Dynamic look at DNA unwinding by a replicative helicase
A prerequisite for DNA replication is the unwinding of duplex DNA catalyzed by a replicative hexameric helicase. Despite a growing body of research, key elements of helicase mechanism remain under substantial debate. In particular, the number of DNA strands encircled by the helicase ring during unwinding and the ring orientation at the replication fork completely contrast in contemporary mechanistic models. Here we use single-molecule and ensemble assays to address these questions for the papillomavirus E1 helicase. We find that E1 unwinds DNA with a strand-exclusion mechanism, with the N-terminal side of the helicase ring facing the replication fork. We show that E1 generates strikingly heterogeneous unwinding patterns stemming from varying degrees of repetitive movements, which is modulated by the DNA-binding domain. Correlation measurements in high-multiplicity events
Requirements for correlation measurements in high--multiplicity events are
discussed. Attention is focussed on detection of so--called hot spots,
two--particle rapidity correlations, two--particle momentum correlations (for
quantum interferometry) and higher--order correlations. The signal--to--noise
ratio may become large in the high--multiplicity limit, allowing meaningful
single--event measurements, only if the correlations are due to collective

IWIN: the Isfjorden Weather Information Network
In an effort led by the University Centre in Svalbard (UNIS), with support from the Norwegian Meteorological Institute (MET Norway), the Isfjorden Weather Information Network (IWIN) is under development in the Isfjorden region in central Svalbard. The network substantially expands upon the relatively sparse existing operational network of weather stations and consists of compact and cost-efficient all-in-one weather stations permanently installed at lighthouses around Isfjorden and on board small tourist cruise ships trafficking the fjord from spring to autumn. All data from the network until June 2023, as presented in this paper, can be found at https://doi.org/10.5281/zenodo.8137588 (Frank et al., 2023b). New data become freely available in near-real time via MET Norway's data portals (https://doi.org/10.21343/ebrw-w846, Frank et al., 2023a). The IWIN data are highly valuable for scientific purposes such as atmospheric boundary layer research, the validation and development of numerical weather prediction models, and assimilation in these, as well as the planning and safe conduction of outdoor activities in the region.</p
Viral-E1 and Viral-E2 Proteins Support Replication of Homologous and Heterologous Papillomaviral Origins
We have shown that E1 and E2 proteins of human papillomavirus type 11 (HPV-11) were essential to support the replication of the homologous viral origin (ori) in a transient replication assay, similar to reports on bovine papillomavirus type 1 (BPV-1). Unexpectedly, matched or even mixed combinations of E1 and E2 proteins from HPV-11 or BPV-1 replicated either ori in human, monkey, and rodent cell lines of epithelial or fibroblastic lineage, albeit with varied efficiencies. Either set of viral proteins was also able to initiate replication of ori-containing plasmids from many other human and animal papillomaviruses. Thus the interactions among the cis elements and trans factors of papillomaviruses are more conserved than expected from the other members of the papovavirus family, simian virus 40 and polyomavirus, for which large tumor antigen does not replicate a heterologous ori in either permissive or nonpermissive cells. We infer that the stringent species and tissue specificities observed for papillomaviruses in vivo are not entirely due to direct restrictions on viral DNA replication. Rather, transcriptional control of viral gene expression must play a dominant role
Azimuthal Correlations in the Target Fragmentation Region of High Energy Nuclear Collisions
Results on the target mass dependence of proton and pion pseudorapidity
distributions and of their azimuthal correlations in the target rapidity range
are presented. The data have been taken with the
Plastic-Ball detector set-up for 4.9 GeV p + Au collisions at the Berkeley
BEVALAC and for 200 GeV/ p-, O-, and S-induced reactions on
different nuclei at the CERN-SPS. The yield of protons at backward rapidities
is found to be proportional to the target mass. Although protons show a typical
``back-to-back'' correlations, a ``side-by-side'' correlation is observed for
positive pions, which increases both with target mass and with impact parameter
of a collision. The data can consistently be described by assuming strong
rescattering phenomena including pion absorption effects in the entire excited

