    The Allele Distribution for the rs7250346 SNP in SLC5A5 among Saudis

    Background: The sodium/iodide cotransporter solute carrier family 5 member 5 (SLC5A5) is found in the basolateral cell membrane of thyroid follicular epithelial cells as a sodium iodide symporter. It helps in the secretion of triiodothyronine (T3) and tetraiodothyronine (T4). Polymorphisms in SLC5A5 result in hypothyroidism. The aim of the study is to estimate the frequency of rs7250346, a single nucleotide polymorphism (SNP) associated with thyroid disease, in a Saudi population and to compare it to other populations. Methods: Two hundred and forty Saudi patients from King Faisal Specialist Hospital and Research Centre provided samples for the genetic analysis. Samples were genotyped for target SNPs by real time polymerase chain reaction (PCR), and the resultant rs7250346 frequency in the Saudi population was compared to other populations using HapMap. Results: Out of the 240 Saudi samples, 64% had C genotype and 35% had G genotype in the rs7250346. Conclusion: The Saudi frequency of the rs7250346 SNP of SLC5A5 differs from that of European, Chinese, Japanese, or Sub-Saharan populations in HapMap (http://hapmap.org)

    Attitudes and Perceptions among Dublin International Foundation College Students towards Taking Conventional Medications and Herbal Medicines

    Background: The use of both conventional medications and herbal medicines has increased recently. Consequently, the chance of misusing medications has also increased, leading to unwanted complications and economic burdens. This study examined the attitudes and perceptions of international students at Dublin International Foundation College (DIFC) towards conventional medication and herbal medicine. Methods: A cross-sectional quantitative method was used to collect information from international students at DIFC. A total of 85 questionnaires were distributed to the students, and 54 completed questionnaires were received from them. Chi-square test was used to examine the possible relationships between students’ background, gender and region of origin and their attitudes and perceptions toward using conventional medications and herbal medicines. Results: Participants from different regions of origin showed significant differences in the responses to questions about informing their healthcare providers about their drug allergies. While most students did not usually mix herbal medicines with either conventional medications or other types of herbs, students aged between 21 and 25 were more likely to combine different types of medications compared to students from other age groups. No significant differences were observed in relation to students' educational backgrounds and gender. Conclusion: This study demonstrated the need of educating students from different regions and backgrounds about the use of conventional medications and herbal medicines. Information regarding the proper use of medications and the dangers of drug interactions should be included in the curricula of formal education and disseminated to the public using culturally sensitive strategies

    Knowledge, Attitudes and Practices towards Medication Use among Health Care Students in King Saud University

    Background: Health sciences students are expected to have appropriate knowledge and attitudes toward medication use. However, literary evidence of such expertise among health sciences students of King Saud University is unknown. This study was completed to assess the knowledge about medicines and behavior of health science students towards safe use of medications. It also aims to assess the health knowledge, attitude and practices of the students. Methods: This cross-sectional study used a questionnaire consisting of 24 questions. This was administered by the researcher between October and December 2009 in the colleges of medicine, dentistry, pharmacy, applied medical science and nursing of the King Saud University. The survey consisted of three parts: Ten questions assessed the students’ knowledge on drug safety (Part 1). Four questions assessed student attitude toward medication consultations by the pharmacist (Part 2) and ten questions involved medication use practices and consultation with pharmacists (Part 3). A stratified sampling method was used to select participants. Results: Pharmacy students had better medication knowledge compared to other health sciences students especially regarding antihypertensive drugs, antibiotics, paracetamol and antacids (p<0.05). Pharmacy students showed a positive attitude regarding the trustworthiness of a pharmacist to give a consultation. Nearly all other health science students showed a negative attitude about dispensing and consultation concerning nutritional supplements by a pharmacist. All health sciences students had a similar per­ception toward medication use and practice.     An intelligent System for Diagnosis Schizophrenia and Bipolar Diseases based on Support Vector Machine with Different Kernels

