    Taming the beast: a revised classification of Cortinariaceae based on genomic data

This article is licensed under a Creative Commons Attribution 4.0 International License, http://creativecommons.org/licenses/by/4.0/.Abstract: Family Cortinariaceae currently includes only one genus, Cortinarius, which is the largest Agaricales genus, with thousands of species worldwide. The species are important ectomycorrhizal fungi and form associations with many vascular plant genera from tropicals to arctic regions. Genus Cortinarius contains a lot of morphological variation, and its complexity has led many taxonomists to specialize in particular on infrageneric groups. The previous attempts to divide Cortinarius have been shown to be unnatural and the phylogenetic studies done to date have not been able to resolve the higher-level classification of the group above section level. Genomic approaches have revolutionized our view on fungal relationships and provide a way to tackle difficult groups. We used both targeted capture sequencing and shallow whole genome sequencing to produce data and to perform phylogenomic analyses of 75 single-copy genes from 19 species. In addition, a wider 5-locus analysis of 245 species, from the Northern and Southern Hemispheres, was also done. Based on our results, a classification of the family Cortinariaceae into ten genera—Cortinarius, Phlegmacium, Thaxterogaster, Calonarius, Aureonarius, Cystinarius, Volvanarius, Hygronarius, Mystinarius, and Austrocortinarius—is proposed. Seven genera, 10 subgenera, and four sections are described as new to science and five subgenera are introduced as new combinations in a new rank. In addition, 41 section names and 514 species names are combined in new genera and four lecto- and epitypes designated. The position of Stephanopus in suborder Agaricineae remains to be studied. Targeted capture sequencing is used for the first time in fungal taxonomy in Basidiomycetes. It provides a cost-efficient way to produce -omics data in species-rich groups. The -omics data was produced from fungarium specimens up to 21 years old, demonstrating the value of museum specimens in the study of the fungal tree of life.     Physical Conditions of Fast Glacier Flow 1:Measurements From Boreholes Drilled to the Bed of Store Glacier, West Greenland

    Marine-terminating outlet glaciers of the Greenland ice sheet make significant contributions to global sea level rise, yet the conditions that facilitate their fast flow remain poorly constrained owing to a paucity of data. We drilled and instrumented seven boreholes on Store Glacier, Greenland, to monitor subglacial water pressure, temperature, electrical conductivity and turbidity along with englacial ice temperature and deformation. These observations were supplemented by surface velocity and meteorological measurements to gain insight into the conditions and mechanisms of fast glacier flow. Located 30 km from the calving front, each borehole drained rapidly on attaining 600m depth indicating a direct connection with an active subglacial hydrological system. Persistently high subglacial water pressures indicate low effective pressure (180 - 280 kPa), with small amplitude variations correlated with notable peaks in surface velocity driven by the diurnal melt cycle and longer periods of melt and rainfall. The englacial deformation profile determined from borehole tilt measurements indicates that 63-71% of total ice motion occurred at the bed, with the remaining 29-37% predominantly attributed to enhanced deformation in the lowermost 50-100 m of the ice column. We interpret this lowermost 100m to be formed of warmer, pre-Holocene ice overlying a thin (0-8m) layer of temperate basal ice. Our observations are consistent with a spatially-extensive and persistently-inefficient subglacial drainage system that we hypothesize comprises drainage both at the ice-sediment interface and through subglacial sediments. This configuration has similarities to that interpreted beneath dynamically-analogous Antarctic ice streams, Alaskan tidewater glaciers, and glaciers in surge.This research was funded by UK National Environment Research Council grants NE/K006126 and NE/K005871/1 and an Aberystwyth University Capital Equipment grant to B.     The study was carried out in four European schools (pupils age 9 to13 years old) within four countries (Portugal, Spain, UK and Ireland). The adopted methodology was based in action-research procedures and it included multiple methods and techniques: systematic classroom observation of teachers and pupil work and learning activities; documentary evidence (teachers working sheets, schemes of work) interviews (with selected pupils); questionnaires to all pupils and staff at intervals. This methodology implied the construction and testing of data collection instruments. Data was submitted to a qualitative and quantitative analysis.     Reclassification of Parapterulicium Corner (Pterulaceae, Agaricales), contributions to Lachnocladiaceae and Peniophoraceae (Russulales) and introduction of Baltazaria gen. nov.

