    Geometric uncertainty models for correspondence problems in digital image processing

    Many recent advances in technology rely heavily on the correct interpretation of an enormous amount of visual information. All available sources of visual data (e.g. cameras in surveillance networks, smartphones, game consoles) must be adequately processed to retrieve the most interesting user information. Therefore, computer vision and image processing techniques gain significant interest at the moment, and will do so in the near future. Most commonly applied image processing algorithms require a reliable solution for correspondence problems. The solution involves, first, the localization of corresponding points -visualizing the same 3D point in the observed scene- in the different images of distinct sources, and second, the computation of consistent geometric transformations relating correspondences on scene objects. This PhD presents a theoretical framework for solving correspondence problems with geometric features (such as points and straight lines) representing rigid objects in image sequences of complex scenes with static and dynamic cameras. The research focuses on localization uncertainty due to errors in feature detection and measurement, and its effect on each step in the solution of a correspondence problem. Whereas most other recent methods apply statistical-based models for spatial localization uncertainty, this work considers a novel geometric approach. Localization uncertainty is modeled as a convex polygonal region in the image space. This model can be efficiently propagated throughout the correspondence finding procedure. It allows for an easy extension toward transformation uncertainty models, and to infer confidence measures to verify the reliability of the outcome in the correspondence framework. Our procedure aims at finding reliable consistent transformations in sets of few and ill-localized features, possibly containing a large fraction of false candidate correspondences. The evaluation of the proposed procedure in practical correspondence problems shows that correct consistent correspondence sets are returned in over 95% of the experiments for small sets of 10-40 features contaminated with up to 400% of false positives and 40% of false negatives. The presented techniques prove to be beneficial in typical image processing applications, such as image registration and rigid object tracking

    This paper addresses the possibilities of implementing Computer Integrated Manufacturing in the process industry, and the chemical industry in particular. After presenting some distinct differences of the process industry in relation to discrete manufacturing, a number of focal points are discussed. They identify the adaptations to be made to a conventional CIM system, so that it will be applicable to the process industry.     A production model and maintenance planning model for the process industry

    In this paper a model is developed to simultaneously plan preventive maintenance and production in a process industry environment, where maintenance planning is extremely important. The model schedules production jobs and preventive maintenance jobs, while minimizing costs associated with production, backorders, corrective maintenance and preventive maintenance. The formulation of the model is flexible, so that it can be adapted to several production situations.     ‘Leven is scheiden van degene die we waren om op te gaan die we moeten worden, een altijd vreemde toekomst.’ -Octavio Paz (1914-1998) Synopsis Met dit vanuit kritisch humanistisch, hermeneutische invalshoek verrichte kwalitatieve onderzoek aangevuld met literatuuronderzoek is gepoogd om de tot achtergronddissensus verworden communicatieve onafgestemdheid binnen AAAS in kaart te brengen en in een theoretisch perspectief te plaatsen. Het verkennende karakter van dit onderzoek is gericht op het sensibiliseren en het vergroten van de speelruimte in het individuele respectievelijk organisatorische denken en handelen. Het zij benadrukt dat voor deze communicatieve onafgestemdheid of dissensus noch een eenduidige causale verklaring noch een eenduidige oplossing gegeven kan worden. Overeenkomstig de humanistisch levensbeschouwelijke insteek is het onderzoek juist gericht op het op zinvolle manier creëren van de organisatorische voorwaarden om gezamenlijk tot overeenstemming en activiteiten te komen. Vanuit een synthetiserend theoretisch perspectief, te weten de concepten besefcontext, communicatief handelen, is deze achtergronddissensus te duiden als een communicatieve blokkering tussen de directie en het managementteam enerzijds en de werknemers anderzijds. In het licht van een ontbrekend communicatief management en toenemend strategisch handelen gepaard aan ontbrekend metabesef is binnen AAAS een vicieuze cirkel van niet communiceren met alle gevolgen van dien ontstaan. De directie en het management zullen deze cirkel enkel kunnen doorbreken vanuit een gesensibiliseerd metabesef over de besefcontextuele verschillen tussen henzelf en de werkvloer. Door het implementeren van door de concepten normatieve professionaliteit en cocreatie gedragen horizontale leerprocessen kan gewerkt worden aan een breed gedragen organisatorische situatiedefinitie en naar een bedrijfscultuur op basis van transparante gedeelde waarden. Met zo’n insteek wordt bovendien ook recht gedaan aan het beginsel van verantwoord ondernemen waarin human capital het leidende beginsel is. Door invulling te geven aan humaniserend waardegebonden arbobeleid binnen de eigen organisatorische geledingen zou AAAS zelfs tot innoverend voorbeeld kunnen gaan strekken

