    Historical Evolution of Design and Supply of Drinking Water Treatment Plant for the City of Bethlehem, 1741-Present

    The purpose of this document is to provide a detailed account of the history of the water supply for the city of Bethlehem. This report begins with the settlement of the Moravians in the area, their need for water and the solutions they developed to meet their needs. It explains why Bethlehem water sources were switched, and how contamination, increasing population and water demands affected Bethlehem and its water supply. The document also introduces South Bethlehem and their water system. New projects that were pursued by Bethlehem are discussed, with specific reasons for the change in technology. Specific characteristics of the systems and technologies are discussed from an engineering standpoint. The characteristics are then further analyzed in regard to how specifications met the requirements and needs of the people of the city. Overall this report concludes that throughout the history of Bethlehem, water resources provided a significant role in the development of the city. Moravians chose to settle in the Bethlehem area because of its abundant supply of water. The growth of the city in population and industry was guided on multiple occasions by the availability of water. For the most part, the city engineers and city officials have met the water needs and standards required by the government and Bethlehem’s citizens

    Long term post-fire vegetation dynamics in north-east Mediterranean ecosystems. The case of Mount Athos Greece

    Fire is an ecological and disturbance factor with a significant historical role in shaping the landscape of fire-prone environments. Despite the large amount of literature regarding post-fire vegetation dynamics, the north-east Mediterranean region is rather underrepresented in the literature. Studies that refer to the early post fire years and long term research are rather scarce. The current study is conducted in the socially and geographically isolated peninsula of Mount Athos (Holly Mountain) in northern Greece, and it studies vegetation dynamics over a period of 30 years since the last fire. Field data were collected 11 years since the event and were used to identify the present plant communities in the area, using TWINSPAN, and the factors affecting their distribution using CART. Four Landsat (TM, ETM, OLI) images are employed for the calculation of NDVI, which was found effective in detecting the intercommunity variation in the study area, and it is used for long term monitoring. The study includes four communities, from maquis to forest which are common in the Mediterranean region covering a wide altitudinal range. The results suggest that fire affects the various communities in a different way and their recovery differs significantly. While forest communities recover quickly after fire, maintaining their composition and structure, the maquis communities may need several years before reaching the pre-fire characteristics. The dry climatic conditions of the study area are probably the reason for the slow recovery of the most fire prone communities. Given that climate change is expected to make the conditions even drier in the region, studies like this emphasize the need to adopt measures for controlling wildfires and preventing ecosystem degradation

    Adaptive diversity of beech seedlings under climate change scenarios

    The ability of beech (Fagus sylvatica L.) populations to adapt to the ongoing climate change is especially important in the southern part of Europe, where environmental change is expected to be more intense. In this study, we tested the existing adaptive potential of eight beech populations from two provenances in N.E. Greece (Evros and Drama) that show differences in their environmental conditions and biogeographical background. Seedling survival, growth and leaf phenological traits were selected as adaptive traits and were measured under simulated controlled climate change conditions in a growth chamber. Seedling survival was also tested under current conditions in the field. In the growth chamber, simulated conditions of temperature and precipitation for the year 2050 were applied for 3 years, under two different irrigation schemes, where the same amount of water was distributed either frequently (once every week) or non-frequently (once in 20 days). The results showed that beech seedlings were generally able to survive under climate change conditions and showed adaptive differences among provenances and populations. Furthermore, changes in the duration of the growing season of seedlings were recorded in the growth chamber, allowing them to avoid environmental stress and high selection pressure. Differences were observed between populations and provenances in terms of temporal distribution patterns of precipitation and temperature, rather than the average annual or monthly values of these measures. Additionally, different adaptive strategies appeared among beech seedlings when the same amount of water was distributed differently within each month. This indicates that the physiological response mechanisms of beech individuals are very complex and depend on several interacting parameters. For this reason, the choice of beech provenances for translocation and use in afforestation or reforestation projects should consider the small scale ecotypic diversity of the species and view multiple environmental and climatic parameters in connection to each other

    Introduction of apple ANR genes into tobacco inhibits expression of both CHI and DFR genes in flowers, leading to loss of anthocyanin

