    ELEVATE-TN Study. New data of acalabrutinib in first-line treatment of chronic lymphocytic leukemia. Resolution

    Over the past decade, we have seen a significant change in modern approaches in the first-line treatment of chronic lymphocytic leukemia (CLL). The CLL-10 study data established the FCR regimen as the treatment of choice for younger patients with limited comorbidities, while for patients older than 65 years, the BR regimen is more often considered as less toxic one. According to published data, 46% of patients with newly diagnosed CLL have comorbidities. Moreover, high-risk patients with del(17p) and/or TP53 mutation do not have response on immunochemotherapy (ICT) most often. Thus, about 1/2 of the patients cannot be treated or will not respond to standard ICT regimens. Targeted therapy with Brutons tyrosine kinase (BTK) inhibitors is an important option of the first-line treatment of patients with CLL. Acalabrutinib is a highly selective second-generation BTK inhibitor that does not inhibit EGFR, ITK or TEC targets. Acalabrutinib in combination with obinutuzumab or as monotherapy can be considered as a highly effective and safe option of the first line of CLL therapy. Based on the hight selectivity of the agent, acalabrutinib can be considered as the preferable option for patients who are not eligible for ICT, including patients with commodities, such as cardiovascular diseases or risk factors for their development

    Wip1 phosphatase: between p53 and MAPK kinases pathways

    IF 5.008International audienceCells undergoing oncogenic transformation frequently inactivate tumor suppressor pathways that could prevent their uncontrolled growth. Among those pathways p53 and p38MAPK pathways play a critical role in regulation of cell cycle, senescence and cell death in response to activation of oncogenes, stress and DNA damage. Consequently, these two pathways are important in determining the sensitivity of tumor cells to anti-cancer treatment. Wild type p53-induced phosphatase, Wip1, is involved in governance of both pathways. Recently, strategies directed to manipulation with Wip1 activity proposed to advance current day anticancer treatment and novel chemical compounds synthesized to improve specificity of manipulation with Wip1 activity. Here we reviewed the history of Wip1 studies in vitro and in vivo, in genetically modified animal models that support Wip1 role in tumorigenesis through regulation of p53 and p38MAPK pathways. Based on our knowledge we propose several recommendations for future more accurate studies of Wip1 interactions with other pathways involved in tumorigenesis using recently developed tools and for adoption of Wip1 manipulation strategies in anti-cancer therapy

    The paper looks into the problem of finding methods and educational technologies that reveal the creative potential of students, ensure their involvement in the educational process, and ensure the prevention and correction of psychological problems the students might have. The authors review the value of gaming technologies, the peculiarities of their use in teaching foreign languages, especially in connection with the emergence of language anxiety of students, and analyze the types of language anxiety and the role of gaming technologies in the prevention and correction of language anxiety of students. The paper focuses on the causes of language anxiety in teenage students. The study aimed to look into the influence of game forms of education on the reduction of language anxiety of school students learning foreign languages. To achieve the goal and confirm the hypothesis put forward, the authors used the method of measuring language anxiety during the study of a foreign language by Horwitz (Foreign Language Classroom Anxiety Scale). At the formative stage, a set of games was developed, which was used in German lessons with students of the experimental group for one month. Further, the paper analyzes the results of the study of the influence of game forms of learning on the reduction of language anxiety in the study of foreign languages. The paper has not only theoretical significance in terms of considering language anxiety, but also practical significance and novelty because the results of the study showed the high efficiency of gaming technologies and confirmed the fundamental possibility of prevention and psychocorrection of language anxiety with their help