    Primate population dynamics over 32.9 years at Ngogo, Kibale National Park, Uganda

    We present census data for eight primate species spanning 32.9 years along the same transect at Ngogo, Kibale National Park, Uganda, demonstrating major changes in the composition of the primate community. Correlated with an estimated decline of ∼89% in the red colobus population was an increase in encounter rates with chimpanzee parties. Our data, along with the unusually high rates of predation by chimpanzees on red colobus at Ngogo and the fact that the chimpanzee community at Ngogo is the largest ever recorded, support the conclusion that the red colobus decline was caused primarily by chimpanzee predation. This seems to be the first documented case of predation by one nonhuman primate causing the population decline in another. We evaluated disease and interspecific competition as other possible causes of the red colobus decline, but judged them to be relatively insignificant compared with predation by chimpanzees. Notable changes in encounter rates with other primate species may have resulted from forest expansion. Those for mangabeys, redtails, and black and white colobus increased significantly. Encounter rates increased for l'Hoest's monkeys too, but the increased sightings may have been an artifact of increased habituation. Sightings of blue monkey and baboon groups declined. There was no significant change in encounter rates for all species combined. The Ngogo primate community seemed to be in a nonequilibrium state, changing from one dominated by two species, a folivore (red colobus) and a frugivorous omnivore (redtails), to one dominated by three species of frugivorous omnivores (redtails, mangabeys, and chimpanzees).     Where there’s smoke, there’s more smoke: The social settings and friendship interactions that encourage young adults to smoke cigarettes

    Despite widespread public health campaigns in Western countries people continue to smoke cigarettes and more worryingly, young people continue to take up the habit. In this thesis it is argued that cigarette smoking practices for young adults can be understood in terms of contributing to their sense of identity construction through friendship interactions and sociability. Data collected from email administered surveys and snowball sampling techniques, alongside secondary data from the National Drug Strategy Household Survey (2007), inform the research undertaken in this thesis and the results support the social benefits hypothesis in explaining why young adults smoke cigarettes. This study thereby suggests that in order for anti-smoking initiatives to be more successful in tackling the smoking habits of young adults additional research is required in identity formation, interactive factors and sociability factors that affect cigarette smoking practices of young adults

    Semi-high-throughput detection of Plasmodium falciparum and Plasmodium vivax oocysts in mosquitoes using bead-beating followed by circumsporozoite ELISA and quantitative PCR.

    BACKGROUND: The malaria infection status of mosquitoes is commonly determined by microscopic detection of oocysts on the dissected mosquito midgut. This method is labour-intensive, does not allow processing of large numbers of mosquitoes and can be challenging in terms of objective classification of oocysts. Here, a semi-high-throughput bead-beating ELISA method is proposed for detection of the circumsporozoite protein (CSP) followed by confirmation by quantitative PCR (qPCR). METHODS: Cultured Plasmodium falciparum gametocytes were offered to Anopheles stephensi mosquitoes and examined by microscopy. After bead-beating, mosquito homogenate was examined by CSP-ELISA and 18S qPCR. As negative controls, mosquitoes that were offered a heat-inactivated gametocyte blood meal were used. The CSP-ELISA/qPCR methodology was applied to high and low-intensity infections of cultured P. falciparum gametocytes. A similar methodology optimized for P. vivax was used on mosquitoes that were offered blood from Ethiopian donors who were naturally infected with P. vivax. RESULTS: There was considerable variation in CSP-ELISA signal and qPCR values in mosquitoes with low oocyst intensities. There was a strong agreement mosquito positivity by CSP-ELISA and by qPCR in mosquitoes that fed on cultured P. falciparum material (agreement 96.9%; kappa = 0.97) and naturally infected P. vivax parasite carriers [agreement 92.4% (kappa = 0.83)]. CONCLUSIONS: The proposed bead-beating CSP-ELISA/qPCR methodology considerably increases throughput for the detection of mosquito infection. qPCR remains necessary to confirm infections in mosquitoes with low CSP-ELISA signal. This methodology may prove particularly useful for studies where very low mosquito infection prevalence is expected and study sites where experience with oocyst detection is limited

    A randomized feasibility trial comparing four antimalarial drug regimens to induce Plasmodium falciparum gametocytemia in the controlled human malaria infection model.