    Three genes encoding anthocyanidin reductase (ANR) in apple (Malus×domestica Borkh.), designated MdANR1, MdANR2a, and MdANR2b, have been cloned and characterized. MdANR1 shows 91% identity in coding DNA sequences with MdANR2a and MdANR2b, while MdANR2a and MdANR2b are allelic and share 99% nucleotide sequence identity in the coding region. MdANR1 and MdANR2 genes are located on linkage groups 10 and 5, respectively. Expression levels of both MdANR1 and MdANR2 genes are generally higher in yellow-skinned cv. Golden Delicious than in red-skinned cv. Red Delicious. Transcript accumulation of MdANR1 and MdANR2 genes in fruits gradually decreased throughout fruit development. Ectopic expression of apple MdANR genes in tobacco positively and negatively regulates the biosynthesis of proanthocyanidins (PAs) and anthocyanin, respectively, resulting in white, pale pink-coloured, and white/red variegated flowers. The accumulation of anthocyanin is significantly reduced in all tobacco transgenic flowers, while catechin and epicatechin contents in transgenic flowers are significantly higher than those in flowers of wild-type plants. The inhibition of anthocyanin synthesis in tobacco transgenic flowers overexpressing MdANR genes is probably attributed to down-regulation of CHALCONE ISOMERASE (CHI) and DIHYDROFLAVONOL-4-REDUCTASE (DFR) genes involved in the anthocyanin pathway. Interestingly, several transgenic lines show no detectable transcripts of the gene encoding leucoanthocyanidin reductase (LAR) in flowers, but accumulate higher levels of catechin in flowers of transgenic plants than those of wild-type plants. This finding suggests that the ANR gene may be capable of generating catechin via an alternative route, although this mechanism is yet to be further elucidated

    Light is one of the most important environmental factors affecting flavonoid biosynthesis in plants. The absolute dependency of light to the plant development has driven evolvement of sophisticated mechanisms to sense and transduce multiple aspects of the light signal. Light effects can be categorized in photoperiod (duration), intensity (quantity), direction and quality (wavelength) including UV-light. Recently, new information has been achieved on the regulation of light-controlled flavonoid biosynthesis in fruits, in which flavonoids have a major contribution on quality. This review focuses on the effects of the different light conditions on the control of flavonoid biosynthesis in fruit producing plants. An overview of the currently known mechanisms of the light-controlled flavonoid accumulation is provided. R2R3 MYB transcription factors are known to regulate by differential expression the biosynthesis of distinct flavonoids in response to specific light wavelengths. Despite recent advances, many gaps remain to be understood in the mechanisms of the transduction pathway of light-controlled flavonoid biosynthesis. A better knowledge on these regulatory mechanisms is likely to be useful for breeding programs aiming to modify fruit flavonoid pattern

    Metabolic effects of elevated temperature on organic acid degradation in ripening <em>Vitis vinifera</em> fruit

    First published online: September 1, 2014Berries of the cultivated grapevine Vitis vinifera are notably responsive to temperature, which can influence fruit quality and hence the future compatibility of varieties with their current growing regions. Organic acids represent a key component of fruit organoleptic quality and their content is significantly influenced by temperature. The objectives of this study were to (i) manipulate thermal regimes to realistically capture warming-driven reduction of malate content in Shiraz berries, and (ii) investigate the mechanisms behind temperature-sensitive malate loss and the potential downstream effects on berry metabolism. In the field we compared untreated controls at ambient temperature with longer and milder warming (2-4 °C differential for three weeks; Experiment 1) or shorter and more severe warming (4-6 °C differential for 11 days; Experiment 2). We complemented field trials with control (25/15 °C) and elevated (35/20 °C) day/night temperature controlled-environment trials using potted vines (Experiment 3). Elevating maximum temperatures (4-10 °C above controls) during pre-véraison stages led to higher malate content, particularly with warmer nights. Heating at véraison and ripening stages reduced malate content, consistent with effects typically seen in warm vintages. However, when minimum temperatures were also raised by 4-6 °C, malate content was not reduced, suggesting that the regulation of malate metabolism differs during the day and night. Increased NAD-dependent malic enzyme activity and decreased phosphoenolpyruvate carboxylase and pyruvate kinase activities, as well as the accumulation of various amino acids and γ-aminobutyric acid, suggest enhanced anaplerotic capacity of the TCA cycle and a need for coping with decreased cytosolic pH in heated fruit.C.