    Bipolar disorder and schizophrenia overlap in symptoms and may share some underlying neural substrates. The discrimination between the two diseases is one of the problems that face psychiatric experts. This paper will propose some solutions to this problem based on the artificial methods. The support vector machine (SVM) is used for discrimination based on measuring of the patient EEG rhythms. The large set of features included in the EEG rhythms is reduced into smaller set of features after Fast Fourier Transform (FFT) segmentation. Different kernels are applied on the SVM which are linear, polynomial, quadratic and radial basis function. The application of SVM with different kernels for the EEG discrimination of the patients suffering from schizophrenia and bipolar diseases is the core of this work. Experimental results have shown that the proposed algorithms will solve the discrimination between the two diseases using EEG rhythms and the support vector machine with linear and quadratic kernels have achieved a high performance rate equal to 98 % and 97.667% respectively compared to the other kernels

    Polydimethylsiloxane-based giant glycosylated polymersomes with tunable bacterial affinity

    A synthetic cell mimic in the form of giant glycosylated polymersomes (GGPs) comprised of a novel amphiphilic diblock copolymer is reported. A synthetic approach involving a poly(dimethylsiloxane) (PDMS) macro-chain transfer agent (macroCTA) and postpolymerization modification was used to marry the hydrophobic and highly flexible properties of PDMS with the biological activity of glycopolymers. 2-Bromoethyl acrylate (BEA) was first polymerized using a PDMS macroCTA (Mn,th ≈ 4900 g·mol–1, Đ = 1.1) to prepare well-defined PDMS-b-pBEA diblock copolymers (Đ = 1.1) that were then substituted with 1-thio-ÎČ-d-glucose or 1-thio-ÎČ-d-galactose under facile conditions to yield PDMS-b-glycopolymers. Compositions possessing ≈25% of the glycopolymer block (by mass) were able to adopt a vesicular morphology in aqueous solution (≈210 nm in diameter), as indicated by TEM and light scattering techniques. The resulting carbohydrate-decorated polymersomes exhibited selective binding with the lectin concanavalin A (Con A), as demonstrated by turbidimetric experiments. Self-assembly of the same diblock copolymer compositions using an electroformation method yielded GGPs (ranging from 2–20 ÎŒm in diameter). Interaction of these cell-sized polymersomes with fimH positive E. coli was then studied via confocal microscopy. The glucose-decorated GGPs were found to cluster upon addition of the bacteria, while galactose-decorated GGPs could successfully interact with (and possibly immobilize) the bacteria without the onset of clustering. This demonstrates an opportunity to modulate the response of these synthetic cell mimics (protocells) toward biological entities through exploitation of selective ligand–receptor interactions, which may be readily tuned through a considered choice of carbohydrate functionality

    Dual-Band Transmitter and Receiver with Bowtie-Antenna in 0.13 ÎŒm SiGe BiCMOS for Gas Spectroscopy at 222 - 270 GHz

    This paper presents a transmitter (TX) and a receiver (RX) with bowtie-antenna and silicon lens for gas spectroscopy at 222-270 GHz, which are fabricated in IHP’s 0.13 ÎŒm SiGe BiCMOS technology. The TX and RX use two integrated local oscillators for 222 – 256 GHz and 250 – 270 GHz, which are switched for dual-band operation. Due to its directivity of about 27 dBi, the single integrated bowtie-antenna with silicon lens enables an EIRP of about 25 dBm for the TX, and therefore a considerably higher EIRP for the 2-band TX compared to previously reported systems. The double sideband noise temperature of the RX is 20,000 K (18.5 dB noise figure) as measured by the Y-factor method. Absorption spectroscopy of gaseous methanol is used as a measure for the performance of the gas spectroscopy system with TX- and RX-modules

    DGGE-RAPD analysis as a useful tool for cultivar identification

    Denaturing gradient gel electrophoresis-random amplified polymorphic DNA (DGGE-RAPD) was used to overcome the main drawbacks of RAPD (i.e., the low levels of reproducibility and polymorphism). As a model, six barley cultivars of known origin were tested for RAPD markers using DGGE methodology with 29 arbitrary primers. Among a total of 418 bands observed, as high as 99 were polymorphic. Comparison between agarose-RAPD and DGGE-RAPD revealed that the latter was highly reproducible and gave higher level of polymorphism and consequently more markers. The relationships among barley cultivars derived from this study based on DGGE-RAPD are consistent with the known lineage of these cultivars. In conclusion, we recommend the use of DGGE-RAPD as an alternative tool to the more costly DNA-based analysis in cultivar identification in laboratories with limited funds.Key words: Denaturing gradient gel electrophoresis-random amplified polymorphic DNA, artificial heteroduplex, dendogram, lineage