    Background: Malaria elimination strategies require a thorough understanding of parasite transmission from human to mosquito. A clinical model to induce gametocytes to understand their dynamics and evaluate transmission-blocking interventions (TBI) is currently unavailable. Here, we explore the use of the well-established Controlled Human Malaria Infection model (CHMI) to induce gametocyte carriage with different antimalarial drug regimens. Methods: In a single centre, open-label randomised trial, healthy malaria-naive participants (aged 18–35 years) were infected with Plasmodium falciparum by bites of infected Anopheles mosquitoes. Participants were randomly allocated to four different treatment arms (n = 4 per arm) comprising low-dose (LD) piperaquine (PIP) or sulfadoxine-pyrimethamine (SP), followed by a curative regimen upon recrudescence. Male and female gametocyte densities were determined by molecular assays. Results: Mature gametocytes were observed in all participants (16/16, 100%). Gametocytes appeared 8.5–12 days after the first detection of asexual parasites. Peak gametocyte densities and gametocyte burden was highest in the LD-PIP/SP arm, and associated with the preceding asexual parasite biomass (p=0.026). Male gametocytes had a mean estimated circulation time of 2.7 days (95% CI 1.5–3.9) compared to 5.1 days (95% CI 4.1–6.1) for female gametocytes. Exploratory mosquito feeding assays showed successful sporadic mosquito infections. There were no serious adverse events or significant differences in the occurrence and severity of adverse events between study arms (p=0.49 and p=0.28). Conclusions: The early appearance of gametocytes indicates gametocyte commitment during the first wave of asexual parasites emerging from the liver. Treatment by LD-PIP followed by a curative SP regimen, results in the highest gametocyte densities and the largest number of gametocyte-positive days. This model can be used to evaluate the effect of drugs and vaccines on gametocyte dynamics, and lays the foundation for fulfilling the critical unmet need to evaluate transmission-blocking interventions against falciparum malaria for downstream selection and clinical development.     Modest heterologous protection after Plasmodium falciparum sporozoite immunization: a double-blind randomized controlled clinical trial.

    BACKGROUND: A highly efficacious vaccine is needed for malaria control and eradication. Immunization with Plasmodium falciparum NF54 parasites under chemoprophylaxis (chemoprophylaxis and sporozoite (CPS)-immunization) induces the most efficient long-lasting protection against a homologous parasite. However, parasite genetic diversity is a major hurdle for protection against heterologous strains. METHODS: We conducted a double-blind, randomized controlled trial in 39 healthy participants of NF54-CPS immunization by bites of 45 NF54-infected (n = 24 volunteers) or uninfected mosquitoes (placebo; n = 15 volunteers) against a controlled human malaria infection with the homologous NF54 or the genetically distinct NF135.C10 and NF166.C8 clones. Cellular and humoral immune assays were performed as well as genetic characterization of the parasite clones. RESULTS: NF54-CPS immunization induced complete protection in 5/5 volunteers against NF54 challenge infection at 14 weeks post-immunization, but sterilely protected only 2/10 and 1/9 volunteers against NF135.C10 and NF166.C8 challenge infection, respectively. Post-immunization plasma showed a significantly lower capacity to block heterologous parasite development in primary human hepatocytes compared to NF54. Whole genome sequencing showed that NF135.C10 and NF166.C8 have amino acid changes in multiple antigens targeted by CPS-induced antibodies. Volunteers protected against heterologous challenge were among the stronger immune responders to in vitro parasite stimulation. CONCLUSIONS: Although highly protective against homologous parasites, NF54-CPS-induced immunity is less effective against heterologous parasite clones both in vivo and in vitro. Our data indicate that whole sporozoite-based vaccine approaches require more potent immune responses for heterologous protection.
