The recently collected material is compared with the original description and other subsequent descriptions of the species. The phylogenetic position of S. lindsayae is inferred using a secondary structure-based alignment of a partial sequence of nuclear Large Subunit (LSU) ribosomal DNA. Phylogenetic trees were inferred using base-paired substitution models implemented in PHASE 2 software and Bayesian inference, and show S. lindsayae as the sister group to Stegelletina taxa

    Host records and geographical distribution of Corynosoma magdaleni, C. semerme and C. strumosum (Acanthocephala: Polymorphidae).

    A literature survey was conducted to investigate the host and geographical distribution patterns of three Corynosoma species (Acanthocephala: Polymorphidae), viz. C. magdaleni, C. semerme and C. strumosum. All three species appear to be restricted to the Northern Hemisphere. Occurrence records of C. magdaleni are limited to the Northern Atlantic coasts, while C. semerme has a circumpolar distribution. The geographical range of Corynosoma strumosum encompasses the distributions of the other two species, but also extends into warmer southern regions. Some Corynosoma populations are living with their definitive hosts in very isolated locations, such as in the brackish Baltic Sea or different freshwater lakes (e.g. Lake Saimaa). All three species have a heteroxenous life cycle, comprising a peracaridan intermediate host, a fish paratenic host and a mammalian definitive host. Occasionally, an acanthocephalan may enter an accidental host, from which it is unable to complete its life cycle. The host records reported here are categorised by type, i.e. intermediate, paratenic, definitive or accidental. While most of the definitive hosts are shared amongst the three Corynosoma species, C. strumosum showed the broadest range of paratenic hosts, which reflects its more extensive geographical distribution. One aim of this study and extensive literature summary is to guide future sampling efforts and therewith contribute to throw more light on the on-going species and morphotype discussion for this interesting parasite species

    Sealworm (Pseudoterranova decipiens) infection in grey seals (Halichoerus grypus), cod (Gadus morhua) and shorthorn sculpin (Myoxocephalus scorpius) in the Baltic Sea

    The anisakid nematode Pseudoterranova decipiens, known as the sealworm or cod worm, can infect the flesh of several fish species. The parasite causes cosmetic problems for the fish industry and can cause abdominal discomfort if consumed by humans. There are only scattered studies on the abundance or distribution of the sealworm in fish and seals in the Baltic Sea. To remedy this situation, the extent of sealworm infection was investigated in cod (Gadus morhua) and shorthorn sculpin (Myoxocephalus scorpius) collected along the Swedish coast. A relative presence of the sealworm was also investigated in samples from grey seal (Halichoerus grypus) stomachs. Up to 100 % of the fish were infected in some of the areas. Sculpin were generally worse infected than cod, both in abundance and prevalence of parasites. General linear models showed a significant correlation between the number of seals in an area and the prevalence of sealworms in cod. There was a sharp decrease of infected fish in areas with salinity lower than 7 aEuro degrees. Even though the northern Baltic proper and the southern Bothnian Sea have a high number of grey seals, only one sealworm was found in a sculpin in that region, and none in cod. In grey seal stomachs the sealworm was only found in samples from the central Baltic proper; further north, all anisakid nematodes identified in seals were Contracaecum osculatum. The results indicate that seal presence drives the distribution in the southern parts of the Baltic and that low salinity, or some other variable which correlates with salinity, limits the distribution in the northern part

    Cross-Cultural Adaptation of the KONTAKT Social Skills Group Training Program for Children and Adolescents with high-functioning Autism Spectrum Disorder: A feasibility Study

    Background: Social skills group training is an intervention method that has demonstrated moderate evidence of improvement among children and adolescents with autism spectrum disorder (ASD). KONTAKT is a manualized social skills group training program that was developed in Germany and that has demonstrated preliminary evidence of positive effect. In this study, we describe its adaptation to Scandinavian settings. Objective: The aim of this study was to evaluate the clinical feasibility of the Swedish version of KONTAKT. The program was piloted in two outpatient departments in Stockholm County. Method: A convergent mixed-method approach that involved both quantitative (ratings scales for ASD, clinical severity, and adaptive functioning) and qualitative (semi-structured interviewing) evaluation was applied. Twenty-two children and adolescents with high-functioning ASD between the ages of 8 and 17 years and their parents were enrolled in a one-group trial and completed assessments before and after KONTAKT training. Results: The quantitative evaluation showed improvements in social communication and global everyday functioning; the qualitative evaluations yielded general treatment satisfaction. Twenty of the 22 enrolled adolescents (91%) completed the training. Conclusions: These findings suggest that the Swedish adaptation of KONTAKT is feasible for Scandinavian clinical settings. It is currently being examined for efficacy and effectiveness in the largest multicenter randomized controlled trial of social skills group training in patients with ASD that has ever been undertaken (NCT01854346)