DGAT1 activity synchronises with mitophagy to protect cells from metabolic rewiring by iron depletion
Mitophagy removes defective mitochondria via lysosomal elimination. Increased mitophagy coincides with metabolic reprogramming, yet it remains unknown whether mitophagy is a cause or consequence of such state changes. The signalling pathways that integrate with mitophagy to sustain cell and tissue integrity also remain poorly defined. We performed temporal metabolomics on mammalian cells treated with deferiprone, a therapeutic iron chelator that stimulates PINK1/PARKIN-independent mitophagy. Iron depletion profoundly rewired the metabolome, hallmarked by remodelling of lipid metabolism within minutes of treatment. DGAT1-dependent lipid droplet biosynthesis occurred several hours before mitochondrial clearance, with lipid droplets bordering mitochondria upon iron chelation. We demonstrate that DGAT1 inhibition restricts mitophagy in vitro, with impaired lysosomal homeostasis and cell viability. Importantly, genetic depletion of DGAT1 in vivo significantly impaired neuronal mitophagy and locomotor function in Drosophila. Our data define iron depletion as a potent signal that rapidly reshapes metabolism and establishes an unexpected synergy between lipid homeostasis and mitophagy that safeguards cell and tissue integrity.Peer reviewe
Interferometry of Direct Photons in Central 280Pb+208Pb Collisions at 158A GeV
Two-particle correlations of direct photons were measured in central
208Pb+208Pb collisions at 158 AGeV. The invariant interferometric radii were
extracted for 100<K_T<300 MeV/c and compared to radii extracted from charged
pion correlations. The yield of soft direct photons, K_T<300 MeV/c, was
extracted from the correlation strength and compared to theoretical

Event-by-Event Fluctuations in Particle Multiplicities and Transverse Energy Produced in 158.A GeV Pb+Pb collisions
Event-by-event fluctuations in the multiplicities of charged particles and
photons, and the total transverse energy in 158 GeV Pb+Pb collisions
are studied for a wide range of centralities. For narrow centrality bins the
multiplicity and transverse energy distributions are found to be near perfect
Gaussians. The effect of detector acceptance on the multiplicity fluctuations
has been studied and demonstrated to follow statistical considerations. The
centrality dependence of the charged particle multiplicity fluctuations in the
measured data has been found to agree reasonably well with those obtained from
a participant model. However for photons the multiplicity fluctuations has been
found to be lower compared to those obtained from a participant model. The
multiplicity and transverse energy fluctuations have also been compared to

Centrality Dependence of Neutral Pion Production in 158 A GeV Pb + Pb Collisions
The production of neutral pions in 158AGeV Pb+Pb collisions has been studied
in the WA98 experiment at the CERN SPS. Transverse momentum spectra are studied
for the range 0.3 GeV/c < mT-m0 < 4.0 GeV/c. The results for central collisions
are compared to various models. The centrality dependence of the neutral pion
spectral shape and yield is investigated. An invariance of the spectral shape
and a simple scaling of the yield with the number of participating nucleons is
observed for centralities with greater than about 30 participating nucleons

Measurement of the mid-rapidity transverse energy distribution from GeV Au+Au collisions at RHIC
The first measurement of energy produced transverse to the beam direction at
RHIC is presented. The mid-rapidity transverse energy density per participating
nucleon rises steadily with the number of participants, closely paralleling the
rise in charged-particle density, such that E_T / N_ch remains relatively
constant as a function of centrality. The energy density calculated via
Bjorken's prescription for the 2% most central Au+Au collisions at
sqrt(s_NN)=130 GeV is at least epsilon_Bj = 4.6 GeV/fm^3 which is a factor of
1.6 larger than found at sqrt(s_NN)=17.2 GeV (Pb+Pb at CERN).Comment: 307 authors, 6 pages, 4 figures, 1 table, submitted to PRL 4/18/2001;