    EJP18 peptide derived from the juxtamembrane domain of epidermal growth factor receptor represents a novel membrane-active cell-penetrating peptide

    Membrane-active peptides have been extensively studied to probe protein–membrane interactions, to act as antimicrobial agents and cell-penetrating peptides (CPPs) for the delivery of therapeutic agents to cells. Hundreds of membrane-active sequences acting as CPPs have now been described including bioportides that serve as single entity modifiers of cell physiology at the intracellular level. Translation of promising CPPs in pre-clinical studies have, however, been disappointing as only few identified delivery systems have progressed to clinical trials. To search for novel membrane-active peptides a sequence from the EGFR juxtamembrane region was identified (named EJP18), synthesised, and examined in its L- and D-form for its ability to mediate the delivery of a small fluorophore and whole proteins to cancer cell lines. Initial studies identified the peptide as being highly membrane-active causing extensive and rapid plasma membrane reorganisation, blebbing, and toxicity. At lower, non-toxic concentrations the peptides outperformed the well-characterised CPP octaarginine in cellular delivery capacity for a fluorophore or proteins that were associated with the peptide covalently or via ionic interactions. EJP18 thus represents a novel membrane-active peptide that may be used as a naturally derived model for biophysical protein–membrane interactions or for delivery of cargo into cells for therapeutic or diagnostic applications

    Accelerated hyperfractionation (AHF) compared to conventional fractionation (CF) in the postoperative radiotherapy of locally advanced head and neck cancer: influence of proliferation

    Based on the assumption that an accelerated proliferation process prevails in tumour cell residues after surgery, the possibility that treatment acceleration would offer a therapeutic advantage in postoperative radiotherapy of locally advanced head and neck cancer was investigated. The value of Tpot in predicting the treatment outcome and in selecting patients for accelerated fractionation was tested. Seventy patients with (T2/N1–N2) or (T3-4/any N) squamous cell carcinoma of the oral cavity, larynx and hypopharynx who underwent radical surgery, were randomized to either (a) accelerated hyperfractionation: 46.2 Gy per 12 days, 1.4 Gy per fraction, three fractions per day with 6 h interfraction interval, treating 6 days per week or (b) Conventional fractionation: 60 Gy per 6 weeks, 2 Gy per fraction, treating 5 days per week. The 3-year locoregional control rate was significantly better in the accelerated hyperfractionation (88±4%) than in the CF (57±9%) group, P=0.01 (and this was confirmed by multivariate analysis), but the difference in survival (60±10% vs 46±9%) was not significant (P=0.29). The favourable influence of a short treatment time was further substantiated by demonstrating the importance of the gap between surgery and radiotherapy and the overall treatment time between surgery and end of radiotherapy. Early mucositis progressed more rapidly and was more severe in the accelerated hyperfractionation group; reflecting a faster rate of dose accumulation. Xerostomia was experienced by all patients with a tendency to be more severe after accelerated hyperfractionation. Fibrosis and oedema also tended to be more frequent after accelerated hyperfractionation and probably represent consequential reactions. Tpot showed a correlation with disease-free survival in a univariate analysis but did not prove to be an independent factor. Moreover, the use of the minimum and corrected P-values did not identify a significant cut-off. Compared to conventional fractionation, accelerated hyperfractionation did not seem to offer a survival advantage in fast tumours though a better local control rate was noted. This limits the use of Tpot as a guide for selecting patients for accelerated hyperfractionation. For slowly growing tumours, tumour control and survival probabilities were not significantly different in the conventional fractionation and accelerated hyperfractionation groups. A rapid tumour growth was associated with a higher risk of distant metastases (P=0.01). In conclusion, tumour cell repopulation seems to be an important determinant of postoperative radiotherapy of locally advanced head and neck cancer despite lack of a definite association between Tpot and treatment outcome. In fast growing tumours accelerated hyperfractionation provided an improved local control but without a survival advantage. To gain a full benefit from treatment acceleration, the surgery-radiotherapy gap and the overall treatment time should not exceed 6 and 10 weeks respectively