    The genus Parapterulicium was first introduced to accommodate two Brazilian species of coralloid fungi with affinities to Pterulaceae (Agaricales). Despite the coralloid habit and the presence of skeletal hyphae, other features, notably the presence of gloeocystidia, dichophyses and papillate hyphal ends, differentiate this genus from Pterulaceae sensu stricto. Fieldwork in Brazil resulted in the rediscovery of two coralloid fungi identifiable as Parapterulicium, the first verified collections of this genus since Corner's original work in the 1950s. Molecular phylogenetic analyses of nrITS and nrLSU sequences from these modern specimens revealed affinities with the /peniophorales clade in the Russulales, rather than Pterulaceae. The presence of distinctive hyphal elements, homologous to the defining features of /peniophorales, is consistent with the phylogenetic evidence and thus clearly distinguished Parapterulicium and its type species P. subarbusculum from Pterulaceae, placing this genus within /peniophorales. Parapterulicium was also found to be polyphyletic so Baltazaria gen. nov. is proposed to accommodate P. octopodites, Scytinostroma galactinum, S. neogalactinum and S. eurasiaticogalactinum also within /peniophorales

    Clinical and morphological characteristic of the case of secondary enucleation of the eye with retinoblastoma

    Purpose: To analyze case of secondary enucleation of a child’s eye with recurrent retinoblastoma (RB) developing due to parents’ failure to take the child to surveillance appointments after salvage eye therapy. Material and Methods: Observation and treatment of a 10-month child diagnosed with bilateral RB. The right eye was diagnosed with RB at stage T1N0M0, and the left eye, with RB at stage T3cN0M0 complicated by secondary glaucoma. Results: Salvage eye therapy was performed which included six cycles of combined multiagent chemotherapy (with systemic intravitreal chemoreduction melphalan injection), followed by three courses of laser coagulation of tumor focus in the right eye and additional seventh local chemotherapy in the left eye. As a result, type 4 RB regression pattern in the right eye, and was developed incomplete type 2 RB regression pattern (“fish-flesh” appearance) in the left eye. However, after the child did not visit the medical care facility for surveillance and treatment during seven months, tumor regrowth developed. Secondary enucleation of the left eye was performed due to big tumor size, anterior chamber involvement blindness, and the absence of visual functions without prospects for their improvement. Histopathology found undifferentiated retinoblastoma with invasion into the episclera, the child was administered two cycles of chemoreduction as well as external beam radiotherapy of the left orbit using a medical linear accelerator system. Conclusion: Should tumor recurrence occur or if there is no tumor control, urgent secondary enucleation with eye ball histopathology and, if indicated, further adjuvant therapy are recommended to prevent metastasis and save child’s life

    Binge-Like Eating Is Not Influenced by the Murine Model of OPRM1 A118G Polymorphism

    Impairments in opioid receptor signaling have been implicated in disordered eating. A functional variant of the OPRM1 gene is a guanine (G) substitution for adenine (A) at the 118 position of exon 1 (A118G). The influence of the A118G variant on binge eating behaviors and the effectiveness of pharmacotherapies used to treat binge eating have not been characterized. Mice were generated with A to G substitution at the 112 position on exon 1 to produce a murine equivalent of the human A118G variant. Homozygous female mice (AA or GG) were exposed to intermittent access to a highly palatable sweet-fat food with or without prior calorie deprivation to promote dietary-induced binge eating. There were no genotype-dependent differences in the dietary-induced binge eating. However, GG mice exposed to intermittent calorie restriction (Restrict) had higher body weights compared with GG mice exposed to intermittent sweet fat-food (Binge) and ad libitum feeding (Naive). Acute oral dosing of lisdexamfetamine (0.15, 0.5, and 1.5 mg/kg) or sibutramine (0.3, 1, and 3 mg/kg) did not produce genotype-dependent differences in binge-like eating. In addition, no genotype-dependent differences in binge-like eating were observed with chronic (14-day) dosing of lisdexamfetamine (1.5 mg/kg/day) or sibutramine (3 mg/kg/day). In the chronic dosing, body weights were higher in the GG Restrict compared with AA Restrict. Our findings suggest that the A112G polymorphism does not influence binge eating behaviors or pharmacotherapies for treating binge eating

    Ice core evidence for a recent increase in snow accumulation in coastal Dronning Maud Land, East Antarctica

    Abstract. Ice cores provide temporal records of snow accumulation, a crucial component of Antarctic mass balance. Coastal areas are particularly under-represented in such records, despite their relatively high and sensitive accumulation rates. Here we present records from a 120 m ice core drilled on Derwael Ice Rise, coastal Dronning Maud Land (DML), East Antarctica in 2012. We date the ice core bottom back to 1745 ± 2 AD. δ18O and δD stratigraphy is supplemented by discontinuous major ion profiles, and verified independently by electrical conductivity measurements (ECM) to detect volcanic horizons. The resulting annual layer history is combined with the core density profile to calculate accumulation history, corrected for the influence of ice deformation. The mean long-term accumulation is 0.425 ± 0.035 m water equivalent (w.e.) a−1 (average corrected value). Reconstructed annual accumulation rates show an increase from 1955 onward to a mean value of 0.61 ± 0.02 m w.e. a−1 between 1955 and 2012. This trend is compared to other reported accumulation data in Antarctica, generally showing a high spatial variability. Output of the fully coupled Community Earth System Model demonstrates that sea ice and atmospheric patterns largely explain the accumulation variability. This is the first and longest record from a coastal ice core in East Antarctica showing a steady increase during the 20th and 21st centuries, thereby supporting modelling predictions. </jats:p