    AbstractIn an in vitro synthesizing system programmed with RNA from human liver a polypeptide with an estimated Mr of 80000 (80 kDa)±1400 (mean±SD, n=5) was synthesized. This polypeptide could be precipitated with antiserum to a narrow density cut of LDL (d=1.030-1.055) or antiserum against the high-Mr form of apoB (apoB 100 [4]). The synthesized protein is immunologically related to a 75 kDa protein isolated from LDL. We suggest that the 80 kDa protein represents a primary translation product of apoB synthesized in human liver

    The SNARE Protein SNAP23 and the SNARE-Interacting Protein Munc18c in Human Skeletal Muscle Are Implicated in Insulin Resistance/Type 2 Diabetes

    OBJECTIVE-Our previous studies suggest that the SNARE protein synaptosomal-associated protein of 23 kDa (SNAP23) is involved in the link between increased lipid levels and insulin resistance in cardiomyocytes. The objective was to determine whether SNAP23 may also be involved in the known association between lipid accumulation in skeletal muscle and insulin resistance/type 2 diabetes in humans, as well as to identify a potential regulator of SNAP23. RESEARCH DESIGN AND METHODS-We analyzed skeletal muscle biopsies from patients with type 2 diabetes and healthy, insulin-sensitive control subjects for expression (mRNA and protein) and intracellular localization (subcellular fractionation and immunohistochemistry) of SNAP23, and for expression of proteins known to interact with SNARE proteins. Insulin resistance was determined by a euglycemic hyperinsulinemic clamp Potential mechanisms for regulation of SNAP23 were also investigated in the skeletal muscle cell line L6. RESULTS-We showed increased SNAP23 levels in skeletal muscle from patients with type 2 diabetes compared with that from lean control subjects Moreover, SNAP23 was redistributed from the plasma membrane to the microsomal/cytosolic compartment in the patients with the type 2 diabetes Expression of the SNARE-interacting protein Munc18c was higher in skeletal muscle from patients with type 2 diabetes Studies in L6 cells showed that Munc18c promoted the expression of SNAP23. CONCLUSIONS-We have translated our previous in vitro results into humans by showing that there is a change in the distribution of SNAP23 to the interior of the cell in skeletal muscle from patients with type 2 diabetes.     Randomised double-blind phase 3 clinical study testing impact of atorvastatin on prostate cancer progression after initiation of androgen deprivation therapy : study protocol

    Introduction Blood cholesterol is likely a risk factor for prostate cancer prognosis and use of statins is associated with lowered risk of prostate cancer recurrence and progression. Furthermore, use of statins has been associated with prolonged time before development of castration resistance (CR) during androgen deprivation therapy (ADT) for prostate cancer. However, the efficacy of statins on delaying castration-resistance has not been tested in a randomised placebo-controlled setting. This study aims to test statins' efficacy compared to placebo in delaying development of CR during ADT treatment for primary metastatic or recurrent prostate cancer. Secondary aim is to explore effect of statin intervention on prostate cancer mortality and lipid metabolism during ADT. Methods and analysis In this randomised placebo-controlled trial, a total of 400 men with de novo metastatic prostate cancer or recurrent disease after primary treatment and starting ADT will be recruited and randomised 1:1 to use daily 80 mg of atorvastatin or placebo. All researchers, study nurses and patients will be blinded throughout the trial. Patients are followed until disease recurrence or death. Primary outcome is time to formation of CR after initiation of ADT. Serum lipid levels (total cholesterol, high-density lipoprotein (HDL), low-density lipoprotein (LDL) and trigyserides) are analysed to test whether changes in serum cholesterol parameters during ADT predict length of treatment response. Furthermore, the trial will compare quality of life, cardiovascular morbidity, changes in blood glucose and circulating cell-free DNA, and urine lipidome during trial. Ethics and dissemination This study is approved by the Regional ethics committees of the Pirkanrnaa Hospital District, Science centre, Tampere, Finland (R18065M) and Tarto University Hospital, Tarto, Estonia (319/T-6). All participants read and sign informed consent form before study entry. After publication of results for the primary endpoints, anonymised summary metadata and statistical code will be made openly available.
