 6 h when EHEC grew in co-culture. Data are available via ProteomeXchange with identifier PXD014588. Besides, EHEC showed a decreased adhesion capacity to ECM proteins in the presence of the bioprotective strain. Finally, Ent. mundtii CRL35 did not induce the lytic cycle of W933 bacteriophage, thus indicating its potential safe use for eliminating this pathogen. Overall, this study expands the knowledge of EHEC- Ent. mundtii CRL35 interaction in a meat environment, which will certainly contribute to find out effective biological strategies to eliminate this pathogen.Fil: Orihuel, Alejandra. From Neighborhoods To Wellbeing And Conservation: Enhancing The Use Of Greenspace Through Walkability
In drylands, it is essential to maximize the coupling of social and ecological systems in order to achieve sustainability, particularly in human dominated landscapes such as cities. The enhanced use of greenspace in cities in drylands provides unique opportunities to maximize the coupling of social and ecological systems. It maintains the functioning of ecological systems while involving civil society in the conservation of biodiversity and improving human wellbeing in urban settings. The provision and access to greenspace in cities is determined by neighborhood design. The access for the human use of greenspace can be enhanced through walkability, or the characteristics of the built environment that influence physical activity. Walkable neighborhoods that provide access to greenspace can be catalysts for activity and health and have the potential to increase the level of conservation support in urban residents. The purpose of this research is to look for wellbeing and conservation synergies between walkable neighborhoods and the enhanced use of greenspace. To accomplish this research, first we assess walkability in the built environment through an interdisciplinary literature review that integrates the findings on walkability from several research domains. We create a conceptual framework that organizes the neighborhood design elements that influence physical activity into nine walkability categories: connectivity, land-use, density, traffic safety, surveillance, parking, experience, greenspace, and community. We call this the Walkability Framework. This analysis allows us to identify gaps and strengths of walkability in the Leadership for Energy and Environmental Design for Neighborhood Development (LEED-ND) certification system. After a quantitative and qualitative analysis, we propose an enhanced version for walkability that we call LEED-NDW+ (walkability plus). The next step is to test if the Walkability Framework can be used as a model to measure the interactions between the built environment and physical activity. We accomplish this through the use of a questionnaire (N=486) that captures the perceptions, attitudes and behaviors of residents in Tucson, Arizona. Significant correlations between all the walkability categories and physical activity support the use of the framework as a model. We call this the Walkability Model. The final stage of this research uses the Walkability Model to evaluate walkability in four neighborhood design types in Tucson that include traditional development, suburban development, enclosed community, and cluster housing. We then look for wellbeing and conservation synergies between walkable neighborhoods and the enhanced use of greenspace. Results from this study suggest that neighborhoods with a high level of walkability have the potential to enhance the use of greenspace, which in turn provide important wellbeing and conservation synergies that can contribute to healthier communities and increase the support for conservation of biodiversity within and beyond cities. The enhanced use of greenspace maximizes the coupling of social and ecological systems in cities in drylands, which increases resilience in the face of climate change
Emotion elicitation and subjective feeling’s evaluation in virtual reality
Virtual reality (VR) technology is a highly promising tool for research in psychology and affective sciences. This research aimed to explore to what extent full VR elements (3 dimensions and movement) can be compared with elements related to more traditional methods (e.g., photos or videos in 2 dimensions, static or dynamic), in their impact on emotion elicitation. Sixty participants observed twenty-four stimuli (a koala, a frog and a spider) presented in 2 or 3 dimensions and in a static or a dynamic format, twice. Physiological measurements were recorded during the experiment and, participants evaluated their subjective feeling using three tools (twenty participants used an intensity slider; twenty participants used the Geneva emotion wheel and twenty participants used a self-report manikin registering bodily location of feelings). No interactions between animal, dimension and format were found influencing subjective feeling or physiological responses. Some evidence of an effect of format was found through two of the tools used to evaluate subjective feeling (intensity slider and manikin), with dynamic images that tended to induce more intense feelings than static images
Genomic and proteomic characterization of two strains of Shigella flexneri 2 isolated from infants’ stool samples in Argentina
Background: Shigella specie is a globally important intestinal pathogen disseminated all over the world. In this study we analyzed the genome and the proteomic component of two Shigella flexneri 2a clinical isolates, collected from pediatric patients with gastroenteritis of the Northwest region of Argentina (NWA) in two periods of time, with four years of difference. Our goal was to determine putative changes at molecular levels occurred during these four years, that could explain the presence of this Shigella`s serovar as the prevalent pathogen in the population under study. Results: As previously reported, our findings support the idea of Shigella has a conserved “core” genome, since comparative studies of CI133 and CI172 genomes performed against 80 genomes obtained from the NCBI database, showed that there is a large number of genes shared among all of them. However, we observed that CI133 and CI172 harbors a small number of strain-specific genes, several of them present in mobile genetic elements, supporting the hypothesis that these isolates were established in the population by horizontal acquisition of genes. These differences were also observed at proteomic level, where it was possible to detect the presence of certain secreted proteins in a culture medium that simulates the host environment. Conclusion: Great similarities were observed between the CI133 and CI172 strains, confirming the high percentage of genes constituting the “core” genome of S. flexneri 2. However, numerous strain specific genes were also determined. The presence of the here identified molecular elements into other strain of our culture collation, is currently used to develop characteristic markers of local pathogens. In addition, the most outstanding result of this study was the first description of a S. flexneri 2 producing Colicin E, as one of the characteristics that allows S. flexneri 2 to persist in the microbial community. These findings could also contribute to clarify the mechanism and the evolution strategy used by this pathogen to specifically colonize, survive, and cause infection within the NWA population.Fil: Torrez Lamberti, Monica Florencia. Relationship of Alcohol Consumption With Overweight and Obesity in Adults in an Andean Province and six Amazonian Provinces in Ecuador
The caloric intake of alcohol intake in consumers is underestimated, so its relationship with overweight and obesity is poorly understood. The objective of the study was to determine the relationship between alcohol consumption and overweight and obesity in adults in an Andean province and six amazonian provinces of Ecuador in 2021. Through a virtual survey disseminated through social networks that included: socioeconomic data; anthropometric and health data; physical activity; alcohol and tobacco consumption and dietary intake. A total of 309 surveys were obtained for statistical analysis. The R program version 4.0.1 was used and the Chi square test was used. It was found that alcohol consumption is related to overweight and obesity, in addition to this consumption is higher in adults of amazonian provinces than in the andean province. In conclusion, excessive consumption of alcoholic beverages such as: wine, beer and spirits are related to overweight and obesity, determined with Body Mass Index (BMI). Therefore, the consumption is an important aspect to consider to evaluate the nutritional status of the adult population
